【発明の属する技術分野】
本発明は抗疲労組成物に関し、詳細にはビオプテリン関連物質を有効成分として含有する抗疲労医薬組成物および食品組成物に関する。

【従来の技術】
めまぐるしく動的である現代社会においてはほとんどの老若男女が疲れている。1985年の総理府の健康に関する国民意識調査では疲労感を訴える人が66%であり、そのうちの72%は「一晩の睡眠により疲労感は回復する」と答えている。その後、1999年に厚生省疲労調査研究班(木谷照夫班長)が愛知県豊川保健所管内を対象に15−65歳の男女3,000名以上の回答により疫学調査をおこなったところ、疲労感を感じているヒトの割合は約60%であるものの、実にその半数を超えるヒト(全体の36%)が半年以上続く慢性疲労に悩んでいることが明らかになった。これは、日本の就労人口約8千万人に対し実に3千万人近いヒトが半年以上疲労感に悩んでいるを意味する。
このように、疲労がこれだけ蔓延している状況であるにもかかわらず、わが国においては数年前まで疲労に関する研究はほとんど行われていなかった。
慢性疲労症候群(Chronic Fatigue Syndrome、CFS)という疾患概念が克服すべき難病の一つとして最近話題になってきた。欧米では20年ほど前から発病が認められるが、わが国ではようやく1991年ごろから厚生省の専門研究班が実態調査に乗り出した段階である。これは、日常生活に支障を来すほどの長期的な全身疲労感、倦怠感、微熱、リンパ節腫脹、筋肉痛、関節痛、精神神経症状などの基本的な症状を示す。しかし、現状ではその原因は分からず、診断基準も十分確立していない。
他方、現代日本で広く知られ、不名誉な日本発の国際語としても有名になっている「過労死(Karoshi)」という言葉がある。これは長時間過密の働きすぎによる突然死を指し、大きな社会問題としてクローズアップされている。過労死は、命名者である国立公衆衛生院成人室長・上畑鉄之丞医師によって、医学・生理学的には「過重労働が誘引となって高血圧や動脈硬化が悪化し、脳出血、くも膜下出血、脳梗塞などの脳血管疾患や心筋梗塞などの虚血性心疾患、急性心臓死などを発症し、永久的労働不能や死亡に至った状態」、または「非生理的な労働過程が進行するなかで、労働者の正常な労働リズムや生活リズムが崩壊し、その結果、生体内で疲労蓄積が進み、過労状態に移行、既存の高血圧や動脈硬化が悪化し、破綻をきたした致命的な状態」と定義されている。過労死の問題は医学的、経済的、社会的にも非常に重要であると認識されているにもかかわらず、その病因メカニズムについてはほとんど解明されていない。
過労死が社会問題となっている今日、慢性疲労症候群も早期の原因究明が望まれるところであるが、現状では上記の通り、その診断基準も確立されていない。

慢性疲労症候群を処置する対症療法として、アスコルビン酸(非特許文献1)、アセチル−L−カルニチン(特許文献1)、デヒドロエピアンドロステロンサルフェート(DHEA−S)(特許文献2)が有効であることが報告されている。
6R−L−エリスロ−5,6,7,8−テトラヒドロビオプテリンはBH4と略称され、ドパミンやセロトニン合成の補助因子であることが知られている(非特許文献2)。これらの神経伝達物質の欠乏は神経症状の原因となることから、BH4はいくつかの脳障害を処置するうえで有効であるとの報告がある。そのような報告のなかには、6歳児における自閉症(非特許文献3)やうつ病(非特許文献4)に対する有効例がある。
しかし、BH4が、疲労の処置に有効であることは報告されていない。

【発明が解決しようとする課題】
本発明は、新たな抗疲労組成物、詳細にはビオプテリン関連物質を有効成分として含有する、疲労を回復させるためのまたは疲労を減弱させるための新たな組成物を提供することを目的とする。好ましい態様として、本発明は慢性疲労症候群を処置および過労死を予防する医薬組成物を提供する。

本発明者らは、ある種の疲労モデル動物を作製し、この動物実験系を用い、アスコルビン酸、アセチル−L−カルニチン、DHEA−Sと同様に、BH4が疲労減弱効果を有することを証明し、本発明を完成した。

【発明の実施の形態】
本発明は第1の態様として、ビオプテリン関連物質、即ちビオプテリン、6R−L−エリスロ−5,6,7,8−テトラヒドロビオプテリン(BH4)またはそれらの塩を有効成分として含有する抗疲労組成物を提供する。

本発明組成物が目的とする疲労とは、「身体的または精神的負荷を連続して与えたときにみられる一時的な身体的および精神的なパフォーマンスの低下」と定義され、ここに「パフォーマンスの低下」とは身体的および精神的作業能力の質的または量的な低下を意味する。本発明において、疲労とは以下に説明する慢性疲労症候群や過労死に至る状態を包含する意味を有する。
よって、本発明において「抗疲労組成物」とは、疲労を回復させるためのまたは疲労を減弱させるための組成物、を意味する。

