多くの生物学的過程は、動的で可動的な細胞集団によって行われる。例えば、免疫系の細胞が血流から炎症部位または感染部位まで集められて、罹患部分での免疫細胞の蓄積をもたらす。免疫細胞の顕著な浸潤が、自己免疫疾患、癌、および感染症に罹患した組織において起こる場合が多い。同様に、移植拒絶は、移植された組織に侵入し、そして破壊する宿主の免疫細胞によって媒介される。骨髄に由来する幹細胞が血流を通じて移動し、そして損傷を受けた組織の再生を助けるとの証拠が増えつつある。

1.概観
本開示は、インビボでの検出のための細胞のインビトロ標識のパラメーターの迅速かつ効率的な最適化を可能にする過程について記載している。ここで、「インビボ検出剤」は、ポジトロン放出断層撮影法(PET)技術、γカメラ、SPECT(単光子放射型コンピューター断層撮影法)、またはNMR技術(例えば、MRIまたはMRS)によってヒトまたは他の哺乳動物において検出される薬剤と理解されるであろう。「MRI標識」または「インビボの造影試薬」は、核磁気共鳴技術(例えば、MRIまたはMRS)を使用して検出できる任意の常磁性造影剤または19Fトレーサー薬剤(例えば、ペルフルオロポリエーテル乳剤)であると理解される。「撮像」と「検出」は同義と理解される。

本明細書に記載の通り、核磁気共鳴技術を使用して、標識細胞の集団を検出できる。「検出する」という用語は、特に核磁気共鳴技術によって、標識された分子または細胞の有無を確認するための任意の取り組みを含むように使用される。「検出する」という用語は、定量的測定および二次元または三次画像の作成を含むより洗練された測定を含むことも意図される。例えば、MRIを使用して、このような細胞の画像を作成できる。多くの場合、標識細胞は生きた被験体に投与できる。細胞の投与後、被験体の一部または被験体の全体をMRIで検査して、MRIデータセットを作成できる。本明細書で使用する「データセット」という用語は、被験物質の磁気共鳴プロービング中に収集された生データならびに処理された、変換された、もしくは生データから抽出された情報を含むように意図される。処理された情報の例としては、被験物質の2次元または3次元画像表示が挙げられる。抽出された情報の他の例は、被験物質中の造影試薬または19Fシグナルの量または濃度を示すスコアである。例えば、19Fシグナルの信号雑音比(SNR)を測定し、そして標識細胞の存在量を計算するために使用できる。この種類のデータは、例えば、特に標識細胞に関連する脾臓または別の臓器などの被験体の1つの領域で収集できる。標識細胞を被験体以外との関連で試験できる。培養中の標識細胞を試験することが望まれうる。特定の態様では、標識細胞を組織サンプルまたは組織培養に適用する、もしくはこれらの中で作成させることができ、したがって標識細胞をこれらの関連で撮像することもできる。例えば、臓器、組織、または移植された他の細胞性物質を造影試薬と接触させて、被験体においてこのような移植片の移植前に標識細胞を作成できる。

蛍光検出を可能にする造影試薬は、特に様々な用途で有用である。例えば、蛍光標識によって、蛍光活性化細胞選別(FACS)などの蛍光ベースの細胞選別機序の使用が可能になる。細胞選別は、例えば、標識の成功した細胞集団を濃縮するために望ましい。これは、標識がよりまれな細胞集団を対象とする場合に特に有用でありうる。代用試薬はデュアルモード薬剤でよく、そしてこれは、それらが生きている被験体に移植された後に、標識細胞を見いだし、そして特性付けする際にさらに有用である。この用途では、細胞の生検を行うことができ、または、一定期間にわたり存在した後、被験体から他の手段によって回収できる。そして次に、回収した細胞の生物学的解析を実施できる。例えば、FACS分析を回収した細胞に実施して、ここでは蛍光PFPE標識について細胞を陽性選択後、(異なるカラー蛍光プローブを使用して)特異的な細胞表面マーカーの発現について細胞を分析して、移植後に起こる細胞表現型の変化を研究することができる。蛍光標識は、特に3次元共焦点蛍光顕微鏡を使用して、細胞の蛍光顕微鏡にも使用できる。

造影試薬または代用試薬により細胞を標識するために、様々な方法を利用できる。一般に、造影試薬が細胞に結合するように、細胞は造影試薬と接触させて置かれる。条件は、細胞生存率を維持するように設計された標準的な細胞培養条件である場合が多い。「結合した(associated)」という用語は、インビボまたはインビトロを問わず、造影試薬が細胞の位置に関する有用な情報を提供できるように、造影試薬と細胞が十分な時間にわたり十分に近い物理的近接内にとどまるようにするための任意の方法を含むことが意図される。例えば、食作用、液相エンドサイトーシス、受容体媒介性エンドサイトーシス、陽イオン部分の付加、トランスフェクション、エレクトロポレーション、または界面活性物質媒介性の細胞内侵入後に造影試薬は細胞内に局在化できる。樹状細胞、マクロファージ、およびT細胞などの免疫細胞は高い食作用を持ち、そして本明細書および他の試験で示されるデータによって、このような細胞および他の食細胞型が容易に標識されることが実証される。
a.最適な標識パラメーターを決定するための方法
標準的な無菌組織培養法を使用して、標識試薬を対象の細胞に結合させることができる。一部の態様では、対象の細胞は、細胞サンプルから採取される細胞を含む複数のサブサンプルである。一部の態様では、サンプルはヒト患者などの被験体由来である。

