材料
凍結乾燥されたキングコブラの粗毒液はPT Venom Indo Persada(Jakarta、Indonesia)から得た。キングコブラの毒腺および肝臓は、寛大にもDepartment of Biological Sciences、National University of Singapore、SingaporeのDr Bryan G.Fryから提供された。腺および肝臓を直ちに液体窒素で凍結し、使用時まで−70℃で維持した。全ての化学物質および試薬は、次のものを除いて全てSigma(St.Louis、MO、USA)より購入した:Lys−CエンドペプチダーゼおよびトリプシンはWako Pure Chemicals(Osaka、Japan)より購入し、エドマン分解N末端配列決定用の試薬(Applied Biosystem、Foster City、CA、USA)、アセトニトリル(Merck KGaA、Darmstadt、Germany)、LB培地および寒天はQ.BIOgene(Irvine、CA、USA)より購入し、SDS−PAGEゲル標準物質(Prestained broad range SDS−PAGE標準物質およびPrecision plus prestaineddual−color標準物質)はBio−Rad Laboratories(Hercules、CA、USA)から購入した。Superdex 30 Hiload(16/60)およびμRPC C2/C18(10μ 120Å 2.1mm×100mm)カラムはAmersham Pharmacia(Uppsala、Sweden)から得た。RP−Jupiter C18(5μ 300Å 1mm×150mm)およびRP−Jupiter C18(10μ 300Å 10mm×250mm)カラムはPhenomenex(Torrance、CA、USA)から購入した。ニッケル−NTAアガロースはQiagen GmbH(Hilden、Germany)から購入した。オリゴヌクレオチドは全てBioBasic(Shanghai、China)および1st Base Pte.Ltd.(Singapore)から購入した。Platinum Taqポリメーラゼ、dNTPミックス、およびラダー(50bp、100bp、および1Kb Plus)はGIBCO BRL(登録商標)(Carlsbad、CA、USA)から購入した。使用した全ての制限エンドヌクレアーゼは、New England Biolabs(登録商標)(Beverly、MA、USA)から、またpGEMT−easyベクターはPromega(Madison、WI、USA)から得た。RNeasy(登録商標)Miniキット、QIAGEN(登録商標)OneStep RT−PCRキット、QIAprep(登録商標)Miniprepキット、QIAEX II Gel ExtractionキットおよびDNeasy(登録商標)TissueキットはQiagen GmbH(Hilden、Germany)から購入した。SMART(商標)RACE cDNA AmplificationキットはClontech Laboratories Inc.(Palo Alto、CA、USA)から購入した。ABI PRISM(登録商標)BigDye(商標)Terminator Cycle Sequencing Ready Reactionキット(バージョン3.0)はPE Applied Biosystem(Foster City、CA、USA)から購入した。水はMilliQシステム(Millipore、Billerica、MA、USA)を使って精製した。
動物実験には雄のSwissアルビノマウス(20±2g)を使用した。環境の変化および行動研究中の取り扱いからの影響を軽減するために、それぞれの場合について、マウスをLaboratory Animal Holding Centerおよび研究環境に3日間および少なくとも実験前1時間順応させた。動物は標準条件に維持され、食事(低タンパク質食)および水は自由に摂取させた。動物は照明管理された(12時間の明暗サイクル、07:00に点灯)23℃、60%相対湿度の室の中でケージ当たり4匹ずつ飼育した。全ての行動実験は、08:30時〜13:00時の間に行われた。各試験群は、少なくとも7匹のマウスから成り、各マウスは1回のみ使用した。全ての動物実験は、Laboratory Animal Center of the National University of Singporeが定めるガイドラインに従って行われた(Howard−Jones(10)に適合)。
凍結乾燥された粗毒液(60μg)は、Perkin−Elmer Sciex API300 LC/MS/MSシステム質量分析装置(Thornton、Canada)に接続された、0.1%(v/v)TFA(トリフルオロ酢酸)で平衡化しておいたRP−Jupiter C18分析カラムに直接注入する前に、20μlのMilliQ水に溶解した。粗混合液は、80%(v/v)ACN(アセトニトリル)の0.1%TFA溶液の直線勾配を用いて、流速50μl/分で溶出した。ESI/MS(エレクトロスプレー質量スペクトル)データは、オリフィス電子80Vの陽イオンモードで得た。窒素は、カーテンガスとして流速0.6リットル/分で、また噴霧ガスとしては100psiの圧力設定で使用した。完全スキャンデータを、500〜3000m/zのイオン幅について、0.1Da刻みに獲得した。データ処理は、BioMultiviewソフトウエア(Perkin−Elmer Sciex、Thornton、Canada)の助けを借りて行った。
凍結乾燥された粗毒液(各200mgの複数のバッチ)を2mlのMilliQ水に溶解し、50mMのTris−HCl(pH7.4)で予備平衡化しておいたSuperdex 30カラムにかけた。タンパク質はFPLC(Fast Protein Liquid Chromatography system、Amersham Pharmacia、Uppsala、Sweden)で50mMのTris−HCl(pH7.4)を用いて流速1ml/分で溶出した。タンパク質の溶出は、280nmでモニタリングした。次に関心対象の分画をVision Workstation(PE Applied Biosystem、Foster City、CA、USA)を用いて、0.1%TFA(v/v)で平衡化したRP−Jupiter C18半調製カラムにかけた。結合したタンパク質は、80%ACNの0.1%TFA(v/v)溶液の直線勾配を用いて、流速2ml/分で1時間かけて溶出した。タンパク質の溶出は280nmおよび215nmでモニタリングした。関心対象のタンパク質は、質量を決定して(以下に記載する)同定した。
