Abstract

神経脱髄は、中枢神経系中のミエリンタンパク質の減少によって特徴づけられる有害な病態である。ミエリンは、中枢神経系(CNS)および末梢神経系(PNS)の重要要素であり、ニューロンの軸索を包み込み、髄鞘として公知の絶縁層を形成する。髄鞘の存在により、下にある神経軸索への電位の伝播形態の神経シグナルの速度および完全性を増強する。髄鞘の喪失により、神経シグナルがその標的に非常にゆっくりか、非同期的か(例えば、いくつかの軸索が神経伝導において他の軸索よりも速い)、断続的に(例えば、高周波の伝導のみが損なわれる)に到達するか、全く到達しないので、感覚、運動、および他の機能型が有意に損なわれる。
本発明は、神経脱髄を軽減する生物活性剤の開発方法を提供する。本方法は、(a)候補薬剤を髄鞘形成細胞と接触させる工程と、(b)コントロール細胞と比較して遺伝子もしくは遺伝子産物の発現の変化または前記遺伝子産物の活性の変化を検出する工程と、前記遺伝子または遺伝子産物が小胞体(ER)ストレスに相関することと、(c)前記遺伝子もしくは遺伝子産物の発現レベルまたは前記遺伝子産物の活性レベルが前記コントロール細胞と比較して調整された場合、前記薬剤を候補として選択する工程とを含む。

脱髄に及ぼすIFN−γの影響を、テトラサイクリン調節系を使用してIFN−γの送達を一過性に調節することが可能なトランスジェニックマウスを使用したクプリゾン動物モデルにおいて評価した(Lin,et al.(2004)J.Neurosci.24:10074−10083)。トランスジェニックマウスを、C57BL/6バックグラウンドの110 GFAP/tTAマウス株とC57BL/6バックグラウンドの184 TRE/IFN−γとを交配して、GFAP/tTA;TRE/IFN−γ二重トランスジェニックマウスを得ることによって作製した(Lin,et al.(2004)J.Neurosci.24:10074−10083)。コントロール(DOX+)マウスにおけるtTAによるTRE/IFN−γ導入遺伝子の転写活性化を、飲料水に0.05mg/mlドキシサイクリンを添加し、これを計画日から自由に与えることによって抑制した。DOX+二重トランスジェニックマウスは、クプリゾン固形飼料を与えられ、且つドキシサイクリン溶液から決して開放されないGFAP/tTA;TRE/IFN−γ二重トランスジェニック動物であった。DOX−二重トランスジェニックマウスは、クプリゾン固形飼料を与えられ、且つIFN−γ発現を誘導するためにドキシサイクリンから開放したGFAP/tTA;TRE/IFN−γ二重トランスジェニック動物であった。
実施例1に記載のクプリゾン処置マウスにおける再髄鞘形成に及ぼすIFN−γの影響を、以下の5週間目のクプリゾンの使用中止後に決定した。使用した実験方法は、実施例1に記載の方法と同一である。
再髄鞘形成は、病変部位での乏突起膠細胞前駆体(OPC)による脱髄病変の再増殖によって起こり、乏突起膠細胞前駆体は病変部位で乏突起膠細胞に分化する(Matsushima,et al.(2001)Brain Pathol.11:107−116;Mason,et al.(2000)J.Neurosci.Res.61:251−262)。CC1陽性乏突起膠細胞は、NG2陽性OPCに由来する(Watanabe et al 1998;Mason et al 2000))。したがって、病変の再増殖を、CC1および/またはNG2陽性細胞の存在によって評価することができる。
CNSへのIFN−γの送達を一過性に調節することが可能な二重トランスジェニックマウス(Lin et al.,J Neurosci 24:10074−10083(2004))を使用して、EAEの病理発生におけるIFN−γの役割を評価した。GFAP/tTA;TRE/IFN−γ二重トランスジェニックマウスを実施例1に記載のように使用して、EAEに起因する脱髄に及ぼすIFN−γの影響を決定した。全マウスに、計画日からドキシサイクリンを与えてIFN−γ発現を抑制し、ミエリン乏突起膠細胞タンパク質(MOG35−55)で免疫化してEAEを誘導した。免疫化後7日目(PID7)に、実験群のドキシサイクリンの使用を中止した(PID7 DOX−)。
IFN−γは、MHC抗原を誘導してマクロファージおよびTリンパ球を活性化することが知られている。T細胞およびマクロファージの決定ならびにMHC−1、TNF−a、IL−2、IL−12、およびIL−17の発現の測定によって再髄鞘形成中のCNSにおけるIFN−γの炎症効果を研究した。実施例4に記載のように、EAEを罹患したDOX+およびDOX−マウス由来の組織において免疫組織化学を行った。実験手順は、上記実施例に記載の手順に従った。
乏突起膠細胞は、タンパク質合成の破壊および分泌経路の混乱に高い感受性を示すことが示されている(Pfeiffer,et al.(1993)Trends Cell
Biol.3:191−197;Southwood,et al.(2002)Neuron 36:585−596;Leegwater,et al.(2001)Nat.Genet.29:383−388)。IFN−γが再髄鞘形成乏突起膠細胞における小胞体(ER)機能を妨害するかどうかを決定するために、IFN−γを発現するマウスの脳梁中のERストレスマーカーの発現をモニタリングした。実施例1に記載のトランスジェニックマウスを使用して研究を行った。
