【発明の背景】
現代工業社会においてゴム製品は至るところに使用されており、ゴム製品なくして社会が成り立たないといっても過言ではない。ゴムはそのように重要な素材であるが、2000年のゴム素材の日本での消費実績は、天然ゴムで約750千トン、代表的な合成ゴムのSBRで約1,140千トンである。言うまでもなくSBRは石油由来のものであり、鉱物資源の使用、地球環境保全(炭酸ガスの排出)の観点からはSBRの消費を減少、節減することが望ましい。したがって、SBRの消費分を天然ゴムで代替すればよいが、天然ゴムは南方地域にその資源的偏りがあり、必ずしも代替がスムーズに行われない。

天然ゴムはゴムの木から採取される樹液を原料とするシス型のイソプレンゴムであるが、その他のゴム資源としては温暖域においても生育可能な杜仲も天然ゴムを産生することが知られている。石油系合成ゴムの発明以前には杜仲からもゴム物質が採取され、海底電線の被覆材、ゴルフボールの被覆材、また、歯科材料などに広く利用されていた。ところが、合成ゴムが安価に合成されるようになったため、これらはほとんど合成ゴムにとって替わられた。

そこで、植物系天然ゴムの生産を復活させることは、地球環境規模においてもきわめて意義あることである。植物系天然ゴムの生産には、言うまでもなく植物を育成することが必要である。これは、先に述べたように植生に地域性があり、特にシス型イソプレンゴム(所謂、天然ゴム)は、南米、東南アジアといったごく限られた地域でしか生産されないからである。したがって、世界中のどこでも植物系天然ゴムの生産が達成されれば、これはゴム生産産業に計り知れない利益をもたらすものである。そして、その技術は地球環境保全の観点から人類にとっても重要な意味を持ってくる。

本発明は、このような実状に鑑み、植物の木としての生育を経由せずに、その植物の培養細胞によって、直接植物系のゴムを提供することを企図したものである。したがって、本発明方法によれば、特殊な地域での植物の栽培、広大な育成面積を必要とせず、工場内でのゴム生産が可能である。

【従来の技術】
通常、植物系天然ゴムを得るには、植物を栽培し、その植物体内でゴム物質を生合成させ、それを採取している。しかし、植物体は必ずしも完全な木まで成長させずともカルスなどの植物培養体においても固有の成分(二次代謝物質)を生合成できることは知られている。

しかし、一般的にはカルス等の植物培養組織は植物体と比べて二次代謝物質の生産能力に劣り、従来、植物培養組織においてゴム物質を生合成(二次代謝)させることに関する報告はない。

【発明が解決しようとする課題】
本発明は、植物の培養細胞によって植物系天然ゴムを生産する方法を提供することを課題とする。

【課題を解決するための手段】
本発明は、植物培養組織体において、ゴム物質を生合成させタンク培養によってゴム物質を生産する方法を提供するものである。

すなわち本発明は、植物生長ホルモンおよび炭素源を含む植物組織培養用培地中で培養、育成した杜仲植物体の根組織よりゴムを得ることを特徴とする植物培養によるゴムの生産方法である。

好ましい植物組織培養用培地は、MS培地、LS培地、ガンボーグB5培地、White培地、Nitsch培地またはこれらを微改変した培地である。

好ましい炭素源は、シュークロース、グルコース、フルクトース、キシロース、キシルロース等の糖類である。

好ましい植物生長ホルモンは、オーキシン類およびサイトカイン類の少なくとも一方である。

【発明の実施の形態】
つぎに、この発明を具体的に説明するために、この発明の実施例およびこれとの比較を示すための比較例をいくつか挙げる。

実施例1
愛媛県生名島の杜仲雌株より採集した種子を2重量%の蔗糖を含むゲルライト培地(ゲルライト濃度0.24重量%)に無菌的に播種した。播種後40〜60日経過した幼植物体から根を2〜3cmに切断し、根の切断片を採取した。採取した根切断片を、培地を50mlずつ分注したコニカルビーカーに3本ずつ入れ、無菌的に振とう培養を行った。