凍結乾燥し精製された関心対象タンパク質を、既に報告した方法(11)を用いて還元し、ピリジルエチル化した。タンパク質(500μg)を500μlの変性緩衝液(6M GdnCl(グアニジン塩酸)、50mM Tris−HCl、1mM EDTA pH8.0)に溶解した。β−ME(β−メルカプトエタノール)を10μl加えた後、混合液を37℃で2時間、真空下でインキュベーションした。続いて4−ビニルピリジンを20μl混合液に加えて、室温(〜25℃)で更に2時間、真空下に維持した。還元し、ピリジルエチル化されたタンパク質を、SMART Workstation(Amersham Pharmacia、Uppsala、Sweden)上の0.1%のTFA(v/v)で平衡化したRP−μRPC C2/C18分析カラムにかけた。結合したタンパク質を、80%ACNの0.1%TFA(v/v)溶液の直線勾配を用いて、1時間かけて流速200μl/分で溶出した。タンパク質の溶出は280nmおよび215nmでモニタリングした。
Lys−Cエンドペプチダーゼおよびトリプシンによるピリジルエチル化タンパク質の消化を、37℃で20時間行った。タンパク質(150μg)を150μlの酵素消化緩衝液(50mM Tris−HCl、4M尿素、5mM EDTA pH7.5)に溶解し、プロテアーゼを1:50(w/w)の割合で加えた。
還元ピリジルエチル化タンパク質のギ酸による消化(Asp特異的)は、Inglis(12)が記載した通りに実施した。簡単に説明すると、ピリジルエチル化タンパク質150μgをガラス製バイアルに入った2%のギ酸に溶解し、次に−30℃で凍結した。続いて真空下、室温でバイアルを融解してから開封した。次にバイアルを108℃で2時間加熱し、室温まで冷ました。
酵素消化および化学的消化の両方によって生成されたペプチドを、SMART Workstation(Amersham Pharmacia、Uppsala、Sweden)上のRP−μRPC C2/C18分析カラムを用いて、80%ACNの0.1%TGA(v/v)溶液の直線勾配を用い、1時間かけて分画化した。ペプチドの溶出を215nmおよび280nmでモニタリングした。
ESI/MSを用いて、未変性タンパク質およびペプチド両方の正確な質量および純度(±0.01%)を決定した。RP−HPLC分画をPerkin−Elmer Sciex API300 LC/MC/MSシステム質量分析装置(Thornton、Canada)に直接注入した。イオンスプレー、オリフィス、およびリングの電圧はそれぞれ4600V、50V、および350Vに設定した。窒素を流速0.6リットル/分でカーテンガスとして、また圧力を100psiに設定して噴霧ガスとして使用した。質量はLC−10AD Shimadzu Liquid Chromatographyポンプを溶媒デリバリーシステム(40%ACN、0.1%TFA溶液)として用いて流速50μl/分で直接注入し決定した。BioMultiviewソフトウエアを用いて、未加工の質量スペクトルを分析し、デコンボリューションした。
未変性および消化したペプチドのN末端の配列は、オンライン785A PTH−アミノ酸分析装置と一緒にPerkin−Elmer Applied Biosystem 494パルス液相タンパク質シーケンサー(Procise)を用い、自動エドマン分解によって決定した。次に標準的なクロマトグラムを用いて各分離プロフィールをマッピングして、誘導体化されたPTH−アミノ酸を連続的に同定した。
i.p.(腹腔内投与)経路から注射される容積は200μlであり、タンパク質は水に溶解した。i.c.v.(脳室内)注射は、頭蓋の頂部から2mmまで通るように改良された針の付いた10μlのルアーチップHamiltonマイクロシリンジを使ってブレグマの側方1.5mm、後方1.0mmにある穿刺点を通して、2μlの容積で行った(13)。i.c.v注射用のタンパク質はACSF(人工脳脊髄液)に溶解した。針を回転させながら引き抜いた。これら2種類の投与方法を、移動活動およびホットプレート実験に用いた。
未変性のタンパク質をマウスに0.1mg/kg、1mg/kg、および10mg/kgの用量でi.p.注射した(n=2)。注射後、マウスの行動観察を15分おきに、6時間まで記録した。動物は24時間後に屠殺し、死体解剖を行った。
マウスの移動活動は、赤外線センサー、シグナル増幅回路、および制御回路から構成されるNS−AS01活動モニタリングシステム(Neuroscience,Inc.、Tokyo、Japan)で測定した。マウスの運動は、マウスの体温に付随する赤外線に基づいて、赤外線センサーによって検出された。各マウスを飼育ケージから取り出し、ケージ全体に8チャンネルの赤外線センサーを配置したケージ(12cm×12cm×30cm)に一匹ずつ入れた。ケージの床には、約40mlのおがくずを敷した。別々のケージに入れた8匹の動物の運動活動を同時に測定した。4cm以上の距離の運動は全て赤外線センサーによって検知され、それぞれが注射されたマウスの全身運動の数値となった。動物の活動性は、予備走行実験を実施して評価しておいた。その後の実験に用いられた動物は、最初に行ったこの20分間の予備走行実験中に最低で450カウント、最大で850カウントを示した。次に活動性のマウスにタンパク質を投与して、同じ運動活動モニタリングシステムにかけた。その直後から移動活動のカウントを10分間隔で80分間、コンピュータ接続分析システム(AB System−24A、Neuroscience,Inc.、Tokyo、Japan)を使って集めた。
各マウスを透明なプラスチック製の輪(直径12cm、高さ13cm)を使って拘束した状態でホットプレート(55℃)の上に置いた。ホットプレート装置は滑らかな金属表面15cm×15cmを持つ密封された木製の箱で、水槽(Model Y22 Grant、Cambridge、UK)を使って加熱した。マウスをホットプレートの上にそっと置いてから、次の反応の一つ、すなわち、WoolfeとMacDonaldが記載したような(14)跳躍、なめること、または後ろ脚の踏みつけのうちの一つを初めて示した時間までの潜伏期間を測定した。ホットプレートアッセイは、i.p.またはi.c.v.経路による薬物投与から15分後に行った。
移動活動の変化は、反復測定しながら二元配置分散分析によって分析した。オハニンによって誘導された疼痛過敏作用は、一元配置分散分析を用いて分析した。全ての分散分析は続いて、ボンフェローニ補正により事後分析を行った。統計的有意は、p<0.05の時とした。
