D−アミノ酸は、生物学的に活性な製造物、例えばβ−ラクタム、ペプチドホルモン及び殺虫剤のための重要な基礎単位である。これらの「非自然な」アミノ酸の、一般的に使用される製造法は、対応するDL−5−置換のヒダントインが鏡像体選択的にヒダントイナーゼで加水分解され、対応するN−カルバモイルアミノ酸を製造する方法である。このN−カルバモイルアミノ酸は対応するD−アミノ酸に酵素的に転化される。

公知の方法の欠点は、相当な反応時間を実現すること、相対的に大量の生物触媒が必要であることである、なぜならカルバモイル酸の加水分解の原因である酵素(カルバモイラーゼ、N―カルバモイル−D−アミノ酸アミドヒドラーゼともよばれる)は、反応生成物であるアンモニアに強く阻害されるからである。アンモニアにより引き起こされる阻害が起きる濃度は、非常に低い(〜10mM)と報告されている(Olivieriら、 “Biotechnology and Bioengineering”,23,pp2173〜2183(1981);Runser,“Applied Microbiology and Biotechnology,”33,pp382〜388(1990);Louwrier“Enzyme and Microbial Technology,19,pp562〜571,(1996))。公知の方法について、このことは、10mMのアンモニアの濃度において、カルバモイラーゼは最大速度の半分の活性であることを意味する。20mMのアンモニア濃度において、カルバモイラーゼの速度は最大速度の1/3に過ぎない。高い生成物濃度、例えば>500mMが所望される、工業的に関連する条件下において、カルバモイラーゼの速度は非常に強く阻害されるであろうということは明らかであろう。カルバモイラーゼの活性がほとんど最大である、より低い生成物の濃度(大きさ10mMのオーダー)は、その低い生産能力のために経済的に魅力がない。

文献において、酵素的な転化反応の間においてアンモニアの濃度を低く保つための様々な方法が記載されている。インシチューでのアンモニアの除去は、Kim及びKim“Enzyme and Microbial Technology”17,pp63〜67(1995)において記載されており、ヒダントイナーゼ及びカルバモイラーゼを用いるD−p−ヒドロキシフェニルグリシンの製造の間のアンモニアの除去のために、吸着剤(AD300NS,トミタ薬品(株)(日本)のシリケート錯体)が利用される。この方法の欠点は、吸着剤が高価であり、従って回収されなければならないことである。

日本特許出願公開第61285996号(1986年)において記載される、もう1つの方法に従って、生成されるアンモニアは蒸留により除去される。しかし、アンモニアの除去のためのこの方法は、反応混合物が高いpHを有するときのみ有効である。そのような高いpHにおいては、しかし、カルバモイラーゼ酵素は活性ではない。アンモニアが中性のpH、7〜7.5のpHにおいて蒸留により除去されるとき、生成されるアンモニアの60〜90%は、反応体積の90%より多くが除去されたときでさえ、反応混合物中に残る。従って中性pHにおける効果的な除去は可能ではない。

本発明は、所謂ヒダントインルートによる鏡像体に富むアミノ酸の製造における使用に特に適しており、該ヒダントインルートは、任意的にラセマーゼと組み合わされた、ヒダントイナーゼによる対応するヒダントインからのD−鏡像体に富むN−カルバモイルアミノ酸の製造、続くD−カルバモイラーゼにより脱カルバモイル化を含み、ここで脱カルバモイル化が全体的な律速段階である。燐酸塩及び/又は燐酸アンモニウム塩の存在は、ヒダントイナーゼ、カルバモイラーゼ及び/又はラセマーゼに阻害的又は変性させる効果を有しないことが見出された。公知の方法においては、これらの転化が起きるpHはほとんど7〜8に存在する。なぜならより高いpH値においては、NH3の存在のために実質的に完全な酵素の阻害が起きるからである。およそ中性のpHにおいて反応を行うことの欠点は、ほとんどの全てのアミノ酸、特に脂肪族のアミノ酸の製造において、後に残っているヒダントイン鏡像体の遅いラセミ化が起きることである。

本発明に従う反応がより高いpH、例えば7.0〜9.0、好ましくは7.5〜8.5のpHにおいて行われるとき、酵素阻害はほとんど全く起こらないが、望まれない鏡像体のラセミ化は起きることが今見出された。その結果、ラセミ化がなしで理論的な可能な最大である50%よりずっと高い収率が達成される。

本発明に従う方法は、D−選択性のヒダントイン−加水分解性及びN−カルバモイルアミノ酸―加水分解性酵素を含む微生物を用いて、DL−N−カルバモイルアミノ酸が、D−鏡像体に富む対応するアミノ酸、及び鏡像体に富む転化されなかったL−N−カルバモイルアミノ酸に転化される分割方法においてもまた使用されることができる。該反応は高められたpH(例えば7.5〜9)において行われることができるので、L−Nカルバモイルアミノ酸の損失はより少ない、なぜなら高められたpHにおいてヒダントイナーゼの反応は進行しないからである。
【実施例1】

34gのDL−p−ヒドロキシフェニルグリシンのヒダントイン及び34gのMgHPO4・3H2Oが200mlの水に添加された。反応混合物のpHが5NNaOHによりpH=7.2に調節されたのち、34mlのAgrobacterium radiobacterの細胞サスペンジョンの添加により反応が開始された。反応は窒素雰囲気下40℃において行われた。反応混合物のpHは、5NのNaOHの添加によりpH7.2において一定に保たれた。HPLC分析により、>99%の添加率が10時間の加水分解後にすでに達成されたことが確認された。

比較実験1
実施例1に記載されたものと共通の条件下であるが燐酸塩のない状態において、34gのDL−p−ヒドロキシフェニルグリシンのヒダントインが加水分解された。34mlの同じAgrobacterium radiobacterの細胞サスペンジョンの添加により反応が開始された。反応は窒素雰囲気下40℃において行われた。反応混合物のpHは、1.3MのH3PO4による自動的滴定によりpH=7.2において一定に保たれた。反応の進行はHPLC分析により定期的に確認された。約10時間の加水分解後、約57%の転化率が得られたことが記録された。30時間後に測定された転化率は>99%まで達した。
【実施例2】

DL−p−ヒドロキシフェニルグリシンのヒダントインの加水分解が、122gのこの化合物を122gのMgHPO4・3H2Oともに575mlの水に添加することにより行われた。この混合物のpHは5NのNaOHでpH=7.2に調節され、その後窒素ガスが1時間通された。酵素的転化は、8mlのAgrobacterium radiobacterの細胞サスペンジョンの添加により開始された。反応混合物のpHは、NaOH(25重量%)の添加によりpH=7.2において一定に保たれた。全部で19gのNaOHの消費が記録された。95時間の加水分解後、99.3%の転化率が測定された。

【実施例3】

【実施例4】
