Description
本発明はバチルススブチリス(B.subtilis)等の宿主細胞のチオール酸化能力を高める方法に関する。
TrxA欠失の効果は、ジスルフィド結合含有タンパク質PhoAに特異的であるようだ。実際は、高められたPhoAレベルはPhoA分解生成物の消滅と同時に起こり、培地中に不完全に処理されたpro−PhoAが現れる。このことは、PhoAの折りたたみがTrxA欠失で改善されたことを示している。従来のDNA配列解析は主要な分泌機構成分に関する遺伝子ではないもの又はバチルススブチリスのタンパク質ではないものの発現がTrxA欠失により影響されると示していた(Smits et al.,J.Bacteriol.187:3921−3930,2005)。総合すれば、これらの発見はTrxAがPhoA分泌に特異的に含まれている分泌機構成分の活性(しかし量ではない)に影響を与えていることを示している。即ち、TrxA欠失が酸化されたBdbD分子の細胞レベルを高めることから、TrxAは、細胞質外のチオール−ジスルフィド酸化還元酵素BdbDの酸化還元状態に有意に影響を与えていることを示している。これらの観察は細胞質の還元の能力を限定し、TrxA欠失をさせることは、酸化されたBdbD分子のレベルを高めることになることを示している。従って、TrxA消失はbdbC変異の逆の効果を有し、PhoAの強力な還元された(減少された)折りたたみを生成し(Bolhuis et al.,J.Biol.Chem.274:24531−24538,1999)、同時に還元されたBdbDのレベルを有意に高める。特に、TrxA欠失細胞による改善されたPhoA分泌は依然としてBdbCの存在に依存している。それ故、TrxA欠失細胞中の酸化されたBdbD分子のレベルを高めることが、PhoA等の酸化にさらされたタンパク質の細胞質キャパシティーを高める。PhoAは2つのジシルフィド結合を含むが、ジスルフィド結合はAmyQ及びバチスルスブチリスが分泌する大部分のタンパク質にはないので、このデータはTrxA欠失細胞によるPhoAの改善された分泌が改善されたPhoA膜移行よりも、翻訳後のPhoAにおけるジスルフィド結合の形成に貢献できることを示している。

で定義される核酸配列によりコードされるアミノ酸配列を有するスタフィロコカールカルノーサスDsbA、又はオキシダーゼ活性を有するこれらの変異体である。ジェンバンクにおける相同ポリペプチドを同定することにより、又は例えば、配列番号3をハイブリダイズプローブとして用いる、宿主細胞の核酸ライブラリーの相同核酸配列を同定することにより、他の相同体も同定できる。変異体は既知のTrxAの配列中に変異(挿入、欠失、又は置換)を作成して、生成できる。幾つかの態様において、保存置換が、例えば、触媒領域外に作成される。約約60%、65%、70%、75%、80%、85%、905、95%、98%、又は99%同一な変異体が意図される。
本発明は遺伝的に修飾された宿主細胞及びジスルフィド結合含有タンパク質を生成するための方法を提供する。宿主細胞の語はトランスフォーム、トランスフェクト、インフェクトされ、あるいは核酸配列でトランスフォーム、トランスフェクト、又はインフェクトされ、所望のタンパク質を組換え生成する選択された所望の遺伝子を発現する。この語は親細胞の子孫も含む。選択された遺伝子又は遺伝子修飾が存在する限り、この子孫は親細胞と相同であるか、又は遺伝的に修飾されているかは問題ではない。
本発明は細胞質リダクターゼの低減された活性を有する遺伝的に修飾された宿主細胞も提供する。「低減された活性」の語は宿主細胞の細胞質でのリダクターゼ活性が低められている、又は抑制されている、又は完全に排除されていることを意味する。この低減された活性は対応する野生型又は未修飾バチルス宿主細胞等の未修飾宿主細胞における活性と比較される。低減された細胞質リダクターゼ活性はリダクターゼポリペプチドの発現の低減、細胞質リダクターゼアミノ酸配列をコードしている遺伝子転写の低減、このリダクターゼアミノ酸配列をコードしているRNAの翻訳の低減又は抑制、又は細胞質リダクターゼポリペプチドの不活性化又は抑制により達成することができる。リダクターゼ活性の低減は特定のリダクターゼの活性を直接低減又は欠失させることにより、あるいは特定のリダクターゼのリダクターゼ経路の上流又は下流を修飾することにより間接的に実施することができる。遺伝的に修飾された宿主細胞の低減された活性を測定するためのガイダンスは、実施例2で示すように、4−アセトアミド−4’−マレイミジル−スチルベン−2,2’−ジスルホネート(AMS)でラベルしている細胞抽出物でアッセイするような方法で行うことができる。

