本出願は、米国特許法第119条e項に基づいて、米国仮出願番号60/432,092号(2002年12月10日出願)(その内容は、その全体が本明細書に参考として援用される)の優先権を主張する。
本出願は、イヌRANKリガンド(RANKL)ポリペプチド、イヌRANKLポリペプチドをコードする核酸、イヌRANKLポリペプチドを含む免疫原組成物および/またはワクチン、イヌRANKLのアンタゴニスト、イヌRANKLのアンタゴニストを同定するための方法、ならびにRANKL媒介性の医学的状態を処置するための方法に関する。
骨組織は、骨基質として公知のタンパク質、細胞および鉱物の組成物である。生きている動物では、骨芽細胞と呼ばれる細胞は骨基質を構築し、破骨細胞と呼ばれる細胞は骨基質を破壊して再吸収する。骨芽細胞は、間葉幹細胞から生じて、発達の間、骨損傷後および寿命の全体にわたって生じる正常な骨の再構築の間に骨基質を生成する。破骨細胞は、損傷またはストレスに応答して、そして種々の疾患状態に応答して、単球−マクロファージ系列の造血前駆体から分化して、骨基質を再吸収し、正常な骨再構築を支持する。
これらおよび他の問題は、本発明によって解決され、これによって、イヌRANKLポリペプチドの実質的に全てをコードする核酸、ならびにこれを作製および使用する方法が提供される。詳細には、本発明は、単離された核酸分子、およびその相補体であって、ポリペプチドをコードする核酸配列を含み、このコードされたポリペプチドが配列番号2に記載のアミノ酸残基を含むものを提供する。本発明はまた、ストリンジェントな条件下で単離された核酸分子の相補体にハイブリダイズする核酸分子を、このハイブリダイズ可能な核酸分子がヒトのRANKLもマウスのRANKLもラットのRANKLもコードしないという条件で、提供する。当業者は、本発明の単離された核酸分子が必要に応じてDNAまたはRNAであるということを理解する。
(a)少なくとも1つの外来ヘルパーT細胞リンパ球エピトープ、

従って、本発明は、イヌRANKLポリペプチドの実質的に全てであって、そのタンパク質の細胞外部分(抗原性)部分全体を含むものをコードする核酸分子を提供する。イヌRANKLは、II型膜タンパク質であって、N末端細胞内ドメイン(約48アミノ酸)、続いて膜貫通ドメイン(アミノ酸49〜68)、および細胞外ドメイン(アミノ酸59〜319)を有する。イヌRANKリガンドの細胞外ドメイン、膜貫通ドメインおよび細胞内ドメインの一部をコードする、核酸配列および対応するアミノ酸配列は、それぞれ配列番号1および配列番号2に規定される。詳細には、イヌRANKLの核酸およびアミノ酸配列は、この核酸の最初の129ヌクレオチドおよびこのポリペプチドに対応する43アミノ酸を除けば、完全である。

