本出願は、現在は放棄された1997年12月19日提出の出願番号第60/068,094号仮出願の利益を主張し、後に米国特許第6,291,431号として発行された1998年4月2日提出の米国特許出願第09/054,331号の継続出願である米国特許出願第09/839,695号の継続出願であり、各出願は参照により全体が本明細書中に含まれる。
薬剤耐性ブドウ球菌が主な医療問題となっている
ブドウ球菌(特に、S.aureusやS.epidermids)は、ヒトに対する主要な病原体であり、そして、毎年入院患者の約200万人に院内感染を引き起こし、米国の病院での死亡率の35%増加を引き起こしている米国で報告される院内感染の最大の共通原因である。S.aureusおよびS.epidermidsによる感染は、院内での肺炎、手術部位および血流の感染、医療器具に起因する感染、ならびに骨髄炎、敗血症性関節炎、心内膜炎、髄膜炎など、市中感染の主な原因となっている(Rubin RJなど、Emerg Infect Dis.1999;5:9−17[1])。現在、世界のブドウ球菌感染患者の95%は、ペニシリンまたはアンピリシンなどの第一世代の抗生物質には反応しない。以前、薬剤耐性ブドウ球菌は、概ね病院および養護施設に限られていたが、現在は市中に広がりつつある(Lowy FD.N.Engl.J.Med.1998;339:520−532[2])。
S.aureusは、ヒトの皮膚の正常な細菌フロラの一部であるが、生物膜の形成および毒性外部分子の少なくとも一方の形成による致死的疾患の原因となりうる。毒素は、毒素性ショック症候群を引き起こす発熱性毒素である毒素性ショック症候群毒素−1(TSST−1)、食中毒の主な原因であるブドウ球菌エンテロトキシン、細菌がその代謝産物の環境を利用可能にし、宿主内にその細菌を拡散可能にするタンパク質分解酵素、および、ブドウ球菌によって発現、分泌、結合され、感染過程の結果への影響を示す溶血素、ロイコシジンおよびその他の毒性因子を含んでいる[2]。
生物膜の形成および毒素の生産は、細菌(自己誘導物質)によって生産、分泌された分子が閾値濃度に到達し、形質導入経路の信号を活性化し、さらに毒性因子をコード化している遺伝子を活性化するクオラムセンシング機構によって調節される[2]。
RAPは、その標的分子TRAPのリン酸化を誘発し、未知の様式で細胞接着の増加およびagrの活性化の両方をもたらす。一度agrが(増殖の指数増殖期の中期において)活性化されRNAIIが生産されると、オクタペプチド(AIP)や、その受容体AgrCが生産される。AIPは、TRAPのリン酸化を低減するとともに(その結果接着特性が低下する)[5]、独立してその受容体、AgrCのリン酸化を増大する[6]。これがAgrAのリン酸化をもたらし、その結果、RNAIIIが生産される。RNAIIIは、多数の毒性外部分子の発現をもたらす[10]。
RAP(天然型および組み換え型)のワクチンを接種されたマウスは、S.aureusのチャレンジ試験から守った[3]。これは、S.aureusの病原性におけるRAPの重要な役割を確認し、新しいワクチン開発の機会を開くものである。
表面に接着した細菌は、それら自身が合成する水和重合マトリクスで集塊して生物膜を形成する。これら固着性の群体の形成および抗菌薬に対する先天的な抵抗力は、多くの持続的、慢性的細菌感染の根元となっている(Costerton JWら、Science.1999;21:284:1318−1322[15])。生物膜は、多くの場合、不活性な表面または死んだ組織上で優先的に発育し、そして、医療器具や除去して死んだ骨のような死んだ組織断片でも一般的に起こり、さらに、それらは、心内膜炎の場合などのように生体組織上でも形成し得る。生物膜は、1箇所以上でゆっくりと増殖し、生物膜感染は、しばしば、明確な症状を呈するのが遅い。定着した細菌細胞は、抗原を放出し、抗体の生産を刺激するが、抗体は、生物膜内の細菌の殺滅効果はなく、周囲の組織に免疫複合物による病変を引き起こす可能性がある。すぐれた細胞および体液の免疫反応を有する個体においても、生物膜感染は、宿主の防御機構ではほとんど解消されない。代表的な例としては、抗生物質による治療は、生物膜から放出される浮遊集合体細胞によって引き起こされた症状を緩解させるが、生物膜の殺滅には機能しない。このような理由から、生物膜感染は、代表的な例として、抗生物質治療のサイクルの後、接着性の個体群が体内から外科的に除去されるまで再発症状を呈する。したがって、生物膜が形成された後に生物膜を根絶させようと試みるのでなく、生物膜形成を阻害することが重要である。