例えば、非保存的置換は他のクラスのメンバーの1つのメンバーを交換することを含む。そのようにして置換された残基は非ヒトIL−17様ポリペプチドオーソログと相同なポリペプチド領域へ、又はこの分子の非相同領域へ導入される。
遺伝的に修飾された宿主細胞が対応する未修飾宿主細胞におけるリダクターゼと比較して低減された、抑制された、又は喪失してリダクターゼ活性を有する場合、この修飾及び未修飾宿主細胞は発現されたリダクターゼの量を測定する又は好適な基質の還元能力を測定して、発現されたリダクターゼの相対的な量又は相対的な活性を測定する。発現されたリダクターゼ酵素の相対的な量は、ウエスタンブロット解析、SDSポリアクリルアミド電気泳動、非変異性ゲル電気泳動、高圧液体クロマクグラフィー(HPLC)分離、免疫沈降法、及び/又はDNA結合ゲルシフトアッセイ、及びアルカリフォスファターゼアッセイ等の活性を測定する。
本発明はジスルフィド結合含有タンパク質の生成のための方法を提供する。この方法は宿主細胞が培養培地へタンパク質を分泌するような条件下で遺伝的に修飾された宿主細胞を育成する工程を含む。「ジスルフィド結合形成」の語は1つ又は2つのポリペプチド中に存在する2つのシステインの間に共有結合が形成されるプロセスを意味する。ジスルフィド結合の酸化は酵素の活性部位のシステイン及び標的タンパク質のシステインとの間のチオールジスルフィド交換により仲介される。ジスルフィド結合形成はジスルフィド結合形成を触媒するとの好適な酵素により促進される。
好適な精製方法は、親和クロマトグラフィー、免疫親和クロマトグラフィー、イオン交換クロマトグラフィー、高圧液体クロマトグラフィー(HPLC)、電気泳動(非変性ゲル電気泳動を含む)次いでゲル溶解、及び等電点電気泳動を含むがこれらに限定されない。幾つかのケースにおいて、2つ以上の精製技術を組み合わせて精製度を高める。
本発明は組換えタンパク質の折りたたみを改善する方法も提供する。この方法はジスルフィド結合含有タンパク質を発現させ、折りたたみを改善する条件下で本発明の遺伝的に修飾された宿主細胞を育成する工程を含む。これらの方法は一般的に組み替えタンパク質精製のための前述の方法を用いて行われる。本発明の遺伝的に修飾された宿主細胞は促進されたジスルフィド結合の形成に依存して改善されたタンパク質折りたたみを有するタンパク質を精製することができる。
バチルススブチリスにおける潜在的に還元性のあるTDORの一覧
ジスルフィド結合含有タンパク質のより効果的な分泌のためのバチルススブチリスの酸化力を高めるための最初のアプローチとして、有機体の潜在的な還元システムを解析した。対応する遺伝子の欠失又はそれらの発現の低減はバチルススブチリスをより非還元的にすると仮定される。次に、このことはジスルフィド結合を有するタンパク質の折りたたみを改善すると考えられた。この目的のために、他の微生物から知られている可能性のある3つのシステムが排除できた;第一にバチルススブチリスはグラム陰性バクテリア又は真核生物においてグルタチオンの合成のために必要とされている酵素の相同体を欠失している;第二にバチルススブチリスはストレプトマイセス種及び糸状菌の細胞質に見られる還元剤マイトチオールを欠いている(Newton et al,J.Bacteriol. 178:1990−1995,1996);第三はバチルススブチリスはE.coliに見られるような異性化経路に含まれるタンパク質を欠いている。
PhoA分泌のレベルを決定するTrxAの細胞レベル
ジスルフィド結合含有タンパク質の分泌において前述の可能性のあるTDORの影響を試験するために、trxA、ybdE、ydbP、ydfQ、ykuV、Sto A (ykvVor、spoIVH)、yneN、ytpP、yusEの一部位変異体及びbdbA及びyosR遺伝子をキャリーしているSPβファージを欠いている株をpPSPhoA5で形質転換した。
DNA技術は前述のように実施された(Kouwen, MoI.Microbiol 64:984−999,2007)。可能性のある細胞質TDORの変異株を以下のように構築して、得た。必須trxA遺伝子を劣化させたバチルススブチリス168誘導体株ItrxA(Scharf et al.,J.Bacteriol.180:1869−1877,1998)を前述のように構築した(Smits et al, J.Bacteriol.187:3921−3930,2005)。ItrxA株において、trxA遺伝子をtrxA遺伝子の前のシングルクロスオーバー(キャンベルタイプ)統合pMutin2mcsに依存するIPTG誘発PSPACプロモーターの制御下においた。25μMのIPTGの存在下で育成したとき、Pspacは活性であったが充分には誘発されていなかった。ItrxA dbdC株をdbdC::Kmr変異体の染色体DNAでItrxA株を形質転換し、その後PTG劣化及びKm耐性形質転換の選択を行った。
