Abstract
【解決手段】 電極と、電極を覆うアルブミンと架橋された酵素との複合膜と、疎水性であり、架橋反応を行う為の官能基を有する低分子の電子移動メディエータとを具え、電子移動メディエータが複合膜中に存在することを特徴とする電気化学バイオセンサ。

Description
図1は本発明による電気化学グルコースセンサ構造断面の模式図を示す。図1の電気化学グルコースセンサは、直径1.6mmの金電極1、エポキシ樹脂2、ビニルフェロセン層3、ビニルフェロセン−アルブミン−グルコース酸化酵素複合層4を有する。
白金電極による過酸化水素直接酸化型グルコースセンサを作製し、ウマ血清存在下でのグルコース測定を行った。
オスミウムポリビニルピリジン錯体(Os-gel-HRP)を電子移動メディエータとするグルコースセンサを作製し、ウマ血清存在下でのグルコース測定を行った。
酵素反応により生成した過酸化水素を還元して検出するバイオセンサとして、Os-gel-HRPをメディエータとするグルコースセンサは以下のようにして作製した。まず直径1.6mmの白金電極上にOs-gel-HRPを2μl、キャスト法により修飾した。室温で1時間乾燥させた後、2%牛血清アルブミンに重量比2%になるようにグルコース酸化酵素を混合し、さらに終濃度0.5%になるようにグルタルアルデヒドを加えた溶液を、Os-gel-HRPが修飾された白金電極上に2μl塗布し、積層させた。
直径1.6mmの金電極上に実施例1と同様にビニルフェロセン層を形成した。その後、2%牛血清アルブミンに重量比2%になるように乳酸酸化酵素を混合し、さらに終濃度0.5になるようにグルタルアルデヒドを加えた溶液4μlを、ビニルフェロセン上に塗布した。
実施例1と同様に、電極上にビニルフェロセン層3を形成した。さらに2%の牛血清アルブミン溶液に重量比2%になるようにグルコース酸化酵素を混合した溶液を、ビニルフェロセン層3の上に2μl塗布し、風乾した。この電極を25%グルタルアルデヒド(シグマ社製)1ml底面に入れた容器に入れグルタルアルデヒドの蒸気によって牛血清アルブミンとグルコース酸化酵素の混合膜を不溶化し、ビニルフェロセン−アルブミン−グルコース酸化酵素複合層4を形成した。

Claims (8)
- 電極と、前記電極を覆うアルブミンと架橋された酵素との複合膜と、疎水性であり、架橋反応を行うための官能基を有する低分子の電子移動メディエータとを具え、前記電子移動メディエータが前記複合膜中に存在することを特徴とする電気化学バイオセンサ。
- 前記電子移動メディエータが、前記複合膜中に拡散され、膜内に電極と酵素の間の導電性電子移動経路を形成し、複合固定化されていることを特徴とする請求項1に記載の電気化学バイオセンサ。
- 前記電子移動メディエータが、ビニルフェロセンおよびその誘導体であることを特徴とする請求項2に記載の電気化学バイオセンサ。
- 電極上に電子移動メディエータを塗布する工程と、
酵素およびグルタルアルデヒドを溶解させたアルブミン水溶液を、前記電子移動メディエータ上に塗布して、前記電子移動メディエータをアルブミン中に拡散させる工程と、
前記アルブミン、酵素および電子移動メディエータを反応架橋させて、複合膜を形成する工程と
を具えたことを特徴とする電気化学バイオセンサの製造方法。 - 前記電子移動メディエータを塗布する工程を、キャスト法、ディップコート法およびスピンコーティング法からなる群から選択される方法によって実施することを特徴とする請求項4に記載の電気化学バイオセンサの製造方法。
- 電極と、
前記電極を覆う、ビニルフェロセンおよびその誘導体の疎水性の電子移動メディエータと、アルブミンと架橋された酵素とを含む複合膜と
を含み、前記電子移動メディエータが疎水性相互作用によって前記複合膜中に保持されていることを特徴とする電気化学バイオセンサ。 - 電極上にビニルフェロセンおよびその誘導体の疎水性の電子移動メディエータを塗布する工程と、
酵素およびグルタルアルデヒドを溶解させたアルブミン水溶液を、前記電子移動メディエータ上に塗布して、前記電子移動メディエータをアルブミン中に拡散させる工程と、
を具えたことを特徴とする電気化学バイオセンサの製造方法。 - 前記電子移動メディエータを塗布する工程を、キャスト法、ディップコート法およびスピンコーティング法からなる群から選択される方法によって実施することを特徴とする請求項7に記載の電気化学バイオセンサの製造方法。
