感染に対する処置または保護
本発明に従って、コアグラーゼ陰性ブドウ球菌感染を予防または処置するための方法が提供され、この方法は、感染を処置又は予防するための有効量でこのような処置が必要なヒトまたは動物患者に上記の抗体の有効量を投与することを包含する。さらに、本発明に従う抗体は、種々の細菌のフィブリノゲンへの結合を損なう際に特に有用であり、従って、該結合を阻害することによってコアグラーゼ陰性ブドウ球菌のような細菌由来の感染を処置または予防する際に有効であることが証明された。

本発明の好ましい実施形態の局面を例示する以下の実施例が提供される。以下の実施例において開示される技術は、本発明の実施において十分に機能することが発明者らによって発見される技術を示し、従って、その実施のための好ましい様式を構成すると考えられ得ることが当業者によって理解されるべきである。しかし、当業者は、本発明の開示を考慮して、多数の変化が、開示される特定の実施形態において行われ得、本発明の精神および範囲から逸脱することなく同様の結果を依然として得ることを理解するべきである。
本発明に従って、SdrGタンパク質の関連ドメインから得られるタンパク質を、クローニングし、組み換え発現し、単離および/または精製した。SdrG N1N2N3タンパク質(50〜597)は、SdrG遺伝子の推定Aドメインを示す。SdrG N2N3タンパク質(273〜597)は、ヒトフィブリノゲン結合に必要なサブドメインを示す。SdrG TR2タンパク質(273〜577)は、フィブリノゲン結合を安定化する、C末端部分が除去された、ヒトフィブリノゲン結合に必要なサブドメインを示す。これらのタンパク質についてのヌクレオチドおよびアミノ酸配列を以下に示す。
SdrG N1N2N3(50〜597):
ヌクレオチド配列(配列番号1)
PCRを使用して、SdrGN1N2N3(アミノ酸50〜597を示す)またはSdrGN2N3(アミノ酸273〜597を示す)のようなそのサブドメインもしくはその短縮物TR2(アミノ酸273〜577)を、S.epidermidis K28ゲノムDNA(上記の配列より)から増幅し、E.coli発現ベクターPQE−30(Qiagen)にサブクローニングした。これは、6つのヒスチジン残基を含む組換え融合タンパク質の発現を可能にする。このベクターを、引き続き、E.coil株ATCC55151に形質転換し、0.7の光学密度(OD600)まで15リットルの発酵槽中で増殖し、0.2mMイソプロピル−1−β−Dガラクトシド(IPTG)で4時間誘導した。細胞を、AG Technologies中空糸アセンブリ(0.45□mの孔サイズ)を使用して収穫し、細胞ペーストを−80℃で凍結した。細胞を、French Press@1100psiを二回通して、1×PBS(10mLの緩衝液/1gの細胞ペースト)中で溶解した。溶解した細胞を、30分間17,000rpmでスピンダウンして、細胞破片を除去した。上清を、0.1M NiCl2とともにチャージした5mL HiTrap Chelating(Pharmacia)カラムを通過させた。ローディング後、カラムを、10mM Tris,pH8.0、100mM NaCl(緩衝液A)の5カラム容量で洗浄した。タンパク質を、10mM Tris(pH8.0)、100mM NaCl、200mMイミダゾール(緩衝液B)の0〜100%の勾配を使用して、30カラム容量で溶出した。SdrGN1N2N3、SdrGN2N3またはTR2を、約13%の緩衝液B(約26mMのイミダゾール)で溶出した。280nmにおける吸光度をモニターした。SdrGN1N2N3、SdrGN2N3またはTR2を含む画分を1×PBS中で透析した。
モノクローナル抗体(mAb)を産生および特徴付ける目的で、フィブリノゲンのSdrGへの結合を阻害または制限しそしてインビボにおける治療効果を実証し得る、高い親和性でSdrGに対するmAbを産生するためのストラテジーを形成した。E.coliによって発現されそして精製されたSdrG(N1N2N3、N2N3またはTR2)タンパク質は、マウスモノクローナル抗体のパネルを作製するために使用され得る。手短に言うと、Balb/CまたはSJLマウスの群は、以下の表1に記載されるように、溶液またはアジュバントと混合した溶液中、1〜10mgのタンパク質の一連の皮下免疫化を受けた。

