a.DN-IGF-IR (KR)のHIV媒介性の発現は、培養された脈絡叢上皮細胞上でIGF-Iシグナルを遮断する。IGF-IR (pTyrIGF-IR、二つのウイルス希釈液で試験)およびその下流のキナーゼ Akt (pAkt)のIGF-Iによって誘導されたリン酸化の欠損によって決定されたとき、感染した細胞はIGF-Iに応答しない。IGF-IRおよびAktの合計量は変化しなかった。3つの実験の代表的ブロットを示す。
a.Pan-特異的抗Aβ抗体でのウェスタンブロット分析は、KR注入の3および6月後に皮質において増加したAβレベルを示し(左)、かつCSFにおいて減少したAβレベルを示した(右)。代表的ブロットおよび濃度ヒストグラムが示される。対照n=13、3ヵ月、n=6;6ヶ月n=7;*p<0.05および**p<0.01 vs 対照。

a.3ヶ月前にHIV-KR3を受けたラットに対するHIV-野生型(wt)IGF-IRの注入は、IGF-Iに対する脈絡叢の応答の正常化をもたらす。IGF-Iのic注入後、KR-wtIGF-IR処理ラットは、脈絡叢において対照のpAktレベルを示す(この反応を図1eにおけるKRラットによって示されたものと比較、n=7)。

導入遺伝子内に変異を含むプラスミド領域(lys 1003またはarg 1003)は、pHIV-IGF1-DNの塩基7700および8100の間で構成された領域である。寄託された株については、この領域は、微生物の生存度を確認するために配列決定し得る。
本発明の一実施態様によれば、本発明は、ドミナントネガティブIGF-I受容体または機能的IGF-I受容体のいずれかを発現することができる遺伝子転移ベクターに関する。本発明によって考えられる遺伝子転移ベクターは、HIVまたはアデノ関連ウイルス(AAV)ベクターから好ましくは誘導される。

他の実施態様によれば、本発明は、βアミロイドおよび/またはアミロイドプラークの異常な脳蓄積が関与し、脳からのβアミロイドのクリアランスが減少する疾患のためのモデルとして使用される非ヒト動物に関する。本発明に関わる好ましい疾患は、アルツハイマー疾患である。本明細書中で使用されたとき、用語「非ヒト動物」は、本発明に適した全ての非ヒト動物をいう。これらの非ヒト動物の中では、マウスおよびラットのようなげっ歯類、およびカニクイザル(Macaca fascicularis)のような霊長類が好ましい。列挙された動物は、モデルとしての使用に適した動物、すなわち実験動物を構成するのに適した動物の例である。本発明は、研究または試験において使用または使用を目的とした、このような実験動物を特に対象とする。
他の実施態様によれば、本発明は、βアミロイドおよび/またはアミロイドプラークの異常な脳蓄積が関与する疾患の治療のための分子をスクリーニングするための方法であって、前記方法が、アルツハイマー疾患様障害が回復するのに十分な時間および量において前記分子を上記に定義した動物に投与することを含み、アルツハイマー疾患様障害の回復が、βアミロイドおよび/またはアミロイドプラークの異常な脳蓄積が関与する疾患の治療のための分子の指標となる方法を提供する。
他の態様によれば、本発明は、IGF-1受容体と相互作用する活性分子をスクリーニングするための方法であって、前記方法が、アルツハイマー疾患様障害を修飾するのに十分な時間および量において前記分子を動物に投与することを含み、アルツハイマー疾患様障害の回復がIGF-I受容体活性を増加させる分子の指標となり、かつアルツハイマー疾患様障害の外観がIGF-I受容体活性を減少させる分子の指標となる方法を提供する。有利には、アルツハイマー疾患様障害の回復が、上記に定義した動物において観察される。
