【発明の属する技術分野】
本発明は、粉体原料の表面特性を改質し、有効な薬物含有複合粒子を製造する複合化処理装置に関する。

【従来の技術】
医薬品製剤には、例えば、製造時における取り扱い易さ(ハンドリング性)、苦みのマスキング、溶解性の制御、DDS(Drug Delivery System)特性などの多様な特性が要求されるため、従来より、必要な性質を付与するために、複数の原料を複合化することが行われている。複数の原料が複合化されたものとしては、例えば、賦形剤と薬物とが複合化されたもの、薬物の表面が滑沢剤やコート剤により覆われたもの等が挙げられる。ここで、上記賦型剤は薬物の取り扱い性を向上させて、所望の形態に製剤化することを容易にする役割を担い、滑沢剤は薬物の表面を滑らかでつやのある状態にする役割を担い、コート剤は、薬物の表面を覆うことにより、例えば、薬物の苦みをマスキングする役割を担うものである。

このような、薬効成分を含む原料の複合化においては、安定性を確保することが特に重要であるため、複合化に要する時間が短く、薬効成分の安定性低下の問題も生じない複合化方法が望まれている。

そこで、特許文献1において、薬物粉末を含む2種以上の粉末原料よりなる混合物に圧縮力および剪断力を加えて複合化する方法が提案されている。特許文献1の粉体処理装置を、図6を参照しつつ以下に説明する。

上記粉体処理装置61は、図6に示すように、大略的に、略円筒形状の閉空間を形成するケーシング62、当該ケーシング62の内部に設けられた同じく有底略円筒形状の筒状回転体63、当該筒状回転体63の内周面に対して押圧力および剪断力を発生させて混合物を処理すべく上記筒状回転体3の内部に配設したプレスヘッド64とからなる。上記筒状回転体63を回転させることで、当該筒状回転体63の内周面に形成した受け面65と上記プレスヘッド64とを相対回転させ、上記受け面65と上記プレスヘッド64との間に間隙として形成されているクリアランス60に存在する混合物67に押圧力および剪断力を付与して複合化処理を行うものである。

また、粉体処理装置61では、混合物67はケーシング62の混合物投入口70よりその処理空間69に投入され、筒状回転体63とプレスヘッド64とにより強力な圧縮力・剪断力を受けることにより複合化処理される。

さらに、上記ケーシング62の周囲には、主に上記処理空間69の温度を調節するためのジャケット73を設けている。当該ジャケット73へは、別に設けたタンク(図示せず)からの加熱媒体又は冷却媒体が必要に応じて循環供給される。これにより、上記ケーシング62の内部温度を調節することができる。例えば、混合物67として、温度変化によって変質する虞のある薬物を処理する場合においては、ジャケット73に加熱媒体又は冷却媒体を循環供給することにより、処理中の混合物67の温度を所望の温度として薬物の変質を防止することができる。

【発明が解決しようとする課題】
しかしながら、特許文献1の方法では、循環した混合物がケーシングに高い圧力で押し付けられて付着し、これにより排出性の低下や温度上昇が生じ、この温度変化によって変質する虞のある薬物を複合化する場合に、より一層効率的に混合物や複合化処理部分を冷却する方法が求められている。

そこで、特許文献1の方法を応用しつつより効率的にかつ安定的に薬物粉末を複合化させるためのさらなる改良が課題となっていた。

【課題を解決するための手段】
本発明の複合化処理装置は、上記の課題を解決するために、薬物粉末を含む2種以上の粉末原料よりなる混合物に圧縮力および剪断力を加えて複合化する複合化部と、上記複合化部を密閉空間に閉じこめるケーシングと、を有する複合化処理装置において、上記ケーシングの形成する密閉空間が、上記ケーシングにおける複合化部の高さを1とした場合に、0.7以上1.25以下の高さの、混合物が分散されるための循環空間を含んでいることを特徴としている。

ここで、複合化部とは混合物に圧縮力および剪断力を発生させる部分を示す。複合化部の高さとは、ケーシングの壁面が、複合化部を内包する内径が略一定な垂直部分と、上記垂直部分から連続的に内径が小さくなる部分とからなっている場合の、垂直部分の高さが複合化部の高さである。なお、内径が一定な垂直部分は、例えば円筒形である。

上記複合化部の高さと循環空間の高さとの比は、ケーシングの形成する空間における複合化部の占有割合を示しており、言い換えればケーシングの形成する空間における循環空間の割合を示している。この比を上記範囲とすることで、混合物が適度に循環できる空間が確保され、複合化の進行を妨げずに混合物の冷却が行われる。