RIPは、真核細胞(HEp2細胞で試験)およびプラスチック(ポリスチレン、シリコンおよびポリウレタンで試験(Balaban Nら、Kidney Int. 2003;63:340−345[16]))に対する細菌接着を低減する。RIPは、ブドウ球菌の感染を予防するために、医療器具を被覆するのに用いることができる。
RIPは線形ペプチド[13]であるがAIPは活性化されるチオラクトン構造を含有する[11]こと、RIPのセンサはTRAP[5]であるがAIPのセンサはAgrC[6]であること、RIPは細胞接着と毒素生産[16]の両方を阻害するが、阻害性のAIPは毒素生産を阻害するが細胞接着を活性化する(Vuong C, Saenz HL,Gotz F, Otto M.S.aureus中のポリスチレンに対する接着の対するagrクオラムセンシングシステムの影響、J.Infect Dis 2000;182:1688−1693[17])ということから、RIPは、ATPとは逸脱している。
具体的な分子の機構は完全に理解されていないが、RIPは、agrの発現(RNAIIおよびRNAIII[13])を阻害し、それゆえ毒素の生産を阻害することが知られている(Vieira−da−Motta Oら、Peptides、2001;22:1621−1628[18])。agrヌル細胞の接着が、野生型と同様にRIPの存在下で阻害されることから、RIPは、agr非依存性の機構において細胞接着を調節することが知られている。RIPは、TRAPのリン酸化[5]を阻害し、TRAPが細胞接着、agr発現、そして、病原性に不可欠であることがこれまで実証されてきているため(以下の詳細な説明を参照)、RIPは、S.aureusの病原性を、TRAPを介して、そして、おそらく、付加的な標的を介して調節する。
他の細菌に起因する感染は、TRAPと類似した配列のタンパク質を有しているようである。これら細菌には、Bacillus subtilus、Bacillus anthracis、Bacillus cereus、Listeria innocua、Listeria monoctogenesが含まれる(本発明の詳細な説明を参照)。
本発明は、非病原性のブドウ球菌属から単離、精製されたタンパク質(RAP)も提供する。RAPタンパク質は、約33kDaの分子量を有する。一実施形態において、RAPは、米国特許出願第09/839,695号の配列識別番号12を含み、米国特許出願第09/839,695号の配列識別番号13のアミノ酸配列を含むポリヌクレオチドによってコード化されたタンパク質である。
用語「RAP遺伝子」は、総称的にRAP遺伝子およびその代替形態を意図して使用される。また、「RAP遺伝子」は、特異的なRAPタンパク質をコード化するオープンリーディングフレームと、発現の調節に関与する5’と3’非コード化ヌクレオチド配列(例えばプロモータ領域)を意図している。RAP遺伝子が米国特許出願第09/839,695号の配列識別番号12の配列を含む一実施形態では、遺伝子は、染色体外での維持や、宿主への組み込みを目的とする適切なベクターに導入できる。
RAPタンパク質は、任意の適切な手段、例えば天然にRAPを発現する細菌からの単離、組み換え手段(例えば米国特許出願第09/839,695号の配列識別番号12の配列を有するポリヌクレオチドの発現)、合成手段などによって生産することができる。