次いで、融合から産生した任意のクローンについて、標準的なELISAアッセイを使用して特異的な抗SdrG抗体産生についてスクリーニングした。陽性クローンを拡大し、フローサイトメトリーによる全細菌細胞結合アッセイ中の活性、およびBiacore分析によるSdrG結合についてさらに試験した。
Immulon 2−HB high−binding 96ウェルマイクロタイタープレート(Dynex)を、1×PBS(pH7.4)中、1μg/ウェルのrClfA−(40〜559)でコーティングし、室温で2時間インキュベートした。ELISAにおける全ての洗浄工程を、1×PBS、0.05%Tween−20洗浄緩衝液で3回実施した。プレートを洗浄し、ハイブリドーマ上清サンプルをウェルに添加する前に、室温で1時間、1%BSA溶液でブロックした。プレートを、サンプルおよび培地単独のような関連コントロールと共に室温で1時間インキュベートし、洗浄し、1×PBS、0.05%Tween−20、0.1%BSA中で1:5000に希釈したヤギ抗マウスIgG−AP(Sigma)を、第2の試薬として使用した。プレートを、4−ニトロフェニルホスフェート(pNPP)(Sigma)の1mg/ml溶液の添加によって発色させ、続いて、37℃で30分間インキュベートした。吸光度を、SpectraMax 190プレートリーダー(Molecular Devices Corp.)を使用して405nmで読んだ。バックグラウンド(培地単独、約0.1OD)の3倍以上のOD405を有する抗体上清は、陽性であると考えた。
分析の間ずっと、流速は、10ml/分で一定であった。SdrGN1N2N3またはSdrGN2N3/TR2注射の前に、試験抗体を、RAM−Fc結合を介してチップに吸着させた。0時間において、30mg/mlの濃度のSdrG(N2N3、TR2またはN1N2N3)を、3分間、チップに注射し、続いて2分間解離した。分析のこの段階は、Mab/SdrG相互作用の相対的な結合および/解離速度論を測定した。
細菌サンプル(HB、9142またはSdrG/Lactococcus)を回収し、洗浄し、そしてウサギIgG(50mg/ml)でのブロッキング後、MabまたはPBS単独(コントロール)と共に2mg/mlの濃度でインキュベートした。抗体とのインキュベーション後、細菌細胞を、検出抗体として機能するヤギ−F(ab')2−抗マウス−F(ab')2−FITCと共にインキュベートした。抗体標識後、細菌細胞を、FACScaliberフローサイトメーターを通して吸引し、蛍光放出を分析した(励起:488、放出:570)。各細菌株について、10,000の事象を回収し、測定した。

速度論分析を行って、選択および特徴付けられる抗SdrG mAbの多様性を示した。以下に示されるように、mAbは、オンレート(on-rate)およびオフレート(off-rate)ならびに全体の親和性において異なる。
速度論分析を、ソフトウェアに含まれるリガンド捕獲方法を使用してBiacore 3000で実施した。GAH−F(ab)2チップ。次いで、抗SdrG mAbを、GAM−F(ab)2チップを通過させ、Fc部分への結合を可能にした。次いで、種々の濃度のSdrGタンパク質を、チップ表面を通過させ、データを収集した。Biacore提供の評価ソフトウェア(バージョン3.1)を使用して、konおよびkoffを測定し、KAおよびKDを計算した。

産生され、そして組換えSdrGを用いて選択された抗SdrG mAbが、コアグラーゼ陰性ブドウ球菌に対して発現されるネイティブなSdrGと交差反応するかを決定するために、フローサイトメトリー分析を使用した。全ての場合において、mAbは、L.lactisに対して発現されたSdrGを認識したが、HBおよびF40802に対する反応性において変化した。

高親和性の多数の選択された抗SdrG mAbはまた、SdrG MSCRAMMへのヒトフィブリノゲンまたはβ−フィブリノゲンペプチドフラグメントの結合を阻害する能力を示した。この阻害を、以下に記載の多数のアッセイを使用して特徴付けた。このデータは、SdrG MSCRAMMのヒトフィブリノゲンへの接着特性を阻害することが可能であり得ることを示唆する。
分析の間ずっと、流速は、10ml/分で一定であった。SdrGN1N2N3、SdrGN2N3またはTR2注射の前に、試験抗体を、RAM−Fc結合を介してチップに吸着させた。0時間において、30mg/mlの濃度のSdrG(N2N3またはN1N2N3)を、3分間、チップに注射し、続いて2分間解離した。分析のこの段階は、Mab/SdrG相互作用の相対的な結合および/解離速度論を測定した。この分析の第2段階において、SdrGに結合したMabの、フィブリノゲンと相互作用しそして結合する能力を測定した。100mg/mlの濃度のフィブリノゲンを、チップに注射し、3分後に報告ポイントをとった。本発明に従うmAbのいくつかの結合の例を図1に示す。
フィブリノゲン分子上のSdrGについての正確な結合部位は、β鎖のN末端部分に局在する。さらなる分析および特徴付けのために、本発明者らは、N末端システイン残基を追加したこの部位を含むペプチドを合成した。配列は以下である:

βフィブリノゲンペプチドは、Biacoreによって詳細に説明される手順に従って、N末端システインを介して、研究等級のCM5チップ(Biacore)にチオール結合される。SdrGタンパク質(30μg/ml;完全なAドメイン)を、1:1の割合で、種々の濃度のmAb(90μg/ml〜0.7μg/ml)と混合した。混合物を、室温で20分間インキュベートし、次いで、βフィブリノゲンペプチドチップ上を通過させ、結合のレベルを測定した。緩衝液と1:1で希釈したSdrGは、最大のSdrG結合として機能し、非SdrG mAbをインキュベーションすることは、ネガティブコントロールとして機能する。非インヒビターは、ペプチドへのSdrG/mAb複合体結合の大きい密度に起因して、共鳴単位(Resonance Units)(RU)における大きな増加(最大のSdrG結合を超える)を引き起こすはずである。あるいは、インヒビターは、最大のSdrG未満に結合レベルを減少させるはずである。パーセント結合は、以下のように測定した:生データ(RUについて)を、SdrGコントロールレベルによって除算し、100を掛ける。従って、mAbを有さないSdrGは、常に、所定の実験について100%であり、ランの間の比較を可能にする。Biacoreチップ上のβフィブリノゲンペプチドへのSdrG結合の阻害を示す本発明に従うmAbの例を、図2に示す。
Immulon 2−HB high−binding 96ウェルプレートを、PBS中の1μg/ml SdrG(アミノ酸50〜597)またはコアグラーゼ陰性ブドウ球菌タンパク質(実施例7に記載される)でコーティングし、室温で2時間インキュベートした。プレートを洗浄し、1%BSA溶液で1時間ブロックし、次いで洗浄し、モノクローナル抗体(ハイブリドーマ上清または精製された抗体のいずれか)と共に室温で1時間インキュベートした。抗体とのインキュベーション後、プレートを洗浄するかまたは未処理のいずれかとし、20μg/mlヒトフィブリノゲン(Enzyme Resarch Lab,South Bend,Indiana,USA)を添加した。プレートを、37℃で1時間インキュベートし、洗浄し、ヤギ抗フィブリノゲン−HRP複合体を添加した。複合体とのインキュベーション後、プレートを洗浄し、ABTS基質を添加した。次いで、プレートを室温で10分間インキュベートし、反応を10%SDSの添加で停止させ、そして吸光度を405nmで読んだ。全てのデータを、SOFTmax Pro v.3.1.2.ソフトウェア(Molecular Devices Corp.,Sunnyvale,California,USA)を使用して分析した。ELISAにより、SdrGに結合するヒトフィブリノゲンの阻害を示す本発明に従うmAbを図3に示す。
コアグラーゼ陰性ブドウ球菌に見出される他のタンパク質との潜在的な交差反応性を評価するために、以下のタンパク質(アクセス番号Y17116としてgene bankにおいて同定される)を実施例1に記載される方法と類似の方法を使用してクローニングし、発現し、精製した。興味深いことに、このタンパク質とのかなりの交差反応性が、本発明の抗SdrG mAbの多くについて(従って、このタンパク質を認識する)同定された。しかし、SdrG−フィブリノゲン結合に対して阻害活性を有する1つの抗SdrG mAb(59〜59)は、交差反応せず、フィブリノゲンとの以下に記載されるタンパク質の結合を阻害しなかった。

本発明に従う多数の抗SdrG mAbを、インビボ動物モデルにおける効力について試験し、治療剤としてのそれらの潜在的な有用性を示す。
時期を定めて妊娠した(13〜15日)Sprague−Dawleyラットを、Taconic Farms(Germantown,NY)より購入した。3〜6日齢の新生児ラットに、単回の腹腔内(IP)注射によって0.35mgのモノクローナル抗体を投与した。抗体投与の20時間後、新生児ラットを、2×108CFUのS.epidermidisのHB株の(IP)注射でチャレンジした。次いで、動物の生存を、7日間追跡した。生存曲線のKaplan−Meier分析を実施し、有意性を、logランク試験(Mantel−Haenszel Test)を使用して試験した。試験結果を以下に示す:

