若齢対高齢のp16INK4a−/−動物の連続移植能における差異は、著しかった。p16INK4a発現が、恒常状態下の若齢HSCに見出せなかったので、若齢動物における影響は予期しないものであった。しかし、移植後の若齢マウス由来の骨髄をテストした場合、他のストレス状態下の他により見られるように、低いレベルのp16INK4a発現が見られた(データは示さず)(Chkhotua, A.B. et al. Am. J. Kidney Dis. 41, 1303-1313 (2003))(Chimenti, C. et al. Circ. Res. 93, 604-613 (2003))。この場合のHSCにおけるp16INK4a欠損の有害な影響は、p16INK4aとARFの間の公知のプロモーターの競合によるかもしれないが、結果的にp16INK4a の欠損によりARFのわずかな増加をもたらした(Sharpless, et al. Oncogene 22, 5055-5059 (2003))。ARFの発現は、HSCの死を著しく増加させることが示されている(Park, I.K. et al. Nature 423, 302-305 (2003))。逆に、p16INK4a及びARFの2つの欠如又はARF単独では、若齢動物の連続移植において何れの欠損ももたらさないことが示されている(Stepanova, L. et al. Blood (2005))。現実に、二重に欠損の動物は、自己再生において僅かな増加を有する(Stepanova, L. et al. Blood (2005))。p16INK4aが存在しない状態での年齢に伴う自己再生が目立って改善されたことは、p16INK4aの年齢依存性の増加により誘導される分子の事象が軽減することに起因しているという仮説を立てた。
Claims (19)
- p16INK4aの抑制剤を含有してなる医薬組成物。
- 医薬組成物が、幹細胞の自己再生の維持又は促進するためのものである請求項1に記載の医薬組成物。
- 医薬組成物が、自己再生する幹細胞の量を増加させるためのものである請求項1又は2に記載の医薬組成物。
- 医薬組成物が、幹細胞の移植を増進するためのものである請求項1から3のいずれかに記載の医薬組成物。
- 幹細胞が、p16INK4aを発現する幹細胞である請求項2から4のいずれかに記載の医薬組成物。
- 医薬組成物が、血小板減少症、貧血症、リンパ球減少症、リンパ漏、リンパうっ滞、赤血球減少症、赤血球変性疾患、赤芽球減少症、白赤芽球症、赤血球崩壊、サラセミア、骨髄線維症、血小板減少症、播種性血管内凝固症(DIC)、免疫性血小板減少性紫斑病(ITP)、HIV合併ITP、骨髄異形成症、血小板性疾患(thrombocytotic disease)、血小板増加症、好中球減少症、骨髄異形成症候群、感染症、免疫不全症、リウマチ性関節炎、ループス、免疫抑制、全身性エリテマトーデス、リウマチ性関節炎、自己免疫性甲状腺炎、強皮症及び炎症性腸疾患よりなる群から選ばれる疾病の治療又は予防用である請求項1から5のいずれかに記載の医薬組成物。
- 医薬組成物が、骨髄移植時に投与されるものである、請求項1から6のいずれかに記載の医薬組成物。
- p16INK4aの抑制剤が、p16INK4aの発現を減少させるものである、請求項1から7のいずれかに記載の医薬組成物。
- p16INK4aの抑制剤が、p16INK4amRNAのレベルを不安定化する又は低減することのできる化合物、p16INK4amRNAの翻訳を低減することのできる化合物、並びにp16INK4a、テロメラーゼ逆転写酵素(hTERT)、DNA結合/分化抑制因子(Id又はId−1)、潜伏感染膜タンパク質(LMP1)、へリックス・ループ・へリックス転写因子TAL1/SCL、ダイオキシン及びシクロオキシゲナーゼ 2(COX−2)を高メチル化できる化合物よりなる群から選ばれる化合物である、請求項8に記載の医薬組成物。
- p16INK4aの抑制剤が、p16INK4aの活性を低減するものである、請求項1から7のいずれかに記載の医薬組成物。
- p16INK4aの抑制剤が、p16INK4a抗体、並びにp16INK4a、テロメラーゼ逆転写酵素(hTERT)、皮膚性ヒトパピローマ・ウイルス16型(HPV16)E7タンパク質及びサイクリンD1を高メチル化できる化合物よりなる群から選ばれる、請求項10に記載の医薬組成物。
- 医薬組成物が、対象から得られた幹細胞を包含する単離された細胞集団に接触させるものである請求項1から11のいずれかに記載の医薬組成物。
- 細胞集団が、骨髄細胞又は造血幹細胞を含んでなる、請求項12に記載の医薬組成物。
- 細胞集団が、Lin−、cKit−及びScal+である、請求項12又は13に記載の医薬組成物。
- 幹細胞でhes−1及びgfi−1の発現が増加する、請求項2から14のいずれかに記載の医薬組成物。
- 請求項1から15のいずれかに記載の医薬組成物を、幹細胞にエクスビボで接触させること、それにより幹細胞の自己再生を維持又は促進すること、の段階を含んでなる、
p16INK4aを発現する幹細胞の自己再生を維持又は促進させる方法。 - 幹細胞が、骨髄由来の幹細胞又は造血幹細胞である、請求項16に記載の方法。
- 幹細胞が、間葉、皮膚、神経、腸、肝臓、心臓、前立腺、乳腺、腎臓、膵臓、網膜及び肺の幹細胞よりなる群から選ばれる、請求項16に記載の方法。
- p16INK4aを発現する幹細胞を包含する単離された細胞集団に、p16INK4aの抑制剤であると思われる薬剤を接触させること、及び
長期的に再増殖する細胞の総数の増加を検出すること、それにより幹細胞の自己再生を促進するp16INK4aの抑制剤を特定すること、を含んでなる、
当該薬剤が幹細胞の自己再生を促進する、p16INK4aの抑制剤であることをスクリーニングする方法。
