Description
[0001] 本発明は、概して、骨成長の誘発又は骨損失の抑制のための組成物及び方法に関する。特に、骨成長の誘発又は骨損失の抑制のためのイソフラボン又はその代謝物の使用に関する。
[0002] 骨のリモデリングは継続的なサイクルである。このサイクルは、破骨細胞が媒介する骨吸収とともに始まり、その後、骨芽細胞による骨量回復が起こる。骨のリモデリング過程は、主に性ホルモン、特にエストロゲンによって調節される。ただし、遺伝因子、栄養因子及び環境因子が、骨の代謝回転に影響しうる。女性はもとより、男性における骨のリモデリングでも、エストロゲンが主要な役割を果たすことが示されている。エストロゲンは、骨髄前駆体からの破骨細胞形成及び骨芽細胞形成を抑制することで骨のリモデリングを低下させ、吸収促進性サイトカインの低減により骨吸収を抑制し、骨芽細胞の寿命及び活性を調節する。
[0015] 本明細書では、以下の略語を用いることがある。CLA:共役リノール酸;BCS:ボディコンディションスコア;BMR:基礎代謝率;MER:維持エネルギー要求量;DEXA:二重エネルギーX線吸収法;BMC:骨ミネラル含量;BMD:骨ミネラル密度;BW:体重;SGM:大豆胚芽粉。
[0041] 一態様において、本発明は、動物における骨成長の誘発又は骨損失の抑制に適した組成物を提供する。この組成物は、1種以上のイソフラボン又はその代謝物を、動物における骨成長の誘発又は骨損失の抑制に有効な量で含む。本発明は、イソフラボン及びその各種の代謝物が、動物における健常な骨成長及び骨発達の促進、並びに骨損失の抑制に有効であるという発見に基づく。本発明は、幼若な発育中の動物における健常な骨成長を促進し、成体動物における骨損失を低下させるのに有用である。上記組成物は、閉経後、アンドロポーズ後、性腺摘出後、不妊処置後又は去勢後の動物において、特に動物の骨格系が成長、成熟しつつある場合において、骨成長を誘発し、又は骨損失を抑制するために特に有効であることが判明している。
[0076] イヌと食餌:正常、非肥満で、新規に性腺摘出した、4〜5カ月齢のラブラドールレトリーバー犬30匹を、試験に用いた。仔犬を同腹子に基づき以下の3群に無作為化し、骨格筋の成長及び健康に対する遺伝的影響を最小化した。第1群は、新規に性腺摘出した雌雄のラブラドールレトリーバー犬10匹からなり、これには、29〜30%のタンパク質及び20%の脂肪を含む、バランスの良い仔犬用食餌(対照食餌)を与えた。第2群は、新規に性腺摘出した雌雄のラブラドールレトリーバー犬10匹からなり、これには、対照食餌に10%の大豆胚芽粉(SGM)を補充して与えた。第3群は、新規に性腺摘出した雌雄のラブラドールレトリーバー犬10匹からなり、これには、対照食餌に10%のSGM、100ppmのL−カルニチン、及び1.5%のCLAを補充して与えた。SGMは、以下の大豆イソフラボンを含む。17%のゲニステイン、52%のダイゼイン、及び31%のグリシテイン。食物摂取を調整することにより、仔犬すべてに食事を与え、ボディコンディションスコア5を維持した。試験期間は、70週間であった。
Claims (25)
- 1種以上のイソフラボン又はその代謝物を、動物における骨成長の誘発又は骨損失の抑制に有効な量で含む組成物。
- ヒト用食品組成物、ペットフード組成物又は栄養補助食品である、請求項1に記載の組成物。
- 約0.1%〜約50%のイソフラボン又はその代謝物を含有する、請求項2に記載の食品組成物。
- イソフラボンが、ダイゼイン、6−O−マロニルダイゼイン、6−O−アセチルダイゼイン、ゲニステイン、6−O−マロニルゲニステイン、6−O−アセチルゲニステイン、グリシテイン、6−O−マロニルグリシテイン、6−O−アセチルグリシテイン、バイオチャニンA又はフォルモノネチンの少なくとも1種を含む、請求項1に記載の組成物。
- イソフラボン又はその代謝物が大豆イソフラボン又はその代謝物である、請求項1に記載の組成物。
- 大豆イソフラボンの代謝物がエコールである、請求項5に記載の組成物。
- 前記動物がイヌ又はネコである、請求項1に記載の組成物。
- 前記動物が、閉経後、アンドロポーズ後、性腺摘出後、不妊処置後又は去勢後の動物である、請求項1に記載の組成物。
- DT56a、アンドロステンジオン、デヒドロエピアンドロステロン(DHEA)、ケイ素、L−カルニチン、共役リノール酸及びオルトケイ酸の少なくとも1種をさらに含む、請求項1に記載の組成物。
- 動物における骨成長の誘発又は骨損失の抑制のための方法であって、
1種以上のイソフラボン又はその代謝物を、動物における骨成長の誘発又は骨損失の抑制に有効な量で含む組成物、を動物に投与するステップを含む方法。 - 前記組成物が、ヒト用食品組成物、ペットフード組成物又は栄養補助食品である、請求項10に記載の方法。
- 食品組成物が、約0.1%〜約50%のイソフラボン又はその代謝物を含む、請求項11に記載の方法。
- イソフラボンが、ダイゼイン、6−O−マロニルダイゼイン、6−O−アセチルダイゼイン、ゲニステイン、6−O−マロニルゲニステイン、6−O−アセチルゲニステイン、グリシテイン、6−O−マロニルグリシテイン、6−O−アセチルグリシテイン、バイオチャニンA又はフォルモノネチンの少なくとも1種を含む、請求項10に記載の方法。

- 前記組成物が動物に毎日投与される、請求項10に記載の方法。
- 前記組成物が、DT56a、アンドロステンジオン、デヒドロエピアンドロステロン(DHEA)、ケイ素、L−カルニチン、共役リノール酸及びオルトケイ酸の少なくとも1種をさらに含む、請求項10に記載の方法。
- イソフラボン又はその代謝物が、1種以上の骨作用物質と併用して投与される、請求項10に記載の方法。
- 前記動物がイヌ又はネコである、請求項10に記載の方法。
- 1種以上のイソフラボン又はその代謝物を含む組成物を動物に投与するのに適したキットであって、
キット構成成分に応じて、単一パッケージ中の別々の容器内、又は仮想パッケージ中の別々の容器内に、1種以上のイソフラボン又はその代謝物と、(1)動物による摂取に適した1種以上の成分、(2)骨成長の誘発又は骨損失の抑制に適した1種以上の骨作用物質、(3)イソフラボン又はその代謝物と他のキット構成成分との混合法に関する説明書、及び(4)イソフラボン又はその代謝物及び本発明の他の構成成分の使用法に関する説明書、の少なくとも1つと、を含むキット。 - (1)イソフラボン又はその代謝物を用いて、動物において骨成長を誘発し、又は骨損失を抑制すること、(2)イソフラボン又はその代謝物を本発明の他の構成成分と混合すること、(3)イソフラボン又はその代謝物を単独で、又は本発明の他の構成要素とともに動物に投与すること、及び(4)本発明のキットを用いてイソフラボン又はその代謝物を動物に投与すること、の1つ又は複数に関する情報又は指示を伝達するための手段であって、