上記複合化部の高さを1とした場合の、循環空間の高さが1.25より大きいと、密閉空間が広くなりすぎ、混合物が広範囲に分散して、圧縮力、剪断力を効率的に加えることができない。一方、上記複合化部の高さを1とした場合の、循環空間の高さが0.7より小さいと、密閉空間が狭くなりすぎる。そのため、循環が抑えられたり、循環途中の混合物がケーシングに付着したり、混合物が空間中を分散する時間が短くなって生じた熱が放散されず、混合物の物性が変化してしまうなどのおそれがある。

そこで、この比を上記範囲とすることで、混合物が適度に循環できる空間が確保されるため、混合物の循環効率、つまり複合化部に混合物が供給される率を低下させることなく、混合物のケーシングへの付着が防がれ、また混合物の熱も放散する。

本発明の複合化処理装置は、上記の課題を解決するために、上記複合化部が底部を有する筒形状の回転部材を含んでおり、上記循環空間が複合化部の上記底部と反対側に隣接して設けられてことを特徴としている。

上記複合化部を底部を有する筒形状とすることで、混合物が落ちることなく筒内に集まり、効率よく複合化を行うことができる。このとき、混合物は底部から離れる方向に分散するため、上記循環空間は複合化部の底部と反対側に設けられていることが望ましい。

本発明の複合化処理装置は、上記の課題を解決するために、さらに、上記ケーシングが、上記混合物を投入する、あるいは複合化後の混合物を排出するための投入排出口を有し、上記投入排出口の複合化部に対する鉛直方向の位置が変わるように、上記ケーシングが鉛直方向に回転可能に設置されていることを特徴としている。

ケーシングが鉛直方向に回転するとは水平方向の軸を中心に回転することであり、これにより上記投入排出口の複合化部に対する鉛直方向の位置が変わる。例えば、ケーシングの真上に投入排出口がある場合は、投入排出口は複合化部に対して上の位置にある。ここで、ケーシングを鉛直方向に回転させることで、ケーシングを反転させ、投入排出口がケーシングの真下にくる状態にする。このとき、投入排出口は複合化部に対して下の位置になる。

このように、投入排出口の複合化部に対する鉛直方向の位置が変わるため、複合化部への混合物投入および排出が容易にでき、また回収率が向上する。つまり、投入排出口が複合化部より上にあるときに、混合物を投入排出口から投入することで混合物が容易にケーシング内に投入される。そして、複合化完了後に、投入排出口を複合化部より下に位置させることで、容易に投入排出口から混合物を排出でき、中の混合物を効率よく排出できる。

本発明の複合化処理装置は、上記の課題を解決するために、上記ケーシングが、上記投入排出口に向かって狭まる方向へ傾斜する傾斜部を有することを特徴としている。

上記構成とすることで、複合化部から投入排出口まで、すなわちケーシング壁面から投入排出口まで緩やかな傾斜が形成されるため、混合物がスムーズに投入排出口から排出される。

テーパー角度が80度より小さいと、壁面から投入排出口まで傾斜を形成しようとすると、投入排出口20が必要以上に大きくなるので、装置の大型化やコストアップにつながり、効率的でなくなる。また、投入排出口20を大きくする代わりに、投入排出口20周辺に水平部分を作ると、排出時に混合物が排出されずに残ってしまう。一方、テーパー角度が100度より大きいと混合物がスムーズに投入排出口に導かれない。

一方、円錐のテーパー角度が80度以上100度以下であれば、投入排出口が真下に向いたときに、テーパー角度により40度から50度の範囲で傾斜した斜面が壁面から投入排出口に向かって形成されるため、混合物が投入排出口に誘導され、排出される。

本発明の複合化処理装置は、上記の課題を解決するために、上記投入排出口は容器を接続できるバルブを備えており、上記バルブが容器接続後に開放可能なものであることを特徴としている。

上記構成によれば、毒性の強い混合物であっても、使用者に暴露することなく複合化を行うことができる。また、他の物質の混入が防がれ、汚染されることなく複合化した混合物を排出できるので、特に薬物等の製造に有用である。