病変を再髄鞘形成するためのIFN−γに起因する減少におけるERストレス応答の関与を、膵臓ERキナーゼ(PERK)の機能変異の喪失にヘテロ接合性を示し(Harding,et al.,(2001)Mol Cell 7:1153−1163)、CNS中のIFN−γの発現を一過性に調節するように作製されたトランスジェニックマウスで試験した。TRE/IFN−γマウスを、最初に、C57BL/6バックグラウンドのPERK+/−マウスと交配して(Harding,et al.(2001)Mol.Cell 7:1153−1163)、PERK変異を有するマウスを作製し、得られた子孫をGFAP/tTAマウスと交配させて、PERK変異にヘテロ接合性を示すトランスジェニックマウスを得た。IFN−γの存在下または非存在下での再髄鞘形成に及ぼすPERKの影響を、再髄鞘形成病変中の乏突起膠細胞の髄鞘形成レベルおよび再髄鞘形成病変中に存在する乏突起膠細胞数の関数として評価した。
IFN−γによって誘導された乏突起膠細胞のアポトーシスがERストレスに影響を及ぼすかどうかを試験するために(形態学に及ぼす影響)、アポトーシス度およびERマーカーの発現を、乏突起膠細胞前駆細胞(OPC)培養物において研究した。乏突起膠細胞前駆体を、新生ラット脳から培養した(Baerwald,et al.(1998)J.Neurosci.Res.52:230−239)。混合グリア培養物を、フラスコ中の10%ウシ胎児血清(FBS)を含む培地中で成長させ、星状細胞層がコンフルエントになった場合(10〜14日目)、オービタルシェーカーを使用して、乏突起膠細胞前駆体を星状細胞および小グリアから分離した。その95%を超える細胞がA2B5陽性、GFAP陰性、およびCD11b陰性である細胞を、PDGF(10ng/ml)およびFGF(5ng/ml)(共にR&D Systems,Minneapolis,MN)も含む0.5%FBS含有培地中で培養した。次いで、細胞を、分化のためにPDGFおよびFGFを含まない0.5%FBS培地に変更した。分化培地中で5日後に、約40%の細胞がミエリンタンパク質2’3’−環状ヌクレオチド3’−ホスホジエステラーゼ(CNP)に陽性であった。70U/ml組換えラットIFN−γ(Calbiochem,La Jolla,CA)を、5日間分化させた細胞に添加した。分化培地中で5日後に、約40%の細胞がCNP陽性であった。70U/mlの組換えIFN−γ(Calbiochem)を、5日間分化させた細胞に添加した。一般的なERストレス誘導剤に対する乏突起膠細胞応答を試験するために、5日間または7日間分化培地で培養した前駆細胞を、2μg/mlのツニカマイシン(Sigma Aldrich)で6時間処置した。
マウス発達中の乏突起膠細胞の髄鞘形成に及ぼすIFN−γの影響を研究するために、テトラサイクリン(tet)調節系(Lin,et al.(2004)J.Neurosci.24:10074−10083)を使用したCNSへのIFN−γ送達を一過性に調節したトランスジェニックマウスを作製した。星状細胞中でtTA発現を駆動するために、グリア線維酸性遺伝子(GFAP)遺伝子の転写調節領域を選択した(Brenner,et al.(1994)J.Neurosci.14:1030−1037)。GFAP/tTAマウスをTRE/IFN−γマウスと交配させて、両方の導入遺伝子にヘミ接合性を示す動物を産生した。これらのマウスをドキシサイクリンに対して維持する場合(DOX+)、IFN−γ導入遺伝子の発現は抑制される(図10A)。二重トランスジェニックマウスをドキシサイクリンから開放した場合(DOX−)、IFN−γが発現する。これらの実験の目的のために、発生後14日目(E14)までにドキシサイクリンをマウスに投与し、この時点で、実験動物(DOX−)にドキシサイクリンを投与するのを停止する一方で、コントロール動物(DOX+)に薬物を継続的に与えた。IFN−γのmRNAは、生後10日目の早さでDOX−マウスにおいて検出することができる(データ示さず)。
IFN−γによって誘導されたERストレス応答の関与を試験するために、膵臓ERキナーゼ(PERK)の変異機能の喪失にヘテロ接合性を示すトランスジェニックマウスを使用してPERK酵素の関与を評価した(Harding,et al.(2001)Mol.Cell 7:1153−1163)。PERK+/−マウス中のp−eIF−2αレベルの関数を評価したので、導入遺伝子動物の表現型を分析した。
PERK+/−)は、さらにより重篤な表現型を有していた。これらの動物は、GFAP/tTAおよびTRE/IFN−γ対立遺伝子の組み合わせを受け継がなかったIFN−γ発現PERK+/+同腹仔またはPERK+/−動物よりもかなり小さく、重篤な振戦および運動失調を示さず、強直発作を経験したこれらのマウスの約2/3であった。顕著には、E14でドキシサイクリンから開放されたPERK+/−バックグラウンドの二重トランスジェニックマウスの90%超が生後(PDN)27日までに死亡したのに対して、野生型バックグラウンドの二重トランスジェニックマウスは、正常に生存した(図15)。
CNS中にIFN−γを発現するPERK+/−マウスによって示される強直発作を伴う振戦表現型(実施例3に記載)は、ミエリン変態が示唆される。PERK+/−マウスにおける髄鞘形成レベルを評価するために、髄鞘形成状態を、MBPの免疫染色によって評価し、この評価を、E14でドキシサイクリンから開放された14日齢のGFAP/tTA;TRE/IFN−γ;PERK+/−マウスにおいて行い、野生型バックグラウンドの二重トランスジェニックマウスと比較した(図16)。ミエリン塩基性タンパク質(MBP)についての免疫染色は、野生型バックグラウンドの二重トランスジェニックマウスと比較して、E14でドキシサイクリンから開放された14日齢のGFAP/tTA;TRE/IFN−γ,PERK+/−マウスのCNS中で顕著に減少した(図16)。さらに、超微細構造試験により、CNS中にIFN−γを発現するPERK+/−マウスの脊髄中の軸索の大部分(81%±14.9%)が髄鞘を形成しなかったことが明らかとなった(図17)。対照的に、E14でドキシサイクリンから開放された野生型バックグラウンドの二重トランスジェニック動物は、非有髄軸索が顕著により少なかったのに対して(30%±12.9%)、IFN−γ発現を抑制するためにドキシサイクリンに対して継続的に維持した動物は、非有髄軸索がさらにより少なかった(9.8%±6.1%)。