加齢、アルツハイマー疾患(AD)の主要な危険因子は、インシュリン様成長因子I(IGF-I)、脳βアミロイド(Aβ)レベルの修飾因子の減少した入力に関する。本発明者らは、障害されたIGF-I受容体(IGF-IR)機能が原因の減少したAβクリアランスが、アミロイド症だけではなく、ADの他の病理学的特徴にも影響を与えることを証明した。脈絡叢(IGF-IによるAβクリアランスに関係する脳構造)におけるIGF-IRの特異的な遮断は、脳アミロイド症、認知障害および高リン酸化tau蓄積を、他のAD関連障害、例えば神経膠症およびシナプスタンパク質欠如とともに導く。異常に低い血清IGF-Iレベルを示すAD様障害老齢変異マウスでは、脈絡叢におけるIGF-IR遮断がAD様病理を悪化させる。これらの知見は、遅延発症型アルツハイマー疾患の原因を解明した。つまり、脈絡叢に入力される年齢関与性のIGF-Iの異常な減少が遺伝的易発性患者におけるADの発症の原因になることを示した。
ウィルスベクター
ドミナントネガティブ(DN)および野生型(wt)IGF-I受容体(IGF-IR)cDNAを、HIV-I-ホスホグリセリン酸キナーゼ1(PGK)移行ベクター40のSamI/Xbal部位にサブクローニングした。緑色蛍光タンパク質(GFP)cDNAを、BamHI/Sall部位にサブクローニングした。SamI/Xbal部位をもつHIV-I-PGKベクターを、対照(空のベクター)として使用した。パッケージング構造および小胞性口内炎ウイルスGタンパク質のエンベローブは、それぞれpCMVΔR-8.92、pRSV-RevおよびpMD.Gプラスミド41を含む。移行ベクター(13μg)、エンベロープ(3.75μg)、およびパッケージングプラスミド(3.5μg)は、10% FCS、1% グルタミンおよび1% ペニシリン/ストレプトマイシンを含むダルベッコ改変イーグル培地(DMEM, Gibco, USA)で培養された293T細胞(5×106細胞/デッシュ)にリン酸カルシウムでコトランスフェクトした。培地は、トランスフェクションの2時間前に交換し、その24時間後に交換した。調整された培地は24時間後に回収され、遠心処理を行い(1000rpm/5分)、約100倍程度まで濃縮した(19000rpm/1.5時間)。ペレットは、1%ウシ血清アルブミンを含むリン酸緩衝生理食塩水中に再縣濁し、ウイルスは-80℃で貯蔵した。ウイルス価は、HIV-1 p24 ELISA (Perkin Elmer, USA)によって決定した。
Wistarラット(5〜6月齢、〜300g)、および肝臓-IGF-I-欠乏性(LID)マウス(6〜21月齢、〜25〜30g)は、我々の所有する同系集団に由来する。動物の取り扱いについては以下のEECガイドラインに従った。使用する実験動物を極力少なくするために、本発明者らは、無処置の(ニセの)動物の反応と、空のベクターで治療された動物において得られた反応とを比較し(下記を参照)、差異が認められなかったので(例えば図1d〜fを参照)、本発明者らは、対照として後者のグループのみを使用した。
ウェスタンブロット(WB)および免疫沈降を、文献42に記載されたように行った。Aβ沈着を分析するために、冠状脳部位を連続的に切断し、88%ギ酸中で事前にインキュベートを行い、上述したように免疫染色を行った。ELISAによる合計Aβの検出のために、本発明者らは、下部層において4G8抗体(Sigma)および上部層において抗-Aβ1-40または抗-Aβ1-42(Calbiochem, USA)を使用した。Aβの可溶性および不溶性形態の両方を定量するために、サンプルをギ酸で抽出し、文献43に記載されたようにアッセイを行った。ヒトAD脳部位はNovagen (USA)から入手し、APP/PS2マウス脳はH. Loetscher (Hoffman-La Roche, Switzerland)の寄贈を受けた。げっ歯類およびヒトN-末端Aβ形態を認識するマウス抗-Aβ(MBL、日本)、抗アルブミン(Bethyl, USA)、抗トランスチレチン (Santa Cruz, USA)、抗アポリポタンパク質J (Chemicon, USA)、抗シナプトフィジン (Sigma)、抗-ダイナミン 1 (Santa Cruz)、抗-GFAP (Sigma)、抗-カルビンジン(Swant, Switzerland)、抗-βIII-チューブリン (Promega, USA)、抗-PHF-tau (AT8, Innogenetics, Belgium)、抗-ユビキチン(Santa Cruz)、抗-pSer9 および 抗-pTyr216 GSK3β (New England Biolabs, USA)、抗-pAkt (Cell Signalling, USA)は全て、1:500〜1:1000の希釈液で使用した。第二の抗体は、Alexa-結合(Molecular Probes, USA)またはビオチン化した(Jackson Immunoresearch, USA)。