大量のポリペプチドが入手可能であれば、発現宿主を利用することによって、RAPタンパク質は、従来の方法、例えばHPLC、排除クロマトグラフィ、ゲル電気泳動、アフィニティクロマトグラフィを使用して、または、その他精製技術などによって単離、精製できる。精製されたタンパク質は、一般に、純度が少なくとも約80%、望ましくは少なくとも約90%であり、最高純度100%にもできる。
本発明は、RAPと特異的に結合し、免疫反応性を有する抗体も提供する。抗体は、モノクローナル抗体、ポリクローナル抗体またはヒト化抗体であり、当業者に周知の方法を使用して調製される。一般に、抗体は、従来の方法で調製され、タンパク質またはその抗原性部分を、単独でまたはKLH、pre−S、HBsAg、そして、その他ウイルス性または真核細胞タンパク質などの既知の抗原性基材に結合して免疫原として使用される。種々のアジュバントを、適宜一連の注射で利用することもできる。モノクローナル抗体の場合、1回または複数回のブースタ注射の後、脾臓が単離され、リンパ球が細胞融合によってイモータライズされ、高親和性抗体の結合のためにスクリーニングされる。望ましい実施形態では、脾臓またはリンパ節細胞と骨髄腫細胞とは、約20:1から約1:1の比率で、望ましくは約2:1の比率で混合される。融合処理で使用される体細胞および骨髄腫細胞のソースとして、同種の動物を使用することが望ましく、動物は、ラット、マウス、ウサギ、ウシ、ニワトリ、シチメンチョウまたはヒトから選択される。体細胞と骨髄腫細胞との融合によってハイブリドーマを生産し、このハイブリドーマは、所望のモノクローナル抗体を標準的な手法で生産するため、培養で育成される。さらに詳しい説明は、例えば、モノクローナル抗体は、実験室マニュアル、HarlowとLane eds、Cold Spring Harbor Laboratories、コールドスプリングハーバー、ニューヨーク、1988を参照。所望の場合、H鎖およびL鎖をコード化するmRNAは、Escherichia coliでのクローニングによって単離および変異し、H鎖およびL鎖は、抗体の親和性をさらに高めるために混合することもできる。抗体を生産するための方法としてのインビトロでの免疫法に代わるものとしては、通常、インビトロでのアフィニティマチューション接合といったファージ「ディスプレイ」ライブラリへの結合が含まれる。
細菌中のRAP(RplB)配列
RAPは、S.aureus RN6390Bの培養上清から精製され、RAPのNH2−末端配列をIKKYKPITN(Balaban Nら、Science 2000、287:391a[18])とすることに決定した。この配列は、いくつかのS.aureusデータベースと比較され(TblastNアルゴリズム)、わずか1個のORFが上記ペプチド配列と強く一致していることが見いだされた。このORFは、推定277アミノ酸タンパク質をコードしており、L2(またはRplB)配列のオーソログである(Ban Nら、Science 2000、289:905−920[19])。
rRAPを生産するため、制限部位を加えたrap(RplB)遺伝子5’および3’末端に対応するフォワードプライマおよびリバースプライマが設計された。これらのプライマは、S.aureus RN6390B染色体DNAをテンプレートとして用い、PCRによって完全なRplB遺伝子を増殖するのに使用された。増殖されたDNAは、挿入された遺伝子の5’末端に6ヒスチジン標識を有するpET14bベクター(Novagen、ウィスコンシン)の対応する部位に、分解されて連結された。rap(pET2−5)を含むプラスミドが、E.coli BL−21(DE23)/pLysSを転換するのに使用された。細胞が誘導、採取され、組み換えHis−rRAPタンパク質が、ニッケルカラムを使用して単離され、his標識をトロンビンで除去した。
