Description
本発明は、哺乳動物の神経細胞疾患の分野、及び特に効果的な化合物を同定するための方法、及びヒトにおける知的能力の進行性喪失に関連する疾患の予防及び治療に有用な、そのような化合物を用いた治療並びに組成物に関連する。
知的能力の進行的喪失として最も広く知られている神経疾患はアルツハイマー病(AD)である。世界的に、約2000万人の人々がアルツハイマー病で苦しんでいる。ADは、記憶の初期喪失に続いて見当識障害、一般的に認識機能障害としてみなされる判断並びに論理的思考の障害、及び最終的には完全な認知症によって臨床的に特徴付けられる。AD患者は最終的に該疾患の発症後4〜12年の間、ひどく衰弱して不動状態に陥る。
主要な現在のAD治療は、コリン作動性ニューロンがADの間に変性する最初の神経細胞であるために低下するアセチルコリン神経伝達物質レベルを上昇させるアセチルコリンエステラーゼ酵素を阻害することによって、進行的な記憶の喪失を遅延させることに限定される。この治療は該疾患の進行を止めはしない。
本発明は、特定の既知のポリペプチドが、神経細胞においてアミロイドベータ前駆体のプロセシングの上方調節及び/又は誘導における因子であり、かつそうしたポリペプチドの機能の阻害は、アミロイドベータペプチドのレベルを下げることに効果的であるという発見に基づいている。
本発明は、哺乳動物細胞内において、Gタンパク質共役受容体(「GPCR」)の機能とアミロイドベータ前駆体タンパク質プロセシングとの関係に関連する。
本発明のある態様は、哺乳動物細胞内において、アミロイドベータ前駆体タンパク質のプロセシングを阻害する化合物を同定するための方法であって;
(a)化合物を、配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに接触させること;及び
(b)アミロイドベータタンパク質の産生に関連する化合物−ポリペプチド特性を測定すること、を含む。
本発明の別の態様は、認識機能障害に苦しんでいる又は該病気に感受性の高い対象に対して、アミロイドベータ前駆体のプロセシングを阻害するのに効果的な量のGPCR拮抗薬(antagonist)又は逆作動薬(inverse agonist)を含む医薬組成物を投与することによって、認識機能障害を伴う病気又は該病気に対する感受性を治療又は予防する方法である。
下記の用語は、以下に示された意味を有することが意図され、また本発明の記述及び意図された範囲を理解することにおいて有用である。
(定義)
用語「作動薬(agonist)」は、作動薬が結合する受容体の細胞内反応を活性化させるリガンドをさす。
用語「拮抗薬」は、作動薬と同じ部位で受容体に競合的に結合するが、該受容体の活性形態によって開始される細胞内反応を活性化させない部分を意味し、それによって作動薬による細胞内反応を阻害することができる。拮抗薬は、作動薬又は部分的作動薬の不在下で、細胞内反応のベースラインを減少させない。
用語「接触(contact)」又は「接触すること(contacting)」は、インビトロ(in vitro)システム内又はインビボ(in vivo)システム内を問わず、少なくとも2つの部分を1つに合わせることを意味する。
用語「病気(condition)」又は「疾患(disease)」は、症状の明白な表出(すなわち、疾病(illness))若しくは異常な臨床指標の表出(例えば、生化学的指標)を意味し、あるアミロイドベータタンパク質前駆体プロセシングの欠陥に起因する。あるいは、用語「疾患」は、そのような症状又は異常な臨床指標を発現する遺伝的危険性又は環境的危険性若しくは傾向に関連する。
用語「発現性核酸」は、タンパク質分子、RNA分子又はDNA分子をコードする核酸を意味する。
用語「ハイブリダイゼーション」は、それによって核酸の鎖が塩基対を介して相補鎖に結合する任意のプロセスを意味する。用語「ハイブリダイゼーション複合体」は、相補的塩基間での水素結合の形成の効力によって、2つの核酸配列間で形成される複合体をさす。ハイブリダイゼーション複合体は、溶液中(例えば、C. sub. 0t又はR.sub.0t解析)で形成してもよく、又は溶液中に存在するある核酸配列と、固形支持体(例えば、紙、膜、フィルター、チップ、ピン、又はガラススライド、若しくは細胞又はそれらの核酸が固定される任意の他の適切な基質)上に固定された別の核酸配列との間で形成してもよい。用語「厳しい条件(stringent condition)」は、ポリヌクレオチドと本特許請求の範囲に記載されたポリヌクレオチドとの間のハイブリダイゼーションを可能にする条件をさす。厳しい条件は、塩濃度、有機溶媒、例えばホルムアミドの濃度、温度、及び当業者に周知の他の条件によって定義され得る。特に、塩濃度を減少させること、ホルムアミド濃度を増加させること又はハイブリダイゼーション温度を上げることは、厳しさを増加させ得る。
