Abstract
Description
本発明は、クエン酸リアーゼ及び/又はトリカルボキシル酸輸送体の阻害剤を含む組成物及びそのような組成物を用いて癌と診断された患者を治療する方法に関する。
各年に、百万を超える侵入性の癌が米国のみで報告される。治療における改善及び多くの成功にも拘わらず、癌は、死及び数え切れない苦痛の顕著な原因であり続ける。
多数の証拠が示唆することは、癌細胞が、大多数の体細胞のそれらとは明らかに異なる代謝要求性を有することである。癌細胞の代謝要求性の違いは、ある特定の悪性腫瘍、例えば、幼少期の白血病におけるL−アスパラギナーゼにおいて治療上の利点を生かしてきたが、癌細胞の唯一の代謝を生かすための一般化された機構は考案されて(deviced)いない。
癌を標的化するための追加の薬剤及び方法に関する要求が残されている。
本発明は、癌を有する個体(individual)を治療する方法に関し、高率の好気性解糖を有する癌細胞を含む癌のような癌を同定し、そして治療上有効な量のATPクエン酸リアーゼ阻害剤を個体に投与する工程を含む。
本発明は、さらに、癌細胞においてアポトーシスを誘導する方法に関し、細胞においてアポトーシスを誘導するのに有効な量のトリカルボキシル酸輸送体阻害剤を癌細胞に投与する工程を含む。
定義
本明細書にて使用される用語「癌を有すると認定された個体」及び「癌患者」は、交換可能に使用され、そして癌を有すると診断された個体を意味する。癌を有する個体を認定するためのよく知られた手段は多数存在する。いくつかの態様において、癌の診断は、PETイメージングを用いて作成されるか又は確認される。発明のいくつかの態様は、癌を有する個体を認定する工程を含む。
本発明は、多くのヒトの癌が高率の好気性解糖を示すこと、及びこの高率の好気性解糖が生長期の(vegetative)細胞の生存に通常は必要ない特定の代謝酵素に対する癌細胞の増加した依存性を誘導するという観察から生まれる。そのような酵素に対する増加した依存性が、そのような酵素の阻害剤による処理に対して癌細胞を感受性に誘導するか否かを決定する探索が企てられた。それらの環境からのグルコースの過剰な取り込みのために高率の好気性解糖を示す癌細胞の一つの鍵となる特徴は、当該細胞が莫大な量のピルビン酸(pyruvate)を生産することである。そのようなピルビン酸のミトコンドリアマトリックスへの輸送及びピルビン酸デヒドロゲナーゼ複合体によるそのさらなる分解は、トリカルボキシル酸サイクル(TCA)への増加したアセチル−CoAの侵入をもたらす。TCAサイクルにより生産されたNADHが酸化的リン酸化を通した電子の輸送及び細胞性ATPの生産を維持するのに必要な量を超えるなら、連続的なミトコンドリアの高分極及び電子輸送連鎖(electron transport chain)からの増加した酸素種(ROS)の生産が結果として起こり得る。ミトコンドリアの高分極及び電子輸送連鎖からの増加したROS生産は、最終的に、細胞のアポトーシスをもたらす(Vander Heiden,M.G.,et al.Cell,91:627−637,1997;引用により本明細書に編入される)。
発明のいくつかの態様は、癌を治療するためのATPクエン酸リアーゼ阻害剤の使用を含む。
本発明の態様は、高率の好気性解糖を有するとして同定された癌を有する個体を治療するのに特に有用である。いくつかの態様において、癌を有する個体を治療する方法は、高率の好気性解糖を有するとして癌を最初に同定し、そして次に治療上有効な量のATPクエン酸リアーゼ阻害剤をそのような個体に投与する工程を含む。いくつかの好ましい態様において、高率の好気性解糖を有するという癌の同定は、PETイメージング、好ましくは18フルオロ−デオキシグルコースにより実施される。いくつかの態様において、ATPクエン酸リアーゼ阻害剤は、1mM未満の濃度においてインビトロにおいてアポトーシスアッセイにて50%より多い細胞にアポトーシスを誘導するのに有効である。好ましい態様において、ATPクエン酸リアーゼ阻害剤は、50μM未満の濃度においてインビトロのアポトーシスアッセイにて50%より多い細胞にアポトーシスを誘導するのに有効である。