文献45に詳細に記載された水迷路試験44を用いて空間的記憶について評価を行った。簡潔には、一日目の習慣付け訓練(1日)後、タンククオドラント間の優先度は認められなかったが、続く2〜5/6日後、動物は禁止されたプラットフォームを見つけ出すことを学習した(習得)。プラットフォームのない試験訓練の一日後、遊泳速度は全てのグループにおいて類似していることが解り、プラットフォームクオドラントの優先度が評価された。9〜10日後、動物に対して、最初の位置に配置されたプラットフォームを用いて長期間の(記憶)保持についての試験を行った。最後の日、キューバージョンプロトコル(cued version protocol)は、試験動物間の感覚運動および欲求の差異の可能性を排除するように行われた。行動データを、ANOVAおよびStudentのt試験によって分析した。
脈絡叢におけるIGF-Iシグナルの遮断
IGF-I受容体のドミナントネガティブ(DN)形態の発現は、IGF-Iシグナルを障害する。実際には、脈絡叢上皮細胞内におけるDN IGF-IR(KR)のウイルス誘導性の発現が、その受容体およびその下流のキナーゼAktのIGF-I誘導型のリン酸化を無効にする(図1a)。本発明者らは、IGF-Iが脈絡叢を通したCSFへのアルブミンの流入を促進することを以前に見出した4。脈絡叢細胞がHIV-KRベクターによる感染を受けると、上皮性単層を通したアルブミンのIGF-I誘導型トランスサイトーシスが阻害される(図1b)。これは、IGF-IR機能の遮断がアルブミンのようなAβキャリアの脈絡叢細胞通過を障害することを示す。従って、本発明者らは、HIV-KRベクターの脳室内注入によってインビボ脈絡叢内のIGF-Iシグナルを阻害した。
本発明者らは、脈絡叢におけるIGF-IRの遮断が、Aβキャリアの脳への流入を減少させ、脳Aβの増加を導くという仮説を立てた4。実際、HIV-KRのicv注入後、小脳(図示せず)では観察されなかった、皮質(図2a)および海馬(図示せず)におけるAβ1-xレベルの段階的増加と、CSF中のAβ1-xレベルの同時的減少(図2a)とが全特異的(panspecific)な抗Aβを使用して観察された。Aβ1-40およびAβ1-42のELISAによる定量化は、皮質中の増加したβA1-40を示す一方、βA1-42がKR注入6ヶ月後まで変化しないことを示した(図2b)。Aβ1-xまたはAβ1-42特異的抗体を使用したKR注入ラットでは、アミロイドの堆積は観察されなかった(図示せず)。Aβキャリア、例えばアルブミン、アポリポタンパク質Jおよびトランスチレチンの脳およびCSFレベルにおける並行的減少がまた観察された(図2c)。
正常な成体KR処理ラットは、たとえ高い脳Aβ1-40レベルを有していてもプラークを発達させない。プラークの欠損は、KRラットが変化のないレベルのAβ1-42、好ましいプラーク形成Aβペプチドを有するためであるか15、あるいは脳内の年齢関連変化がプラークを発達させるために必要であることによる。しかしながら、老齢のげっ歯類が認知障害および増加した脳Aβレベルのより大きな徴候を示す一方、Aβプラークを発達させないことが知られている16、17。後者に関わらず、発明者らは、KRベクターで老齢変異LIDマウスを処理した18。これらのマウスは、Aβ1-40およびAβ1-42の高い脳内レベルを示し4、脳内においてAD様アミロイド症の一因となるおそれのある低血清IGF-Iおよびインシュリン抵抗性を含む、生命における早期の他の年齢関連変化を示す。この動物モデルを用いて、本発明者らは、脈絡叢IGF-IRの遮断後のAD様変化を誘導するプロセスにさらなる見識を得るために老齢のヒト脳において見出された条件をより精巧に再現することを目指した。