CNS中にIFN−γを発現するPERK+/−マウスによって示される髄鞘低形成を説明する細胞機構を洞察するために、これらの動物における乏突起膠細胞機能の状帯を試験した。本発明者らは、ミエリンマーカーであるMBP、PLP、およびセラミドガラクトシルトランスフェラーゼ(CGT)をコードするmRNAの定常状態のレベルを決定した。リアルタイムPCR分析は、E14でドキシサイクリンから開放された野生型バックグラウンドの14日齢の二重トランスジェニックマウスの脳内のMBP、PLP、およびCGTのmRNAレベルが正常よりわずかに低いことを示した。E14でドキシサイクリンから開放されたGFAP/tTA;TRE/IFN− 7;PERK+/−マウスのCNS中のこれらのmRNAレベルはさらに低かった(図18)。ミエリンタンパク質コードmRNAの定常状態レベルの減少が髄鞘形成細胞数の減少に起因するかどうかを決定するために、本発明者らはこれらのマウス中の乏突起膠細胞数を決定した。コントロールマウスと比較して、E14でドキシサイクリンから開放された野生型バックグラウンドの14日齢のGFAP/tTA;TRE/IFN−γトランスジェニックマウスにおけるCC1免疫染色によって同定された乏突起膠細胞がわずかにより少なかった(図19A)。対照的に、PERK+/−バックグラウンドのIFN−γ発現トランスジェニックマウスの脳梁および小脳中で非常に少数の乏突起膠細胞しか検出することができず、これらのマウスの脊髄中の乏突起膠細胞数は、50%を超えて減少した(図19A)。
幼若な成長中の動物の能動的に髄鞘形成する乏突起膠細胞と比較して、成体マウス中の乏突起膠細胞は、ミエリン構造におけるホメオスタシスの維持に過不足のないより低いレベルの膜タンパク質および脂質を産生する(Morell,et al.(1999)Basic Neurochemistry:Molecular,Cellular,and Medical Aspects(Philadelphia,PA:Lippincott−Raven Publishers):69−93)。したがって、成体動物中の乏突起膠細胞のERは、増加したタンパク質負荷を処理するためのより簡素な能力を有し得、そのようなものとして、タンパク質分泌経路の破壊により小さな感受性を示し得る。この可能性を試験するために、二重トランスジェニックマウスを成熟するまで成長させ、その時点で、CNS中のIFN−γ発現が開始された。リアルタイムPCR分析は、4週目でドキシサイクリンから開放された二重トランスジェニック動物が約6週齢でIFN−γを発現し始め、CNS中のIFN−γmRNAおよびタンパク質レベルがE14でドキシサイクリンから開放された成長中のマウスのレベルに匹敵することを示した(データ示さず)。IFN−γは、成体マウス、PERK+/−バックグラウンドのマウスにおいてさえも乏突起膠細胞の生存に影響を及ぼさなかった(図20B、20C、20D、および20E)。さらに、超微細構造試験により、4週齢でドキシサイクリンから開放された野生型またはPERK+/−バックグラウンドの10週齢の二重トランスジェニックマウスのCNS中の正常なミエリンが明らかとなった(図20F、20G、20H、および20I)。IFN−γによるBIPおよびCHOPの小さな誘導は、4週齢でドキシサイクリンから開放された10週齢の二重トランスジェニックマウスの小脳中で認められた(図20A)。それにもかかわらず、共存分析は、成熟乏突起膠細胞がBIP発現を有意に増加させないことを示した(図20B、20C、20D、および20E)。
CNSへのIFN−γの送達を一過性に調節することが可能な二重トランスジェニックマウス(Lin et al.,J Neurosci 24:10074−10083(2004))を使用して、EAEの病理発生におけるIFN−γの役割を評価した。GFAP/tTA;TRE/IFN−γ二重トランスジェニックマウスを上記実施例に記載のように使用して、EAEに起因する脱髄に及ぼすIFN−γの影響を決定した。全マウスに、計画日からドキシサイクリンを与えてIFN−γ発現を抑制し、実験群をその後にMOG35−55ペプチドで免疫化してEAEの発症を誘導した。EAEの誘導のために、マウスの脇腹および尾の根元(tail base)に、600μgの結核菌(H37Ra株;Difco)を補足した完全フロイントアジュバント(Difco)中に乳化した200μgのMOG 35−55ペプチドを皮下注射した。400ngの百日咳毒素(List Biological Laboratories)の2回の腹腔内注射を、24時間後および72時間後に行った。臨床スコア(0=健康、1=弛緩した尾、2=運動失調および/または後肢の不全麻痺、3=後肢の麻痺および/または前肢の不全麻痺、4=四肢の麻痺、および5=瀕死または死亡)を毎日記録した。コントロール動物に継続的にドキシサイクリンを与えた一方で(DOX+二重マウス)、実験動物(DOX−二重マウス)の抗生物質異を枯渇させてIFN−γを発現させた。IFN−γの影響を以下のようにモニタリングした。