当業界においては、ATPクエン酸リアーゼ阻害剤の例が多数ある。Gribbleらに対して1995年9月5日に発行された米国特許第5,447,954号、Chengらに対して2002年7月2日に発行された米国特許第6,414,002号、Chengらにより2003年5月8日に公開された米国出願公開番号20030087935 A1、Chengらにより2003年4月10日に公開された米国出願公開番号20030069275 A1、及びBarrow et al.,”Antimycins,Inhibitors of ATP−Citrate Lyase,from a Streptomyces sp.”,Journal of Antibiotics,vol.50,No.9,pp.729(1977)の各々は、引用により本明細書に各々編入され、ATPクエン酸リアーゼ阻害剤である化合物を開示する。好ましい態様において、ATPクエン酸リアーゼ阻害剤は、米国特許第5,447,954号において構造Iとして示された式により定義される構造を有する化合物からなる群から選択され、好ましくは、そこに開示されて特別に記載された構造の一つを有する化合物、好ましくはそこに示された実施例中の特別の化合物、好ましくはその中の図4において示された構造を有する化合物である。他の公知の阻害剤は、(−)ヒドロキシクエン酸、(R,S)−S−(3,4−ジカルボキシ−3−ヒドロキシ−3−メチル−ブチル)−CoA、及びS−カルボキシメチル−CoAを含む。
発明のいくつかの態様は、癌を治療するためのトリカルボキシル酸輸送体阻害剤の使用を含む。
本発明のいくつかの態様において、癌を有するとして同定された個体を治療する方法は、治療上有効な量のトリカルボキシル酸輸送体阻害剤をそのような個体に投与することを含む。いくつかの態様において、本発明の方法は、内部で合成される脂肪酸に依存しない癌細胞を有する癌を有する患者を治療するのに特に有用である。いくつかの好ましい態様において、トリカルボキシル酸輸送体阻害剤の投与前に、癌が高率の好気性解糖により特徴付けされる癌であることを確認する。そうすることの好ましい方法は、PETイメージングであり、好ましくは、18フルオロ−デオキシグルコースを用いる。
当業界においては、トリカルボキシル酸輸送体阻害剤の例が多数ある。そのような例は、1,2,3−ベンゼントリカルボキシル酸、イソクエン酸、マレイン酸、ホスホエノールピルビン酸、n−ブチルマロン酸、スルフィドリル試薬、ジエチルピロカルボン酸、2,3−ブタンジオン、フェニルグリオキサル、ピリドキサル、5−ホスフェートジカルボキシル酸、琥珀酸、マレイン酸、オキサロ酢酸、トリカルボキシル酸、イソクエン酸、トリカルバリル酸、及びパルミトイル−CoAを含む。
アポトーシスは、当業者にはよく知られた多くの手法により検出できる。アポトーシスを検出する方法の例は、限定ではないが、TUNELアッセイ、カスパーゼ活性を測定すること、アネキシン−V染色、等を含む。アポトーシス活性は、実施例1に記載されたアッセイにおいて示されたとおりに測定される。
いくつかの態様においては、ATPクエン酸リアーゼ阻害剤及び/又はトリカルボキシル酸輸送体阻害剤を他の治療剤及び/又は放射線治療を含む治療養生法の一部と同時に投与することができる。
いくつかの好ましい態様において、発明による薬学組成物による治療は、外科医の介入が先行する。
薬学組成物は、当業者により、選択された投与様式に依存して選択された組成物を用いて製剤化してよい。適切な薬学上の担体は、Remington’s Pharmaceutical Science,A.Osolの最近の版に記載されており、この分野の標準の参照テキストである。
本発明によれば、癌を有すると同定された個体において癌を治療する方法は、そのような個体に、個体において腫瘍細胞内でアポトーシスを誘導するのに十分な量のATPクエン酸リアーゼ阻害剤又はトリカルボキシル酸輸送体阻害剤を送達することにより実施される。そうすることにより、腫瘍細胞は、アポトーシスを経て、腫瘍自体がサイズを減少させるか又は完全に除去される。即ち、アポトーシス誘導投薬においてATPクエン酸リアーゼ阻害剤又はトリカルボキシル酸輸送体阻害剤の投与を含まない治療に比較して、患者の生存が伸びるか、及び/又は生活の質が改善される。本発明は、癌細胞内でアポトーシスを誘導する方法を提供し、アポトーシスを誘導するのに有効な量にて、クエン酸リアーゼ阻害剤又はトリカルボキシル酸輸送体阻害剤をそのような細胞に送達する工程を含む。