用語「逆作動薬」は、受容体の内因性形態に結合する部分を意味し、内因性リガンド又は作動薬、若しくは膜へのGTPの結合の減少の不在下で観察される通常の基準レベルの活性以下で、受容体の活性型内因性形態によって開始される基準細胞内反応を阻害する。好ましくは、該基準細胞内反応は、逆作動薬の不在下における基準反応に比較して、少なくとも30%、より好ましくは少なくとも50%、及び最も好ましくは少なくとも75%が逆作動薬の存在下で減少させられる。
本明細書で用いられているように、用語「医薬として許容し得るプロドラッグ」は、本発明において有用な化合物のプロドラッグを意味し、これは健全な医学的判断の範囲内で、過度の毒性、刺激、妥当な利益/危険性比率に相応なアレルギー反応を有する患者の組織と接触する使用に適しており、かつ本発明の化合物のそれらの使用目的に対して効果的である。用語「プロドラッグ」は、本発明、又はその医薬として許容し得る塩、水和物、又は溶媒和物において有用な、効果的な化合物を生じさせるために、インビボで形質転換された化合物を意味する。形質転換は、血中での加水分解を介するなどの、様々な機構によって発生することがある。代謝的に切断可能な基を有する化合物は、代謝的に切断可能な基の存在の特性によって、親化合物に与えられた促進された可溶性及び/又は吸収速度の結果として生体利用効率の向上が示されることがあり、それゆえそのような化合物はプロドラッグとして機能するので有利である。徹底した議論は、『プロドラッグのデザイン』、H. 用語「ポリヌクレオチド」は、一本鎖又は二本鎖形態の、かつセンス又はアンチセンス方向のポリ核酸、厳しい条件下において特定のポリ核酸にハイブリダイズする相補的ポリ核酸、及びその塩基対の少なくとも約60パーセント、及びより好ましくはその塩基対の70%が共通であり、最も好ましくは90パーセント、及び特別な実施態様ではその塩基対の100パーセントで相同であるポリヌクレオチドを意味する。該ポリヌクレオチドは、ポリリボ核酸、ポリデオキシリボ核酸及びそれらの合成類似物を含む。該ポリヌクレオチドは、約10〜約5000塩基、好ましくは約100〜約4000塩基、より好ましくは約250〜約2500塩基の範囲の長さで変化する配列によって記載される。好ましいポリヌクレオチドの実施態様は約10〜約30塩基の長さを含む。ポリヌクレオチドの特別な実施態様は、約10〜約22ヌクレオチドのポリリボ核酸であり、より一般的には低分子干渉RNA(siRNA)として記載される。別の特別な実施態様は、ペプチド核酸(PNA)、ポリシロキサン、及び2´−O−(2−メトキシ)エチルフォスフォロチオエートなどの修飾された骨格を有する核酸であり、又は非天然の核酸残基、若しくはメチル、チオ、硫酸、ベンゾイル、フェニル、アミノ、プロピル、クロロ及びメタノカルバヌクレオシドなどの1以上の核酸置換基、又はその検出を促進するためのレポーター分子を含む。
用語「溶媒和物」は、1以上の溶媒分子を有する、本発明において有用な化合物の物理的結合を意味する。この物理的結合は水素結合を含む。特定の例において溶媒和物は、例えば、1以上の溶媒分子が結晶性固体の結晶格子に組み込まれた場合に単離可能であろう。「溶媒和物」は、液相、及び単離可能な溶媒和物の両方を含む。代表的な溶媒和物は、水和物、エタノレート及びメタノレートを含む。
用語「有効量」又は「治療的に効果的な量」は、医師又は他の臨床医によって探求される対象の生物学的若しくは医学的反応を誘導するであろう化合物の量又は作用物質の量を意味する。特に、神経疾患の治療に関して、用語「有効量」は対象の脳組織中の、アミロイドベータペプチドのレベルの生物学的に有意な減少を引き起こすであろう化合物又は作用物質において、効果的にアミロイドベータ前駆体プロセシングを阻害する量を意味することを意図する。
本発明の発見の背景は、以下において手短に記載される。