【発明の実施の形態】
本発明の薬物含有複合粒子の製造に用いることができる粉体処理装置を、図1および図5を参照しつつ以下に説明する。

図2は、粉体処理装置の外観を示す斜視図である。粉体処理装置は、粉体処理部21と、コントローラー26とからなる。

また、図3は、粉体処理部21の正面図である。粉体処理部21は、処理部1、軸および軸受で構成される駆動部22、これに連結するモーター23からなる本体27と、支持体24・25からなる。

粉体処理部21は、処理部1に投入された混合物を、剪断力および圧縮力が加えて、複合化するものである。処理部1については後に詳細に説明する。モーター部23は、駆動部22を介して処理部1に接続しており、複合化の駆動源となる。

また、コントローラー26は、図4に示すように、電源スイッチ31、メインパネル32、流量モニター36、流量設定スイッチ33、圧力モニター34、圧力設定スイッチ35からなり、処理部1内の環境を表示したり、使用者が環境設定を指示するためのものである。電源スイッチ31は、粉体処理部21の電源のオンオフを行う。メインパネル32は、筒状回転体3の回転数やモーター23の消費電力量を表示したり、使用者が設定を指示するボタンがある。流量モニター36はセンサーで測定した筒状回転体3の軸封用エアーパージの流量を表示し、流量設定スイッチ33は使用者が所望の流量設定を指示する。圧力モニター34は軸封用エアーパージの圧力を表示し、圧力設定スイッチ35は使用者が所望の圧力設定を指示する。

次に、処理部1への混合物の投入・排出について説明する。
粉体処理部21には、ポット29が接続可能となっており、ポット29に含有された混合物を粉体処理部21に投入したり、複合化された混合物をポット29に排出したりする。このときの粉体処理部21とポット29との間の混合物の移送は、混合物が毒性の強い物質であった場合を鑑み使用者に暴露しないように、あるいは混合物への他の物質の混入を防ぐために、ハイコンテインメントとする、すなわち、粉体処理部21とポット29との間を移送される間、混合物の外部との接触を防ぐことが好ましい。

そのために、例えば接続用のバルブとして、特にスプリットバタフライバルブを用いることが好ましい。この場合、2つの部品からなるスプリットバタフライバルブの一方を、ポット29の口に取り付け、粉体処理部21の投入排出口20に他方を取り付ける。バルブは、この時点では両者の口を塞ぐ蓋として機能する。

そして、粉体処理部21にポット29を接続させると、ポット29に取り付けたバルブの部品と、粉体処理部21に取り付けたバルブの部品とがドッキングし、ふた部分が開いて、ポット29から粉体処理部21へ、あるいは粉体処理部21からポット29へ中身を流入させる。なお、スプリットバタフライバルブとしては、PSLチャージングポイント(PSL社製)を用いることができる。また、ポット29としては、ポリプロピレン等からなる使い捨ての容器を用いることで、コンタミネーションを防ぐことができる。

その他には、ポット29として、柔軟性のあるプラスチックバッグ、例えばGEA BUCK(登録商標)Bag(GEA Buck Valve)を使用し、バルブとしてプラスチックバッグを挟み込んで密閉する、例えばhicoflex(登録商標)(GEA Buck Valve)を用いてもよい。

また、粉体処理部21は、本体27全体がY軸(水平軸)を中心に鉛直方向(A方向)に回転(縦回転)することで、ポット29からの混合物の投入、およびポット29への混合物の排出の両方を容易に行うことができる。本体27の縦回転は、処理部1に接続した駆動部22からY軸方向に伸びた軸を、支持体24・25の軸受部に接続させることで行われている。なお、本体27の縦回転は、使用者が支持体25に取り付けられた回転レバー28を操作することで制御される。

このように本体27が縦方向に回転可能なので、ケーシング2に投入排出口20から混合物を投入し、粉体処理を行った後、本体27を回転させて混合物を投入排出口20から排出することができる。

次に、処理部1について、図1を用いて説明する。
処理部1は、図1に示すように、大略的に、略円筒形状の閉空間を形成するケーシング2、当該密閉容器2の内部に設けられた同じく有底略円筒形状の筒状回転体3、当該筒状回転体3の内周面に対して押圧力および剪断力を発生させて混合物を処理すべく上記筒状回転体3の内部に配設したプレスヘッド4とからなる。上記筒状回転体3を回転させることで、当該筒状回転体3の内周面に形成した受け面5と上記プレスヘッド4とを相対回転させ、上記受け面5と上記プレスヘッド4との間のクリアランス100に存在する混合物に押圧力および剪断力を付与して複合化処理を行うものである。