モノクローナル抗体は、マウスから生産され、rRAPによってプライミングされた。ハイブリドーマ上清は、注射した抗原に対する特異的な抗体の存在をELISA法で試験された。陽性ハイブリドーマは、クローン化され、腹水を得るために用いられた。組み換えタンパク質に対する抗体が天然型分子を認識するかどうかを試験するため、rRAPと、部分的に精製された天然型RAP(野生型S.aureus RN6390>10kDaの指数増殖後の上清)とが、SDS12.5%のPAGE、ウェスタンブロットにかけられ、膜がポンソーで染色された(図2、レーン1、2)。膜は、ブロッキングされ、陽性抗rRAPハイブリドーマから作成した腹水とともにインキュベートされた(希釈率1:1000)。結合された抗体は、ペルオキシダーゼ抱合抗マウスIgGを用いて検出され、化学発光を用いて可視化した(図2、レーン3、4)。図2に示すように、rRAPに対して生じたモノクローナル抗体は、上清に分泌された天然型分子を特異的に認識する(図2、レーン4)。
組み換えRAPは、活性型でありRNAIII合成を誘発できるか否かを試験するため、rnaiii::blaZ融合構成物を含有するS.aureus細胞が、rRAPの増量のために培養された。対照として、細胞が、増殖培地のみで培養された(ネガティブコントロール)。RNAIII(□−ラクタマーゼ活性として)は、黄色の物質で、□−ラクタマーゼの存在時にはピンク色になるニトロセフィンの添加によって検出した。この方法は、実験群と対照群との間の差が、少なくとも2倍ある場合有用であり、そして、片方はピンク色になるが、残りの他方は黄色のままである。図3Aに示すように、rRAPは、濃度依存性にRNAIII合成を活性化し、細菌数が2.25nmol/107の閾値に到達する。組み換えRAPによるRNAIIIの活性化は、部分的に精製されたRAPの活性化と類似していた。2.25nmolのrRAPの存在下でのRNAIII合成(□−ラクタマーゼ活性)の増加は、rRAPの添加の内場合と比較して有意な(P<0.00169)差がある。また、本発明者らは、ノーザンブロット法によるRNAIII合成の誘発試験も行い、ここでは、細胞は、rRAPあり、または、なしで30分間増殖し、細胞を採集し、ノーザンブロットした後、RNAIIIの存在を、RNAIIIに特異的な、放射線標識したDNAを使用して検出した。膜は、オートラジオグラフィーにかけられ、バンドの密度が検出された。図3Bに示すように、rRAPは、RNAIIIの合成を誘発した。
TRAP
TRAPはS.aureusの病原性に必要とされる
RAPは、TRAPのリン酸化反応を誘発し、そして、RIPは、TRAPのリン酸化反応を阻害することが示されてきている[5]。TRAPがS.aureusの病原性に重要であることを示すため、traP遺伝子は、S.aureus 8325−4で分離されて、親株(TRAP+)と変異株(TRAP−)が、増殖、RNAIII合成、毒素生産、そして、病原性について試験された。細胞増殖およびRNAIII合成を試験するため、細胞は、指数増殖期の早期から指数増殖期の後まで増殖された。間隔を置いて、細胞密度が決定され、各増殖段階からの細胞試料が、収集され、ノーザンブロットにかけられ、RNAIIIの存在をRNAIIIに特異的な、放射線標識されたDNAを用いて検出された。図6Aに示すとおり、TRAP+株とTRAP−株との間では、細胞増殖の差が観察されなかった。TRAP+株のRNAIIIの合成は、指数増殖期の中期から観察されたが、TRAP−株(図6B)には存在せず、これは、TRAPがRNAIII合成の誘発における重要な因子であることを確認するものである。
S.aureusおよびS.epidermidsの種々の臨床単離株におけるtraP遺伝子が、PCR法で増幅され、その配列が決定された。これら配列と、異なるデータベース(NBBI微生物ゲノムデータベース[http://www.Ncbi.nlm.nih.gov/Microb_blast/unfinisihedgenome.html]など)におけるBLAST検索との比較では、TRAPがブドウ球菌に固有のものであることが示されている。導き出されたS.aureusとS.epidermidsとのTRAPタンパク質のアミノ酸配列の比較では、配列がブドウ球菌全体にわたって高度に保存されていることを示している。種々の株からのS.aureusTRAPタンパク質の配列のマルチシークエンスアライメント解析(ClustalW)は、配列が2つのサブグループに分類されることを示している(図9)。