369bpを含む完全長の合成遺伝子は2つの断片からアセンブルしたが、各断片は、特異的アニーリングを促進するために50%を超える高いGC含有量を持つ21bpの重複領域をそれぞれ持つ、96bp〜117bpの2つの重複オリゴヌクレオチドから組み立てられた。プライマー1(5’−GGAATTCGTCGACGGATCCATGGCTAGCCCGCCGGGTAACTGGCAGAAAGCGGACGTCACCTTCGATAGCAACACCGCGTTCGAAAGCCTGGTGGTGAGCCCGGAC−3’)およびプライマー2(5’−TCCCCCCGGGCTGCCTAGGACGCACGGGCTCGAGGAGAAGCGTTCCGGGCTATCCGGCACACCTTTCGGCACACCAACGTTTTCCACGGTTTTTTTGTCCGGGCTCACCACCAGGCT−3’)を用いて、第1断片を調製した;プライマー3(5’−TCCCCCCGGGTTTCCGTTCCGGAAAACACTTCTTCGAGGTGAAATACGGTACCCAGCGTGAATGGGCGGTGGGGCTAGCGGGTAAAAGCGTGAAGCGTAAGGGTTAC−3’)およびプライマー4(5’−GACTAGTAAGCTTGCGGCCGCCTACAGCCACCACAGACCTTTCTGCCAGATACGTTCTTCCGCACCAGCCTTAAGTAACCCTTACGCTTCACGCT−3’)を用いて第2断片を調製した。下線を付したヌクレオチドは、XmaI制限部位のフランキング配列である。両断片を生成するPCR混合物は、合計容積25μlの中に最終濃度0.3UのPlatinum Taqポリメラーゼ、0.2mM dNTP混合物、および0.2μMのプライマーを含んでいた。増幅条件は次の通りである:94℃/1分間を1サイクル;94℃/30秒、55℃/30秒、72℃/1分間を20サイクル;72℃/5分間の最終伸長。2つの断片はXmaIで消化し、一つに連結して完全長の合成遺伝子を得た。この連結産物をpGEMT−easyベクターにクローニングし、配列を決定した。
発現ベクター内にクローニングするために、369bpの合成遺伝子断片を制限エンドヌクレアーゼBamHIおよびNotIで二重消化した。ベクターM(pET32Aの改良版)を用いて、大腸菌BL21/DE株にて合成オハニンを発現させた。サブクローニングの結果、N末端がヘキサヒスチジンタグから成る融合タンパク質が発現した。発現させるために、ベクターM/オハニンを含む単一コロニーを100μg/mlのAmp(アンピシリン)を含むLB培地に接種し、37℃、200rpmで14時間インキュベーションした。一晩培養したものを100μg/mlのAmpを1:50の希釈率で含む新鮮なLB培地に接種した。再度、細菌培養物を37℃、200rpmで、培養物のA600が約0.6に達するまでインキュベーションした。次にイソプロピル−β−D−チオガラクトピラノシド(IPTG)を最終濃度0.1mMになるように加えて発現を誘導し、さらに細菌を集める前に16℃、200rpmで16時間インキュベーションした。細菌細胞は使用時まで−80℃で保管した。大腸菌での組換え体タンパク質の発現は、Laemmli(15)の方法に従った、15%のアクリルアミドゲルを用いたSDS−PAGE(ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動)によって分析した。
沈殿物溶解のプロセスは、変性条件の下、結合緩衝液を用いて4℃で少なくとも4時間続けた。結合緩衝液で溶解できなかった細胞破砕物は、遠心分離によって取り除いた。上清を結合緩衝液で事前に平衡化しておいた荷電Ni−NTA樹脂カラムにかけた。アフィニティークロマトグラフィーは製造元のガイドラインに従って実施した。洗浄緩衝液(10mMのIMD(イミダゾール)、6M GdnCl、10mM Tris−HCl、5mM β−ME pH8.0)を用いて徹底的に洗浄した後、結合したタンパク質を最小容積の溶出緩衝液(250mM IMD、6M GdnCl、10mM Tris−HCl、5mM β−ME pH8.0)を用いて溶出した。融合タンパク質の溶出および濃度は280nmでモニタリングした。
未変性および組換え体オハニンの二次構造は、経路長2mmのセルとJasco J810分光偏光計(Jasco Corporation、Tokyo、Japan)を用いて、260nmから190nmの波長域について22℃でUV CDスペクトルを記録して測定した。キュベットチャンバーは、実験前および実験中、持続的に窒素でパージした。未変性および組換え体タンパク質の測定は共にMilliQ水の中で行い、3回のスキャンの平均を取って、良好なシグナル対ノイズ比を得た。結果はdeg.cm2.dmol−1を単位とした平均残留楕円率(θ)として表した。αヘリックス、βシート、およびランダムコイル含有量は、http://www.embl−heidelberg.de/〜andrade/k2d/に記載されている方法を用いて見積もった。
トータルRNAは、RNeasy(登録商標)Miniキットの製造元のプロトコールに従ってキングコブラの毒液から単離した。簡単に説明すると、毒液腺組織(30mg)を液体窒素中で、モーターおよび−80℃に前もって冷却しておいた乳棒を用いて粉砕し、さらにHeidolph DIAX600ホモジェナイザー(Schwabach、Germany)を用いて600μlの緩衝液RLTと共に更にホモジェナイズした。完全に均一になった溶解物を20〜30秒後に得て、溶解物を最大速度で3分間遠心分離した。透明になった溶解物を新しい1.5mlのエッペンドルフチューブに移した。70%エタノール(550μl)を溶解物に加え、ピペッティングによってよく混合した。次に混合液を、15秒間、13,000rpmの遠心分離にかけるための2mlの収集用チューブの上にセットしたRNeasyミニスピンカラムに連続的にかけた。ピペットを使って緩衝液RW1(700μl)をRNeasyカラムに加え、カラムを15秒間、13,000rpmで遠心分離して洗浄した。RNeasyカラムを新しい採取用チューブに移した。緩衝液RPE(500μl)を用いてRNeasyカラムを15秒間、13,000rpmで洗浄した。ピペットを使って更に500μlの緩衝液RPEをRNeasyカラムに加え、2分間、最大速度で遠心分離してRNeasy膜を乾かした。RNeasyカラムを新しい1.5mlのエッペンドルフチューブに移した。無RNase水(40μl)をRNeasy膜の上に直接加えた。室温で1分間インキュベーションした後、1分間、14,000rpmで遠心分離を行い膜からRNAを溶出した。RNAの完全性は、変性アガロースゲル電気泳動によって調べた。