これらの方法の実施可能性の例として、蛍光MRIデュアルモード薬剤を合成できるか否か、非蛍光バージョンとして類似の特性を持つ細胞を標識するために使用できるか否か、ならびに蛍光強度と細胞の19F含量の間に相関関係があるか否かについて分析する。従って、標識細胞用に開発される組織培養プロトコルでは、PFPEナノ粒子の蛍光および非蛍光バージョンのいずれでも使用できる。さらに、細胞標識プロトコル開発および/またはバリデーションの一部として重要パラメーターであるFcのインビトロ測定において、蛍光アナログまたはデュアルモード薬剤を使用できる。例えば、高価な19F NMR機器よりもむしろ安価な蛍光光度計を使用して、Fcにより測定された平均細胞負荷量を評価でき、そして非蛍光バージョンのPFPEを使用して、その後のインビボ実験においてFcの結果を使用できる。
19F NMRによって測定された取り込み量と線形相関を示すデュアルモード薬剤または他の代用薬剤を使用して、出願の方法によるハイスループットフォーマットにおいて過去に未試験の細胞型および患者のサンプルにおける細胞標識を評価する。

Claims (34)
- 患者への投与が意図された細胞を標識することを評価するための方法であって、前記方法が:
a)実質的に同じ複数の細胞サンプルの細胞をインビボの造影試薬と接触させること;
b)前記サンプル中の前記インビボの造影試薬を検出すること;ならびに
c)前記インビボの造影試薬での細胞の標識を評価すること;
を含む前記方法。 - 患者への投与が意図された細胞を標識することを評価するための方法であって、前記方法が:
a)実質的に同じ複数の細胞サンプルの細胞をインビボの造影試薬および代用試薬と接触させること;
b)前記サンプル中の前記代用試薬を検出すること;ならびに
c)前記代用試薬で細胞の標識を評価し、これにより前記インビボの造影試薬での細胞の標識を評価すること;
を含む前記方法。 - 少なくとも2個の実質的に同じ細胞サンプルを互いに異なる所定の条件にさらし、そして前記方法は、異なる条件がインビボの前記造影試薬での細胞の標識に及ぼす効果を評価することをさらに含む、請求項1または2に記載の方法。
- 少なくとも10個の実質的に同じ細胞サンプルを互いに異なる所定の条件にさらす、請求項3記載の方法。
- 自動化ハイスループット技術の使用により、前記細胞をインビボの造影試薬またはインビボの造影試薬および代用試薬と接触させる、請求項4記載の方法。
- 自動化ハイスループット技術を利用してインビボの造影試薬での前記細胞の標識を評価する、請求項5記載の方法。
- 前記インビボの造影試薬が核磁気共鳴技術によるインビボの検出に適した薬剤である、請求項1〜6のいずれか1項に記載の方法。
- 前記インビボの造影試薬が、常磁性造影剤、超常磁性酸化鉄粒子、磁鉄鉱粒子、フルオロカーボン造影試薬、Gdキレート、およびMnキレートからなる群より選択される、請求項7記載の方法。
- 前記インビボの造影試薬がPFPE乳剤を含む、請求項8記載の方法。
- 前記インビボの造影試薬が、ポジトロン放出断層撮影法(PET)、γ線検出、またはSPECT(単光子放射型コンピューター断層撮影法)からなる群より選択される非侵襲性の撮像技術による検出に適した薬剤である、請求項1〜6のいずれか1項に記載の方法。
- 前記代用試薬が、蛍光タンパク質または発光タンパク質、蛍光アナログ剤または発光アナログ剤、蛍光色素または発光色素、比色剤、ならびに放射性医薬品からなる群より選択される、請求項2記載の方法。
- 前記インビボの造影試薬が代用試薬と安定的に結合して、デュアル造影試薬を形成する、請求項2記載の方法。
- 前記インビボの造影試薬が核磁気共鳴技術によるインビボの検出に適した薬剤である、請求項12記載の方法。
- 前記代用試薬が、蛍光タンパク質または発光タンパク質、蛍光アナログ剤または発光アナログ剤、蛍光色素または発光色素、比色剤、ならびに放射性医薬品からなる群より選択される、請求項12記載の方法。
- 互いに異なる前記所定の条件が、温度、大気組成、湿度、インビボの標識薬剤の培養液中濃度、インビボの標識薬剤への曝露時間、薬剤取り込み促進剤の濃度、および薬剤取り込み促進剤への曝露時間からなる群より選択される1またはそれ以上の局面について異なる、請求項3〜6のいずれか1項に記載の方法。