プラスミドpPSPhoA5はDarmon et al (Appl.Environ. Microbiol 72:6876−6885,2006)に記載のようなStaphylococcus hyicus由来リパーゼパー請求項のプレプロ領域及び成熟E.coliPhoAタンパク質の間の融合を含む。このタンパク質は2つのジスルフィド結合を含みプロテアーゼ耐性構造への折りたたみのための酸化力のあるTDORを必要とすることから、E.coliPhoAはバチルススブチリス(B.subtilis)におけるTDOR活性について感受性のあるレポーターである。特に、BdbC及び/又はBdbSの欠失において、折りたたみのないPhoAはバチルススブチリス(B.subtilis)細胞壁及び/又は細胞培地の高い細胞分解性の環境において、容易に分解される(Sarvas et al,Biochim.Biophys.Acta.1694:311−327、2004)。このことは基本的に酸化能力のあるTDOR活性により分泌されたPhoAの折りたたみが効果的に提供するためのin vivoプロテアーゼアッセイを提供する。興味深いことに、インタクトbdbA、ybdE、ydbP、ydfQ、ykuV、stoA、yneN、yosR、ytpP、又はyusE遺伝子を欠失している株はE.coliの活性PhoAの分泌に有意に影響していなかった。
TrxA及びBdbCの細胞レベルはBdbDの酸化還元状態に影響する
TrxAの存在又は欠失がBdbCチオール酸化経路の活性化に影響するかどうかを試験するために、BdbDの酸化還元状態をKobayashi et al., (Pr oc.Natl.Acad.Sci. U.S.A 100:4678−4683,2003)に記載の方法により、チオール特異的架橋剤4−アセトアミド4’−マレイミジル−スチルベン−2,2’−ジスルホネート(AMS;モレキュラープローブ)を用いて評価した。AMSの分子量マスに依存して、タンパク質中でシステイン残基を還元するこの試薬の架橋はSDS−PAGEの間のこのタンパク質の移動性を有意に減らす(Kobayashi et al, Proc. Natl. Acad. Sci. U.S.A 100:4678−4683,2003)。BdbDはCxxC活性部位である2つのシステイン残基しか含まない。BdbDの酸化還元部位を研究するために、一晩培養を行って、ItrxA株(25μMIPTG)、bdbC一部位変異体、ItrxA bdbC二重変異体、又は親株168由来の新しい細胞溶解液を調製した。
DsbAの発現はPhoAの分泌を高める
チオール酸化のためのバチルススブチリスの能力を高めるためのほかの可能性のあるアプローチは既知のチオールオキシダーゼの過剰生成を誘発することである。しかしながら、BdbC及びBdbDのレベルを高めるための試みは、今日まで成功していない。それ故、異種オキシダーゼを発現している可能性を評価した。
分泌されたPhoAの最適なレベルは酸化還元活性培地化合物のより促進される
E.coliのPhoAの細胞外地区製を促進しているスタフィロコカールDsbAの活性は育成培地における酸化還元活性化合物に依存していることが報告されている。このことはシステイン/シスチンを添加又は無添加の合成培地においてDsbAを生成するバチスルスブチリスを育成することにより示された。それ故、このDsbA活性が細胞がリッチなLB培地において育成された場合、このDsbA活性がシステイン又はシスチンのような追加的な酸化還元活性化合物の添加により高めることができるかを評価した。この目的のために、すべてのTrxA欠失及び/又はDsbA−発現株を育成し、並行培養で、100μg/mlのシステイン(即ち、酸化されたシスチン)/シスチンの存在下で行った。システイン又はシスチンをItrxA X−ScdbsA株に添加すると、PhoAの細胞外蓄積が強く高められた。この増加はシステインが添加された場合が顕著であった。更に、このポジティブトレンドはX−ScdsbA株でも見られた。しかし、刺激の程度はItrxA X−ScdsbA株よりも低かった。育成培地にシステイン又はシスチンを添加した他の株については、細胞外PhoAレベルは統計的に有意な増加は示さなかった。このPhoA活性データはウエスタンブロッティングで確認され、観察された全ての増加はPhoAタンパク質の高められたレベルに関係していることを示している。特に、システインの添加はPhoA分解生成物の蓄積を親は部168よりも減らした。