1994年にMarcheseとその同僚は、GPR3遺伝子(Marcheseらの文献、1994)をクローニングし、1年後に、単一エキソンが330アミノ酸を有するこの受容体タンパク質をコードすることが見出され、アデニル酸シクラーゼ構成的活性化因子(ACCA)と呼ばれた。そのアミノ酸配列に基づいて、GPR3を、GPR6及びGPR12と同じサブファミリーに分類することができた:GPR3とGPR6は58%の同一性、及びGPR3とGPR12は57%の同一性を示す(図5)。

先に記載されたように、本発明は、Gタンパク質共役受容体(「GPCR」)が、哺乳動物、及び基本的には神経細胞におけるアミロイドベータ前駆体プロセシングの上方調節及び/又は誘導における因子であること、並びにそのようなポリペプチドの機能の抑制はアミロイドベータタンパク質ペプチドのレベルを減少させることに効果的であるという、本発明者の発見に基づいている。
(a)化合物を配列番号:4〜6からなる群から選択されるアミノ酸配列を含むポリペプチドに接触させること、
(b)該化合物の該ポリペプチドに対する結合親和性を決定すること、
(c)該化合物を発現している哺乳動物細胞の集団を、少なくとも10マイクロモラーの結合親和性を示す化合物と接触させること、及び
(d)哺乳動物細胞内で、アミロイドベータ前駆体タンパク質のプロセシングを阻害する化合物を同定することを含む。
これらの組成物及び方法の特別な態様は、GPCRポリペプチドに選択的に相互作用することができる細胞内結合タンパク質をコードするポリヌクレオチドの誘導された発現による、GPCRポリペプチドの発現の下方調節又は阻止に関連する。細胞内結合タンパク質は、それが発現している細胞内でポリペプチドに選択的に相互作用又は結合し、かつ該ポリペプチドの機能を中和することができる任意のタンパク質を含む。好ましくは、細胞内結合タンパク質は配列番号:4〜6の該GPCRポリペプチドの細胞内ドメインに結合親和性を有する中和抗体又は中和抗体のフラグメントである。より好ましくは、細胞内結合タンパク質は一本鎖抗体である。

生物学的に適合性を有する組成物は、固体、液体、ゲル、又は他の形態であることもある組成物であり、組成物中に本発明の化合物、ポリヌクレオチド、ベクター及び抗体が、活性な形態、例えば、生物学的活性に影響を与え得る形態で維持される。例えば本発明の化合物は、GPCR上で逆作動薬活性又は拮抗薬活性を有し;核酸は複製、伝達内容の翻訳、又はGPCRの相補的mRNAにハイブリダイズすることができ;ベクターは標的細胞にトランスフェクト、及び先に記載されたようなアンチセンス、抗体、リボザイム又はsiRNAを発現することができ;抗体はGPCRポリペプチドドメインに結合するであろう。
(実施例1):GPR3はアミロイドベータ1〜42のレベルを上昇させる
APPプロセシングを変化させる新規薬剤標的を同定するために、APPを過剰発現する安定細胞株を作成する。この安定細胞株は、pcDNA3.1にクローン化されたAPP770wtのcDNAをHEK293細胞にトランスフェクトし、続いて3週間、G418を用いて選抜することによって作成する。この時点でコロニーを選別し、安定なクローンを増幅させ、かつ分泌されたアミロイドベータペプチドのレベルをテストする。高レベルでアミロイドベータを分泌するクローンであるHEK293 APPwtを、薬剤標的を同定する実験のために選別する。これは、アデノウイルスcDNAライブラリーを用いてHEK293 APPwtに形質導入すること、及び結果として生じたアミロイドベータ1〜42のレベルの変化をELISAを介して測定することによって達成される。
ヒトGPR3受容体のアミノ酸配列は、GPR3の最も近い類似体を見つけるために、全てのヒトGPCRに対して、BLASTサーチにおける問い合わせ(query)として使用した。表2(配列番号:5〜6)は、2つの最も近いGPR3受容体である。ClustalWを用いて配列比較を構築して、GPR3とその最も近い類似体であるGPR6並びにGPR12との間の相同性の度合いを示した(図5)。
拮抗薬の効果は、標的タンパク質の発現レベルの減少をもたらすsiRNAに基づいた方法の使用を介して模倣し得る。例えば、GPR3 siRNAを用いたトランスフェクションはアミロイドベータ1〜42を減少させる。