本明細書において用いる場合、「ポリヌクレオチド(polynucleotide)」または「核酸(nucleic acid)」という用語は、一連のヌクレオチドで、例えば、ポリマー骨格に結合した、デオキシリボ核酸またはリボ核酸塩基をいう。これらとしては、例えば、ゲノムDNA、cDNA、RNA、mRNA、任意の合成および遺伝子操作されたポリヌクレオチド、ならびにセンスポリヌクレオチドおよびアンチセンスポリヌクレオチドの両方が挙げられる。この用語はまた、一本鎖および二本鎖分子、すなわち、DNA−DNA、DNA−RNAおよびRNA−RNAハイブリッドを包含する。代表的なヌクレオチドとしては、イノシン、アデノシン、グアノシン、シトシン、ウラシルおよびチミジンが挙げられる。しかし、核酸はまた、修飾されたヌクレオチド塩基、例えば、チオ−ウラシル、チオ−グアニンおよびフルオロ−ウラシルを含んでもよい。
本発明のタンパク質、ポリペプチドおよび抗原性フラグメントは、限定はしないが、塩またはアルコール沈殿、分取ディスクゲル電気泳動、等電点電気泳動、高圧液体クロマトグラフィー(HPLC)、逆相HPLC,ゲル濾過、陽イオンおよび陰イオン交換、分配クロマトグラフィーおよび向流分配を含む標準的な方法によって精製され得る。このような精製方法は当該分野で周知であり、例えば、Guide to Protein Purification,Methods in Enzymology,第182巻、M.Deutscher編、1990、Academic Press,New York,NYに開示されている。
本発明によって、炎症、骨疾患、変形性関節炎、関節リウマチ、骨粗しょう症および疼痛を含む種々の疾患の処置および管理に有用であり得るイヌRANKリガンドの選択的アンタゴニストの開発が可能になる。従って、本発明のリガンドは、RANKLのアンタゴニストを同定するためのスクリーニングシステムにおいて使用することができる。本質的に、これらのシステムはRANKレセプター、イヌRANKリガンド自体を含む適切なリガンド、およびイヌRANKLアンタゴニストの存在下で試験されるべきサンプルを一緒にするための方法を提供する。
(a)RANKレセプターまたはその配列を、既知の量のイヌRANKLの存在下において、イヌRANKLアンタゴニストの存在について試験すべきサンプルと接触させる工程と;
(b)レセプターに結合したイヌRANKLの量を測定する工程と;
を包含し、これによってサンプル中のイヌRANKLアンタゴニストは、このようなアンタゴニストの非存在下で測定される結合に比べて、RANKレセプターに対するRANKLの実質的に低下した結合を測定することによって特定される。前に言及したとおり、RANKLが標識されてもRANKが標識されてもよい。

本発明は、他の種からRANKリガンドをクローニングするための方法を提供する。要するに、サザンブロットおよびノーザンブロット分析を行なって、RANKリガンドをコードする遺伝子を発現する他の種由来の細胞を同定することができる。相補的DNA(cDNA)ライブラリーは、このような細胞から単離されたmRNAから標準的な方法によって調製可能であり、そして本明細書に提供される核酸およびアミノ酸の配列に基づくプローブまたはPCRプライマーを変性することによって、RANKリガンドをコードするクローンを同定することができる。
本発明はまた、配列番号2のアミノ酸配列によって規定されるポリペプチドをコードするメッセンジャーRNAの局在化のための組成物および方法を提供する。
「免疫原性組成物(immunogenic composition)」は、物質または物質の組み合わせであって、共有結合的にまたは非共有結合的に組み合わされ、生物体においておよび/または単離された免疫系細胞から免疫応答を誘発するのに有効である。免疫応答は、外来物質に対する身体の反応であって、このような反応の生理学的なまたは病理学的な結果を導くことなく、すなわち生物体に対して防御免疫を付与する必要のない反応である。免疫応答は、以下の1つ以上を含んでもよい:抗原に対する曝露に応答した胸腺によるリンパ球の産生を含む、細胞媒介性免疫応答;および/または引き続く抗体産生をともなう抗原暴露に応答した血漿リンパ球の産生を含む、体液性免疫応答。免疫原性組成物は、抗体の産生を惹起するための抗原として有用である。

本発明のイヌRANKリガンドアンタゴニストは、RANKリガンドの活性をブロックするために、そしてこれによって、RANK/RANKL系によって生じるかもしくは媒介される任意の医学的状態を処置するために、治療上用いることができる。治療適用に関与する投薬レジメンは、治療物質の作用を改変し得る種々の要因、例えば、患者の状態、体重、性別および食事、投与のタイミング、ならびに他の臨床要因を考慮して、担当する獣医師によって決定される。
本発明はまた、配列番号2に規定されるアミノ酸配列またはそのサブ配列を有するイヌRANKリガンドをコードするmRNAに対して特異的にハイブリダイズして、そのmRNAの翻訳を妨げることができるアンチセンスオリゴヌクレオチドを包含する。さらに、本発明は、配列番号2に規定されるアミノ酸配列またはそのサブ配列を有するイヌRANKLをコードするゲノムDNA分子に対して特異的にハイブリダイズできるアンチセンスオリゴヌクレオチドを意図する。