Strand Synthesis System for RT−PCRキット(Invitrogen)を使用して逆転写を行った。TaqmanリアルタイムPCRを、以前に記載のように、iQ supermix(Bio−Rad)を使用して、Bio−Rad iQ リアルタイムPCR検出システムにて行った12。図21(b)は、DOX−動物の脊髄中のIFN−γレベルがEAE発症時(PID14)に検出できるようになり、疾患のピーク時(PID17)にピークに到達し、その後の疾患の回復期(PID22)に減少した。
EAE病理発生中、T細胞が末梢神経系で刺激され、臨床的疾患発症の十分に以前にCNSに入る(Hickey et al.,(1991)J Neurosci Res
28:254−260)。EAEの脱髄の影響からのCNSの防御におけるIFN−γの影響がIFN−γの抗炎症活性に起因するかどうかを決定するために、T細胞および単球の浸潤レベルを評価し、既知の炎症性サイトカインの発現を評価した。
脱髄および乏突起膠細胞の喪失は、多発性硬化症(MS)およびその動物モデル(実験的自己免疫性脳髄膜炎(EAE))の特質である。重篤な細胞損傷に起因せずに下流シグナル伝達経路を活性化する低レベルのストレスがより重篤なストレスの多い事象へのその後の曝露を防御することができることが長期にわたり知られている。膵臓小胞体キナーゼ(PERK)は、ERストレス誘導性キナーゼをコードし、これは真核細胞翻訳開始因子2α(eIF2α)をリン酸化して多数の細胞防御遺伝子のストレス誘導性発現を増強する(Harding et al.,(1999)Nature397:27l−274;Lu et al.,(2004) EMBO J 23:169−179;Harding et al,(2003)Mol Cell 11:619−633)。
脱髄におけるGADD−34の役割を、GADD−34を欠損し、EAEが誘導されたマウスにおいて研究した。
種々のストレスの多い容態は、eIF2αと関連する。以下の4つの哺乳動物eIF2αキナーゼが同定されている:PERK、GCN2、RNA活性化タンパク質キナーゼ(PKR)、およびHRI(Proud,2005)。上記実施例に記載されるように、PERK−eIF2α経路を、免疫サイトカインIFN−γによってEAE中の乏突起膠細胞中で活性化させる。Chakrabarty et al.(2004)は、PKR−eIF2α経路の活性化が活性なEAEを罹患した動物のCNS中の炎症細胞で生じることを示した。
乏突起膠細胞で認められ、且つGADD34欠失によって増加する防御効果を評価するために、GADD34の発現を遮断するためにsiRNAを使用する実験を、精製ラット培養乏突起膠細胞に対して行う。実験プロトコールは、Lin et al.(投稿中)に記載されている。新生ラット脳由来の乏突起膠細胞前駆体(OPC)を、以下のように培養する。混合グリア培養物を、フラスコ中の10%FBSを含む培地中で成長させ、星状細胞層がコンフルエントになった場合(10〜14日目)、オービタルシェーカーを使用して、乏突起膠細胞前駆体を星状細胞および小グリアから分離した。その95%を超える細胞がA2B5陽性、GFAP陰性、およびCD11b陰性である細胞を、0.5%FBS、PDGF(10ng/ml)、およびFGF(5ng/ml)を含む培地中で最初に培養した。次いで、細胞をPDGFおよびFGFを含まない0.5%FBS培地に移して分化を誘導した。
Vectors Insert Design Tool(Ambion)と組み合わせて使用して、siRNA標的配列由来のDNAオリゴヌクレオチドインサートをコードするヘアピンsiRNAを作製する。プログラムは、クローニングのために使用されるループ配列およびオーバーハングを付加する。ヘアピンsiRNAコードDNAオリゴヌクレオチドインサートを、製造者の説明書にしたがってpSilencer 4.1−CMVベクター(Ambion)にクローン化し、GADD34に特異的なヘアピンsiRNAをコードするpSilencer 4.1−CMVベクターを、製造者のプロトコールに従ってNucleofectorキット(Amaxa)を使用してOPCにトランスフェクトする。コントロール乏突起膠細胞を、GADD34siRNAを欠く4.1−CMVベクターであるコントロールベクターでトランスフェクトする。トランスフェクション後、細胞を、2.5μg/mLピューロマイシンを使用して10〜14日間にわたって選択する。トランスフェクトしたOPCを、TaqmanリアルタイムPCRを使用してGADD34mRNAの存在について分析し、ウェスタンブロット分析を使用してGADD34タンパク質の存在について分析する。
PERKの活性化は、PERKの二量体形成によって開始され、それにより、トランス自己リン酸化され、その基質eIF2αのリン酸化能力が増加する。通常、この二量体形成事象は、PERKのストレス感受性ERルーメンドメインによって駆動される(Harding 1999)。他の研究者は、PERKのeIF2αキナーゼエフェクタードメインを2修飾FK506結合ドメイン(Fv2E)を含むポリペプチドと融合して、融合タンパク質(Fv2E−PERK)を生成し、この人工的eIF2αキナーゼ(Fv2E−PERK)の活性が二量体AP20187に従属し、ERストレスの上流シグナル伝達から脱共役される(Lu et al.,2004)。
乏突起膠細胞に及ぼすPERK経路の活性化の防御効果を説明する機構を決定するために、mRNAマイクロアレイ分析を使用して、上方制御された細胞防御遺伝子を同定する。
PERK−eIF2α経路の活性化が乏突起膠細胞に及ぼす細胞防御効果によってEAE誘導性組織損傷を防御することを直接証明するために、マウス中のPERK活性を、乏突起膠細胞中のPERK経路の活性化を調節可能にするように調整する。