オリゴフェクタミンと共に、GPR3(Dharmacon, USA:表3)、eGFP、ルシフェラーゼ及びBACEのsiRNAのスマートプールを用いて、HEK293 APPwt細胞をトランスフェクトさせる。トランスフェクション後24時間で培地を新しくし、該細胞は先に記載したように、ELISA解析に先立つ24時間の間、馴化培地中でアミロイドベータペプチドを蓄積することが可能になる。データは、GPR3 RNAレベルに対して標的化されたsiRNAは、コントロール条件に比較してアミロイドベータ1〜42レベルを減少させることを明らかに示す(図4)。結論として、これらのデータは、GPR3が分泌されるアミロイドベータのレベルを調節することを示す。同手順は、GPR6及びGPR12によるAPPプロセシングの解析に対して用いる。
APPプロセシングのモジュレーターの同定の際、モジュレーターが関心を引く組織及び細胞で発現しているかどうかを検討することは重要である。これは組織並びに細胞における、RNA及び/又はタンパク質レベルを測定することによって達成され得る。近年、RNAレベルはリアルタイムPCR技術を使用して定量され、それによってRNAは最初にcDNAへ転写され、続いて、関心を引くcDNAの増幅がPCR反応の間監視される。増幅プロット及び結果のCt値は、サンプル中に存在するRNA量についての指標である。ハウスキーピング遺伝子のレベルの決定は、異なるRNAサンプル間での標的遺伝子のRNAレベルの標準化を可能にし、ΔCt値として表される。
特異的な細胞性発現へのさらなる洞察を得るために、免疫組織化学(タンパク質レベル)及び/又はインサイチュウハイブリダイゼーション(RNAレベル)を、ヒト正常並びにアルツハイマーの脳海馬、皮質及び皮質下構造からの切片で実行する。これらの結果は、発現が、神経細胞、マイクログリア細胞、又はアストロサイトで起こるかどうかを示す。疾患組織と健康組織との比較は、GPR3が疾患組織で発現しているか、かつその発現レベルが、非病的状況に比較して変化しているかを示す。同手順は、GPR6及びGPR12の発現プロファイリングに対して使用する。
実際のニューロンにおいて、GPR3がアミロイドベータ産生に影響するかどうかを調べるために、ヒト又はラットの初代海馬若しくは皮質ニューロンに、GPR3 cDNAを含むアデノウイルスを用いて形質導入する。アミロイドベータのレベルをELISAで決定する(実施例1を参照されたい)。齧歯類のAPP遺伝子は、該ヒト配列に比較してAPP中に多くの突然変異を有しているため、齧歯類のAPP遺伝子はより少ないアミロイドベータ1〜40及び1〜42を産生する。より高いアミロイドベータレベルを達成するために、GPR3と、ヒト野生型APP又はヒトスウェーデン変異型APP(これはアミロイドベータ産生を増幅する)cDNAとの共形質導入を実行する。
データ(図6)は、初代ニューロンにおけるGPR3の過剰発現の増加レベルによって、ネガティブコントロールウイルスに比較して、アミロイドベータの1〜42レベルが用量依存的に増加して対応したことをはっきりと示している。
(レポーター遺伝子選別)
HEK293細胞又はCHO−K1細胞などの哺乳動物細胞は、cAMP依存性プロモータ(CREエレメント)の制御下にルシフェラーゼ遺伝子を保持しているプラスミドを用いて安定的にトランスフェクトするか、又はcAMP依存性プロモータの制御下でルシフェラーゼ遺伝子を保持しているアデノウイルスを用いて形質転換するかのどちらかである。さらに、レポーターコンストラクトを、ルシフェラーゼ遺伝子と共に、Ca2+依存性プロモータ(NF−ATエレメント)の制御下又はNF−κBによって制御されるプロモータの制御下で使用することができる。該レポーターコンストラクトを発現しているこれらの細胞をそれから、本発明のGPCRのcDNAを保持しているアデノウイルスを用いて形質導入する。形質導入40時間後、該細胞を以下を用いて処理する:
a)受容体に対する作動薬(例えば、スフィンゴシン−1−リン酸)、及びペプチド(LOPAP, Sigma Aldrich社)、脂質(Biomol, TimTech社)、炭水化物(Specs)、天然化合物(Specs, TimTech社)、小さな化合物(Tocris)、市販のスクリーニングライブラリー、並びに配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに結合親和性を有することが実証された化合物を含む対照化合物の多数のコレクションに対して選別された作動薬であって、アリールオキシジチオウレア(米国特許第6,420,563号を参照されたい)、その塩、水和物、又は溶媒和物を含む化合物を含む作動薬、又は、
