Abstract

Description
本発明は一般に医薬グレードの製品を作製するための材料に関し、そしてより特定すれば、プラスチック容器および医薬チューブのような物品に使用され得るフィルム製品に関する。
医学分野では、容器中に有益な試薬を集め、処理しそして保存し、輸送し、そして最後にはチューブを通して、注入により、患者に送達して、治療効果を得る場合、容器を製造するのに用いられる材料は独特な特性の組合せを有さなければならない。例えば、溶液の粒子混入物を視覚的に検査するためには、容器は光学的に透明でなければならない。容器内への空気の取り入れなしに容器の壁を壊すことにより容器から溶液を注入するためには、壁を形成する材料は十分に柔軟でなければならない。この材料は広範囲の温度にわたって機能的でなければならない。この材料は低温にてその柔軟性および頑丈さを維持することによって、機能しなければならない。なぜなら、特定の溶液(例えば、ある種のプレミックスされた薬物溶液)は、薬物の分解を最小限にするために-25℃〜-30℃のような温度で容器に保存され、運搬される。材料はまた、滅菌(ほとんどの医薬容器および栄養製品に対して出荷の前に行われるプロセス)時の熱に耐えるため、高温においても機能的でなければならない。滅菌プロセスは、通常、容器を、代表的には121℃の温度および高圧の蒸気にさらす工程を包含する。したがって、材料は、温度および圧力に顕著な歪みなしに耐える(「熱歪み抵抗」)必要がある。

(項目4)前記第1のポリオレフィンがポリプロピレンであり、そして前記第2のポリオレフィンが、超低密度ポリエチレン、ポリブテン-1、ブテンエチレンコポリマー、酢酸ビニルの含量が約18〜50%の間であるエチレン酢酸ビニルコポリマー、メチルアクリレートの含量が約20〜40%の間であるエチレンメチルアクリレートコポリマー、n-ブチルアクリレートの含量が20〜40%の間であるエチレンn-ブチルアクリレートコポリマー、アクリル酸の含量が約15%より多いエチレンアクリル酸コポリマーを包含するエチレンコポリマーの群から選択される、項目3に記載の構造物。
(項目5)前記高周波受容可能ポリマーが、ポリアミド、酢酸ビニルの量が18〜50重量%であるエチレン酢酸ビニル、メチルアクリレートの量が20%〜40重量%であるエチレンメチルアクリレートコポリマー、ビニルアルコールの量が15%〜70%であるエチレンビニルアルコールの群から選択される、項目3に記載の構造物。
(項目6)前記スチレンおよび炭化水素ブロックコポリマーがスチレン-エチレン-ブテン-スチレンブロックコポリマーである、項目5に記載の構造物。
(項目7)さらに、2つの結合層を含有し、1つの結合層が前記スキン層と前記バリアー層との間に配置され、もう1つの結合層が該バリアー層と前記高周波受容可能層との間に配置される、項目6に記載の構造物。
(項目8)前記結合層が、修飾されたポリオレフィン、および修飾されたポリプロピレンおよびポリエチレンコポリマーの群から選択される、項目7に記載の構造物。
(項目9)前記高周波ポリマーが脂肪酸ポリアミドダイマーである、項目7に記載の構造物。
(項目10)前記スキン層が0.5mil〜4.0milの範囲内の厚みを有し、前記バリアー層が0.3mil〜5.0milの範囲内の厚みを有し、前記高周波受容可能層が2.0mil〜8.0milの範囲内の厚みを有し、そして前記結合層がそれぞれ0.3mil〜1.0milの範囲内の厚みを有する、項目7に記載の構造物。
(項目11)多層構造物であって、以下の:
スキン層;
バリアー層;
30〜60重量%の範囲内の量の第1のポリオレフィン、25〜50重量%の範囲内の量の第2のポリオレフィン、3〜40重量%の範囲内の量の高周波受容可能ポリマー、5〜40重量%の範囲内の量のスチレンおよび炭化水素ブロックコポリマーを有する高周波受容可能層;および
該高周波受容可能層を第1および第2の高周波受容可能層に分割する第1のポリプロピレン層であって、ここで該第1および第2の高周波受容可能層のそれぞれが、所定範囲内の厚みを有する、第1のポリプロピレン層を含む、構造物。
(項目12)前記第1および第2の高周波受容可能層が実質的に同じ厚みである、項目11に記載の構造物。