遺伝子特異的配列を生成するために、QIAGEN(登録商標)OneStep RT−PCRキットを用いてRT−PCRを行った。簡単に説明すると、RT−PCR混合液は合計容積50μlの中に2μlのQIAGEN OneStep RT−PC Enzyme Mix、ならびに最終濃度250ngのトータルRNA、0.4mMのdNTP混合物、および0.6μMの縮重プライマーを含んでいた。使用した縮重プライマーは:RT1センスプライマー5’−GGNAAYTGGCARAARGCNGAY−3’およびRT2アンチセンスプライマー5’−CCACCANARNCCYTTYTGCCA−3’であった。逆転写および増幅条件は次の通りである:逆転写、50℃/30分間;初期PCR活性化段階、95℃/15分間;直後に94℃/1分間の変性、50℃/1分間のアニーリング、72℃/2分間の伸長の3段階サーマルサイクリングを30サイクル;および最終伸長、72℃/10分間。次にPCR産物を1.5%TAEアガロースゲル電気泳動で分離した。pGEMT−easyベクターに連結する前に最も強いバンドを切り出し、精製した。pGEMT−easyベクター内の挿入体の配列を、T7およびSP6プライマーを用いて両鎖について、ABI PRISM(登録商標)3100Genetic Analyzer(Applied Biosystem、Foster City、CA、USA)を用いてジデオキシチェインターミネーション法により決定した。ABI PRISM(登録商標)BigDye(商標)Terminator Cycle Sequencing Ready Reactionキット(バージョン3.0)を用いてサイクルシークエンシング反応を行った。配列データの分析は、Sequencing Analysis 3.7(Sample Manager)ソフトウエア(Applied Biosystem、Foster City、CA、USA)を用いて行った。
5’−および3’−RACE−Ready cDNAライブラリーは、SMART(商標)RACEキットを、製造元プロトコール通りに使用して構築した。cDNA増幅の場合はPCR反応混合液を調製した。5’−RACE反応混合液は、合計容積25μlの中に、2.5μlの5’−RACE Ready cDNA、5μlのUniversal Primer Mix(UPM)、ならびに最終濃度1.5UのPlatinum Taqポリメラーゼ、1.5mMのMgCl2、0.2mMのdNTP混合物、および0.2μMのアンチセンスプライマー(GSP2)(5’−CTTCCCAGCTAACCCAACAGCCCATTCCC−3’)を含んだ。3段階のサーマルサイクリングプロフィールは次の通りである:94℃/1分間のホットスタートを1サイクル;94℃/30秒間の変性、67℃/30秒間のアニーリング、72℃/2分間の伸長を30サイクル、それに続いて72℃/10分間の最終伸長である。完全長のcDNA配列を生ずる3’−RACE反応混合液は、合計容積25μl中に、2.5μlの5’−RACE Ready cDNA、5μlのUniversal PrimerMix(UPM)、および最終濃度1.5UのPlatinum Taqポリメラーゼ、1.5mMのMgCl2、0.2mMのdNTP混合物、および0.2μMのセンスプライマー(GSP1)(5’−GATCATTTGATCCAGAGAAGACACAGTCTC−3’)を含んだ。3段階のサーマルサイクリングプロフィールは次の通りである:94℃/1分間のホットスタートを1サイクル;94℃/30秒間の変性、68℃/30秒間のアニーリング、72℃/2分間の伸長を30サイクル、それに続いて72℃/10分間の最終伸長である。PCR産物は1.5%TAEアガロースゲル電気泳動で分離した。pGEMT−easyベクターに連結する前に最も強いバンドを切り出し、精製した。完全長のRACEクローンの配列を決定し、DNAsis For Windows(バージョン2.5)から入手できるコンティグ結合方法を用いてアセンブルした。
シグナルペプチド領域を除く全オープンリーディングフレームをカバーしたcDNAに、PCRを用いて両端に制限部位を加えた。cDNA断片の導入および増幅に使用したプライマーは次の通りである:センスプライマー(19K1)5’−GTCGACGGATCCATGTCACCTCCTGGGAATTGGCAG−3’およびアンチセンスプライマー(19K2)5’−AAGCTTGCGGCCGCTTAAAGATTTGCGAGTGAAACACG−3’。PCR反応混合液は、合計容積25μl中に、最終濃度1.5UのPlatinum Taqポリメラーゼ、1.5mMのMgCl2、0.2mMのdNTP混合物、および0.2μMを含んだ。3段階のサーマルサイクリングプロフィールは次の通りである:94℃/1分間のホットスタートを1サイクル;94℃/1分間の変性、70℃/30秒間のアニーリング、72℃/1分間の伸長を30サイクル、それに続いて72℃/5分間の最終伸長である。ゲル精製したPCR産物を、発現ベクター内にクローニングするために制限エンドヌクレアーゼBamHIおよびNotIで消化した。発現ベクターであるベクターMを用いて、プロ−オハニンを大腸菌BL21/DE3株で発現した。サブクローニングの結果、N末端領域がヘキサヒスチジンタグから成る融合タンパク質が発現した。
上清を荷電Ni−NTA樹脂カラムにかけた。アフィニティークロマトグラフィーは製造元のガイドラインに従って実施した。洗浄緩衝液(10mM IMD、10mM Tris−HCl、5mM β−ME pH8.0)を用いて徹底的に洗浄した後、結合したタンパク質を最小容積の溶出緩衝液(250mM IMD、10mM Tris−HCl、5mM β−ME pH8.0)を用いて溶出した。融合タンパク質の溶出および濃度は280nmでモニタリングした。
ゲノムDNAは、DNeasy(登録商標)Tissueキットの製造元のプロトコールに従ってキングコブラの肝臓組織から単離した。簡単に説明すると、肝臓組織(25mg)を液体窒素中で、モーターおよび−80℃に前もって冷却しておいた乳棒を用いて粉砕した。組織粉末を新しい1.5mlのエッペンドルフチューブに移した。緩衝液ATLおよびプロテイナーゼKをそれぞれ180μlおよび20μl加えて、細胞を溶解した。溶解物を振盪中の水槽の中で、55℃でインキュベーションした。3時間後、400μgのRNaseAを加え、ゆっくり混合し、16℃で2分間インキュベーションしてRNA汚染を防止した。70℃で10分間インキュベーションする前に、緩衝液AL(200μl)を加えて、ゆっくり混合した。100%エタノール(200μl)を溶解物に加えてゲノムDNAを沈殿させた。1分間、8,000rpmで遠心分離するために、溶解物および白色の沈殿の混合物を2ml採取チューブ上に設置したDNeasyミニスピンカラムにかけた。DNeasyスピンカラムを新しい採取用チューブに移した。緩衝液AW1(500μl)を用いてDNeasyカラム中でゲノムDNAを洗浄し、1分間、8,000rpmで遠心分離した。さらに500μlの緩衝液AW2を用いてゲノムDNAを洗浄してからスピンカラムをさらに14,000rpm、3分間の遠心分離にかけ、次の溶出時に残留エタノールが確実に持ち込まれれないようにした。DNeasyカラムを新しい1.5mlのエッペンドルフチューブに移した。緩衝液AE(200μl)をDNeasy膜に直接かけた。室温で1分間インキュベーションした後、1分間、8,000rpmで遠心分離を行い膜からゲノムDNAを溶出した。ゲノムDNAの完全性は、0.8%アガロースゲル電気泳動によって調べた。