b)GPR3が構成的に活性なGPCRであると考えられる場合においてのみ、ペプチド(LOPAP, Sigma Aldrich社)、脂質(Biomol, TimTech社)、炭水化物(Specs)、天然化合物(Specs, TimTech社)、小さな化合物(Tocris)、市販のスクリーニングライブラリー、及び配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに結合親和性を有することが実証された化合物を含む対照化合物の多数のコレクションであって、アリールオキシジチオウレア(米国特許第6,420,563号を参照されたい)、その塩、水和物、又は溶媒和物を含む化合物を含む対照化合物の多数のコレクション。
HEK293細胞又はCHO−K1細胞などの哺乳動物細胞に、本発明のGPCRのcDNAを保持する発現プラスミドコンストラクトを用いて安定的にトランスフェクトする。十分に安定な細胞を得ることができるまで、細胞をまき、成長させ、選別する。細胞はFura3又はFura4などのCa2+依存性蛍光物質と共にロードする。余剰の蛍光物質を洗い流した後、該細胞へ同時に作動薬(あるいは、該受容体の構成的活性が使用されている場合には、作動薬を添加する必要はない)及び化合物を添加することによって、ペプチド(LOPAP, Sigma Aldrich社)、脂質(Biomol, TimTech社)、炭水化物(Specs)、天然化合物(Specs, TimTech社)、小さな化合物(Tocris)、市販のスクリーニングライブラリー、及び配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに結合親和性を有することが実証された化合物を含み、アリールオキシジチオウレア(米国特許第6,420,563号を参照されたい)、その塩、水和物、又は溶媒和物を含む化合物含む、対照化合物の多数のコレクションを対照に、該細胞を選別する。該受容体の活性化は、放出される蛍光物質とCa2+との相互作用に起因するほとんど瞬間的な蛍光の増加として測定する。作動薬によって誘導される蛍光の増加(又は構成的な蛍光)を減少させる又は阻害する化合物は、それらを選別するのに対照する受容体に対する拮抗薬又は逆作動薬であると考えられる。これらの化合物を再び選別し、GPCRへの結合と同様に、分泌されるアミロイドベータペプチドの量を測定する。
安定的にアポエクオリン(Apoaequorin)を発現しているCHO細胞に、GPCRのcDNAを保持しているプラスミドコンストラクトを用いて安定的にトランスフェクトする。十分に安定な細胞が得られることができるまで、細胞をまき、成長させ、選別する。該細胞をアポエクオリンの補因子であるセレンテラジン(coelenterazine)とともにロードする。受容体活性化の直後に細胞内Ca2+貯蔵は枯渇し、そしてエクオリン(aequorin)は発光プロセスにおいてセレンテラジンと反応する。該細胞に同時に作動薬(あるいは、該受容体の構成的活性が使用されている場合には、作動薬は添加する必要はない)及び化合物を添加することによって、ペプチド(LOPAP, Sigma Aldrich)、脂質(Biomol, TimTech)、炭水化物(Specs)、天然化合物(Specs, TimTech)、小さな化合物(Tocris)、市販のスクリーニングライブラリー、及びアリールオキシジチオウレア(米国特許第6,420,563号を参照されたい)、その塩、水和物、又は溶媒和物を含む化合物を含む配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに対して結合親和性を有することが実証された化合物を含む、対照化合物の多数のコレクションを対照に、アポエクオリン及び該受容体の両方を安定的に発現しているCHOを選別する。該受容体の活性化を、アポエクオリン、セレンテラジン、及び放出されるCa2+の相互作用に起因する、ほとんど瞬間的な発光として測定する。作動薬が誘起する光、又は構成的活性の増大を低下若しくは阻害する化合物は、それらが選別された該受容体に対する拮抗薬又は逆作動薬であると考えられる。これらの化合物を再び選別し、該GPCRへの結合に加えて、分泌されるアミロイドベータペプチドの量を測定する。