第1群は、8325のTRAPを含み、これはCOL、MSSA476、Mu50/ATCC700699およびN315(16)や本発明者らの臨床単離株#7および#11と同一である。第2群は、本発明者らの臨床単離株#12および#15や、MRSA252のTRAPを含む(Sanger Centre Database)。第1群のTRAPは、第2群のTRAPに対して約97%の相同性を有する(blastPアルゴリズムによって決定したE値が2e−84である)。第1群のTRAPおよび第2群のTRAP配列のS.epidermidsの配列への類似性は、約86%である(E値は9e−65)。
TRAPは、ブドウ球菌に極めて固有のものであるが、Bacillus subtilisの仮説タンパク質であるYhgCタンパク質に対しいくらかの配列の類似性を有する(GenBank受入番号Z99109)(E値が9e−15である)。興味深いことに、Bacillus subtilisに加えて、160を超える真正細菌のゲノムのうち、B.cereus(TIGRデータベース)の配列と97%相同のBacillus authracis、/Listeria innocuaおよびListeria monocytogenesのみのORFが、TRAPとある程度の配列の類似性を有すると同定される(E値はそれぞれ0.005、0.035、0.1である)。TRAP同様、全てのORFは、166AAであるYhgCを除き、確実に167AAである(図10)。
TRAP遺伝子領域の同じ構造が、全てのブドウ球菌属(S.aureusおよびS.epidermidsの両方)ゲノムにおいて見いだされる(図11)。TRAP遺伝子は、2つのポリシストロン性のオペロンが並んでおり、2つのうち1つ(traPの上流側)は、プロトヘムIX生合成経路(ヘムEHY遺伝子)の後期段階の酵素をコード化しており、2番目(traPの下流側)は、推定的な複数タンパク質の輸送システム(ecsAB(C)遺伝子)をコードしている。traP遺伝子転写の方向は、hemおよびecsオペロンの両方と反対になっている。同一の組織は、B.subtilis、B.anthracisのyhgC領域にも、リステリアのTRAP様ORFにも見出される(図11)。これら結果は、TRAPが細菌のシグナル伝達物質のクラスを示し、TRAPがブドウ球菌に加えて多くの細菌種の治療に対する標的部位であることを示唆している。
TRAP様分子は、ブドウ球菌に加えて、他の細菌種で見いだされることから、本発明者らは、種々のリステリア(Listeria monocytogenesおよびListeria ivanovii)株、種々のバチルス菌株(B.anthracis、B.subtilis、および、B.cereus)を、P32を伴って増殖させ、SDS PAGEとその後のオートラジオグラフィーによってTRAP(21kDa)のリン酸化を試験した。図12に示すように、リステリア菌属は、21kDaのタンパク質は、予想通り、インビボでリン酸化された。バチルス菌属に対しても類似の結果が得られた(図示しない)。これらの結果は、病原性が、細菌発現TRAP様タンパク質のTRAPリン酸化を介して調節されることもまた示唆している。
RIP(天然型または合成、YSPWTNFまたは誘導体)は、RNAIIおよびRNAIII合成を阻害や、これによる毒素生産を阻害する[13、18]だけでなく、S.aureusのヒト細胞への接着を阻害し、プラスチック上の生物膜の形成も阻害することが実証されてきている[16]。また、S.epidermidsの毒性も、その宿主細胞への接着能力や、医療器具への生物膜の形成能力に関連していることが多い。RIPが、S.epidermidsによる宿主細胞でのコロニー形成を予防できるかどうか、それにより、治療および予防の候補となるかどうかを試験するため、FITC標識化S.epidermidsが、RIPの存在または非存在下のいずれかにおいて、ケラチノサイト(HaCat細胞)の融合層とともに30分間培養された。図14Aに示すように、RIPはHaCat細胞へのS.epidermids接着を有意に(p<0.05)低減した。