本発明は、さらに、ATPクエン酸リアーゼ又はトリカルボキシル酸輸送体の発現を阻害することにより癌を治療する方法に関する。発現の阻害は、アンチセンス、RNAi技術又はリボソームを用いて、癌の中のATPクエン酸リアーゼ又はトリカルボキシル酸輸送体の生産を阻害することができる。当業者は、本明細書に記載されたとおり、発明の様々な態様を実施するために、本明細書に記載された阻害剤の代わりに治療上有効な量のそのような化合物を作成して使用することができる。
ヒトに加えて、本発明の方法は、癌を罹患した他の種の動物を治療するために使用してよく、例えば、イヌ、ネコ、ウマ、ブタ、ウシ及びヒツジ上を含む。
実施例1:インビトロアポトーシスアッセイ
処理されたか又は未処理の(0.5−1x106細胞/mlの濃度にて)細胞500μlをFACSチューブに取り出し、50μlの10XアネキシンV結合バッファー(BDバイオサイエンス ファーミンゲン、サンディエゴ、CA)を各チューブに加えた。アネキシンV−FITC(BDバイオサイエンス ファーミンゲン、サンディエゴ、CA)により、4μlのアネキシンV−FITC及びヨウ化プロピジウム(Molecular Probes,Eugene,OR)を最終濃度3μg/ml細胞にて細胞を染色。当該細胞は、好ましくは、造血IL−3依存性細胞系、例えば、異なるトランス遺伝子(Bcl−xL,myrAkt)又はベクター対照プラスミドをトランスフェクトされたIL−3依存性FL5.12 pro−B細胞及びBax−/−Bak−/−ノックアウトマウスの骨髄から単離されたIL−3依存性細胞である。37℃において30分間インキュベートする。初期アポトーシス細胞はアネキシンVに陽性であり、ヨウ化プロピジウムには陰性であるが、後期アポトーシス細胞はアネキシンV及びヨウ化プロピジウムの両方に陽性である。
ATPクエン酸活性は転写/翻訳及び翻訳後の機構の両方により増殖細胞内でアップ制御される
細胞増殖の有糸分裂促進物質の刺激に応答したATPクエン酸リアーゼの可能な誘導を調査するために、細胞抽出物中のATPクエン酸リアーゼ(ACL)活性を定量するアッセイを開発した。陽性対照として、細胞増殖ノックアウト誘導のためにIL−3に依存する細胞系の安定なトランスフェクト体も生成した。野生型細胞及びATPクエン酸リアーゼをトランスフェクトされた細胞からのサイトゾル抽出物を、NADHによるオキサロ酢酸のマレイン酸デヒドロゲナーゼにより触媒された還元により測定された、106細胞あたりのATPクエン酸リアーゼ活性のレベルを測定することにより、分析した。NADHの消費は、340nmにおいて反応を監視することにより測定した。5mMのATP及び300μMのCoAの存在下で100μMにおいてクエン酸を提供することによりアッセイを走らせた。陽性対照として、ACL−トランスフェクトされた細胞はATPクエン酸リアーゼ活性において3.5倍の増加を示したことから、アッセイの有効性が証明された(図2)。成長因子IL−3を培養物から取り出したとき、停止(withdrawal)の12時間後に細胞から細胞周期停止を誘導すると、ACL活性の降下がネクター対照とACL−トランスフェクト細胞の両方において観察された。合わせると、これらは、ACL活性が増殖細胞中で誘導されること、及びACLをトランスフェクトされた細胞中のACL活性は翻訳後に有糸分裂活性化細胞に関連した事象の結果として修飾できることを示唆する。