PERK経路の活性化によってEAE誘導性脱髄病変中の乏突起膠細胞の再髄鞘形成が促進されるかどうかを決定するために、PLP/Fv2E−PERマウスにEAEを誘導し、AP201187の存在下および非存在下でのPERKの役割を評価する。
MS患者のCNS組織を、Human Brain and Spinal Fluid Resource Center(Los Angeles,CA)およびThe Rocky Mountain MS Center(Englewood,CO)から入手する。組織は、その生存期間にMSを罹患していたヒト由来の脳および脊髄組織、髄液、および他の組織標本を含み得る。サンプルを凍結するか、そうでなければ、ドナーの死亡直後に保存し、ドナーの病歴についての情報と共に慎重に貯蔵組織の目録を作成する。
for RT−PCRキット(Invitrogen)を使用して逆転写を行った。TaqmanリアルタイムPCRを使用して、IFN−γおよびERストレスマーカーBIPおよびCHOPの発現を定量した。PERK、eIF2α、p−PERK、およびp−eIF2αのタンパク質レベルを、ウェスタンブロット分析を使用して決定した。
1.in vivo研究
eIF2αのリン酸化は、シグナル伝達経路の間で高度に保存された収束点(point of convergence)である(Jousse et al(2003)J.Cell.Biol.163:767−775;Proud CG(2005)Semin.Cell.Dev.Biol.16:3−12)。4つのタンパク質キナーゼは、ER(PERK)中のタンパク質不良折り畳み(malfolding)、アミノ酸欠乏(GCN2)、ウイルス感染(PKR)、およびヘム欠損(HRI)の他の非関連ストレスにeIF2αのリン酸化をつなぎ合わせることが公知である(Proud CG(2005)Semin.Cell.Dev.Biol.16:3−12)。このeIF2αリン酸化依存性ストレス誘導性経路は、総合的ストレス応答(ISR)と呼ばれている(Harding,et al(2003)Mol.Cell 11:619−633;Proud CG(2005)Semin.Cell.Dev.Biol.16:3−12)。ストレスから回復させるために、eIF2αを、酵素ホスファターゼ(PP1)およびその不可欠な非酵素補因子である成長停止DNA損傷34(growth arrest
and DNA damage 34)(GADD34)を含む複合体によって迅速に脱リン酸化する(Connor,et al.(2001)Mol.Cell.Biol.21:6841−6850;Novoa et al(2001)J.Cell BM.153:1011−1022;Novoa et al(2003)EMBO J.22:1180−1187)。GADD34の発現を、サイトゾル転写因子ATF4の誘導によって調節し、その翻訳を、eIF2αリン酸化の存在下で上方制御し、それにより、強固な自己フィードバックループが作製される(Novoa et al(2003)EMBO J.22:1180−1187;Jiang et al(2004)Mol.Cell.Biol.24:1365−1377)。GADD34欠失により、ストレス細胞中のp−eIF2αレベルが増加し、細胞がストレスから防御される(Jousse
et al(2003)J.Cell.Biol.163:767−775;Marciniak et al(2004)Genes Dev.18:3066−3077)。
脊髄組織を調製し、コントロールマウスが疾患重症度のピークにある時点で分析した。予想通り、GADD34の発現は、EAEを罹患したマウスの腰髄中の乏突起膠細胞中で上方制御され、これは、年齢適合ナイーブマウスの乏突起膠細胞中で検出不可能であった(図27aおよび27b)。さらに、CC1およびp−eIF2αの二重標識により、GADD34欠失によってコントロールマウスと比較して、EAEを罹患したGADD34ヌルマウスの腰髄中の乏突起膠細胞中のp−eIF2αレベルが顕著に増加することが明らかとなった(図27cおよび27b)。組織学的分析により、コントロールマウスの腰髄中に典型的なEAE脱髄病変(髄鞘の破壊(図28aおよび28c)、乏突起膠細胞の喪失(図8e)、軸索損傷(図28g))が明らかとなった。対照的に、この時点でのMBP免疫染色およびトルイジンブルー染色によって、GADD34ヌルマウスの腰髄中に明らかな脱髄病変は認められなかった(図28bおよび28d)。重要には、GADD34ヌルマウスの腰髄中の乏突起膠細胞は、ほとんどインタクトなままであった(図28f)。さらに、GADD34欠失により、コントロールマウスと比較してGADD34ヌルマウスの腰髄中の軸索損傷は劇的に減少した(図28gおよび28h)。まとめると、これらのデータは、GADD34欠失が乏突起膠細胞中のp−eIF2αレベルを増加させ、したがって、総合的ストレス応答を延長し、EAE誘導性脱髄から防御することを示す。
細胞のストレスからの防御におけるeIF2αのリン酸化の重要性により、いくつかのヒト疾患の治療に潜在的に有用であり得るeIF2αのリン酸化の小分子モジュレーターを探索した。Boyce et al.(Boyce,et al.(2005)Science 307:935−939)は、薬物スクリーニングを使用し、このスクリーニングは、ERストレス誘導性アポトーシスに対する細胞防御を付与する小分子を同定するであろう。彼らは、eIF2α脱リン酸化の小分子インヒビターであるサルブリナール(SAL)を同定した。これは、PP1−GADD34ホスファターゼ活性を特異的に阻害し、それによりeIF2αリン酸化が保持される。SAlでの処置により、ERストレスおよびウイルス感染から細胞が防御される(Boyce,et al.(2005)Science 307:935−939)。
