このように、第一の局面において、本発明は、抗体と該抗体に対する抗原の存在下でCD16受容体発現細胞を反応培地において接触させる段階、およびCD16受容体発現細胞によって産生された少なくとも一つのサイトカインの量を測定する段階を含むことを特徴とする、モノクローナル抗体(MoAb)またはポリクローナル抗体による、形質転換されてもされなくてもよい免疫系に属するエフェクター細胞の活性化を測定する方法に関する。
抗体:
WinRhoポリクローナル抗体、DF5-EBVモノクローナル抗体、DF5-YB2/0モノクローナル抗体。
このアッセイ法は、抗D抗体がJurkat CD16細胞において発現されたCD16受容体(FcγRIII)に結合して、IL-2分泌を誘導するか否かを評価する。
陽性対照抗体:ポリ-D WinRho、DF5-YB2/0。
陰性対照抗体:DF5。
Rh陽性赤血球。
JurkatCD16細胞。
IL2アッセイキット:R/DのQuantikine。
赤血球のパパインによる処置
PBSにおいて希釈した赤血球沈降物1 mlをパパイン溶液(1 mg/ml)1 mlと共に37℃で10分インキュベートする。H2O-0.15 M NaClにおいて洗浄を3回行う。
−抗体:IMDM 5%SVFにおける150 ng/mlの希釈液50 μl、
−PMA:IMDM 5%SVCにおける40 ng/mlの希釈液50 μl、
−パパインによって処理した赤血球。IMDM 5%SVFにおいて8×106個/mlを50 μl、
−Jurkat CD16。IMDM 5%SVFにおいて2×106個/mlを50 μl。
37℃で一晩インキュベーション。
次に、プレートを遠心して、上清100 μlを採取して、市販のキットによってIL2に関してアッセイする。450 nmで読み取る。
この試験に関して、三つの抗Dモノクローナル抗体を比較した。
試験モデル:末梢血から精製したNK細胞
適用:抗腫瘍反応の増強
IL2は、T-リンパ球およびNK細胞自身の活性化を誘導し、これによって、細胞増殖の刺激まで起こりうる。IFNγはCTLsの活性を刺激して、マクロファージの活性を増強することができる。
適用:貪食の増強および抗炎症特性の誘導
TNFは、腫瘍浸潤リンパ球およびマクロファージの増殖を刺激する。IL-1Raは、その受容体のレベルでIL1と競合するマクロファージによって産生されるサイトカインであり、このように、抗炎症作用を発揮する。
適用:特定の抗原に対して特異的な寛容の誘導
IL10は、様々なエフェクター細胞の活性化およびサイトカインの産生を阻害する分子である。
三つの細胞集団を調べた:多形核細胞、単核球、およびNK細胞。サイトカイン合成は、標的の存在に依存する。抗体R297およびポリクローナル抗D抗体によって誘導されるサイトカインプロファイルにはほとんど差がない。AD1は、非常にしばしばサイトカイン分泌を誘導しない。
6.1
モノクローナル抗体R297およびポリクローナル抗体WinRhoは、単核球の存在下でIL8のかなりの分泌を誘導する。この分泌は、抗体濃度および抗原性標的の存在に依存する。抗体AD1は、サイトカイン産生の分泌の誘導に関してかなり有効性が低く(図11)、すなわちサイトカイン産生をあまり誘導することができない。
モノクローナル抗体R297およびポリクローナル抗体WinRhoは、多形核細胞による、非常に弱いがAD1より大きいIL2、IFNγ、IP10、およびTNFの分泌を誘導する。この分泌は抗体濃度依存的である(図13)。
モノクローナル抗体R297およびポリクローナル抗体WinRhoは、NK細胞による、IFNγ、IP10、およびTNFのかなりの分泌を誘導する。この分泌は抗体濃度依存的である(図14)。
緒言
本発明者らの最初の結果は、YB2/0において産生された抗D抗体および臨床的に用いられるポリクローナル抗体も同様に、精製NK細胞、または単核球からのサイトカイン、特にTNFαおよびインターフェロンγ(IFNγ)の産生を誘導することを示した。一方、他の細胞株において産生される他の抗D抗体は、ADCCにおいて陰性であり、この分泌を誘導できないことが証明された。
抗体
抗CD20:YB2/0にトランスフェクトしたキメラ抗CD-20抗体を、CHOにおいて産生される市販の抗CD20抗体(リツキサン)と比較する。
抗HLA-DR:キメラ抗HLA-DR抗体をコードする同じ配列をCHO(B11)またはYB2/0(4B7)にトランスフェクトする。
標的細胞:その表面上でCD20およびHLA-DR抗原を発現するRaji細胞。
エフェクター細胞:ヒト血液バッグから負の選択によって精製したヒトNK細胞。
様々な濃度の抗CD20または抗HLA-DR抗体をRaji細胞(標的)およびNK細胞(エフェクター細胞)と共にインキュベートする。16時間インキュベートした後、細胞を遠心する。上清をTNFαおよびIFNγに関してアッセイする。

TNFα:結果を上清においてアッセイしたTNFαのpg/mlで表記する。反応混合物に加えた抗体の様々な濃度を、X-軸に示す(図15)。

Claims (24)
- CD16受容体発現細胞を、抗体および該抗体に対する抗原の存在下で反応培地において接触させること、ならびにCD16受容体発現細胞によって産生された少なくとも一つのサイトカインの量を測定することを含むことを特徴とする、モノクローナル抗体(MoAb)またはポリクローナル抗体による、形質転換されてもされなくてもよい免疫系に属するエフェクター細胞の活性化を測定する方法。
- エフェクター細胞がCD16受容体発現Jurkat細胞であることを特徴とする、請求項1記載の方法。
- IL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10等、TNFα、TGFβ、IP10およびIFNγから選択される少なくとも一つのサイトカインが定量されることを特徴とする、請求項1および2のいずれかに記載の方法。