化合物はGPCRポリペプチドへの結合について選別する。GPCRポリペプチドへの化合物の親和性は置換実験で決定する。手短に言うと、GPCRポリペプチドは、GPCRポリペプチドに結合することが既知である標識された(放射性同位体で標識された、蛍光標識された)リガンド(例えば、スフィンゴシン−1−リン酸、又はジヒドロスフィンゴシン−1−リン酸)、及びラベルされていない化合物と共にインキュベートする。GPCRポリペプチドからの標識されたリガンドの置換は、現在GPCRポリペプチドに結合している標識されたリガンドの量を測定することによって決定する。GPCRポリペプチドに結合した量は化合物の濃度に対してプロットし、IC50値を計算する。この値は、その標的、すなわち化合物のGPCRポリペプチドへの結合親和性を反映する。強力な結合剤は、ナノモラー及びピコモラーの範囲においてさえIC50を有する。少なくとも10マイクロモル又はそれ以下の(nmolからpmol)のIC50を有する化合物をベータアミロイド分泌アッセイに適用し、ベータアミロイド分泌及びプロセシングにおけるそれらの効果をチェックする。GPCRポリペプチドは、アッセイが細胞、細胞画分又は生化学的に精製されたタンパク質のどれで行われるかによって多くの方法で調製可能である。
Gタンパク質受容体が構成的に活性になる場合、それはGタンパク質(Gq、Gs、Gi、Go)に結合し、Gタンパク質へのGTPの結合を促進する。Gタンパク質はそれからGTPaseとして機能し、GTPをGDPへとゆっくりと加水分解し、それによって通常条件下の該受容体は不活化された状態になる。しかしながら、構成的に活性化された受容体はGDPからGTPへの交換を続ける。GTPの非加水分解性類似体である[35S]GTPγSを用いて、構成的に活性化された受容体を発現する膜への促進された結合をモニターすることができる。[35S]GTPγSを用いて、リガンド不在及び存在下で膜に共役するGタンパク質をモニターすることができることが報告されている。さらに、好ましいアプローチは、GPCR−Gタンパク質融合タンパク質の使用である。GPR3−Gタンパク質融合タンパク質を産生するための方法は当業者に周知である。GPR3−Gタンパク質融合タンパク質を発現している膜を、逆作動薬などの候補化合物の直接的な同定における使用のために調製する。ホモジナイズされたGPR3−Gタンパク質融合タンパク質を有する膜を、96ウエルプレート中に移す。ピンツール(pin-tool)を使用して候補化合物を[35S]GTPγSを加えたそれぞれのウエルに移し、続いて室温で60分間、シェーカー上でインキュベーションする。該アッセイは、22℃で15分間、4000RPMで該プレートを回転させることによって停止させる。該プレートをそれから吸引し、続いて放射活性を読み取る。同手順をGPR6及びGPR12の解析に用いる。
受容体は、アデノウイルス形質導入細胞(米国特許第6,340,595号)によって、哺乳動物細胞(HEK293、CHO、COS7)で発現させる。該細胞を、10pM〜10μMにわたる範囲の様々な濃度で、標識されたリガンド(ヨウ素化された、トリチウム化された、又は蛍光)、及びラベルされていない化合物の両方と共にインキュベートする(4℃で3時間:pH7.4に調整した、25 mM HEPES, 140 mM NaCl, 1 mM CaCl2, 5 mM MgCl2 及び0.2% BSA)。反応混合物を、細胞収穫機(Packard)を用いてPEI処理したGF/Bガラスフィルター上に吸引する。該フィルターを、氷冷洗浄バッファー(pH7.4に調整した、25 mM HEPES, 500 mM NaCl, 1 mM CaCl2, 5 mM MgCl2)で2回洗浄する。シンチラント(Scintillant)(MicroScint−10; 35 μl)を乾いたフィルターに加え、そして該フィルターを(Packard Topcount)シンチレーションカウンターで計測する。データをプリズムソフトウエア(GraphPad Software, San Diego, Calif)を用いて解析し、プロットする。競合曲線を解析し、かつIC50値を計算する。1以上のデータ点が、競合曲線のシグモイド範囲内、又は該シグモイド範囲の近傍にこない場合、該アッセイを繰り返し、そして標識されたリガンド及びラベルされていない化合物の濃度を、該曲線のシグモイド範囲内又はその近傍により多くのデータ点を有するように適合化させた。
