Abstract
【解決手段】 2つの官能基Aとそれと反応し結合することのできる官能基Bを1つもつアミノ酸、これらから構成されるペブチド、もしくは、2つの官能基Aとそれと反応し結合することのできる官能基Bを1つもつペブチドをビルディングブロックとして用い、官能基Aを有する物質を中心核として用いて、水系で官能基AとBの結合反応を行わせることによりデンドリマーを製造する方法。

Description
Magnetospirillum magneticum AMB−1は、Appl. Microbiol. Biotechnol.35(2003)651−655に記載されるようにして培養した。菌体を回収し、リン酸緩衝液(PBS)で洗浄した後、フレンチプレスに3回通して破砕した。次に、フレンチプレス破砕試料から磁気を用いて磁気微粒子を回収し、PBSで洗浄した。
デンドリマー表面のカルボキシル基は、カルボキシル基にLysをペプチド結合させた後に表面のアミノ基を測定することにより定量した。実施例1で得られた4種類の磁気微粒子デンドリマー1mgを、それぞれMESバッファーで3回洗浄し、2mM EDCおよび5mM Sulfo−NHSを含む溶液1ml中に分散させ、室温で2時間インキュベートした。次にMESバッファーで3回洗浄し、20mM 2−メルカプトエタノールおよび1mM Lysを含む溶液1ml中に分散させ、室温で4時間インキュベートした。次にこれをPBSで洗浄しそれぞれの磁気微粒子デンドリマー100μgに対し、200μlの5mM スルホスクシンイミジルピリジルジチオプロピオンアミドヘキサノエート(Sulfo−LC−SPDP)溶液に懸濁し、室温で30分間インキュベートした。磁気微粒子デンドリマーをPBSで3回洗浄し、次に200μlの20mMジチオスレイトール中で25℃で30分間インキュベートした。上清を回収し、2−スルフィドリルピリジンの343nmの吸収を測定することにより、磁気微粒子デンドリマー表面の1級アミン分子数を定量した。対照としては、アミノ酸を付加させていない磁気微粒子を用いた。結果を図2に示す。磁気微粒子数を算出するために、粒子を直径100nmの球形のマグネタイトとした時の粒子1個の重量、2.72x10−15gを基に、乾燥重量から換算した。

- 2つの官能基Aとそれと反応し結合することのできる官能基Bを1つもつアミノ酸、これらから構成されるペブチド、もしくは、2つの官能基Aとそれと反応し結合することのできる官能基Bを1つもつペブチドをビルディングブロックとして用い、官能基Aを有する物質を中心核として用いて、水系で官能基AとBの結合反応を行わせることによりデンドリマーを製造する方法。
- 請求項1記載の方法において、官能基Aがアミノ基でありかつ官能基Bがカルボキシル基であるか、または官能基Aがカルボキシル基でありかつ官能基Bがアミノ基であることを特徴とする方法。
- 請求項1記載の方法において、官能基Aがアミノ基でありかつ官能基Bがチオール基であるか、または官能基Aがチオール基でありかつ官能基Bがアミノ基であることを特徴とする方法。
- 請求項1ないし3に記載の方法において、リン脂質膜を中心核として用いることを特徴とする方法。
- 請求項1ないし3に記載の方法において、無機粒子を中心核として用いることを特徴とする方法。
- 請求項1ないし3に記載の方法において、有機粒子を中心核として用いることを特徴とする方法。