ヒドロキシクエン酸の効果がATPクエン酸リアーゼ活性を阻害することにおいてその証明された活性に相当し、ACL活性及び細胞増殖及びアポトーシスの阻害を誘導するのに必要な相対的に高い濃度によりもたらされる混交の(promiscuous)効果ではないことを調査するため、我々は、ATPクエン酸リアーゼ阻害剤として作用すると最近報告されている一連の化合物による二次的な結果を確認した(図4)。ヒドロキシクエン酸とは対照的に、これらの化合物は、細胞抽出物中でATPクエン酸リアーゼ活性のマイクロモラー阻害剤として作用する。3−カルボキシ−11−(2,4−ジクロロフェニル)−3,5−ジヒドロウンデカン酸(構造に関しては図4を参照)を有糸分裂促進物質で刺激されて約3μMのIC50を有する細胞抽出物に加えたとき、及び細胞を細胞浸透性γ−ラクトン誘導体で処理したとき、約10μMの濃度において細胞増殖が阻害され、そして細胞増殖と細胞生存性の両方が30μMの濃度において阻害された(図6)。固定された造血細胞内の細胞増殖の阻害の再現性を、さらに、独立の細胞クローンを調査することにより調査したところ、細胞増殖が約50%各クローンにおいて10から30μMの間の投薬量にて阻害されたことが証明された(図7)。30μMの濃度の細胞浸透性化合物による処理は、有糸分裂促進物質により刺激された細胞及び増殖形質転換された細胞の培養物のG1抑止に関連しており(図8)、そしてアネキシン−V陽性の素早い誘導(図9)はアネキシンの誘導を示唆する。
我々の研究室における以前の研究は、Aktの活性化された変異又は腫瘍サプレッサー機能、PTENの損失の何れかにより形質転換された細胞がそれらの解糖率の劇的なアップ制御を示すことを証明した(Plas et al.2001;Frauwirth et al.2002)。これは、それらが侵入性で転移性の表現型に進むように多くのヒトの悪性腫瘍により証明される特徴であり、そしてWarburgらにより1929年に最初に記載されて、フルオロデオキシグルコースを用いてPETスキャニングの臨床使用の増加により最近広範囲に確認された。Aktによる形質転換がATPクエン酸リアーゼ活性をさらに増加させるか否かを決定するため、我々は、IL−3依存性細胞系の安定なトランスフェクト体、FL5.12を作成し、Aktの構成的に活性化されたバージョンがN末端におけるミリストイル化の添加により生成された。我々は、IL−3存在下で成長させた場合のAkt−形質転換された細胞が、ACL−トランスフェクトされた細胞内で観察されたよりもさらに高いレベルのACL活性を有したことを見いだした(図10)。これは、ACLがAktリン酸化の基質であるということ(Berwick et al.2002)及びAktがACL活性の翻訳後活性化のための一つの分子機構であるかもしれないという他者の最近の報告と一致する。さらに、ACL−及びBcl−xL−トランスフェクトされたクローンとは対照的に、ミリストイル化されたAktによりトランスフェクトされたクローンは、細胞周期からの停止を通常は伴うACL活性の下降に対して耐性である。Akt形質転換された細胞は、非増殖状態においてさえ、高率の解糖を維持する。これが増殖性応答の不在下でさえもACL阻害にそれらを感受性にし続けるか否かを決定するために、我々は、有糸分裂促進物質IL−3の存在又は不在下で、ミリストイル化されたAkt細胞のACL阻害に体する感受性を調査した(図11)。ミリストイル化されたAktをトランスフェクトされた細胞は、細胞がIL−3存在下で成長したか又は細胞周期のG0/G1期がIL−3に対する応答として停止したか否かについて細胞生存性により測定したところ、ACL阻害に対するそれらのIC50に差異は示さない。対照的に、ACL活性を増加させないし且つ成長因子停止(withdrawal)に際してACL活性の下降を示す癌遺伝子Bcl−xLをトランスフェクトされた細胞は、有糸分裂促進物質IL−3の存在又は不在下でACLの阻害に際して細胞死に対する感受性において多大な差異を示す。ATPクエン酸リアーゼのγ−ラクトン阻害剤30μMにおいて、有糸分裂促進物質により誘導された細胞増殖のほぼ完全な阻害があり、そしてアポトーシス性細胞死の誘導がある。対照的に、非成長細胞中の30μMの同じ化合物は相対的に非毒性である。これは、解糖を活性化する癌遺伝子の添加による細胞の癌遺伝子形質転換が、形質転換された細胞のACL活性化に対してのさらに増強された感受性を導くことを示唆する。さらに、活性化に対するこの感受性は細胞周期に非依存性である。
