Description
本発明は、動植物の免疫機能を高め、病気の予防・改善を行うのに好適な免疫活性化剤及び該免疫活性化剤が配合されている免疫活性化剤配合物に関する。
現代社会において、若年層ではアレルギー、中高年ではメタボリックシンドローム、高齢層では癌や感染症、アルツハイマーなどの罹患率が高くなっている。これら疾患の発症には、直接的・間接的、あるいは習慣的・遺伝的要因があるが、共通して言えることは、ストレスや加齢によって免疫力が落ちた時に発症や進行が加速されることである。したがって、免疫力を活性化する技術は、環境が変化し、食生活が欧米化し、運動量が低下している現代社会において、「健康で長生き」を支えるために必要とされる。
免疫活性化素材としては、ラクトフェリンなど蛋白性の因子もあるが、乳酸菌を主体とした発酵食品、キノコ類、酵母等、微生物が主役になっているものが圧倒的に多い。実は、動植物の免疫系は微生物など環境中の外来異物が自然な形で体内に入ることで活性化される。活性化の仕組みは「自然免疫」と呼ばれ、この10年の間に、分子レベルでの細胞内シグナルトランスダクション経路の詳細が明らかになってきた。
さて、酵母やキノコの免疫賦活の有効成分はβグルカンである。βグルカンは、グルコースがβ1,3結合あるいはβ1,6結合で連なったものである。分子量は加水分解しないものでは数百万ダルトンであり、水溶物は高い粘性を持つ。一方、グラム陰性細菌由来のLPS(リポ多糖)も自然免疫を活性化する(例えば、特許文献1参照。)。LPSは脂質にオリゴ糖ユニットが連なった構造で、オリゴ糖ユニットの結合数によって、分子量は5千から数万ダルトンのラダー分布を見せる。ちなみにパントエア菌のLPSは、分子量5000と分子量45000付近にピークを持つ分子量分布を示す。
免疫活性化作用を免疫担当細胞であるマクロファージの活性化によって評価すると、LPSがβグルカンに比較して優位性が高い。比活性が高いことは、微量で有効であることを意味する。
例えば動物用飼料として与える場合、LPSでは10μg〜20μg/kg体重という微量で有効性が見られるが、βグルカンでは50〜200mg/kg体重で用いられる。
ところで、βグルカンは、マクロファージなど自然免疫担当細胞の細胞膜上の外来異物レセプターデクチン1に結合することで、免疫担当細胞を活性化する。一方LPSは、TLR4と呼ばれる別のレセプターに結合することで、やはり免疫担当細胞を活性化する。両者は別々のレセプターに結合することより、両者を同時に用いれば、免疫担当細胞の活性化は相加的に上がることが予想される。しかし、本発明者らによって、LPSとβグルカンはマクロファージの活性化について相乗効果を示すことが明らかとなった。
本発明は、優れた免疫活性化作用を有する免疫活性化剤及び該免疫活性化剤が配合されている免疫活性化剤配合物を提供することを目的とする。
本発明の免疫活性化剤は、LPS及びβグルカンが配合されていることを特徴とする。
本発明によれば、飼料、食品、化粧品、農業用資材及び医薬品などに優れた免疫活性化作用を付与することができる。
以下、添付図面を参照しながら本発明を実施するための形態について詳細に説明する。
LPS(パントエア・アグロメランス由来、特許文献1参照)とβグルカン(パン酵母由来)のマクロファージ活性化能をマクロファージからの一酸化窒素(NO)誘導を指標として比較した。
マクロファージ細胞として、ラットの肺胞マクロファージ細胞株NR8383を用いた。
パン酵母抽出物の粉末品を50mg秤量し、生理食塩水にて50mg/mlのパン酵母抽出物懸濁液を調製した。この懸濁液に、酸アルカリ処理したガラスビーズを約500mg加え、ボルテックスミキサーを用いて攪拌(1分間の攪拌を20回程度)し、パン酵母抽出物の凝集塊を粉砕したものを調製した。具体的には、凝集塊の粉砕の程度は顕微鏡観察(400倍)により確認した。顕微鏡観察により、凝集塊が認められず、約5〜10ミクロン程度のパン酵母の均一懸濁液になるまで粉砕した。これを1分間放置し、上清を粉砕処理したパン酵母抽出物懸濁液原液とした。
LPSは、パントエア・アグロメランスより精製したものを用いた。生理食塩水にて希釈した。
NOの量は、グリース試薬にて測定した。3%(w/v)スルファニルアミドを含む7.5%リン酸水溶液(A液)と、0.15%(w/v)ナフチルエチレンジアミン水溶液(B液)をそれぞれ調製し、A液を1に対してB液を2の割合で混合して用いた。
8×105個/mlに細胞数を調整したNR8383細胞を100μlずつ96穴平底プレートの各ウエルに加えた。37℃5%CO2のインキュベータにて2時間前培養を行った。βグルカンとLPSの希釈溶液を100μlずつウエルに加えた。添加後、プレートシェーカーを用いて10秒間攪拌した後、37℃5%CO2のインキュベータにて、24時間培養を行った。培養終了後、各ウエルの培養上清中の亜硝酸(Nitrite)濃度をグリース試薬にて測定した。
図1に示すように、マクロファージからのNOの誘導(ここでは亜硝酸として検出)は、βグルカン又はLPS単独よりも組み合わせることにより、相加を著しく上回る相乗効果が得られた。
なお、本発明は上記実施例に限定されるものではない。

- LPS(リポ多糖)及びβグルカンが配合されていることを特徴とする免疫活性化剤。
- 前記LPSは、グラム陰性細菌由来であることを特徴とする免疫活性化剤。
- 前記グラム陰性細菌は、パントエア・アグロメランスであることを特徴とする請求項2記載の免疫活性化剤。
- 請求項1乃至3いずれかに記載の免疫活性化剤が配合されていることを特徴とする免疫活性化剤配合物。
- 前記免疫活性化剤配合物は、飼料、食品、化粧品、農業用資材又は医薬品であることを特徴とする請求項4記載の免疫活性化剤配合物。
