Abstract
Description
<好ましい形態の記述>
本発明は、電解質と、電解質中に懸濁した対象のコンジュゲート分子の存在下で組織に電場を印加することを含んでなる組織の中にコンジュゲート分子を導入する方法に関する。コンジュゲート分子は、相補的結合部位の近傍に運ばれてきた場合にこの部位に結合する相補的結合部分を有するいかなる分子であってもよい。相補性決定領域を有する抗体と標的DNAへのマッチング配列を有するDNAオリゴマーがコンジュゲート分子の2つの例である。対象のコンジュゲート分子は、適正なpHの電解質の水溶液に溶解した場合すべて荷電している。この正味電荷は電場による電解質溶液中でのこれらの移動を促進する。組織は、サイトスピン(cytospin)またはThin Prepsなどの慣用の方法により作製された組織切片と無傷の細胞の両方を含む。
次の実験を行って、抗体を組織の中に電気泳動で導入することができるかどうかを決定した。組織を親水性ポリテトラフルオロエチレン(PTFE)膜(TEFLON(登録商標)、配管用テープ)に付着させて、ゲル中での組織の操作と位置付けが可能なようにし、次に以降の電気泳動のためにアガロースゲル中に埋め込んだ。
結論:
1.電気泳動は、扁桃組織の中に、そしてPTFE膜上に載せた扁桃組織の中に抗CD34抗体を導入する能力がある。
2.この抗体はこれらの条件下でその抗原に結合する。
3.組織を通り抜けた抗体が多いほど、染色は濃い。
4.バックグラウンド着色は許容できる。
Claims (13)
- 電解質と、電解質中に懸濁した対象のコンジュゲート分子の存在下で組織に電場を印加することを含んでなる組織の中にコンジュゲート分子を導入する方法。
- 前記コンジュゲート分子が相補的結合部分を有する任意の分子である請求項1に記載の方法。
- 前記コンジュゲート分子が抗体とポリヌクレオチド分子からなる群から選択される請求項1に記載の方法。
- 前記組織が固体組織試料を含んでなる請求項1に記載の方法。
- 前記組織が細胞調製物を含んでなる請求項1に記載の方法。
- 前記組織に前記電場を印加する段階が交流を使用することを含んでなる請求項1に記載の方法。
- 前記組織に前記電場を印加する段階が直流を使用することを含んでなる請求項1に記載の方法。
- 前記組織に前記電場を印加する段階が前記組織の面に直交して前記電場の向きを決めることを含んでなる請求項1に記載の方法。
- 前記コンジュゲート分子が検出可能に標識化されている請求項3に記載の方法。
- 前記標識がハプテン、発蛍光団、放射性標識および発色団からなる群から選択される請求項9に記載の方法。
- (a)前記組織を位置決めし、保持するように改造された試料表面を有する第1の電極;
(b)前記第1の電極から間隔のとられた第2の電極であって、前記試料表面と前記第2の電極の間のギャップを規定する第2の電極;
(c)組織試料の両側に配設された電解質溶液を保持するのに適する貯槽;および
(d)前記試料表面に電流を印加することにより、それに応じてコンジュゲート分子を前記組織の中に導入するのに充分な電場を形成するための手段
を含んでなる組織試料の中にコンジュゲート分子を電気泳動的に導入するための装置。 - (a)前記組織を位置決めし、保持するように改造された試料表面を有する第1の電極;
(b)前記第1の電極から間隔のとられた第2の電極であって、前記試料表面と前記第2の電極の間に電解質流体のメニスカスを支持する能力のあるギャップを規定する第2の電極;および
(c)前記試料表面に電流を印加することにより、それによってそれに応じてコンジュゲート分子を前記組織の中に導入するのに充分な電場を形成するための手段
を含んでなる組織試料の中にコンジュゲート分子を電気泳動的に導入するための装置。 - (a)可動型の電気的に絶縁されたブロックであって、その上に位置決めされた反対極性の少なくとも2種の電極を有し、可動型であることにより前記組織試料の中に電場の方向を決める可動型の電気的に絶縁されたブロック;
(b)前記組織を位置決めおよび保持するように改造された試料表面であって、前記ブロックから間隔がとられていることによって前記試料表面と前記ブロックの間に電解質流体のメニスカスを保持する能力のあるギャップを規定する試料表面;および
(c)前記試料表面に電流を印加することにより、それに応じてコンジュゲート分子を前記組織の中に導入するのに充分な電場を形成するための手段
を含んでなる組織試料の中にコンジュゲート分子を電気泳動的に導入するための装置。