gDNA増幅のためにPCR反応混合液を調製した。PCR反応混合液は、合計容積25μl中に、鋳型となるgDNAを1μl、ならびに最終濃度として1.5UのPlatinum Taqポリメラーゼ、1.5mMのMgCl2、0.2mMのdNTP混合物、および0.2μMのプライマーを含んだ。使用したプライマーは:図15−Aに示すセンス(gDNA1)5’−TCACCTCCTGGGAATTGG−3’およびアンチセンス(gDNA2)5’−AAGATTTGCGAGTGAAAC−3’である。3段階のサーマルサイクリングは、94℃/1分間のホットスタート、続く94℃/1分間、60℃/30秒間、72℃/3分間を30サイクル;および72℃/10分間の最終伸長を含んだ。PCR産物は1.5%アガロースゲルで分析し、強いバンドを切り出して精製した。挿入体を担持する16個のクローンの配列を、T7およびSP6プライマーを使って両鎖について決定した。16個全てのクローンについて、追加の内部プライマーを用いて配列を決定して配列を完成させ、DNAsis For Windows(バージョン2.5)から入手できるコンティグ結合方法を用いてアセンブルした。
これらの実験で使用したオハニンおよびプロ−オハニンをコードする遺伝子は、前述したようにベクターMにクローニングされ、大腸菌株BL21(DE3)に形質転換された。精製は若干変更を加えたPungら(34)の方法に従って行った。精製されたN末端オリゴヒスチジンタグ付きオハニンおよびプロ−オハニン(His−オハニンおよびHis−プロ−オハニンとそれぞれ称する)を免疫蛍光検出に直接使用した。
体重20〜25gのSwissアルビノマウスを用いた。脳室内および腹腔内注射は共に、Pungら(34)が記載した方法を用いて行った。次に動物を全採血して、外科的に脳を取り出した。
マウスの脳を、ロボトミー外科技術を用いて取り出し、30%ショ糖、50mM Tris−アセタート、5mM EDTA(pH7.4)を含み、完全プロテアーゼインヒビターカクテル錠剤(Roche)を補充した溶液の中に、4℃で12時間入れた。
タンパク質は0.05μMの濃度でPBSに溶解した。次にタンパク質溶液(1ml)を、注射していない脳の切片を載せた各スライドに掛けて4℃で一晩インキュベーションした。次にスライドを氷冷したPBSと共に振盪インキュベータの中に入れてすすぎ洗いし、振盪インキュベータで室温にて30分間振盪した。競合アッセイについては、最初のすすぎ洗い段階が終わった後に、1mlの第2タンパク質溶液を各スライドに加えて4℃にて一晩、再度インキュベーションし、すすぎ洗い処置を繰り返した。
Anaspec Inc製のウサギ抗His抗体(α−His、ヤギ)を、PBS中に1:1000の割合で使用した。抗体溶液(1ml)を調製済みの各スライドに掛けて、一晩、4℃でインキュベーションした。次にスライドを、振盪インキュベータの中の氷冷PBS中に入れて、室温で15分間振盪インキュベータで振盪し、3回洗浄した。Molecular Probes製Alexa−fluo488で標識した抗ウサギ二次抗体を1:500の割合で使用した。二次抗体溶液をスライドの上に掛けて、暗所、4℃で一晩インキュベーションした。スライドを一次抗体洗浄時と同様にして暗所で洗浄し、半乾燥状態で放置した。次にProlong Gold Antifadeマウンティング媒体をDAPI(Molecular Probes)と共にスライドの上に加え、カバーガラスをその上に載せた。脳の切片は、Axiocam HRcデジタルカメラアタッチメントの付いたZeiss Axiovert 200M顕微鏡で観察した。連続写真は、Axiovisionバージョン4.3.0.101を用いて、405および480nmの波長で撮影した。写真はPhotoshopバージョン5.5を使って編集し、重ね合わせた。
キングコブラ毒液からの新規タンパク質の同定
Ophiophagus hannah(キングコブラ)の粗毒液のプロフィールをLC/MC(図1)を使って調べ、毒液中に新規の興味深いタンパク質成分を同定した。LC/MSで検出したペプチドおよびタンパク質は、保持時間別に系統だててまとめた(図1−A)。50分後に溶出したタンパク質は比較的ノイズの多いm/zスペクトルを示したことから、これらの分子についてはその質量を決定しなかった。これは、タンパク質のサイズが大きいこと、ならびに糖化およびその他の翻訳後修飾によるものと思われた。このようにしてLC/MSを用いたキングコブラ毒液の質量プロファイリングからは、この技術が有効でなく、限界があることが明らかになった。我々はLC/MSプロフィールを用いて、まずよく知られている毒素ファミリーの質量と異なる質量を持つタンパク質を探した。我々は分子量11951.35±3.92Daを持つタンパク質を同定したが、これは確立されたどのファミリーとも異なっており、それ故に我々はこの新規タンパク質を更に研究することとした。
前記新規タンパク質を、2段階の精製手順を介してキングコブラ毒液から精製した。第1段階は、ゲル濾過クロマトグラフィーを用いた粗毒液の分離を含む。新規タンパク質の分子量はおおよそ12kDaであることから、ゲル濾過クロマトグラフィーには、Superdex30(Hiload 16/60)カラムを選択した。粗毒液のゲル濾過では5つの主要ピークが得られた(図2−A)。我々はゲル濾過クロマトグラフィーの先頭の3つのピークをRP−HPLCにかけた。RP−HPLCから出てきた個々の分画をESI/MSを用いて評価した(データ未提示)。ゲル濾過のピーク1bから、38〜40%の緩衝液B(80%ACNの0.1%TFA溶液)(図2−B)の勾配に溶出されたタンパク質分画が均一であり、分子量が11951.47±0.67Daであることが見出された(図2−C)。新規タンパク質の全体収量は、粗毒液1gからおおよそ1mgであった。
未変性タンパク質のN末端配列をエドマン分解によって決定し、その結果、先頭40個の残基が同定された。前記N末端配列は、既知のヘビ毒素ファミリーのタンパク質のいずれとも配列相同性を示さなかった。全配列を決定するために、ピリジルエチル化タンパク質をLys−Cエンドペプチダーゼ、トリプシン、およびギ酸で消化した。各消化物からのペプチドは逆相HPLCによって分離した(図3)。精製ペプチドの分子量およびアミノ末端配列を得て、全長のアミノ酸配列を完成させた(図4)。ペプチドおよびタンパク質全体の配列は、消化ペプチドの計算質量と観察質量とを比較して確認した(図3−D)。観察分子量は計算分子量とよく一致した。新規タンハク質は1個の遊離システインと共に107個のアミノ酸残基を含み、翻訳後修飾は含んでいなかった。我々はこのタンパク質を、それがキングコブラOphiophagus hannahの毒液から精製されたことからオハニンと命名した。