初期ヒト腫瘍由来の細胞系における我々の結果を確認するため、我々は、患者から単離された神経膠芽細胞腫の細胞系を分析した(図12)。2つのそのような神経膠芽細胞腫の細胞系は、LN18とLN229である。これらの2つの腫瘍は培養物及びインビボにおいて類似の成長速度を示すが、LN18はAktの構成的に活性な形態を有するらしく、一方LN229は有糸分裂促進物質の刺激に応答してのみAktを活性化する。血清の不在下で成長させると、増殖する細胞培養物の両方が、ATPクエン酸リアーゼの阻害剤に応答して細胞蓄積の投薬量依存性抑止と細胞生存性の阻害を呈する。Akt形質転換された細胞系に関しては、活性化されたAktを有する神経膠芽細胞腫LN18が、増加した感受性を示した。これがAkt活性に相関するか否かを決定するため、LN229神経膠芽細胞腫の細胞を構成的形態のAktでトランスフェクトし、ATPクエン酸リアーゼの阻害に対するそれらの感受性に関して再度試験した。ミリストイル化されたAktをトランスフェクトされたLN229細胞は、ベクターのみをトランスフェクトされた細胞に比較して、ATPクエン酸リアーゼ阻害に応答してアポトーシスを経ることに対する感度の増加を示した。これらのデータは、ヒトの腫瘍がATPクエン酸リアーゼに応答して細胞成長抑止及びアポトーシスを経ることに感受性を示すこと、及びこの感受性がそのような細胞の好気性解糖を刺激する癌遺伝子によりさらに増強され得ることを確認する。
ヒトにおける細胞の大多数は生長期であるか又は非増殖状態である。癌治療が治療ウインドーを有するためには、ドラッグが非増殖細胞に対するよりも増殖クローン又は形質転換された細胞に対して高い効果を有することが重要である。これらの問題に、インビトロ培養システムにおいて接近するために、我々は、前アポトーシス(proapoptotic)分子であるBax及びBakの枯渇によりアポトーシスを欠損させた有糸分裂促進物質依存性細胞系の有用性を利用した。IL−3の存在下では、これらの細胞系は、培養において連続して生長するが、IL−3の剥奪に際して、当該細胞は細胞周期から撤退する(withdraw)が、数週は生長期にて生存する。細胞増殖がATPクエン酸リアーゼ阻害に対する細胞の感受性を増加させるか否かを決定するために、我々は、IL−3の存在下で生長させるときに、Bax/Bak−欠損、IL−3依存性細胞系を、ATPクエン酸リアーゼ阻害剤で処理した(図13)。IL−3に応答した細胞生長はACL阻害に対する細胞の感受性を劇的に増加させた。これらの細胞は、我々が、細胞生長に対するATPクエン酸リアーゼ阻害の効果を二次的に試験することを可能にもさせた。休止しているBax/Bak−欠損細胞をIL−3で処理することにより、それらの生長及び細胞周期への再侵入を誘導した。我々は、投薬量依存性様式のATPクエン酸リアーゼ阻害剤が投薬量依存性様式において有糸分裂促進物質誘導性細胞生長を阻害したことを証明することができた。30μMのSB20499において、これらの細胞系の有糸分裂促進物質誘導性細胞の生長をほぼ完全に阻害した(図14)。さらに、我々は、それらの生長期の対応物に比較したときに、有糸分裂促進物質刺激が、ATPクエン酸リアーゼ阻害剤の効果に対して静止期の細胞をより感受性にしたことを証明できた。有糸分裂促進物質により処理された生長期細胞は、細胞の生存により測定したところ、ATPクエン酸リアーゼ阻害剤の処理に対して、選択的に、より感受性になった(図15)。合わせると、これらのデータが示唆するのは、形質転換された細胞が高率の好気性解糖を呈し、そしてアポトーシス性細胞死の誘導の細胞増殖の阻害により明らかにされた、ATPクエン酸リアーゼの阻害に対して高い感受性を示すことである。さらに、非増殖性細胞は、そのような阻害剤の効果に対して相対的な耐性を示し、そして非形質転換細胞は中間の感受性を示すことから、癌の治療においてATPクエン酸リアーゼ阻害剤の効果に関する治療ウインドーが存在することを示唆する。
ATPクエン酸リアーゼ阻害剤の作用の分子上の根拠をさらに決定するために、追加の研究に着手した。我々は、Akt−形質転換された細胞がそれらの増殖性及び細胞生存性を維持するために高率の解糖代謝に依存するようになったことを以前に証明した。ATPクエン酸リアーゼ阻害の一つの予測される効果は、サイトゾル中のクエン酸の蓄積(build−up)であり、解糖経路の負のアロステリック制御因子として作用し得る。ATPクエン酸リアーゼ阻害剤による処理がAkt−形質転換された細胞の高率解糖の阻害を導くか否かを試験するために、我々は、ACL阻害剤による簡単な処理が投薬量依存性様式にてAkt−形質転換された細胞の解糖率を抑圧できるか否かを試験した(図16)。高率の解糖を示す形質転換細胞においては、サイトゾル中のATPクエン酸リアーゼの阻害及びクエン酸の蓄積が、これらの細胞に高率の解糖を通してそれらのバイオエネルギーを維持させないかもしれず、即ち、選択的毒性をもたらさないかもしれない。
