本発明者らは、鋭意検討した結果、以下の方法が本課題を解決することがわかった。
エオシンを必要量溶媒に溶かす。溶媒は水、エタノール等の水溶性有機溶媒あるいはこれらの混液を用いる。
しかしながら、通常エタノール:水=50:50〜99:1を用いる。
これに対して、容量で5〜25%、好ましくは8〜20%の乳酸を加えて撹拌する。
さらに最初の水と水溶性有機溶媒との混液に溶かしたエオシン液を基準としてときの量として、酢酸を0.5〜10%加える。
発明者らが検討した結果、乳酸だけでは染色の状態が悪く少量の酢酸を加えることによって、酢酸臭が許容範囲で且つ染色状態のよい液を見出すことができた。
この染色液を用いて迅速にまた、染色液も少量ですむ方法を考えた。
これはテープストリッピング法で得た角層細胞の付着したテープをそのテープの付着力も利用して、用意した筒の或いは穴の有するホルダーの一方をこのテープに密着固定した。

また、筒の一方、又は両面の断面にフランジがあってもよい。
筒の高さ(長さ)は3〜20mm程度がよく、短すぎると必要量の染色液を筒に入れれないし、染色液がこぼれたりする。