BLASTPアルゴリズム(http://www.ncbi.nlm.nih.gov/BLAST/)(16)を用いてオハニンの完全長アミノ酸配列と他のタンパク質の配列とを比較し、モノクルコブラ(Naja kaouthia)から単離されたタイ(Thai)コブリン(SP:P82885)との間に93%の配列同一性があることを示した。その配列はタンパク質データベースに寄託したが、タイコブリンに関する発表文献はない。かくしてオハニンとタイコブリンは、ヘビ毒素の新規ファミリーの最初のメンバーを形作っている。タイコブリンとの間を除いてオハニンはGenBankデータベース内のその他タンパク質との間に有意な配列類似性を示さなかった(E値>10−5)。
オハニンを、マウスでのin vivo毒性試験に用いた。全てのマウスについて、実験開始前には活動的であることが観察された。タンパク質をi.p.注射すると、1mg/kgおよび10mg/kgの用量が注射されたマウスについては静かになり、動きが緩慢になることが観察された。しかしながらマウスは注射2時間後には回復し、四肢および呼吸器系に麻痺の顕著な兆候は認められなかった。10mg/kgの用量でさえ死亡例は無かった。注射後24時間目の肉眼による病理分析からは、コントロール動物から得た病理分析と比較して、脳、心臓、肺、腎臓、脾臓、および肝臓に出血または壊死を示す証拠は認められなかった(データ未提示)。
in vivo毒性試験での我々の観察を定量的に検証するために、注射したマウスの移動活動に対するオハニンの作用を調べた。図6−Aに示すように、オハニンは0.1mg/kg、1mg/kg、および10mg/kgの用量では、i.p.注射後に用量依存的な移動低下を誘導した(F3,30=5.787、p<0.01)。移動活動の低下は、図6−Aに示すように10mg/kgの用量と0.1mg/kgの用量の間(p=0.030);10mg/kgの用量とコントロール(p=0.004)の間で統計学的に有意であった。10mg/kgの用量では、全運動カウント数がコントロール(1942±147)に比べて742±190まで低下した。図6−Bに示すように、この用量依存的な阻害は、注射後10分の時点でさえ異なっていた。2元配置反復測定分散分析が示すように、1時間の実験期間中、同一用量内には統計学的に有意な時間効果は存在しておらず、阻害作用からまだ回復していないことが示唆された。実験期間(1時間)を超えてこの作用を測定することはしなかった。長時間の身体的活動および食事を与えないことはマウスを消耗させる。それ故に、実験期間を超えた場合の動きの緩慢化は、タンパク質の作用だけによるものではない可能性がある。
オハニン投与したマウスに熱痛刺激が起こす痛覚について、ホットプレートアッセイを用いて評価した。ホットプレートアッセイで使用した投与量は、移動活動に使用したものと同じであった。疼痛刺激に対するオハニンの作用は、i.p.およびi.c.v.注射15分後に評価した。図7−Aおよび図7−Bに示すように、i.p.およびi.c.v.注射は共に同様のU字型の用量反応曲線を示した。オハニンをi.p.注射した場合については、全投与量について有意な作用は認められなかった。(F3,28=0.867、p>0.05)(図7−A)。しかしながら、0.3μg/kgおよび1μg/kgの用量でi.c.v.注射した時には、オハニンは用量依存的な疼痛過敏作用を示した(F3,50=6.390、p<0.01)。しかし、より高い用量の10μg/kgでは、有意な疼痛過敏作用は存在しなかった。図7−Bに示すように、潜伏期間は、1μg/kgとコントロールの間(p=0.002);1μg/kgと10μg/kgの間(p=0.015)で統計学的に有意であった。
粗毒液中に元々存在しているオハニンの量は少ないため、オハニンをコードする合成遺伝子を、そのタンパク質配列に基づき、帰納的(recursive)PCR法(22)によって構築した。オハニンは糖化またはジスルフィド架橋のような翻訳後修飾を含んでいないことから大腸菌発現システムを選択した。第2に、大腸菌発現系でのオハニンの過剰発現は、組換え体タンパク質を適切量提供して、その構造−機能相関についての我々の今後の研究を容易にする上でも有利である。
ベクターM/オハニン構築体を含む大腸菌を用いて組換え体オハニンを発現させた。0.1mM IPTGを用いて、16℃で一晩誘導した後に細菌培養物から調製した全タンパク質のSDS−PAGE分析から、見かけ上の分子量が約14kDaのタンパク質が大量にあることが証明された。未誘導および誘導した培養物から抽出した全タンパク質と、融合タンパク質を可溶性および不溶性タンパク質に分画化したものを、一つにまとめて比較したものを図9−Aに示す。融合タンパク質に相当する14kDa(1として矢印で示す)の明瞭なバンドが不溶性分画に見られた。発現ベクター、細菌株、培養時の細胞密度、インキュベーション時間、緩衝液、および使用したIPTGのような様々なパラメータを変えても、組換え体タンパク質の発現について有意な差は観察されなかった(データ未提示)。
融合タンパク質内のHisタグによって、変性条件下において、単一のアフィニティーカラムを用いて迅速に精製することができる。精製段階を図9−Bに示す。レーン3は1つの主要種(〜14kDa)を示し、レーン5はリフォールディング後に見かけ上の分子量14kDaを持つ融合タンパク質を示す。未変性タンパク質と比較した時に、組換え体タンパク質に追加された2kDaは、N末端のHisタグ、トロンビン、およびCNBr切断部位に対応している(図8−A)。細菌培養物1リットルから、Ni−NTAアフィニティークロマトグラフィーを用いて、25mgのHisタグ付き融合タンパク質が精製された。
未変性および組換え体タンパク質の二次構造をCD分光分析によって評価した(図11−A)。未変性タンパク質のCDスペクトルは、200nmおよび215nm近くで極端に負の楕円率を示し、βシートおよび複数のβシート構造を持つランダムコイル構造の存在が示唆された。組換え体タンパク質のCDスペクトルは未変性タンパク質のそれに近く、200nmおよび215nmに負の楕円率を示した。CDスペクトルから計算した二次構造の骨格を図11−Bに示す。
キングコブラ毒腺から抽出されたトータルRNAは微量であったが(抽出当たり〜4μg)、RNAの質は比較的良好であった。我々はRT−PCRとRACE技術を組み合わせて用い、完全長のオハニンのcDNAを得た。遺伝子特異的な配列を単離するために、RT−PCRをまずトータルRNAを鋳型にして行った。縮重プライマーRT1およびRT2は既知のアミノ酸配列に基づいてデザインした。増幅された長さ〜200bpの断片をゲル精製し、pGEMT−easyベクターに連結して配列を決定した。配列決定分析からは、8つのクローン全てが、オハニンの部分配列と完全に相同であるアミノ酸配列をコードしていることが明らかにされた。