- 患者に投与する細胞用量の調整を可能にする前記インビボの造影試薬での細胞の標識条件を決定することをさらに含み、前記用量がインビボの造影試薬に関連する既知の安全性パラメーターに準拠して、インビボでの標識細胞の検出を可能にする適切な標識を提供する、請求項1〜6のいずれか1項に記載の方法。
- 前記患者に細胞用量を投与することをさらに含み、前記用量がインビボの造影試薬に関連する既知の安全性パラメーターに準拠して、インビボでの標識細胞の検出を可能にする適切な標識を提供する、請求項1〜6のいずれか1項に記載の方法。
- 生死判別試験、細胞数、細胞周期アッセイ、遊走アッセイ、および機能アッセイからなる群より選択される追加アッセイをさらに含む、請求項3〜6のいずれか1項に記載の方法。
- 前記患者が哺乳動物である、請求項1〜6のいずれか1項に記載の方法。
- 前記患者がヒトである、請求項1〜6のいずれか1項に記載の方法。
- 前記細胞が前記患者にとって自己由来である、請求項1〜6のいずれか1項に記載の方法。
- 前記細胞が前記患者にとって同種異系である、請求項1〜6のいずれか1項に記載の方法。
- 細胞が、血液細胞、筋原細胞、骨髄細胞、末梢血細胞、臍帯血細胞、心筋細胞、軟骨細胞、免疫細胞、胎児の神経細胞、神経系前駆細胞、線維芽細胞、肝実質細胞、膵臓の膵島細胞、ケラチノサイト、および前述のいずれかの前駆細胞からなる群より選択される、請求項1〜6のいずれか1項に記載の方法。
- 前記細胞が、胚性幹細胞、胚性幹細胞から培養した細胞、成体幹細胞、および成体幹細胞から培養された細胞からなる群より選択される、請求項1〜6のいずれか1項に記載の方法。
- 前記実質的に同じサンプルが、ドナーから得られた細胞のサンプルの各一部分である、請求項1〜6のいずれか1項に記載の方法。
- 前記実質的に同じサンプルが、ドナーに由来する細胞から培養した細胞の各一部分である、請求項1〜6のいずれか1項に記載の方法。
- タンパク質代用試薬と安定的に結合したインビボの造影試薬を含む、細胞を標識するための組成物。
- 前記タンパク質代用試薬が、蛍光タンパク質、発光タンパク質、着色タンパク質、蛍光発生タンパク質、発光タンパク質、および色素生成タンパク質からなる群より選択される、請求項27記載の組成物。
- 前記インビボの造影試薬での細胞の標識を評価することが、細胞に結合したインビボの造影試薬の量を定量することを含む、請求項1記載の方法。
- 前記インビボの造影試薬での細胞の標識を評価することが、細胞に結合した代用試薬の量を定量することを含む、請求項2記載の方法。
- 患者に安全で有用な用量の標識細胞を投与し、そしてインビボで前記標識細胞を検出する方法であって、前記方法は:
a)実質的に同じ複数の細胞サンプルの細胞をインビボの造影試薬と接触させること;
b)前記サンプル中の前記インビボの造影試薬を検出すること;
c)前記インビボの造影試薬での細胞の標識を評価すること;
d)患者に投与する細胞用量の調製を可能にするインビボの造影試薬での前記細胞の標識条件を決定すること、ここで前記用量はインビボの標識薬剤に関連する既知の安全性パラメーターに準拠して、インビボでの標識細胞の検出を可能にする適切な標識を提供する;
e)前記患者に細胞用量を投与すること;ならびに
f)非侵襲性の撮像技術によってインビボの前記標識細胞を検出すること、
を含む、方法。 - 前記非侵襲性の撮像技術が核磁気共鳴技術である、請求項31記載の方法。
- 患者に安全で有用な用量の標識細胞を投与し、そしてインビボで前記標識細胞を検出する方法であって、前記方法は:
a)実質的に同じ複数の細胞サンプルの細胞をインビボの造影試薬および代用試薬と接触させること;
b)前記サンプル中の前記代用試薬を検出すること;
c)前記代用試薬で細胞の標識を評価し、これにより前記インビボの造影試薬での細胞の標識を評価すること;
d)患者に投与する細胞用量の調製を可能にするインビボの造影試薬での前記細胞の標識条件を決定すること、ここで用量はインビボの標識薬剤に関連する既知の安全性パラメーターに準拠して、インビボでの標識細胞の検出を可能にする適切な標識を提供すること;
e)前記患者に細胞用量を投与すること;ならびに
f)非侵襲性の撮像技術によってインビボの前記標識細胞を検出すること、
を含む、方法 - 前記非侵襲性の撮像技術が核磁気共鳴技術である、請求項33記載の方法。