膜分画は下記のように行い、受容体を過剰発現している哺乳動物細胞(HEK293、CHO、COS7)から単離する:培地を該形質導入細胞から吸引し、そして細胞を穏やかに剥離することによって1×PBS中に収集する。細胞を沈殿させ(2500rpm、5分)、そして50mMトリスpH7.4に再懸濁した(10×106細胞/ml)。該細胞沈殿物は、5秒で3回超音波処理することによって(UP50H; sonotrode MSl; 最大振幅: 140 μm; 最大超音波密度: 125W/cm2)ホモジナイズする。膜画分を、最大速度(13000〜15000から20000g、又はrcf)で20分遠心分離することによって調製する。結果として生じた沈殿物を500μlの50mMトリスpH7.4に再懸濁し、そして再び5秒で3回超音波処理した。該膜画分を遠心分離によって単離し、そして最終的にPBS中に再懸濁した。結合競合及びIC50値の導出を、先に記載したように決定する。
GPCR関連性シグナル転換経路の活性化は一般的に、細胞質から原形質膜、又は細胞質から核への特定のシグナル転換分子の転移を導く。Norak社は、細胞質から原形質膜への受容体−β−アレスチン−GFP複合体の作動薬誘導性転移、及びそれに続く受容体脱感作の間に起こるこの複合体の内在化に基づいて、自社のトランスフロー(transfluor)アッセイを発展させてきた。同様なアッセイは、β−アレスチンの代わりにGFP標識された受容体を用いる。HEK293細胞を、GPR3−eGFP融合タンパク質を翻訳するGPR3−eGFPベクターを用いて形質導入する。形質導入後48時間で、該細胞を60分間、未使用無血清培地に置き、37℃かつ5%CO2で15分間、30分間、60分間又は120分間リガンド(例えば、100nMスフィンゴシン−1−リン酸)で処理する。表示の曝露時間後、細胞をPBSで洗浄し、そして室温で20分間、5%パラホルムアルデヒドを用いて固定する。GFP蛍光を、デジタルカメラがついたツァイス(Zeiss)顕微鏡で可視化する。この方法は、細胞内区画への該融合タンパク質のリガンド介在性(構成的活性仲介型)転移を阻害する化合物の同定を目的とする。同手順は、GPR6及びGPR12の解析に用いる。
β−アレスチン及びβ−ガラクトシダーゼ酵素相補性、並びにエネルギードナーとしての受容体及びエネルギーアクセプターとしてのβ−アレスチンを有するBRETに基づいたアッセイを使用することで、転移アッセイには様々なバリエーションが存在する。またpH感受性色素を用いて標識された特異的受容体抗体の使用は、作動薬によって誘導された酸性リソソームへの受容体転移を検出するために使用する。それらの転移アッセイの全ては、作動薬性及び拮抗薬性作用リガンドの両方のスクリーニングのために使用する。
メラニン細胞アッセイは、ゼノパスメラノフォアズ(Xenopus melanophores)におけるメラノソームを含むメラニンの分布を変化させるGPCRの能力に基づいている。メラノソームの分布は、Gi/o又はGs/qのどちらかに共役する外因性受容体に依存する。メラノソームの分布(分散した又は凝集した)は、吸光度を測定することによって容易に検出される。このタイプのアッセイを、拮抗薬化合物選別と作動薬化合物選別の両方に用いる。
Claims (37)
- 哺乳動物細胞内で、アミロイドベータ前駆体タンパク質のプロセシングを阻害する化合物を同定するための方法であって、
(a)化合物を配列番号:4〜6、289〜333からなる群から選択されるアミノ酸配列を含むポリペプチドに接触させること;及び
(b)アミロイドベータタンパク質の産生に関連する化合物−ポリペプチド特性を測定することを含む、前記方法。 - 前記ポリペプチドが、インビトロ無細胞調製において配列番号:289〜333を含む、請求項1記載の方法。
- 前記ポリペプチドが膜結合型である、請求項1記載の方法。
- 前記ポリペプチドが、哺乳動物細胞内で膜貫通型細胞レセプターとして存在する、請求項2記載の方法。
- 前記特性が、前記化合物の前記ポリペプチドへの結合親和性である、請求項1〜4のいずれか1項記載の方法。