RIPが移植に関連する感染保護できるかどうかを試験するため、ラット移植モデルが使用された。
移植に関連する感染の誘発
ラット(ウィスター系成体オス(300−350g)、n=15/実験グループ)は、麻酔され、背中の毛を剃られ、そこの皮膚を10%ポビドン−イオジン溶液で洗浄された。皮下のポケットが、1.5cm切開することによって、正中線の側部に作成された。無菌状態で、1cm2の無菌のコラーゲンでシールされたダクロン移植体(Albograftの登録商標、Sorin Biomedica Cardio、S.p.A.、Saluggi VC、イタリア)が、そのポケットに移植された。移植の直前に、ダクロンの移植体は、抗生物質(ムピロシン100mg/L、キヌプリスチン−ダルフォプリスチン50mg/L、レボフロキサシン30mg/L、リファンピン5mg/L)とともに、または、抗生物質なしで、生理食塩水の10mg/LのRIPの滅菌溶液に、また、対照として、生理食塩水のみか、不活性RIPペプチドアナログ(YKPETNF)のいずれかに、20分間浸漬された。ポケットは、皮膚クリップで閉じられ、2×107個の細菌を入れたか、または、入れていない1mLの滅菌生理食塩水が、液で満たされた皮下のポケットを作成するため、ツベルクリン注射器を用いて移植体に接種された。RIPまたは生理食塩水に浸漬された移植体を有する動物のうち数体は、抗生物質(セファゾリン30mg/kg、イミペネム30mg/kg、テイコプラニン10mg/kg、レボフロキサシン10mg/kg)を腹腔内に投与された。移植体は、移植から7日後に摘出された。移植体に接着した概算のRIP量は、10−26□gであった。
摘出した移植体は、滅菌チューブに入れられて滅菌生理食塩水で洗浄され、10mLのリン酸緩衝生理食塩水が入ったチューブに入れられ、移植体から付着した細菌を除去するため、5分間超音波処理された。生菌の定量は、細菌懸濁液の10倍連続希釈液(0.1mL)を血液寒天プレートに植えることによって実行された。全てのプレートが、37℃で48時間培養され、株の存在を評価された。細菌は、1プレートあたりのコロニー形成単位(CFU)の数を数えることによって定量された。細菌が効果的に移植体から除去されたかどうかを決定するため、洗浄され、超音波処理された移植体は、ニコンエクリプスE600光学顕微鏡(ニコン Y−THS、日本)で観察した。
インビボでの実験における培養結果の定量は、平均値の□標準偏差の平均値で表された。結果の比較は、対数変換されたデータでの分散の解析(ANOVA)によって行われた。統計解析が、マイクロソフトエクセル(マイクロソフト、ワシントン)によるStudent’s t−検定を用いて、インビトロでの付着の分析のために行われた。P値が□0.05の場合、有意差があるとした。
RIPが移植体に関連した感染を予防するか否かについて試験をするため、ダクロン移植体が、RIPで被覆されるか、または、被覆されず、そして、種々の種類の抗生物質で被覆されるか、または、被覆されなかった(局所的な予防実験用に)。被覆された移植体は、ラットに移植され、細菌が移植組織に注射され、1週間後、移植組織が除去され、そして、細菌の負荷が決定された。これに代えて、ダクロン移植体は、最初にRIPで被覆され、細菌が注射され、そして、抗生物質が腹腔内経路で投与された(非経口の予防実験用に)。陰性対照として、移植体は、局所または非経口のRIP/抗生物質による予防を行わずに移植され、細菌も注射されなかった。陽性対照として、移植体が移植され、細菌が注入されたが、RIP/抗生物質の予防は行われなかった。RIPに対する陰性対照として、RIPアナログ(YKPETNF)不活性形態が、RIPの替わりに使用された。
1)非汚染陰性対照群に含まれる動物はいずれも、移植体感染の微生物学的証拠を示さなかった。