BLASTNアルゴリズム(http://www.ncbi.nlm.nih.gov/BLAST/)(15)を用いた完全長cDNA配列の比較からは、今までのところGenBankデータベースに寄託されている他のヌクレオチド配列と配列相同性は確認されていない。しかしながら、導出されたアミノ酸配列は、完全なPRY−SPRYおよびB30.2類似ドメインと相同性を示し、プロ−タンパク質にはHenryら(20、21)が提案している3rdモチーフが存在していた。導出されたタンパク質配列と、B30.2類似ドメインとのアラインメントを図13に示す。タンパク質配列はPRY−SPRYドメインと、38%の全体同一性および49%の類似性を共有した。B30.2類似ドメインを持つタンパク質の概略図を図14に示す。
プロ−オハニンを、大腸菌を用いて発現させるために発現ベクターにクローニングした。プライマー19K1および19K2を用いて増幅し、同時にMetおよび停止コドンだけでなくベクターMにクローニングするための制限部位も加えた。増幅された、制限部位をフランキングした配列を消化して、BamHI部位およびNotI部位で発現ベクターと連結した。連結のための特異的制限部位を使用することで、プロ−オハニンが適切な向きで確実に挿入されるようにし、一方Metを第2の代替切断部位として挿入した。増幅断片を示すアガロースゲルを図15−Aに示す。プロ−オハニン発現ベクターの構造の概略図を図15−Bに示す。
オハニンとプロ−オハニン両方の二次構造含有率を、12.5μM濃度について、CD分光法を用いて測定した(図18−A)。オハニンのCDスペクトルは、215nm付近で極端な負の楕円率を示し、βシート構造の存在を示した(7%αヘリックス、48%βシート、および45%ランダムコイル)(図18−B)。オハニンと異なり、プロ−オハニンのCDスペクトルは、22%αヘリックス、29%βシート、および49%ランダムコイルから成る混合型の二次構造プロフィールを示した。従って、図18に見られるように、プロ−オハニンのC末端プロペプチドセグメントの存在が、そのαヘリックス含有量を上げていることは明らかである。オハニンおよびプロ−オハニンは共に〜50%のランダムコイル構造を持っている。
精製プロ−オハニンについて、マウスでのin vivo毒性を調べた。それはi.p.投与時には、10mg/kgの用量まで非致死性であった。成熟オハニンを注射した時に見られたような、緩慢化の明瞭な兆候は見られなかった。これに加えて24時間後にマウスを屠殺した時の目視検査では、脳、心臓、肺、腎臓、脾臓、および肝臓に、検知可能な出血または壊死は見られなかった(データ未提示)。
前節で記載したように、オハニンはi.p.注射後に用量依存的、且つ統計有意に移動低下を誘導する(F3,30=5.787、p<0.01)(34)。マウスの移動活動に及ぼすプロ−オハニンの作用を、0.1mg/kg、1mg/kg、および10mg/kgの用量をi.p.注射して調べた。最高用量(10mg/kg用量)時のプロ−オハニンの全運動カウントは約2048±225であり、コントロールのカウント(1942±147)と同等であった。従ってプロ−オハニンは、オハニン(図19−A)に比べると、i.p.注射後、実験マウスの運動性に有意に阻害をもたらさないことは明らかである(F3,28=0.251、p>0.05)(図19−B)。
オハニンと同様にして、ホットプレート実験に及ぼすプロ−オハニンの作用を注射15分後に評価した。プロ−オハニンは、腹腔内投与時には、オハニン注射したマウスのような用量依存性もU字型の用量反応曲線も示さなかった(F3,28=0.922、p>0.05)(図20−Bおよび図20−A)。脳室内注射時には、プロ−オハニンは使用した全ての用量(0.3μg/kg、1μg/kg、および10μg/kg)についてコントロールに比べて比較的短い潜伏期間を示した(F3,39=3.275、p<0.05)(図20−D)。疼痛過敏作用は、0.3μg/kg用量とコントロールとの間でのみ有意であった(p=0.026)。
上記した様に、オハニンには2種類のmRNAサブタイプが存在している。これら2種類のmRNAが2つの独立した遺伝子の産物であるか、または異なるスプライシングに由来するものであるか明らかにすることは興味深い。第1段階として我々はゲノムサザンハイブリダイゼーション実験を行った。キングコブラのゲノムDNAをEcoRI、HindIII、BamHI、およびNdeIで別々に消化した。これらゲノムDNA消化物を、そのcDNAのヌクレオチド319〜616からデザインした297bpのDIG標識プローブでハイブリダイゼーションした。我々は、4種類全ての消化物について単一のバンドを観察し(図21)、オハニンがキングコブラゲノムの中では単一の遺伝子にコードされていることが示唆された。
オハニン遺伝子のゲノム構造を決定するために、ゲノムDNAのPCRおよび「ゲノムウォーキング」法を用いた。オハニンのcDNAの配列を用いて、エクソン−イントロンの境界をマッピングした。最初の増幅では、gDNAsigpepおよびgDNAstopを用いて、そのコーディング領域を増幅した。得られた断片は〜1.9kbであった(データ未提示)。ゲノムDNAの3’−UTR領域をPCR増幅しようとした我々の試みからは、別の〜750bpのバンドが得られた。
マウスでの移動活動に関するこれまでの研究から、オハニンとプロ−オハニンは共にi.c.v.注射によって強力な移動低下作用を示すことが明らかになった。そこでN末端にHisタグを融合させたプロ−オハニン(His−プロ−オハニンと呼ぶ)について、まずin vitroでの脳切片結合能力を試験した(図24)。我々の結果は、図25に示すようにHis−プロ−オハニンが脳の海馬および小脳領域に特異的に結合することを示した。脳切片の他の領域は染色されずに残った。同様にHis−プロ−オハニンと共に予備インキュベーションしなかったコントロール実験も蛍光染色を示さなかった(図25)。
次に我々はオハニンおよび/またはプロ−オハニンが血液脳関門を通過して観察された薬理作用を発揮するか確認することを試みた(図28)。かくしてHis−オハニンおよびHis−プロ−オハニンをi.p.およびi.c.v.経路から注射し、3種類の濃度について、脳内のタンパク質の存在を免疫蛍光を用いて検出した。図29は、His−オハニンがin vivoにおいて小脳および海馬に用量依存的な様式で結合することを示している。しかしながら、His−プロ−オハニンを投与すると、同じ領域の染色が顕著に低下することが見られた(図30)。このことは、プロ−オハニンが血液脳関門を通過できることを示してはいるが、適切な免疫蛍光染色を可能にするために投与したタンパク質の量がこれまでに報告した最高濃度(10mg/kg)よりも1000倍高いことを指摘しておかねばならない。