(イヌRANKLのクローニング)
イヌRANKLは、一連のネスト化PCRストラテジーを用いてクローニングした。
(イヌRANKLの発現および精製)
標準的な分子生物学的技術を用いて、連続してプレプロトリプシンシグナルペプチドの1〜15残基、FLAGTM配列DYKDDDD、KL(構築に用いられるHindIII部位をコードする)、VA残基、イヌRANKL由来のエクトドメインの155〜319残基、残基PRPPTPGNL(タンパク質分解性切断部位をコードする)、およびヒトIgGγ1の定常領域由来の99〜330残基をコードする、キメラDNA構築物を作製する。このキメラコード領域を、Hoekら「Down−regulation of the macrophage lineage through interaction with OX2」Science、第290巻、1768〜1771頁(2000年12月1日)によって以前に記載されたとおり、改変したpQB1−AdCMV5−GFPアデノウイルス移入ベクター(Quantum Biotechnologies,Montreal,Canada)に挿入して、組み換えアデノウイルスを作製するのに用いた。コントロールのアデノウイルスは、同じキメラ構築物からイヌRANKLエクトドメインがないものをコードする。Oppmannら「Novel p19 protein engages IL−12p40 to form a cytokine,IL−23,with biological activities similar as well as distinct from IL−12」、Immunity,第13巻、715〜25頁(2000年11月)に以前に記載された方法を用いて、組み換えIg融合タンパク質を調製した。
(相同なRANKL遺伝子の単離)
イヌRANKL cDNAは、所望の供給源由来のライブラリー、例えば、霊長類細胞cDNAライブラリーをスクリーニングするためのハイブリダイゼーションプローブとして用いることができる。多くの異なる種を、容易なハイブリダイゼーションのために必要なストリンジェンシーについて、そしてプローブを用いて存在についてスクリーニングすることができる。適切なハイブリダイゼーション条件を用いて、交差ハイブリダイゼーションの特異性を示すクローンを選択することができる。
イヌRANKL.Ig融合タンパク質で免疫した8週齢の雌性Lewisラット(Harlan Sprague−Dawley,Indianapolis,IN)の脾細胞からラット抗イヌRANKL mAbを産生する。完全フロイントアジュバントに含まれる25μgの融合タンパク質を腹腔内(i.p.)で用いてラットをプライムして、引き続き、不完全フロイントアジュバントに含まれる、それぞれ10μg(25日)、5μg(40日)および10μg(54日)を腹腔内で用いて3回追加免疫する。それぞれ生理食塩水および不完全フロイントアジュバントに含まれる10μgの融合タンパク質を83日目にi.v.(静脈内)およびi.p.(腹腔内)で用いて、最終の追加免疫を行なった。脾細胞を、87日目に、PEG 1500(Roche Diagnostics,Mannheim,Germany)を用いて、マウス骨髄腫P3X63−AG8.653と融合する。間接的なELISAによって、融合タンパク質およびコントロールのIgタンパク質の両方に対して、ハイブリドーマ上清をスクリーニングして、特定のmAb産生ハイブリドーマを同定する。これらはさらに、ウエスタンブロット、免疫沈降およびFACS分析(例えば、イヌ活性化T細胞で)のような方法によって特徴付けられる。選択された陽性のハイブリドーマ株をサブクローニングして、SITE(Sigma,St.Louis,Missouri)を補充した無血清培地において増殖させる。HiTrap SPおよびQカラム(Amersham Pharmacia Biotech)を介して抗体を精製して、その抗体がRANKL誘導性生物学的応答、例えば、NF−κBの活性化または破骨細胞の活性化を阻害する能力について、例えば、Yasudaら、によって記載されるOCL形成アッセイを用いてスクリーニングする。破骨細胞分化因子は、破骨細胞形成阻害因子(オステオプロテグリン)/破骨細胞形成阻害性因子(osteoclastogenesis−inhibitory factor)のリガンドであって、TRANCE/RANKLと同一である、Proc.Natl.Acad.Sci.第95巻、3597〜3602頁(1998)。

- 哺乳動物においてRANKL活性を下方制御するためのアジュバントを含むワクチンの調製のための、NF−κBリガンド免疫原性組成物のイヌレセプター活性化因子の使用。
- 骨粗鬆症または過剰な骨の再吸収によって特徴付けられる他の状態の処置、予防または寛解のためのアジュバントを含むワクチンの調製のための、NF−κBリガンドのイヌレセプター活性化因子の使用。