- インターロイキンIL-2が定量されることを特徴とする、請求項1〜3の一項に記載の方法。
- 産生されたサイトカインの量がエフェクター細胞の活性化または阻害のマーカーであることを特徴とする、請求項1〜4の一項に記載の方法。
- 分泌されたインターロイキンIL2の量が、その抗原結合完全性(Fc機能)および有効性(抗原性部位)に関してCD16受容体によって結合される抗体の質を反映することを特徴とする、請求項1〜5の一項に記載の方法。
- 分泌されたインターロイキンIL2の量がADCC型活性と相関することを特徴とする、請求項1〜6の一項に記載の方法。
- 免疫系のCD16受容体発現エフェクター細胞を、抗体および該抗体に対する抗原の存在下で反応培地において接触させること、ならびにCD16受容体発現細胞によって産生された少なくとも一つのサイトカインの量を測定することを含むことを特徴とする、モノクローナル抗体またはポリクローナル抗体の有効性を評価する方法。
- 形質転換されてもされなくてもよい免疫系のCD16受容体発現エフェクター細胞を、抗体および該抗体に対する抗原の存在下で反応培地において接触させること、ならびにCD16受容体発現細胞によって産生された少なくとも一つのサイトカインの量を測定することを含むことを特徴とする、細胞が有効なモノクローナル抗体を産生できるか否かを評価する方法。
- 抗体を産生する細胞が、CHO、YB2/0、ヒトリンパ芽球様細胞、昆虫細胞、およびマウス骨髄腫細胞、または他の任意の発現細胞から選択されることを特徴とする、請求項9記載の方法。
- 形質転換されてもされなくてもよい免疫系のCD16受容体発現エフェクター細胞を、抗体および該抗体に対する抗原の存在下で反応培地において接触させること、ならびにCD16受容体発現細胞によって産生された少なくとも一つのサイトカインの量を測定することを含むことを特徴とする、一つまたはそれ以上の精製段階後のポリクローナル抗体の有効性および完全性を評価する方法。
- 抗体の非存在下、または陰性参照物質としての所定の抗体の存在下における対照と比較して、CD16発現細胞によるIL-2放出量の100%、250%、500%、または1000%より多い増加が認められる抗体が選択されることを特徴とする、請求項1〜11の一項に記載の方法。
- 反応混合物がヒト免疫グロブリン(IVIg)を含むことを特徴とする、請求項1〜12の一項に記載の方法。
- 同様にADCCアッセイ法を含むことを特徴とする、請求項1〜12の一項に記載の方法。
- CD16-受容体発現エフェクター細胞によるIL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10等、TNFα、TGFβ、IP10およびIFNγから選択される少なくとも一つのサイトカインの産生を誘導することができるキメラ、ヒト化、またはヒトモノクローナル抗体を選択するために、請求項1〜14の一項に記載の方法の使用。
- トランスジェニック植物またはトランスジェニック哺乳類によるMoAbsの産生を評価するために請求項1〜14の一項に記載の方法の使用。
- 治療的治療、特に自己免疫および炎症疾患、癌、ならびに病原性物質による感染症の治療にとって有効である抗体を選択するために、請求項1〜14の一項に記載の方法の使用。
- 免疫系に属するエフェクター細胞による少なくとも一つのサイトカインの産生を誘導する薬剤を調製するために、請求項15〜17の一項に記載の方法から得ることができるキメラ、ヒト化、またはヒトモノクローナル抗体の使用。
- 免疫系に属するエフェクター細胞による少なくとも一つのサイトカインの産生を誘導する薬剤を調製するために、ラット骨髄腫細胞株、特にYB2/0およびその誘導体の細胞によって産生されるキメラ、ヒト化、またはヒトモノクローナル抗体の使用。
- CD16受容体発現エフェクター細胞による、IL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10等、TNFα、TGFβ、IP10およびIFNγから選択される少なくとも一つのサイトカインの産生を誘導する薬剤を調製するための、請求項19記載の使用。
- 免疫系に属するエフェクター細胞による少なくとも一つのサイトカインの産生を誘導することが意図される薬剤を調製するために、短い鎖、低い程度のシアリル化、非インターカレート末端結合点マンノースおよびGlcNAc、ならびに低い程度のフコシル化を有する両触角型のグリカン構造を有するキメラ、ヒト化、またはヒトモノクローナル抗体の使用。
- CD16受容体発現エフェクター細胞による、IL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10等、TNFα、TGFβ、IP10およびIFNγから選択される少なくとも一つのサイトカインの産生を誘導する薬剤を調製するための、請求項21記載の使用。
- 免疫系に属するエフェクター細胞による少なくとも一つのサイトカインの分泌を誘導することが意図される薬剤を調製するために、G0F+G1F型が、50%未満、好ましくは30%未満であると理解される、G0+G1+G0F+G1F型に関して60%より高い、好ましくは80%より高いグリカン含有量を有する抗体の組成物の利用。
- CD16受容体発現エフェクター細胞による、IL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10等、TNFα、TGFβ、IP10およびIFNγから選択される少なくとも一つのサイトカインの産生を誘導する薬剤を調製するための、請求項23記載の使用。
