Abstract
Description
各包帯材構成要素は、好都合にはモノマーマトリックスの形態またはポリマーマトリックスの形態のどちらかである、キャリアまたは支持体を含む。各包帯材構成要素は、液体、非晶質ゲルの形態、または例えばシート、スラブまたは乾燥フィルムの形態などの層の形態であることができる。
親水性ポリマー材料の水和ヒドロゲル中では、親水性ポリマー材料は、ゲル総重量を基準として、望ましくは少なくとも1%、好ましくは少なくとも2%、より好ましくは少なくとも5%、なおもより好ましくは少なくとも10%、または少なくとも20%、望ましくは少なくとも25%、そしてなおもより望ましくは少なくとも30重量%の濃度で存在する。ゲル総重量を基準として約40重量%までのさらに多くの量を使用してもよい。
化学薬品およびその他の材料
水(導電率<10μScm−1;分析試薬等級、FisherまたはSanyo Fistreem MultiPureのどちらか)
亜硝酸ナトリウム、Sigma製(S2252)
チオグリセロール、Fluka製(88641)
ヒドロキノン、Sigma製(H9003)
塩酸、Fisher製(J/4310/17)
水溶液中のS−ニトロソチオール濃度の測定
次の試薬を調製した。
試薬1:Na−リン酸緩衝液(pH7.4、0.1M)
試薬2:Griess試薬:2mLのDMSOに溶解された20mgのN−(1−ナフチル)エチレンジアミン二塩酸塩(NADD)+500mgのスルファニルアミド(注記:この溶液は感光性であり、できる限り暗所に保存すること)
試薬3:DMSO中の塩化第二水銀(10mM)(5mLのDMSO中に13.58mgのHgCl2)
次に下述の通りである6段階の手順が続いた。
1.5mLの試薬1をプラスチックキュベットに分注する。
200μLのサンプル(すなわちその中でGSNO濃度を判定するサンプル)を添加する。
1.17mLのDI水を添加する。
100μLの試薬2を添加する。
30μLの試薬3を添加して溶液をよく混合する。
得られた混合物の吸光度を496nmで10分間読み取る。
実施例1:ヒドロキノンおよび緩衝されていない亜硝酸ナトリウムの酸性化溶液を含有する混合物中の酸化窒素発生に起因するpHの変化
pHは、ヒドロキノン(すなわちモデル非チオール還元性化合物)の不在下および存在下の双方において、酸性化された亜硝酸塩溶液中で測定された。亜硝酸塩を塩酸の添加によって酸性化し、10mM(図1)または4mM(図2)の最終濃度を得た。反応混合物を酸性化するのに10mM塩酸を使用した場合、pHは還元剤の不在下でおよそ3.8に保たれた。対照的にヒドロキノンの存在はpHの約5.0への迅速な増大をもたらし、さらなる緩慢な増大がそれに続いた。混合物を酸性化するのに4mM塩酸を使用した場合は、最初のpHがより高かった(約4.2)こと以外は、同様のpHプロフィールが観察された(図2)。どちらの場合もpHの増大は、混合物中の酸化窒素の形成を反映する混合物中の気泡形成を伴った。還元剤の不在下では、気泡形成は観察されなかった。
Claims (9)
- プロトン源を含む第1の構成要素、亜硝酸塩を含む第2の構成要素を含み、前記第1および第2の構成要素が、一緒にされて皮膚部位に適用された際に、亜硝酸塩が反応して酸化窒素を発生し、皮膚と接する包帯材のpHを酸性値からより中性の値に増大させるように、非チオール還元剤を含む、皮膚包帯材。
- 前記第1の構成要素のpHが2.0〜5.0である、請求項1に記載の皮膚包帯材。
- 1.0〜4.0のpKaを有するいかなる追加的材料も含まない、請求項1または請求項2に記載の皮膚包帯材。
- 前記プロトン源が4.5〜7.0のpKaを有する緩衝液を含む、請求項1〜3のいずれか一項に記載の皮膚包帯材。
- 亜硝酸塩が反応して酸化窒素または酸化窒素供与体を発生するにつれて、皮膚と接する包帯材のpHが5.0未満から5.0超に増大する請求項1〜4のいずれか一項に記載の皮膚包帯材。
- 前記第2の構成要素のpHが5.0〜12.0である、請求項1〜5のいずれか一項に記載の皮膚包帯材。
- 前記第1および第2の構成要素が非晶質である、請求項1〜6のいずれか一項に記載の皮膚包帯材。
- 前記第1の構成要素および/または第2の構成要素がポリマー支持体を含む、請求項1〜7のいずれか一項に記載の皮膚包帯材。
- 前記第1の構成要素がポリアクリル酸ベースのポリマー支持体を含む、請求項8に記載の皮膚包帯材。