(項目13)前記第1および第2の高周波受容可能層が異なる厚みである、項目11に記載の構造物。
(項目14)前記各層が連続的な順序で積層される、項目11に記載の構造物。
(項目15)さらに、前記スキン層と前記バリアー層との間に配置される第1の結合層;および該バリアー層と前記第1の高周波受容可能層との間に配置される第2の結合層を含む、項目14に記載の構造物。
(項目16)前記バリアー層がエチレンビニルアルコール、および高ガラス状結晶性ポリアミドからなる群から選択される、項目11に記載の構造物。
(項目17)前記第1のポリオレフィンが第2のポリプロピレンであり、そして前記第2のポリオレフィンが、超低密度ポリエチレン、ポリブテン-1、ブテンエチレンコポリマー、酢酸ビニルの含量が約18〜50%の間であるエチレン酢酸ビニルコポリマー、メチルアクリレートの含量が約20〜40%の間であるエチレンメチルアクリレートコポリマー、n-ブチルアクリレートの含量が20〜40%の間であるエチレンn-ブチルアクリレートコポリマー、アクリル酸の含量が約15%より多いエチレンアクリル酸コポリマーを包含するエチレンコポリマーの群から選択される、項目16に記載の構造物。
(項目18)前記スキン層が、ポリプロピレンコポリマーを、該ポリプロピレンの量の0〜20重量%の範囲内の、スチレン-エチレン-ブテン-スチレンブロックコポリマーとともに含有する、項目17に記載の構造物。
(項目19)多層構造物であって、以下の:
スキン層;
2から10ユニットのバリアー層および結合層を有するバリアーサンドイッチ層;および

(項目20)前記スキン層と前記バリアーサンドイッチ層との間にさらに結合層を含有する、項目19に記載の構造物。

(項目22)前記バリアー層がエチレンビニルアルコールおよび高ガラス状結晶性ポリアミドからなる群から選択される、項目21に記載の構造物。
(項目23)前記結合層が、修飾されたポリオレフィンおよび修飾されたポリプロピレンおよびポリエチレンコポリマーの群から選択される、項目22の構造物。
本発明に従って、特定の多層ポリマーを基礎とした構造物を開示する。この構造物から、医薬物品(例えば、医薬溶液または血液生産物のための容器、血液バッグ、および関連する製品)または多層構造物から構成される他の製品を製造し得る。
本発明は、多くの様々の形式での実施態様が可能であり、そして本明細書中に詳細に記載されているが、本発明の好ましい実施態様を、この開示が本発明の原理の例と考えられ、そして本発明の広範な局面を例示の実施態様に限定することを意図しない、との理解のもとに、開示する。
(1)オートクレーブ性能
耐オートクレーブ性はサンプルのクリープ、すなわち、121℃、27psiの負荷で1時間での、サンプルの長さの増加によって測定した。耐オートクレーブ性は60%以下でなければならない。
(2)低温および室温での延性
(A)低温での延性
液体窒素で冷やされる低温環境チャンバーを装着した衝撃試験機にて、7inch×7inch(18cm×18cm)のフィルムサンプルを直径約6inch(15cm)の円形のサンプルホルダーの上に載せる。応力センサーを備えた半球状の衝撃ヘッドを高速(代表的には約3m/sec)で、予めコンディショニングしたフィルムへ、ヘッドを中心に導きながら、打ち込む。応力-変形曲線をプロットし、そして衝撃のエネルギーを積分により計算する。衝撃エネルギーが劇的に上昇し、そして破壊された部分が脆性高歪みモルホロジーから延性高歪みモルホロジーに変化するときの温度を、フィルムの低温性能(「L.TMP」)の測定値とする。
オートクレーブ処理された既知の寸法のフィルムサンプルを、サンプルを伸ばすためのクロスヘッドを有する、サーボ水圧的に運転される機械的試験機に取り付ける。毎分10inch(25cm)のクロスヘッド速度にて、サンプルを、約20%の伸びまで伸ばす。この時点で、クロスヘッドは動いて、そして次いで最初にサンプルを伸ばすために使用された方向と逆方向に動いて戻る。応力と歪みの挙動をデジタル記録計に記録する。弾性率(「E(Kpsi)」)は、応力-歪み曲線の初期の傾きから得、そして回復は、サンプルの伸びのパーセントとしての過剰なサンプル長さから得る。