上記のデータは、癌細胞が、解糖経路の制御、ミトコンドリアの生理機能、及び細胞成長においてATPクエン酸リアーゼ活性の生成物に関する要求に対しての、機構に基づく効果を通して、ATPクエン酸リアーゼ阻害剤に対して選択的感受性を示すことを示唆する。さらに、ATPクエン酸リアーゼ阻害に対する感受性は増殖する細胞の方が非増殖細胞よりもはるかに高く、そして好気性解糖の誘導を導く癌遺伝子形質転換を経た細胞において劇的に増強される。我々は、よって、癌の治療におけるATPクエン酸リアーゼ阻害剤の用途を提案し、そしてAktの活性化又はPTENの枯渇を通して形質転換された細胞、PETイメージングにより測定された好気性解糖の増強率を示す細胞、及び転移部位において成長する細胞においてそれが特に有用であることを示唆する。

Claims (57)
- 癌を有する個体を治療する方法であって、上記の癌を、高率の好気性解糖を有する癌細胞を含む癌細胞として同定し、そして次に、治療上有効な量のATPクエン酸リアーゼ阻害剤を上記個体に投与することを含む方法。
- PETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項1記載の方法。
- 18フルオロ−デオキシグルコースを用いてPETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項1記載の方法。
- ATPクエン酸リアーゼ阻害剤が1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、請求項1乃至3の何れか1項記載の方法。
- ATPクエン酸リアーゼ阻害剤が0.1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、請求項1乃至3の何れか1項記載の方法。
- ATPクエン酸リアーゼ阻害剤が50μMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、請求項1乃至3の何れか1項記載の方法。
- 癌細胞が内部で合成された脂肪酸に依存しない癌細胞を含む、請求項1乃至6の何れか1項記載の方法。
- ATPクエン酸リアーゼ阻害剤が米国特許第5,447,954号に記載された式又は実施例の一つにより定義される構造を有する化合物及び(−)ヒドロキシクエン酸からなる群から選択される、請求項1乃至7の何れか1項記載の方法。
- ATPクエン酸リアーゼ阻害剤が図4に示すSB−204990である、請求項1乃至7の何れか1項記載の方法。
- 癌を有すると同定された個体を治療する方法であって、上記癌は内部で合成された脂肪酸に依存しない細胞を含み、そして上記方法は治療上有効な量のATPクエン酸リアーゼ阻害剤を個体に投与する工程を含む方法。
- 上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項10記載の方法。
- PETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項11記載の方法。
- 18フルオロ−デオキシグルコースを用いてPETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項12記載の方法。
- ATPクエン酸リアーゼ阻害剤を別の抗癌化合物の投与と共に投与する、請求項10乃至13の何れか1項記載の方法。
- ATPクエン酸リアーゼ阻害剤を抗癌放射線治療と共に投与する、請求項10乃至13の何れか1項記載の方法。