IFN−γ曝露の7日後、MBPレベルは、HOCにおいて約80%減少した(図29)。SAL処置のみではMBP発現に影響を及ぼさなかった。しかし、SAL処置により、IFN−γ曝露によって誘発されたMBP発現の減少が顕著に弱められた(図29)。IFN−γの存在は、脱髄病変中の再髄鞘形成を阻害することが示されている(実施例2〜7を参照のこと)。まとめると、これらのデータは、eIF2αリン酸化を促進するか延長する薬剤での処置によって免疫媒介性脱髄疾患においてミエリン修復を促進することができることを示す。
1.トランスジェニックマウスの作製および評価
a)乏突起膠細胞中でSOCS1を発現するトランスジェニックマウスの作製
トランスジェニックマウス株PLP/SOCS1を、PLP発現カセットおよびSOCSl cDNAを含む構築物を使用して精製した(図30A)。PLP発現カセットは、他の場所に記載されており、多数の導入遺伝子の乏突起膠細胞特異的発現のために使用されている(Wight et al.,1993;Fuss et al.,2000;Doerflinger et al.,2003;Gonzales et al.,2005)。ポリメラーゼ連鎖反応(PCR)ベースまたは抗Flag抗体ベースの検出方法におけるSOCS1発現のためのマーカーとして役立つFlag−エピトープ配列を含んでいたので、SOCSl cDNAクローン(Starr et al.,1998)を使用した(Einhauer and Jungbauer,2001)。簡潔に述べれば、Flag−SOCSl cDNAを、XbaIを使用して元の発現ベクターpEF−FLAG−I/m4A2から切り出した。フラグメントにクレノウを充填し、中間ベクター(修飾pNEB/193ベクター)のSmaI制限部位にサブクローン化した。得られたpNEB193/SOCSlベクターを、AscIおよびPacIでさらに消化して(部分消化)、Flag−SOCS1フラグメントを放出し、PLP発現カセットのポリリンカー領域の同一の制限部位にサブクローン化した。PLP/SOCS1ベクターを、ApaIおよびSacIIで消化し(部分消化)、線状の15kb導入遺伝子を、受精(C57BL/6J×DBA/2J)卵母細胞への微量注入のために単離した。導入遺伝子に陽性の子孫を、導入遺伝子特異的プライマーを使用したPCRによるテールDNAの増幅によって同定した。同定された樹立系統を、次に、C57BL/6Jマウスと交配させて(Jackson Laboratories,Bar Harbor,ME)、トランスジェニック系統を確立した。
CNS中にIFN−γを過剰発現するトランスジェニックマウスMBP/IFN−γ(172系統)およびGFAP/tTA×TRE/IFN−γ(184/110系統および184/67系統)は、別の場所に記載されている(Corbin et al.,1996;Gao et al.,2000;Lin et al.,2004,2005)。簡潔に述べれば、MBP/IFN−γ(172系統)マウスは、IFN−γ発現がミエリン塩基性タンパク質(MBP)転写調節領域によって駆動されるトランスジェニック動物である(Gao et al.,2000)。GFAP/tTA×TRE/IFN−γは、単一トランスジェニックGFAP/tTA(184系統)と単一トランスジェニックTRE/IFN−γ(110系統および67系統)マウスとの交配によって得られた二重トランスジェニックマウスである。実験で使用した2つのTRE/IFN−γマウス系統(110系統および67系統)は、GFAP/tTAマウスと交配した場合に異なる量のIFN−γを産生する(184/110および184/67)(Lin e al.,2004,2005)。GFAP/tTA×TRE/IFN−γは、テトラサイクリンオフ誘導系(tetracycline−off−inducible system)であり、この系は、グリア線維酸性遺伝子(GFAP)転写調節領域がtTAの発現を駆動し、次いで、TRE(tet応答エレメント)に結合してIFN−γの発現を開始する。ドキシサイクリンの投与により、tTADNA結合およびIFN−γ発現が抑制され、ドキシサイクリンの除去によってIFN−γ発現の一過性調節誘導が可能である(Gao et al.,1999)。
IFN−γ過剰発現マウスを、二重トランスジェニック交配系(MBP/IFN−γ×PLP/SOCS1)および三重トランスジェニック交配系(GFAP/tTA×TRE/IFN−γ×PLP/SOCS1)でPLP/SOCS1マウスと交配させた。MBP/IFN−γ×PLP/SOCS1(172×PLP/SOCS1)交配を、標準的な交配プロトコールにしたがって行った。GFAP/tTA×TRE/IFN−γ×PLP/SOCS1交配を、以下の2工程交配過程で行った:GFAP/tTAマウス(184系統)を、PLP/SOCS1マウスと最初に交配し、次いで、二重陽性(184×PLP/SOCS1)子孫を、TRE/IFN−γの110および67系統と個別に交配させた。この第2の交配工程を、前に記載した「tetオフ」プロトコールにしたがって行った。ドキシサイクリン0.05mg/ml(Sigma−Aldich)を、胚齢14日目まで妊娠した雌マウスの水に添加し、その後、動物を正常な水に戻し、それにより、IFN−γ転写が開始され、生後にピークになる(Lin et al.,2005)。