- i)宿主細胞において細胞質リダクターゼの活性を低めるための遺伝的修飾及びii)異種スタフィロコカール(Staphylococcal)オキシダーゼ又はオキシダーゼ活性を有しているそれらの変異体をコードしている核酸配列を含む、遺伝的に修飾されたバチルス宿主細胞。
- 前記細胞性リダクターゼがTrxAポリペプチドである、請求項1の宿主細胞。
- TxrAポリペプチドが、
i)配列番号2のアミノ酸配列
ii)配列番号1の核酸配列によりコードされるアミノ酸配列

- バチルス宿主細胞中でジスルフィド結合含有タンパク質を生成する方法であって、
i)配列番号2のアミノ酸配列を含むTrxAポリペプチドの宿主細胞中での発現を減少させる工程、
ii)配列番号4のアミノ酸配列を含む異種スタフィロコカール(Staphylococcal)DsbAポリペプチドを宿主細胞中で発現させる工程、
iii)酸化還元活性化合物を含む培地中で前記宿主細胞を育成する工程であって、前記宿主細胞はジスルフィド結合含有タンパク質を生成し、培地中にこのタンパク質を分泌する工程、からなる方法。 - 組換えタンパク質の折りたたみを改善する方法であって、
a)遺伝的に修飾されたバチルス宿主細胞を酸化還元活性化合物を含む培地中で育成して、ジスルフィド結合タンパク質を発現させ、タンパク質の折りたたみを改善する工程であって、前記宿主細胞が(i)細胞質リダクターゼ活性が減少しており、及び(ii)異種スタフィロコカールオキシダーゼ、又はオキシダーゼ活性を有するそれらの変異体の発現が増加しており、異種ジスルフィド結合含有タンパク質を分泌する工程、並びに
b)任意で、前記タンパク質を培地から単離する工程、より構成される、方法。 - 前記細胞質リダクターゼがTrxAポリペプチドである、請求項31の方法。
- 前記TrxAポリペプチドが
i)配列番号2のアミノ酸配列、
ii)配列番号1のアミノ酸配列によりコードされるアミノ酸配列、
iii)配列番号2のアミノ酸配列に少なくとも90%同一なアミノ酸配列、
iv)配列番号1の核酸配列に少なくとも90%同一な核酸配列によりコードされるアミノ酸配列、及び
v)配列番号1の相補体に過酷な条件下でハイブリダイズする核酸配列によりコードされるアミノ酸配列、からなる群より選択される、請求項32の方法。 - 前記オキシダーゼがオキシダーゼをコードしている遺伝子を含むプラスミドを用いた形質転換により導入される、請求項31、32、及び33のいずれか1項に記載の方法。
- オキシダーゼがDsbAポリペプチドである、請求項35の方法。
- 前記DsbAポリペプチドが
i)配列番号4のアミノ酸配列、
ii)配列番号3の核酸配列によりコードされるアミノ酸配列、
iii)配列番号4のアミノ酸配列に少なくとも90%同一であるアミノ酸配列、
iv)配列番号3の核酸配列に少なくとも90%同一である核酸配列によりコードされるアミノ酸配列、
v)配列番号3の相補体に過酷な条件下でハイブリダイズする核酸によりエンコードされるアミノ酸配列、からなる群より選択される、請求項35の方法。 - 前記宿主細胞がバチルススブチリス(Bacillus subtilis)である、請求項31乃至36のいずれか1項に記載の方法。
- 前記無病誘発スタフィロコッカス(Staphylococcus)バクテリウムが、スタフィロコカールカルノーサス(Staphylococcal carnosus)である、請求項31乃至36のいずれか1項に記載の方法。
- 細胞質リダクターゼ活性の減少をサ細胞質リダクターゼ遺伝子の変異により引き起こす、請求項31乃至36のいずれか1項に記載の方法。
- 前記変異が欠失変異、置換変異、又は挿入変異である、請求項39の方法。
- 前記リダクターゼ活性の減少が誘発プロモーターにより制御される、請求項31乃至36のいずれか1項に記載の方法。
- 前記酸化還元活性化合物が、システイン、シスチン、グルタチオン、2−メルカプトエタノール(BMT)1,4−ジチオトレイトール(DTT)、チオスルファート、亜ジチオン酸塩、メタビスルフィット、スルフィット、N−エチルマレイミド、又はマイコチオールからなる群より選択される、請求項31乃至41のいずれか、1項に記載の方法。
