Description
本発明は、ヒトPKCθ触媒ドメインの構造座標を提供する。用語「構造座標」または「原子座標」とは、PKCθ触媒ドメインを含む結晶の原子(散乱中心)によりX線の単色光線で得られた回析パターンに関する数式(フーリエ変換)から算出される数値座標を意味する。回析データを用いて、結晶単位の繰り返しの電子密度マップを計算する。電子密度マップを用いて、結晶の単位格子内の個々の原子の位置を定める。
PKCθに対する化学物質の可能性のある阻害効果または結合効果を、コンピュータ・モデリング技術の使用により、その実際の合成の前に分析し試験することが可能である。与えられた化学物質の理論的構造が、それとPKCθ間の不十分な相互作用および結合を示唆する場合、その化学物質の合成および試験の必要性が除かれる。しかしながら、コンピュータ・モデリングが強い相互作用を示す場合、その後、前記分子を合成し、そしてPKCθと結合するその能力について試験して良い。故に、お金と時間をかけた無効な化合物の合成を、避けることができる。
用語「分子置換」とは、未知の結晶の観察された回析パターンを最も良く説明するために、構造座標が未知である結晶の予備的構造モデルを、構造座標が既知である分子、例えば、未知の結晶の単位格子内のPKCθ触媒ドメイン座標の方向付けおよび位置決めにより作製することを含む、方法を意味する。その後、位相をこのモデルから計算することができ、観察された振幅と合わせ、座標が未知である構造の近似したフーリエ合成を得る。これは次に、最終的に正確な構造を得るために、いくつかの精緻の形態のいずれかに付し得る。本発明により供される構造座標を用いて、分子置換を、共複合体、未知のリガンド、変異体または相同体の結晶、またはPKCθの異なる結晶形態の構造座標を決定するために用いることができる。さらに、特許請求の範囲の結晶およびその座標を、PKCθと結合する化学物質の構造座標を決定するために用いることができる。
実施例4:バキュロウイルス細胞および昆虫細胞におけるPKCθの発現
バキュロウイルスおよび昆虫細胞培養物の調製方法は、当技術分野で公知である(O’Reilly et al.,1994)。大部分の組換えバキュロウイルスを、CLONTECHのBacPAK6トランスフェクション系、および前に記載のPKCθ構築物を有する伝達ベクターを用いたSf9細胞のトランスフェクションにより作製する。PKCθ−FLa、PKCθ−M2aおよびPKCθ−M1aの発現用の組換えバキュロウイルスの作製を、バクミド(bacmid)により行う。すべてのバキュロウイルス株をプラーク−クローン化し、そして25cm2−TCフラスコ中10mlのTC100培地(Invitrogen)+10%FCS(Invitrogen)にて27℃で培養した付着Sf9細胞を、2回目の増殖中に増殖させる。3回目の増殖中に、浮遊Sf9細胞を、500ml三角フラスコ中100mlのTC100培地+10%FCS+0.1%プルロニックF−68(Invitrogen)と一緒に90rpmで振とうしながら27℃で培養し、プラーク分析により、結果として、0.3−2.0×108pfu/mlの範囲の濃度で組換えバキュロウイルス懸濁液が生じる。