図1に示すように、上記ケーシング2の内部には、略円筒形状であって鉛直方向の回転軸Xを中心にB方向に回転自在な筒状回転体3を備えている。当該筒状回転体3は、回転軸部12と当該回転軸部12に連接した底部14、及び当該底部14に連接した円筒壁部15とにより構成されている。

上記筒状回転体3の底部14は、上記回転軸部12と筒状回転体3の円筒壁部15とを連結する機能、および、上記混合物を保持する保持手段としての機能を有する。即ち、当該底部14は、後述する円筒壁部15との関係において互いの面が折れ曲がった関係にあり、筒状回転体3が回転する際に、混合物が充分に処理されずにクリアランス100から下方に逃げてしまうのを防止する。

上記円筒壁部15の内周面は、遠心力を受けて外向きに移動しようとする混合物の受け面5となる。即ち、混合物を上記クリアランス100に留めておき、上記受け面5と上記プレスヘッド4との協働によって混合物に押圧力および剪断力を付与して粉体処理を行う。

このとき、粉体処理をされている混合物の一部は、円筒壁部15の下部に設けられた円筒壁部15を貫通する開孔43を通って、プレスヘッド4側からケーシング2側へと移動する。ケーシング2側に移動した混合物は掻き揚げ羽根42により上方に掻き上げられ、円筒壁部15を越えて再びクリアランス100に入る。このようにすることで、効率的に粉体処理が行える。

粉体処理装置21は、上記ケーシング2の内部の雰囲気を、混合物の種類等に応じて適宜変更することができるものであってもよい。例えば、不活性ガスや加熱ガス等の各種のガスを投入排出口20からケーシング2の内部に投入したり、加圧・真空ポンプ等を用いてケーシング2の内部を加減圧する構成としてもよい。この場合には、例えば、ケーシング2と筒状回転体3の回転軸部12との間に、図示しないシール部材を設けることにより、処理空間の内部の雰囲気を確実に調整することができる。シール部材は、また軸回転により発生するカス等が処理空間に侵入することも防ぐ。

また、上記ケーシング2の周囲には、主に処理空間の温度を調節するためのジャケット(図示せず)を設けてもよい。当該ジャケットへは、別に設けたタンク(図示せず)からの加熱媒体又は冷却媒体が必要に応じて循環供給される。これにより、上記ケーシング2の内部温度を調節することができる。例えば、混合物として、温度変化によって変質する虞のある薬物を処理する場合においては、ジャケットに加熱媒体又は冷却媒体を循環供給することにより、処理中の混合物の温度を所望の温度として薬物の変質を防止することができる。

さらに、例えば、図5に示すように、プレスヘッド4内に管(内部空間)41を形成しておき、冷却水等の冷却媒体を通過させて、プレスヘッド4を冷却させても良い。この場合、プレスヘッド4内部に通された管41に対して、タンクから給水口を介して冷却水を供給し、管41の水を排出口を介して排出する構成とすればよい。

ここで、ケーシング2の形状について説明する。

粉体処理装置21における粉体処理は、投入された混合物が筒状回転体3とプレスヘッド4によって挟まれ、圧縮力、剪断力を加えられることによって行われている。このとき、ケーシング2内の筒状回転体3の上部空間が広いと、混合物が上部空間に分散してしまい、クリアランス100を通過する効率が悪くなる。言い換えると、混合物を効率的にクリアランス100に導き、効率的な複合化を起こさせるには、ケーシング2における筒状回転体3より上部が、できるだけ狭い空間となっていることが望ましいと言える。

また、処理部1では混合物の物性を保つためには、混合が高温とならないようにする必要がある。複合化はモーター部23の駆動力による筒状回転体3の回転により行われることから、筒状回転体3に熱が発生する。さらに、複合化が、筒状回転体3とプレスヘッド4との間に挟まれた混合物に圧縮力と剪断力とを加えて行われるため、この処理箇所でも摩擦熱が発生する。混合物が加熱に弱い物であった場合、この熱が混合物に悪影響を与え、複合化によって混合物の本来の性質を損なってしまうことになる。また、低融点の粉体などでは融着を起こして安定運転が出来なくなる可能性もある。

そのために、装置や混合物に蓄えられる摩擦熱等を効率的に逃がす形状であることが望ましい。この放熱のしやすさ、言い換えると冷却効率の観点からは、混合物が分散して放熱できるように、あるいは装置が空間に分散するように、ケーシング2における筒状回転体3の上部に、大きな空間が形成されていることが望ましい。