本発明の有効成分であるビオプテリン関連物質とはビオプテリン、BH4、およびにそれらに類似した構造を有しかつBH4と同等の抗疲労活性を有している化合物を意味する。これらは当業者に良く知られており、そして容易に調製可能である。
ビオプテリンは化学名2−アミノ−4−ヒドロキシ−6−[1, 2−ジヒドロキシプロピル]プテリジンで表され、昆虫・両生類の皮膚・ロイヤルゼリーなどに広く分布する天然物質である(生化学辞典第3版p1088:東京化学同人)。ビオプテリンは、Crithidia Fasciculataの増殖因子であり、人尿からも分離されている。ビオプテリンは商品名L−Biopterinとして和光純薬工業(株)から市販されている。BH4はビオプテリンを酸性条件下に接触還元して得られる物質であり、Cayman Chemical Companyより品名「Tetrahydro−L−biopterin, Dihydrochloride」として市販されている。
ビオプテリンおよびBH4の塩としては、塩酸塩、硫酸塩等が挙げられる。

本発明の抗疲労組成物は医薬品や食品として使用できる。医薬品とは薬事法に規定される医薬品を意味する。食品とは「口から摂取されるもの」と定義され、機能性食品、健康補助食品、栄養機能食品、特別用途食品、特定保健用食品等あらゆる経口投与可能なものが包含される。

本発明の組成物中、有効成分の含有量はそれが医薬品、食品の場合でそれぞれ変動する。食品では一般に、0.001〜5重量%、好ましくは0.01〜3重量%、さらに好ましくは0.05〜1重量%である。医薬品では一般に、0.1〜20重量%、好ましくは1〜15重量%、さらに好ましくは3〜10重量%である。有効成分の具体的な投与量は、性別、年齢、症状の程度などに応じて選択できる。例えば成人一日あたり、10〜2000mg、好ましくは30〜500mg、さらに好ましくは100〜300mg程度である。

本発明の抗疲労組成物の形態は特に制限されず、例えば錠剤、顆粒剤、細粒剤、粉末剤、錠剤、スティック状剤、丸剤、散剤、カプセル剤、トローチ剤、口中で舐めて溶かす舐薬やチュアブル剤などの経口固形製剤、およびエリキシル剤、シロップ剤、などの経口液剤などであってもよい。食品の場合、ドリンク剤などの飲用物であってもよい。医薬品の場合、非経口投与製剤、例えば軟膏剤、パップ剤などの経皮投与製剤、および乳剤、注射剤などの静脈投与製剤などであってもよい。

本発明の医薬組成物が目的とする慢性疲労症候群とは、日常生活に支障を来すほどの長期的な全身疲労感、倦怠感、微熱、リンパ節腫脹、筋肉痛、関節痛、精神神経症状などの基本的な症状を示す病状を意味する。本発明の医薬組成物は慢性疲労症候群を処置、即ち慢性疲労症候群の各症状を緩解し、正常な状態に移行させることができると考えられる。

本発明の医薬組成物が目的とする過労死とは、重度の過労状態にあり、身体的活力を保つことができないにもかかわらず、疲労を十分に感じることができなくなり、その結果、高血圧や動脈硬化が新たに招来あるいは悪化し、脳血管疾患や心疾患を発症して永久的労働不能や死亡に至った状態を意味する。
本発明の医薬組成物は慢性疲労症候群を処置することができ、それにより過労死を予防、即ち過労死に至る過程を妨害できると考えられる。

本発明組成物が抗疲労作用を示すことは、負荷強制水泳試験により確かめた。負荷強制水泳試験について簡単に説明する。十分な睡眠や休息姿勢をとることができない環境、例えば水温23±1℃の1.5cm水深ケージにて7週齢の雄SDラットを一定期間飼育(水浸飼育)する。この環境は、ラットにとって精神的にも肉体的にも常に休息できない状態である。この水浸飼育により、疲労動物モデルと過労死に至る動物モデルとを作成できる。水浸飼育したラットを、例えば、体重の8%のおもりを負荷させた状態で遊泳させ、10秒以上鼻が水没してしまうまでの時間を測定することにより、その疲労度が評価される。この評価法に従えば、2日および5日水浸飼育群ではおもりに抗った遊泳が十分にできなる。これにより、2日以上水浸飼育したラットは疲労動物モデルになりうると考えられ、5日水浸飼育したラットは疲労困憊した過労死に至る動物モデルであると考えられる。疲労に関連した脳モノアミン代謝の研究として、運動負荷により脳内セロトニンおよびドパミン神経系が活性化されることが知られている(Meeusen, R. et al., Sports Med., 20: 160−188, 1995)。ラットを使った実験では、セロトニンアゴニストであるクイパジン(quipazine)の投与で運動耐容能が低下する一方、セロトニンアンタゴニストであるLY−53,857の投与では上昇し(Bailey, S.P. et al., Int. J. Sports Med., 14: 330−333, 1993)、他方ドパミンアゴニストであるアンフェタミン投与で運動耐容能が上昇し(Bhagat, B. et al, Neuropharmacology, 12: 711−713, 1973)、6−ヒドロキシドーパミンで線条体を破壊することでは低下が認められている(Heyes, M.P. et al., Life Sci., 42: 1537−1542, 1988)。これらのことから、セロトニン神経系の活性化は疲労感に、ドパミン神経系の活性化は疲労した状態のもとで身体的活力を保つことに、それぞれ関与すると考えられる。上記水浸飼育ラットにおける脳モノアミン代謝を調べると、急性ストレス状態にあると考えられる1日飼育群ではセロトニン神経系活性化の指標である5−HIAA/5−HTの値が前頭葉皮質と視床下部で上昇し、ドパミン神経系活性化の指標であるDOPAC+HVA/DAの値が線条体と視床下部で上昇した。他方、重度の過労状態と考えられる5日飼育群では5−HIAA/5−HTおよびDOPAC+HVA/DAの値がコントロール群と比べて差がないレベルまでに低下していた。これは、5日飼育群では重度の過労状態により身体的活力を保つことができないにもかかわらず、疲労を十分に感じることができなくなることを示している。ヒトの場合でも同様のことが起こっていると考えると、疲労困憊に至った状態では過労状態の当人が自分が疲労していることを認識できていない可能性があり、なぜ過労死するまで働きつづけるのか、なぜ疲れているのに十分な休息をとらないのか、といった質問の答えのひとつになると考えられる。よって、この5日飼育群は過労死に至る動物モデルと考えられる。