これらの結果は、脈絡叢におけるIGF-IR遮断が、認知障害、アミロイド症、過剰リン酸化tau堆積、シナプス小胞タンパク質欠失および神経膠症を含む、げっ歯類におけるAD様障害の引き金を引くことを示す。これらの障害の大半は、学習が障害されたままであったが、IGF-IR遮断を回復させることによって救出され得る。反対に、AD様徴候、特に認知喪失は、IGF-IR遮断が正常な血清IGF-Iレベルよりも少ない値を示す老齢動物において誘発されたときに悪化した。IGF-IR機能の一般的減少は正常な加齢と関連するが21、これらの結果は、脈絡叢におけるIGF-IRシグナルの喪失が遅延性アルツハイマー疾患と結びつくであろうことを示す22。病因のほんの5%を包含するADの類似態様の原因がゆるやかに解明される一方、散発性ADの病因は解明されていない。従って、脈絡叢でのIGF-Iに対する減少した感受性の機構についての見識は、散発性ADの起源を解明するのに役立つであろう。例えば、ADに関連した危険因子は、影響を受けた個体内の脈絡叢におけるIGF-IR機能の大きな喪失の原因となるであろう。遅延性のAD患者は、IGF-IのAβ還元作用に対する感受性の喪失を示すであろう。興味深いことに、わずかに上昇した血清IGF-Iレベルは、散発性AD患者(IGF-Iに対する感受性の喪失に相当する状態24)の試験的研究において見出された23。

- 前記分子が、脈絡叢を通した脳脊髄液へのβアミロイドのキャリアとして作用するタンパク質の移行を促進する請求項10に記載の方法。
- 前記キャリアが、アルブミンである請求項11に記載の方法。
- 前記キャリアが、トランスチレチンである請求項11に記載の方法。
- 前記キャリアが、アポリポタンパク質Jである請求項11に記載の方法。
- 前記キャリアが、ゲルソリンである請求項11に記載の方法。
- βアミロイドのクリアランスが、脈絡叢上皮細胞のIGF-I受容体の活性を増加させることによって増加する請求項10に記載の方法。
- 前記IGF-I受容体活性を増加させるために動物に投与される分子が、標的細胞内のIGF-I受容体の発現を誘導することができる遺伝子移行ベクターである請求項16に記載の方法。
- 前記遺伝子転移ベクターが、HIVまたはAAVから誘導される請求項17に記載の方法。
- 前記ベクターが、受託番号I-3315のもと2004年11月10日付けでCNCMに寄託されたものである請求項18に記載の方法。
- 受託番号I-3316のもと2004年11月10日付けでCNCMに寄託されたドミナントネガティブなIGF-I受容体を発現することができる遺伝子転移ベクター。
- 受託番号I-3315のもと2004年11月10日付けでCNCMに寄託された機能的IGF-I受容体を発現することができる遺伝子転移ベクター。
- IGF-1受容体と相互作用する活性分子をスクリーニングするための方法であって、前記方法が、アルツハイマー疾患様障害を修飾するのに十分な時間および量において前記分子を動物に投与することを含み、アルツハイマー疾患様障害の回復がIGF-I受容体活性を増加させる分子の指標となり、かつアルツハイマー疾患様障害の外観がIGF-I受容体活性を減少させる分子の指標となる方法。
- 前記アルツハイマー疾患様障害の回復が、請求項1に記載の動物において観察される請求項22に記載の方法。
- βアミロイドおよび/またはアミロイドプラークの異常な脳蓄積が関与する疾患の予防または治療のための、IGF-I受容体をコードするヌクレオチド配列の使用。
- 前記疾患が、アルツハイマー疾患である請求項24に記載の使用。
- βアミロイドおよび/またはアミロイドプラークの異常な脳蓄積が関与する疾患の予防または治療のための、IGF-I受容体の機能に類似する機能を有するポリペプチドをコードするヌクレオチド配列の使用。
- 前記核酸配列が、IGF-I受容体の活性フラグメントをコードする請求項26に記載の使用。
- IGF-I受容体の機能に類似する機能を有するポリペプチドをコードするヌクレオチド配列を含む治療学的組成物。
- 前記ヌクレオチド配列が、IGF-I受容体の活性フラグメントをコードする請求項28に記載の治療学的組成物。
- pHIV-IGF1Rベクターを含む治療学的組成物。