(3)RF加工性:
Callahanの27.12MHz、2Kw高周波発生機に、約0.25inch(6.3mm)×4inch(10cm)の直角の真鍮のダイを、平らな真鍮の電極(これもまた発生機に接続している)と反対側に接続する。候補材料の2枚のシート(溶液側をそれぞれ向かい合わせて)を間に挟んでダイを閉めながら、異なった増幅率および時間のRF電力を印加した。RFサイクルが終了すると、ダイを開け、そして得られたシールを、手で2枚のシートに引き離すことにより試験した。シールの(フィルムの強度に対する)強度および破壊の状況(剥離、破断、または凝集破壊)を材料のRF応答性の評価に用いる。
オートクレーブした後のフィルムサンプルを最初に約2×2インチ(5×5cm)平方に切断し、Hunter Colorimeter に装着し、ASTM D-1003に従い、それらの内部ヘーズを測定する。代表的には、その厚みに対して30%より少ない内部ヘーズのレベル(「ヘーズ%」)が必要であり、20%より少ないことが好ましい。
(5)歪み白化
オートクレーブした後のフィルムを、毎分約20インチ(50cm)の通常の速度で約100%の伸び(もとの長さの2倍)まで歪みをかけ、そして歪み白化の存在(1と表示する)または不存在(0と表示する)を記録する(「歪み白化(S.Whitening)」)。
(6)環境適合性:
環境適合性は、3つの重要な特性を含む:(a)材料が、廃棄されたときに埋め立て地に溶出し得る低分子可塑剤を有しない、(2)材料が、医薬送達という第一の目標を達成したとき、有用な物品に熱可塑的にリサイクルし得る、そして(3)焼却によるエネルギーの再生利用(reclaim)により、廃棄されるときに、環境に悪影響を与える有意な無機酸を放出しない。(「環境適合性(Envir.)」)。この組成物はまた、0.1重量%より少ないハロゲンを含む。溶融加工によるリサイクルを容易にするために、得られる組成物は1MHZ、加工温度にて測定して、1.0より大きい損失正接を有するべきである。
(7)溶液適合性
溶液適合性とは、フィルム内に含まれる溶液が組成物を構成する成分によって汚染されないことを意味する。(「溶液適合性(S.Comp.)」)組成物中の低分子量の水溶性留分は0.1%より少ない。
(8)バリアー特性
図11に示すバリアーフィルム構造物を、独立した実験室にて、gm/100in2/24時間の単位で表される水蒸気透過率(WVTR)、相対湿度(RH)50%および相対湿度80%における酸素透過率、および二酸化炭素透過率について試験し、そして酸素および二酸化炭素の透過率をcc/100in2/24時間の単位で表す。
Claims (5)
- 多層構造物であって、以下の:
スキン層;
2から10ユニットのバリアー層および結合層を有するバリアーサンドイッチ層;および
30〜60重量%の範囲内の量の第1のポリオレフィン、25〜50重量%の範囲内の量の第2のポリオレフィン、3〜40重量%の範囲内の量の高周波受容可能ポリマー、5〜40重量%の範囲内の量のスチレンおよび炭化水素ブロックコポリマーを有する高周波受容可能層を含む、構造物。 - 前記スキン層と前記バリアーサンドイッチ層との間にさらに結合層を含有する、請求項1に記載の構造物。
- 前記第1のポリオレフィンがポリプロピレンであり、そして前記第2のポリオレフィンが、超低密度ポリエチレン、ポリブテン-1、ブテンエチレンコポリマー、酢酸ビニルの含量が約18〜50%の間であるエチレン酢酸ビニルコポリマー、メチルアクリレートの含量が約20〜40%の間であるエチレンメチルアクリレートコポリマー、n-ブチルアクリレートの含量が20〜40%の間であるエチレンn-ブチルアクリレートコポリマー、アクリル酸の含量が約15%より多いエチレンアクリル酸コポリマーを包含するエチレンコポリマーの群から選択される、請求項2に記載の構造物。
- 前記バリアー層がエチレンビニルアルコールおよび高ガラス状結晶性ポリアミドからなる群から選択される、請求項3に記載の構造物。
- 前記結合層が、修飾されたポリオレフィンおよび修飾されたポリプロピレンおよびポリエチレンコポリマーの群から選択される、請求項4の構造物。