- 癌を有すると同定された個体を治療する方法であって、治療上有効な量のATPクエン酸リアーゼ阻害剤を個体に投与することを含むが、但し、ATPクエン酸リアーゼ阻害剤は、1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、方法。
- ATPクエン酸リアーゼ阻害剤が0.1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、請求項16記載の方法。

- ATPクエン酸リアーゼ阻害剤を抗癌放射線治療と共に投与する、請求項16乃至24の何れか1項記載の方法。
- 癌細胞内でアポトーシスを誘導する方法であって、当該癌細胞は内部で合成された脂肪酸に依存せず、そして上記癌細胞中でアポトーシスを誘導するのに有効な量のATPクエン酸リアーゼ阻害剤を上記癌細胞に送達する工程を含む、方法。
- 癌細胞においてアポトーシスを誘導する方法であって、当該癌細胞においてアポトーシスを誘導するのに有効な量のATPクエン酸リアーゼ阻害剤を当該癌細胞に送達する工程を含むが、但し、ATPクエン酸リアーゼ阻害剤は、1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、方法。
- ATPクエン酸リアーゼ阻害剤が0.1mMより低い濃度においてインビトロアポトーシスアッセイにて50%を超える細胞においてアポトーシスを誘導するのに有効である、請求項28記載の方法。

- 上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項34乃至36の何れか1項記載の方法。
- PETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項37記載の方法。

- トリカルボキシル酸輸送体阻害剤を別の抗癌化合物の投与と共に投与する、請求項34乃至39の何れか1項記載の方法。
- トリカルボキシル酸輸送体阻害剤を抗癌放射線治療と共に投与する、請求項34乃至39の何れか1項記載の方法。
- 癌細胞においてアポトーシスを誘導する方法であって、当該細胞においてアポトーシスを誘導するのに有効な量のトリカルボキシル酸輸送体阻害剤を当該癌細胞に送達する工程を含む方法。
- 癌細胞が内部で合成された脂肪酸に依存しない癌細胞を含む、請求項42記載の方法。
- トリカルボキシル酸輸送体阻害剤が、1,2,3−ベンゼントリカルボキシル酸、イソクエン酸、マレイン酸、ホスホエノールピルビン酸、n−ブチルマロン酸、スルフィドリル試薬、ジエチルピロカルボン酸、2,3−ブタンジオン、フェニルグリオキサル、ピリドキサル、5−ホスフェートジカルボキシル酸、琥珀酸、マレイン酸、オキサロ酢酸、トリカルボキシル酸、イソクエン酸、トリカルバリル酸、及びパルミトイル−CoAからなる群から選択される、請求項42又は43記載の方法。
- 癌を有する個体を治療する方法であって、高率の好気性解糖を有する癌細胞を含む癌として上記の癌を同定し、そして次に治療上有効な量のトリカルボキシル酸輸送体阻害剤を上記個体に投与する工程を含む方法。
- PETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項45記載の方法。
- 18フルオロ−デオキシグルコースを用いてPETイメージングにより上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項45記載の方法。

- ATPクエン酸リアーゼ阻害剤を抗癌放射線治療と共に投与する、請求項45乃至50の何れか1項記載の方法。
- ATPクエン酸リアーゼ又はトリカルボキシル酸輸送体の発現を阻害する化合物を治療上有効な量、個体に投与することを含む、癌を有すると同定された個体を治療する方法。
- 上記の癌が高率の好気性解糖を有する癌細胞を含む癌であると決定される、請求項54記載の方法。
- 抗癌活性を有する化合物を同定する方法であって、ATPクエン酸リアーゼ又はトリカルボキシル酸輸送体の阻害剤として試験化合物を同定し、そして当該化合物がアポトーシスを誘導するか否かを決定するアポトーシスアッセイを実施する工程を含むが、その際、ATPクエン酸リアーゼ又はトリカルボキシル酸輸送体の阻害剤であり且つアポトーシスを誘導する試験化合物が抗癌活性を有する化合物である、方法。