以下に本発明を実施例によりさらに詳細に説明するが、本発明はこれら実施例に制限されるものではない。本発明は文部科学省科学技術振興調整費生活者ニーズ対応研究「疲労および疲労感の分子・神経メカニズムとその防御に関する研究」の成果である。

テトラヒドロビオプテリンの過労死予防効果
(1)過労死モデル動物の作成
本実験はAnimal Ethics Committee of Osaka City University(承認番号 00093)に承認されたものであり、Committee and Japanese Government Animal Protection and Management Law (No. 105)およびJapanese Government Notification on Feeding and Safekeeping of Animals (No. 6)のガイドラインに則って施行した。

7週齢の雄SDラットを水温23±1℃の1.5cm水深ケージにて飼育した。次いで、疲労度を評価するため、体重の8%のおもりを負荷させた状態で遊泳させ、10秒以上鼻が水没してしまうまでの時間を測定した(負荷強制水泳試験)。得られた結果は、1日水浸飼育群では変化がみられなかったが、2日および5日水浸飼育群ではおもりに抗った遊泳が十分にはできなくなり、より短時間で断念したことを示した。次いで、得られた水浸飼育ラットにおける脳モノアミン代謝を調べた。1日飼育群ではセロトニン神経系活性化の指標である5−HIAA/5−HTの値が前頭葉皮質と視床下部で、ドパミン神経系活性化の指標であるDOPAC+HVA/DAの値が線条体と視床下部でそれぞれ上昇し、他方5日飼育群では5−HIAA/5−HTおよびDOPAC+HVA/DAの値がコントロール群と差がないレベルまでに低下していた。これは、5日飼育群ラットが、重度の過労状態では身体的活力を保つことができないにもかかわらず、疲労を十分に感じることができなくなっていることを示している。これにより、1日飼育群ラットは急性ストレス状態に、他方5日飼育群ラットは重度の過労状態、即ち過労死に至る状態にあると考えられる。

(2)テトラヒドロビオプテリンの過労死予防効果
疲労減弱効果があると推定されている数種の化合物とともに、テトラヒドロビオプテリン(BH4)の疲労に対する治療および予防効果について、上記(1)の過労死モデルを用いて検討を行った。
5日水浸飼育した状態で1日目より1日1回腹腔内に生理食塩水(コントロール)、アスコルビン酸(300mg/kg)、BH4(20 mg/kg)、アセチル−L−カルニチン(100 mg/kg)、デヒドロエピアンドロステロンサルフェート(DHEA−S, 10 mg/kg)をそれぞれ投与し、上記(1)記載の負荷強制水泳試験を行った。得られた結果を図1に示す。

図1は、アスコルビン酸、アセチル−L−カルニチン、BH4、DHEA−Sがコントロール群と比べおもり負荷強制水泳試験において、より長時間の遊泳を可能とすることを示している。以上より、BH4はアスコルビン酸、アセチル−L−カルニチン、DHEA−Sとともに疲労減弱に有効であると考えられた。

有効成分としての6R−L−エリスロ−5,6,7,8−テトラヒドロビオプテリン二塩酸塩(Cayman Chemical Company)20mgおよびL−アスコルビン酸50mg、賦形剤としてのD−マンニトール330mg、乳糖100mgおよび結晶セルロース20mg、結合剤としてのヒドロキシプロピルセルロース50mg、ならびに滑沢剤としてのステアリン酸マグネシウム30mg、をそれぞれ量り取り、合計600mgの錠剤を、日本薬局方製剤総則、錠剤の項に順じて製造した。

抗疲労組成物を提供することにより、医療費高騰問題等を背景に注目されている予防医学に貢献できる。また、多忙な現代人を襲う過労死を予防し、さらに慢性疲労症候群の処置が実現することから、疲労を原因とする問題解決に大いに貢献できると考えられる。