全実験動物を、単位テール(Biotek 2000 automatic system,Beckman−Coutler,Fullerton,CA)を使用して遺伝子型同定を行った。導入遺伝子検出のためのPCR(Qiagen PCRキット、Valencia,CA)を、以下の導入遺伝子特異的スクリーニングプライマーを使用して行った:
総RNAを、TRIzol試薬(Invitrogen Corp.,Carlsbad,CA)を使用して実験動物から単離した。1.2%変性アガロースゲル中で20μgの総RNAを分離することによってノーザンブロットを行った。サンプルをナイロン膜に移し、PCR(GenAmp2400;Perkin−Elmer,Welleslay,MA)によって[γ−32P] dCTPおよび [γ−32P] dATP(New
England Nuclear/Perkin−Elmer,Welleslay,MA)で無作為に標識したSOCS1プローブを使用して一晩ハイブリッド形成した。Kodakフィルムを、ハイブリッド形成した膜に−80℃で48時間曝露し、M7B Kodak処理装置(Kodak,Rochester,NY)を使用して現像した。各レーンに存在する総RNAの相対レベルを評価するために、膜を剥ぎ取り、28SリボゾームRNAに特異的な放射性標識プローブを使用してハイブリッド形成させた(Baerwald et al.,1998)。
以下の実験下でBioRad I−サイクラーリアルタイムPCRユニットを使用して反応を行った:95℃で3分間を1サイクル、95℃で30秒間を40サイクル、および60℃で30秒間(Bio−Rad Laboratories)。Flag−SOCS1およびIFN−γのmRNAレベルを、閾値サイクル(threshold cycles)(Flag−SOCS1/GAPDHおよびIFN−γ/GAPDH比)に基づいたGAPDHの発現レベルに正規化した(Lin et al.,2005)。
いくつかのPLP/SOCS1マウスおよび野生型マウスの脳および脾臓由来の総溶解物を、RIPA緩衝液(Santa Cruz Biotechnology,Santa Cruz,CA)中での組織均質化によって得た。氷上で15分間のインキュベーション後、溶解物を、14,000rpmで30分間遠心分離し、上清を回収した。タンパク質サンプル(50μg)を、15%SDS−ポリアクリルアミドゲルにて電気泳動し、PVDF膜(Trans−blot SD apparatus,Bio−Rad Laboratories)に移し、マウス抗Flag抗体(M2,1000倍希釈)(Sigma−Aldrich,St.Louis,MO)と一晩インキュベートし、ECL Western blot検出試薬(Amersham Biosciences,Piscataway,NJ)で検出した。Flagタンパク質(Sigma−Aldrich)(Flagオリゴペプチドのポリマー)を、反応のためのポジティブコントロールとして使用した。
動物を、腹腔内投与した0.01ml/gの2.5%アベルチン(Sigma−Aldrich)で麻酔し、生理食塩水およびその後の2%パラホルムアルデヒドで10分間潅流した。脳を取り出し、2%パラホルムアルデヒドで1時間後固定し、30%スクロース中で48時間低温保存し、凍結ブロックとして調製し(OCT compound,Sacura,Torrance,CA)、−20℃で厚さ7μmの切片にした(Leica
CM1800 cryostat,Leica Microsystems)。免疫染色前に、切片を0.1%TritonX−100(Sigma−Aldrich)で10分間処置し、10%ウシ血清アルブミン(Sigma−Aldrich)またはヤギ血清(Invitrogen)と30分間インキュベートした。一次抗体(室温で2時間または4℃で一晩)およびFITC抱合またはCy3抱合二次抗体(30分間)との連続インキュベーションによって、間接的免疫染色を行った。本研究で使用した全ての以下のプライマーおよび二次抗体は市販されている:マウスおよびウサギ抗Flag抗体(100倍希釈;Sigma−Aldrich)、マウス抗CC1抗体(20倍希釈;Oncogene)、マウスMHCクラスI抗体(100倍希釈;Chemicon International,Temecula,CA)、マウス抗PLP、プロテオリピドタンパク質、高アイ(100倍希釈;Chemicon International)、マウスおよびウサギ抗SOCS1抗体(100倍希釈;Santa Cruz Biotechnology,Santa Cruz,CA)、ウサギ抗Stat1抗体(100倍希釈;Santa Cruz Biotecnology)、抗マウスまたは抗ウサギFITC抱合二次抗体(100倍希釈;Jackson Immunoresearch,Bar
Harbor,ME)、および抗マウスまたは抗ウサギCy3抱合抗体(500倍希釈;Jackson Immunoresearch)。免疫染色切片を、DAPI核スタチン(Vector Laboratories,Burlingame,CA)を含むVectorshieldマウント培地を使用してマウントし、蛍光顕微鏡(Axoplan;Carl Zeiss Microimaging)を使用して試験した。
電子顕微鏡法研究のために選択したマウスを、4%パラホルムアルデヒドおよび2.5%グルタルアルデヒドで潅流した。脳を採取し、白質構造を、ステレオタイプ顕微鏡(Wild M3C;Wild AG,Heebrugg,Switzerland)を使用して切片にした。組織を、四酸化オスミウム中でさらに後固定し、新たに調製したエポキシ樹脂(Epon−812;Electron Microscope Sciences,Fort Washington,PA)中に60℃で48時間包埋した。樹脂ブロックを、Leica Ultracut ultramicrotome(Leica Microsystems)を使用して90nmの切片にし、5%酢酸ウラシルおよび2.5%クエン酸鉛で染色した。組織サンプルの超微細構造を、Tecnai−F30透過型電子顕微鏡(FEI Company,Hillsboro,OR)を使用して試験した。
