Abstract
【解決手段】 二重結合を少なくとも二つ以上有する不飽和脂肪酸を含有する豆乳に、共役化処理能を有するビフィドバクテリウム属細菌を接種して培養する。具体的なビフィドバクテリウム属細菌としては、ビフィドバクテリウム・ブレーべ(B. breve)、ビフィドバクテリウム・ビフィダム(B. bifidum)、ビフィドバクテリウム・インファンティス(B. infantis)、ビフィドバクテリウム・シュードカテヌラータム(B. psedocatenulatum)から選ばれる。

実施例1:共役リノール酸の製造方法1
100mMリン酸カリウム緩衝液(pH6.5)1mL中にリノール酸50mgとウシ血清アルブミン(BSA)10mgを溶解し、あらかじめリノール酸−BSA−複合体溶液を調製した。このリノール酸−BSA−複合体溶液200μLをGAM(GAM BROTH、日水製薬(株)社製)培地15mLに添加し、ビフィドバクテリウム属細菌(3種)をそれぞれ接種して35℃、120rpm、48時間振塗培養し、ビフィドバクテリウム属細菌培養液を調製した。
100mMリン酸カリウム緩衝液(pH6.5)1mL中にリノール酸50mgとBSA10mgを溶解し、あらかじめリノール酸−BSA−複合体溶液を調製した。このリノール酸−BSA−複合体溶液200μLをGAM(GAM BROTH、日水製薬(株)社製)培地15mLに添加し、ビフィドバクテリウム・ブレーベ(Bifidobacterium breve) YIT10001を接種して35℃、120rpmで対数増殖期まで培養し、Bifidobacterium breve YIT10001培養液を調製した。
豆乳にリノール酸を15mg/mL添加し、ハンドホモゲナイザーにて均質化した。均質化した各培地をネジ口試験管に5mLずつ分注し、オートクレーブ滅菌(105℃、15分)した。種菌培養用豆乳(誘導剤としてリノール酸を2.5mg/mL添加)で対数増殖期まで培養したB. breve YIT10001 を種菌とし、5mLずつ分注した培地に100μL(生菌数4.3×107CFU/mL )接種後、気相を窒素置換後37℃で72時問静置培養した。得られた培養液の脂質を抽出後、メチルエステル化しGCにて脂肪酸を分析した。
Claims (6)
- 二重結合を少なくとも二つ以上有する不飽和脂肪酸を含有する豆乳に、共役化処理能を有するビフィドバクテリウム属細菌を接種して培養することを特徴とする共役脂肪酸の製造方法。
- ビフィドバクテリウム属細菌が、ビフィドバクテリウム・ブレーべ(Bifidobacterium breve)、ビフィドバクテリウム・ビフィダム(Bifidobacterium bifidum)、ビフィドバクテリウム・インファンティス(Bifidobacterium infantis)、ビフィドバクテリウム・シュードカテヌラータム(Bifidobacterium psedocatenulatum)から選ばれるl種以上であることを特徴とする請求項1に記載の共役脂肪酸の製造方法。
- 二重結合を少なくとも二つ以上有する不飽和脂肪酸を添加したGAM培地で培養して得られたビフィドバクテリウム属細菌を、前記豆乳に接種することを特徴とする請求項1又は2に記載の共役脂肪酸の製造方法。
- ビフィドバクテリウム属細菌を前記豆乳に1×105〜1×1010CFU/mL接種することを特徴とする請求項1〜3に記載の共役脂肪酸の製造方法。
- 二重結合を少なくとも二つ以上有する不飽和脂肪酸として、リノール酸を使用することを特徴とする請求項1〜4の何れか1項に記載の共役脂肪酸の製造方法。
- 請求項1〜5の何れか1項に記載の方法により得られた共役脂肪酸を含有する飲食品。