41−211.3モノクローナル抗体(IgG1サブタイプ)を、プロテインGアフィニティクロマトグラフィーを使用して、無血清ハイブリドーマ培養培地から精製した。材料は、プロテインの0.12EU/mg未満の内毒素濃度を有し、10.4mg/mlの濃度であると報告された。該材料を4℃で冷蔵貯蔵した。注射の日に、該材料を1.75mg/mlに希釈して、0.2mlを腹腔内注射を介して動物の適切な群に投与した。投与される最終用量は、0.35mgのIgGであった。
41−075.3モノクローナル抗体(IgG1サブタイプ)を、プロテインGアフィニティクロマトグラフィーを使用して、無血清ハイブリドーマ培養培地から精製した。材料は、プロテインの0.12EU/mg未満の内毒素濃度を有し、7.6mg/mlの濃度であると報告された。該材料を4℃で冷蔵貯蔵した。注射の日に、該材料を1.75mg/mlに希釈して、0.2mlを腹腔内注射を介して動物の適切な群に投与した。投与される最終用量は、0.35mgのIgGであった。

CRL1771モノクローナル抗体(IgG1サブタイプ)を、プロテインGアフィニティクロマトグラフィーを使用して、無血清ハイブリドーマ培養培地から精製した。材料は、プロテインの3.0EU/mg未満の内毒素濃度を有し、6.6mg/mlの濃度であると報告された。該材料を4℃で冷蔵貯蔵した。注射の日に、該材料を1.75mg/mlに希釈して、0.2mlを腹腔内注射を介して動物の適切な群に投与した。投与される最終用量は、0.35mgのIgGであった。
8〜9週齢のオスSpragu−Dawleyラットを、Charles River Laboratories(Raleigh,NC)から購入した。滅菌したポリエチレン/シリコンカテーテル(カテーテル体−ポリエチレン:0.011”id、0.024”od;カテーテル先端−シリコンゴム:0.012”id、0.025”od)を、頸静脈に外科的に配置し、カテーテル先端を、上大静脈へ進行させた。カテーテルは、所定の場所に配置したままであり、研究の間ずっと開放されていた(kept patent)。モノクローナル抗体を、20mg/kgの用量で、カテーテルを介してIV投与した。24時間後、5×103CFUのメチシリン耐性S.epidermidis MRSE(899株)を、カテーテルを介して導入した。チャレンジ後7日目において、動物を犠牲にし、尾側大静脈血、腎臓およびカテーテル関連組織を収穫した。組織サンプル中に存在するMRSEコロニー形成単位を、定量的なプレーティングによって測定した。群間の感染の発生率の統計学的分析を、フィッシャーの直接確率検定(Fisher's Exact Test)を使用して実施した。群間のCFUにおける定量的差異の統計学的分析を、Dunnの多比較ポスト試験(Dunn's multiple comparison post-test)と共にKruskal−Wallis試験を使用して実施した。
SdrG59−59.4モノクローナル抗体、INH−M02001(LN:IAA2B2032)
41−211.3モノクローナル抗体(IgG1サブタイプ)を、プロテインGアフィニティクロマトグラフィーを使用して、無血清ハイブリドーマ培養培地から精製した。材料は、プロテインの0.12EU/mg未満の内毒素濃度を有し、8.2mg/mlの濃度であると報告された。材料を4℃で冷蔵貯蔵した。注射の日に、該材料を20mg/kgのIgGの最終用量についてカテーテルを介して投与した。

Claims (30)
- SdrG N1N2N3、SdrG N2N3およびSdrG TR2からなる群より選択されるS.epidermidis由来のタンパク質を認識する単離された抗体。
- 抗体がモノクローナル抗体である、請求項1に記載の抗体。
- 抗体が、キメラ、マウス、ヒト化およびヒトモノクローナル抗体からなる群より選択される型のものである、請求項2に記載のモノクローナル抗体。
- 抗体が単鎖モノクローナル抗体である、請求項2に記載のモノクローナル抗体。
- 前記抗体が、ヒトまたは動物におけるコアグラーゼ陰性ブドウ球菌感染を妨げる、請求項1に記載の抗体。
- 前記抗体が、ブドウ球菌のフィブリノゲンへの結合を抑制する、請求項1に記載の抗体。
- 前記抗体が、ヒトまたは動物における非経口、経口、鼻腔内、皮下、エアゾールまたは静脈内投与に適している、請求項1に記載の抗体。