我々は、LC−MSを用いた最初のキングコブラ毒液スクリーニングで、異常な分子量を持つ新規タンパク質の存在を確認した(図1)。ここでは、このタンパク質、オハニンの精製および特徴付けについて記載している。オハニンの完全アミノ酸配列は、エドマン分解によって決定した。それは単一のシスティン残基を含む107個のアミノ酸残基を有している(図3)。それは確立されたどのヘビ毒タンパク質ファミリーとも類似性を有していない。かくしてオハニンおよびタイコブリン(タイコブラについて報告されたアイソフォーム)は、新規のヘビ毒タンパク質ファミリーの最初のメンバーである。このファミリーのメンバーの固有の特徴は、システィン残基の含有量が低いことであると思われる(<1%)。これに対し、他の全てのヘビ毒タンパク質ファミリーは、複数のジスルフィド結合および高いシスティン残基含有量を持つ(一般的に8〜10%より高い)。
CDD研究は、オハニンがPRY−SPRYドメインと類似性を共有していることを明らかにした(図5)。SPRYドメインの3つのコピーは、3種類の哺乳類リアノジン受容体(RyR)サブタイプに最初に確認された。このドメインは、Dictyostelium discoideumの二重特異性キナーゼ、splAにも3コピー存在している。splAおよびRyRに於いて反復していることから、これらの配列はSPRYドメインと呼ばれている(23)。SPRYドメインは、B30.2類似ドメインファミリーのサブドメインであることが確認されている(19)。SPRYドメインは、B30.2類似ドメインと比較するとN末端領域に欠失がある。〜50個の残基を含むPRYドメインが、SPRYドメイン(〜110〜120残基)のN末端領域に常に見いだせることを指摘することは興味深い。それ故に、両PRY−SPRYドメインをB30.2類似ドメインのサブドメインと見なすことができるだろう。
オハニンは、i.p.注射により、実験マウスに用量依存的様式で移動低下を誘導した(図4)。ヘビ毒素中の神経毒は骨格筋の麻痺誘導において特に重要であることを指摘しておく(25)。オハニンが末梢神経筋接合の遮断を誘導するか試験するために、われわれは単離したニワトリの二腹頸神経筋標本(CBCNM)に対する作用について研究した。オハニンはCBCNM刺激の直接単収縮反応だけでなく、ACh、CCh、およびKCIのような外から作用させたアゴニストに対する反応にも作用を示さなかった(Y.F.Pung、J.C.Wickramaratna、N.G.Lumsden、W.C.Hodgson、およびR.M.Kini、未発表の観察)。これらの結果は、オハニンがシナプス前およびシナプス後毒性(筋肉毒性を含む)を持たないことを示している。このことから、我々はオハニンをマウス脳室内に直接注射して、中枢神経系での薬理作用を検証した。オハニンは、i.p.注射した場合に比べて、i.c.v.注射した時に、〜6,500倍より強力な移動低下活性を生じた。それ故オハニンは、おそらくは中枢神経系に対する直接作用を介して移動低下を誘導する。オハニンが血液脳関門を通過できるか明らかにするために、更に研究が行われている。
我々は、ヘビ由来の小型の新規毒液タンパク質の構造機能相関研究に関心がある(7)。毒液タンパク質を薬物デザインおよび抗毒素のモデルとして用いるこれらの研究では、タンパク質を得る確実で安価な方法が必要とされる。タンパク質を獲得するための一つの潜在的方法は、それらを固相ペプチド合成およびコンビナトリアルケミストリーを用いて作成することである。第2の、より安価な方法は、分子生物学的技術を用いて細菌宿主細胞にタンパク質を過剰発現させることである。タンパク質製造のための合成遺伝子は、強力な方法である。この方法では、cDNA配列に対し、単一アミノ酸またはタンパク質ドメイン全体を容易に変更できる(30)。
ヘビ毒素は薬理学的に活性なペプチドおよびタンパク質の複合混合物である。それらは攻撃および防御の両機能に重要な役割を果たしている。神経毒のような、これらのタンパク質のいくつかは、被食者を麻痺させるのに関係するが、一方加水分解酵素を含むその他のものは被食者を消化するのに関係している。我々は、オハニンは被食者の運動性を緩慢にすることで貢献し、その捕獲を助ける。疼痛過敏作用もまた、捕食動物に疼痛を誘導することで防御機能に役立つだろう。毒液中に存在するその他成分と関係してオハニンが果たしている役割を明らかにするには、更なる研究が必要である。
オハニンは毒液腺の中で、プレプロ−タンパク質としてC末端プロペプチドセグメントと共に合成される(図12)。これは、成熟タンパク質のC末端にプロペプチドセグメントを含むヘビ毒タンパク質として報告された、これまでのところ最初のものである。
我々はまた、オハニン遺伝子がPRY−SPRYおよびB30.2ドメインの直前に局在する単一のイントロンを有することも示した(図22および23)。現在イントロンの起源については2つ見解がある。一つの理論、初期エクソンは、エクソンが古代ミニ遺伝子の子孫であり、イントロンはそれらの間の間隔であるというものである(32)。別の理論は、後期イントロンは、イントロンの挿入によって中断されていない遺伝子から分断された遺伝子が生じたというものである(33)。簡単に説明すると、最初の理論は、エクソンがタンパク質の個別の機能または構造単位を表すことを示唆するが、一方第2の理論はイントロンの挿入が若干無秩序であることを示唆している。プロ−オハニン遺伝子の構造上の類似性は、それがB30.2ドメインタンパク質と同一の先祖遺伝子から進化したらしいことを示している。このことは更に、オハニン遺伝子の進化が初期エクソン理論により近いことを示している。
オハニンおよびプロ−オハニンが海馬および小脳に局在することは、移動低下作用を示すこのタンパク質の正確な性質を示唆している。海馬は、動物の移動活動に間接的に影響する代謝に関係することが分かっている。実際、海馬の神経細胞は、フェニルシクリプリン(phenylcycliprin)およびコカインによる移動低下に関係している。オハニンはアンタゴニストと同じ受容体に作用して、その移動低下作用を示す可能性がある。一方、小脳は動物の全体のバランスに関係している。アンフェタミンを用いたこれまでの実験は、小脳の神経細胞に作用することを示し、且つアンフェタミン投与の結果として移動低下を示すことを示した。また、同じ神経細胞がオハニンの相互作用にも関係している可能性もある。バランスに影響する神経細胞がオハニンの影響を受けるかは未だ不明であるが、バランスの欠如は全体的な移動能力の欠如をもたらすだろう。
我々は、キングコブラ毒液から新規タンパク質のオハニンを同定し、精製し、且つ機能的に特徴付けした。オハニンは移動低下および疼痛過敏をマウスに誘導する。i.c.v.経路により投与されたオハニンが全身投与した場合に比べ効果的であることは、末梢神経系の役割を排除することはできないが、その作用が中枢神経系を介していることを強く示唆している。我々は将来の構造および機能相関研究のために、合成遺伝子発現系も確立した。オハニンの分子レベルの詳細な作用メカニズムは現在研究中である。