動物(15体/群)が、皮下ポケットにダクロン移植体の移植を受けた。移植体は、S.epidermids(MSSE、MRSEおよびGISEの試験)で、または、S.aureus(MSSA、MRSAおよびGISAの試験)とともに皮下感染された。ダクロン移植体は、移植前に、RIPを含有する生理食塩水20mg/LまたはRIPを含有しない生理食塩水20mg/Lに浸漬され、一部の動物は、予防治療としてRIP10mg/Lを腹腔内投与された。結果を図16(A)および(B)に要約する。
本発明中において特に重要なことは、RAPおよびTRAPの少なくとも一方の発現および機能の少なくとも一方に影響する活性を有する薬剤の同定である。一般に、対象となる薬剤は、例えばRNAIII合成の活性化に作用するRAPの能力を阻害することによって、RAPまたはTRAP活性を阻害する薬剤である。これらの薬剤は、病原性のブドウ球菌の感染に対する治療法開発にあたって候補薬剤となる。特に重要なものは、ヒト細胞への毒性が低くいか、ブドウ球菌への特異性が高いか、それら両方の特性を有する薬剤のスクリーニング法であり、薬剤の作用に耐性を有する株の選択に対する圧迫がほとんどないか、または、できるだけ少なく、宿主の正常細菌フロラにほとんど影響しないもの(例えば、幅広い効果を有する抗生物質とは明確に区別されるもの)が望ましい。
広範な種々のインビトロでのアッセイが、候補薬剤をスクリーニングするために用いられ、例えばタンパク質−タンパク質結合、タンパク質−DNA結合アッセイ、電気泳動移動度シフトアッセイ、タンパク質を結合させる免疫測定法など、インビトロでの標識結合アッセイを含んでいる。精製された天然由来、組み換え、または合成のRAP、TRAPおよびRIPポリペプチドの少なくとも一つと、合成生産ペプチドまたはRAP、TRAPおよびRIPの断片の少なくとも一つとのうちのいずれか一方、または、両方が、天然タンパク質に、結合、調節(例えば増大または阻害)または天然タンパク質の作用を模倣するリガンドまたは基質を特定するため、種々のスクリーニングアッセイに使用され得る。精製されたタンパク質は、3次元結晶構造の決定にも使用可能であり、この3次元結晶構造は、分子内相互作用、転写調節などのモデリングに使用できる。
上述のアッセイで決定されたような所望の活性を有する薬剤は、非ヒト動物モデルで、さらにブドウ球菌毒性因子生産に対して影響を及ぼす能力や、ブドウ球菌の感染能力に対する影響をさらにスクリーニングされ得る。選択された動物モデルは、含まれる因子の数によって変わり、候補薬剤がスクリーニングされるのに対するブドウ球菌の特有の病原株、候補薬剤が治療薬として使われる最終的な目的の宿主などを含むが、これらに限定されるものではない。スクリーニングアッセイに使用するのに適した動物は、選択されたブドウ球菌種による感染を受ける任意の動物が含まれる。例えば、ブドウ球菌種がS.aureusまたはS.epidermidsである場合、動物モデルは、齧歯類モデルにでき、望ましくはラットまたはマウスモデルである。
本発明は、所定の細菌感染(例えばヒトにおけるS.aureus)による感染を受けるヒトまたは動物の予防または治療の方法を、毒性因子生産を促進するRAPまたはTRAP活性を阻害する薬剤を投与することによって、例えば、RNAIIIのRAPを介する活性や、その後の毒性因子生産を阻害することによって提供する。
本発明は、所定の細菌感染(例えばS.aureusやS.epidermids)による感染を受けるヒトまたは動物に予防接種するためのワクチンを、RAP、TRAP、または、RAPやTRAPの抗原有効部位を、任意でアジュバントを含む場合もある医薬品として許容された担体で投与することにより提供する。免疫反応を消滅させるために適した処方は、当業者には周知である。一般に、宿主は、RAPまたはTRAPなどの抗原に曝露され、このため宿主の免疫系をかく乱し、抗原に対する免疫反応をきたす。抗原に対する免疫反応を増加するため、抗原とともにアジュバントも添加できる。投与されたポリペプチドの量は、宿主の保護のための免疫反応を発現させるために十分な量である。これら適切な量を決定する方法は、当業者は日常的に行っており周知である。例えば、RAP、TRAPおよびその抗原的有効部の少なくとも一つを利用して、ブドウ球菌感染の動物モデルに予防接種できる。これら動物モデルに有効量は、予防接種の有効量を提供するため、他の宿主(例えば家畜、ヒトなど)で推定できる。