混合初代乏突起膠細胞培養物を、以前に記載のように調製した(Baerwald et al.,2000)。簡潔に述べれば、PLP/SOCS1およびC57B1/6J交配の2〜3日齢の新生児から脳組織を採取した。同腹仔がトランスジェニックの仔および野生型の仔を含んでいたので、各動物の脳を個別に処理し、個別に培養し、その後に表現型を適合させた。各脳を、0.25%トリプシンおよび10μg/mlDNAaseI(Invitrogen)を含むダルベッコ改変イーグル培地(DMEM)を使用して37℃で20分間個別に消化し、細胞を、ポリD−リジンコーティング75mm2フラスコ(Sigma−Aldrich)上で培養した。培養物を、10%ウシ胎児血清DMEMを使用して37℃、5%CO2で12日間維持し、次いで、5μl/mlのインスリン、50μl/mlのトランスフェリン、30nMのセレン、10nMのビオチン、10nMのプロゲステロン、15nMのT3、0.1%ウシ血清アルブミン、および1%アンピシリン−ストレプトマイシン(Sigma−Aldrich)を含む合成培地に交換した。分化5日目に、培養物を、100U/mLのIFN−γ(Calbiochem,San
Diego,CA)で30分間処置した。抗PLP抗体および抗Stat1抗体の二重免疫染色ならびにDAPI核染色を、上記のように行った。Stat1核転位置アッセイを、6つの個別の培養調製物で行った。野生型およびPLP/SOCS1培養物中の100個のPLP陽性細胞を手作業で計数した。結果を、Stat1核転位置に陽性を示す細胞の平均±SD%として示す。
全データを、3つの独立した実験から作成した。平均、標準偏差、およびp値を、Microsoft Excel(Microsoft,Redmond,WA)のAverage、Stdev、およびAnovaを使用して計算した。統計的有意差を、0.05未満のp値と定義した。
実施例25に記載のように作製したPLP/SOCS1トランスジェニックマウスを、髄鞘形成細胞中でFlagエピトープタグ化SOCS1を発現する用にデザインした(図30A)。これらのマウスは、表現型異常を示さず、メンデルの法則において交配してトランスジェニック子孫を作製し、通常の寿命である。1歳までの異なる時点で行った組織学的評価(電子顕微鏡法が含まれる)により、トランスジェニック同腹仔と野生型同腹仔との間で、髄鞘形成パターンまたは乏突起膠細胞の数、密度、もしくは形態学(以下を参照のこと)に有意差は認められなかった。
初代混合グリア培養物におけるIFN−γに対するPLP/SOCS1乏突起膠細胞の反応性を研究した。トランスジェニックSOCS1の発現がIFN−γシグナル伝達分子Stat1の核転位置を妨害するかどうかを決定するために、細胞培養物を、100U/mLのIFN−γで30分間処置し、抗PLP抗体および抗Stat1抗体をDAPI核色素と共に使用して免疫染色を行った(図33)。野生型培養物におけるStat1細胞内局在の実験により、全ての細胞(PLP陽性乏突起膠細胞が含まれる)におけるDAPI陽性核との強い共存が明らかとなった。対照的に、PLP/SOCS1培養物中のStat1細胞内局在化により、IFN−γに対する差分応答が明らかとなった。トランスジェニックPLP陽性乏突起膠細胞では、Stat1が細胞質中に残存し、IFN−γの存在下で細胞核と共存しなかった(図33E〜H)。これは、野生型細胞と類似してStat1核局在化を伴ってIFN−γに応答した周囲のPLP陰性細胞の応答と対照的であった。野生型培養物中の実質的に全てのPLP陽性乏突起膠細胞(96±3細胞)がStat1核転位置を伴ってIFN−γ刺激に応答した。対照的に、Stat1核転位置は、IFN−γ刺激後の偶発的(occasional)PLP陽性乏突起膠細胞(6±2細胞)においてのみ検出された(p<0.05)。
成長中のマウスのCNSにおけるIFN−γのトランスジェニック発現により、乏突起膠細胞が喪失し、髄鞘低形成が起こる(Corbin et al.,1996;Lin
et al 2005)。SOCS1発現がIFN−γによる損傷の影響から成長中の乏突起膠細胞を防御することができるかどうかを決定するために、本発明者らは、PLP/SOCS1マウスを、異なるレベルでCNS中でIFN−γを過剰発現する以下の3つのトランスジェニックマウス系統と交配させた:MBP/IFN−γ(172系統)、GFAP/tTA×TRE/IFN−γ(184/110系統)、およびGFAP/tTA×TRE/IFN−γ(184/67系統)。以下の3つの交配系を確立した(材料と方法に詳述):MBP/IFN−γ×PLP/SOCS1(172×PLP/SOCS1、二重トランスジェニック系)およびGFAP/tTA×TRE/IFN−γ×PLP/SOCS1(184/110×PLP/SOCS1および184/67×PLP/SOCS1、2つの三重トランスジェニック系)。各交配系の同腹仔(F1世代)を、その表現型に応じて以下の4つの研究群に分類した:野生型/単一トランスジェニックコントロール、SOCS1のみを発現するマウス、IFN−γのみを発現するマウス、IFN−γおよびSOCS1の両方を発現するマウス。交配系あたり全部で40匹の動物(各研究群あたり10匹)を回収し、生後21日目に臨床的および組織学的に試験した。