そこで、ケーシング2の内部での混合物質の循環効率を損なわずに、冷却効率を向上するためには、筒状回転体3の上部に適切な大きさの空間が設けられていることが必要である。空間の高さの比で言うと、本実施の形態の、筒状回転体3を内包する円筒部分と、それに続く円錐部分とからなるケーシング2では、高さ方向(ケーシング2の筒状回転体3の軸方向)における、上記円筒部分の長さをL1とし、ケーシング2の上記円錐部分の長さをL2とした場合のL1/L2が、0.75以上1.25以下であることが望ましい。L1/L2はさらに下限値が0.85以上であることが望ましく、上限値が1.05以下であることが好ましい。言い換えれば、ケーシング2の形成する空間のうち、高さにおける40%〜55%の、上部の空間が循環空間として確保されていることが好ましい。これによれば、後の実施例に示すとおり、複合化の効率を損なわずに混合物やプレスヘッド4の熱を良好に除くことができた。

また、処理部1は、複合化が完了した混合物を排出する際には、投入排出口20側を下に向けて混合物を排出する。このとき、ケーシング2は壁面の円筒形状が投入排出口20に向かって円錐状に狭まるようなテーパー形状であることが望ましい。これは、ケーシング2から混合物を排出するときに、混合物が投入排出口20に到達するまでに、図1の破線に示すような、鉛直方向(軸X方向)と直交する方向(水平方向)のケーシング2の面があると、混合物が水平面に残ってしまい、全量を排出できなくなってしまうからである。したがって、テーパー角度を調節する際は、投入排出口20の口径を広げるあるいは狭めるようにする。このようにすることで、水平面を作らずにテーパー形状の傾斜が直接に投入排出口20に続く構成とできる。

なお、テーパー角度は80度以上100度以下であることが望ましい。また、下限値は85度以上であることがさらに好ましく、上限値は95度以下であることがさらに好ましい。これによれば、壁面から投入排出口20に向かう傾斜が混合物をスムーズに投入排出口20に導く。テーパー角度が100度より大きいと、傾斜が水平方向に近くなるため、混合物をスムーズに投入排出口20から排出することができない。一方、テーパー角度が80度より小さいと、結果的に投入排出口20までの長さL2が長くなったり、投入排出口の口径が大きくなってしまい装置全体が大きくなってしまう。

なお、kが0.75以上1.25以下であり、かつテーパー角度が80度以上100度以下であると、冷却効率、循環効率、排出効率の度の面から見て非常に良好な結果が得られた。

【実施例】
以下に、図1に示す、実施の形態の粉体処理装置のケーシング2形状を変化させ、その複合化効率、冷却効率、排出率への影響を調べた。

ケーシング2は、内径が95mmΦ円筒形状部分と、これに続く円錐形状(コニカルテーパー構造部分)とからなり、コニカルテーパー構造が投入排出口20に接続されるようにした。このケーシング2形状において、投入排出口20の内径とコニカルテーパー構造部分の大きさとを変更しながら、ケーシングディメンションと、ケーシング角度αとを変化させて実験を行った。

ここで、ケーシングディメンションは、複合化処理を行う部分の上部にどのぐらいの空間を有しているかを示しており、つまりケーシング2の全容積に対するローター(筒状回転体)3以外の空間が占める割合を示している。具体的には、図1のケーシング2における複合化処理を行う円筒形状部分(複合化部)を下部ケーシング、円錐形状部分を上部ケーシングとした場合の、上部ケーシングの高さL1/下部ケーシングの高さL2をケーシングディメンションを表すディメンション比kとした。この場合、kが大きいほど上部の空間が大きいことを示す。本実施例では、kが0.5、0.75、0.9、1.25、1.5の装置について測定した。

また、ケーシング角度αとは、円錐形状部分のテーパー角度α、つまりケーシング2の中心縦軸Xを通る断面の円錐形状部分の傾斜間の角度を示している。ケーシング角度αは、45度、60度、75度、80度、90度、100度、105度、120度とした装置について測定した。なお、角度αが大きい場合は排出口の口径を大きくし、角度αが小さい場合は、排出口の口径を小さくした。これにより、種々の角度α、kの値を有する装置を作製した。

その他の条件は、すべて同等とした。条件としては、ローター3とプレスヘッド4との間隔を1mmに設定し、ローター回転数2000rpmで、30分複合化処理を行った。このとき、ジャケットには0℃のエタノール水溶液を循環(流量:1.5L/min)させた。