4週齢のメスBalb/Cマウス(10体/群)に対し、0、7、21日目に、50μgのrL2(50μg/50μL PBS)を、初回注射時に50μLの完全フロインドアジュバント、第2、3回注射時に不完全フロインドアジュバントとともに皮下注射した。対照動物は、アジュバント/PBSのみを注射した。動物は、35日目に下記のように調製した2×109のSmith Diffuse S.aureus(SD)でチャレンジした。動物は、毎日、死亡率、全般的な健康、病変の進展を観察した。病変の大きさが測定された(面積=0.5n(長さ)(幅))。
抗原に対する抗体の発現は、予防接種された動物は全てで、注射された抗原に対して、抗体力価(>1000)が発現した。対照動物はいずれも、注射された抗原に対する検出可能な抗体値を示さなかった。
結論
rL2を予防接種した動物は、死亡が遅れ、死亡率が50%減少し、病変の大きさが63%減少した。これらの結果、rL2は、S.aureus感染に対する保護をもたらす。チャレンジに使用される細菌の数は、例えば異常に高いことを注目すべきであり、存在する細菌の数がより少ない場合は、感染からの保護レベルはより高くなると予想される。
本発明は、RAP、TRAP、RAP様分子またはTRAP様分子を発現するブドウ球菌またはその他の細菌による生物膜の形成を阻害するために有効な量のRAP阻害剤(例えば、抗RAP抗体、抗TRAP抗体、阻害ペプチド、RIPペプチドまたはRIP誘導体ペプチド)を含有する組成物で表面を被覆された器具を提供する。被覆された器具は、宿主(例えば外科患者、生理中の女性)のブドウ球菌またはその他の細菌の感染または曝露の危険に関連する任意の器具である。
Claims (14)
- RAPが病原性に役割を果たす細菌の種に起因する細菌感染の予防または治療のため、抗体反応を誘発する効果を有する量のRAP型ポリペプチドを被験体に投与する、RAPが病原性おいて役割を果たす細菌の種に起因する細菌感染によって起こる感染に対する予防または治療の方法。
- 前記種がS.epidermidsであることを特徴とする請求項1に記載の方法。
- 前記種がStreptococcus pyogenesであることを特徴とする請求項1に記載の方法。
- 前記種がListeria monocytogenesであることを特徴とする請求項1に記載の方法。
- 前記種がLactococcus lactisであることを特徴とする請求項1に記載の方法。
- 前記種がEnterococcus faecalisであることを特徴とする請求項1に記載の方法。
- 前記種がEscherichia coliであることを特徴とする請求項1に記載の方法。
- 前記種がClostridium acetobutyliciumであることを特徴とする請求項1に記載の方法。
- 前記種がBacillus subtilisであることを特徴とする請求項1に記載の方法。
- 細菌感染に対する治療または予防効果を有する量のRIPまたはRIP誘導体を、RAPが病原性に役割を果たす細菌の種に起因する細菌感染の危険を有する被験体に投与する、病原性おいてRAPが役割を果たす細菌の種に起因する細菌感染による感染に対する予防または治療の方法。
- 前記種がブドウ球菌の全ての種であることを特徴とする請求項1に記載の方法。
- 前記RIPまたはRIP誘導体で器具を被覆する工程をさらに含むことを特徴とする請求項10に記載の方法。
- TRAP型分子が病原性に役割を果たす細菌の種に起因する細菌感染の予防または治療のため、被験体内の抗体反応を誘発する効果を有する量のTRAP型ポリペプチドを被験体に投与する、TRAP型分子が病原性に役割を果たす細菌種に起因する細菌感染に起因する感染に対する予防または治療の方法。
- 前記TRAP型分子がYhgCであることを特徴とする請求項13に記載の方法。