培養には、MS培地(Murashige et a1., 1962)に、2,4−ジクロロフェノキシ酢酸(2,4−D)およびα−ナフタレン酢酸(NAA)、3−インドール酪酸(IBA)、3−インドール酢酸(IAA)(以上オーキシン:植物性ホルモン)と6−ベンジルアミノプリン(BAP)を種々の濃度で加え、それぞれに蔗糖(濃度2重量%)を加えた培地を用いた。

温度を25℃に一定に保ち、光合成有効光量子束密度(PPFD)50μ mo1(/m2・s)、明期16時間/日の照明下で振とう培養し、振とう回数は100〜110回/minとした。

根の培養に適したホルモン濃度を決定するために、まず、表1から分かるようにNAAとBAPを種々の濃度で組み合わせた。この結果、BAPの濃度が高くなると根が膨張する傾向にあった。そして、NAAが0.1μM、1μMで、BAPが0μM、0.1μM、1μMである組み合わせの培地(No.2、No.3、No.6、No.7、No.10、No.11)では根の伸長が見られた。そこで、これらの培地で伸長した根の本数および長さの合計を調べたところ、No.3(NAA:1μM、BAP:0μM)の培地で最も大きな値が得られた。その次に大きな値が得られたのはNo.7(NAA:1μM、BAP:1μM)の培地であるが、この培地では培養後4週間目にカルスが形成される傾向があった。

根の伸長の状況は次の通りである。根は培養開始後10日から2週間目で伸長し始め、No.2(NAA:0.1μM、BAP:0μM)の培地では培養後4週間目に、No.3(NAA:1μM、BAP:0μM)の培地では培養後6週間目に、それぞれ著しい根の伸張が見られた。また、No.6、No.7、No.10、No.11の培地でも根の伸長が見られたが、これらの培地では培養後4週間目にカルスが形成されたり根が膨脹する傾向があり、培養の初期段階において効率のよい根の伸張が見られるのはNo.2とNo.3であった。

培養開始後4週間目におけるNo.2(NAA:0.1μM,BAP:0μM)の培地の伸長した根の重量は、0.05〜0.15g増加した。

以上の結果から、根の振とう培養ではBAPを加えると根が膨張し、根を効率よく伸長させることが出来ないこと、および、低濃度のNAAのみであればカルスを形成しないことが分かる。したがって、根の増殖の初期段階にはオーキシンNAAが最も有効で有り、その濃度は0.1μM〜1μM(試験No.2とNo.3)が最も有効である。

比較例1
実施例1で見たように、根の培養にはBAPは必ずしも必要ではなく、オーキシンのみでもよいと考えられたため、他のオーキシンを用いて根の生育状況を試験した。その結果、2,4−D(表2、試験No.17〜22)、IBA(表3、試験No.23〜22)、IAA(表4、試験No.27〜30)では、全ての試験培地で著しい根の伸長が見られたものは無かった。

実施例2
増殖した根の培養細胞を凍結破砕し、トルエンソックスレー抽出により破砕細胞からゴム成分の抽出を行った。

得られたトルエンソックスレー抽出物をNMR分析に供し、ゴム成分の存在の有無を調べた。その結果、図2に示すようにトランスポリイソプレンに由来するシグナルが認められ、ゴムの存在が確認された。

また、サイズ排除クロマトグラフィー(SEC)により、ゴムの分子量分布を調べた。その結果、図3に示すように、数平均分子量11000、重量平均分子量76000、多分散度6.9の分子量分布を有するゴムが生産したことが確認された。

比較例2
実施例1で述べた試験No.4の条件で得られた杜仲のカルスについて、同様にトルエンソックスレー抽出によりゴムの抽出を試みNMR分析を行った。しかし、図2で認められるようなゴムに由来するシグナルは観測されず、ゴムの存在は磯認できなかった。

培養に、MS培地(Murashige et a1., 1962)に、α−ナフタレン酢酸(NAA)または2,4−ジクロロフェノキシ酢酸(2,4−D)と6−ベンジルアミノプリン(BAP)を種々の濃度で加え、それぞれに蔗糖(濃度2重量%)を加えた培地(表5,表6)を用いた以外、実施例1と同様の操作を行った。

【発明の効果】
この発明によれば、従来難しいとされてきた組織培養系でのゴム生産が初めて可能になった。これにより、タンク培養によるゴム生産が可能である。さらに、遺伝子改変が容易な本培養細胞において、高いゴム生産能を有する形質転換体を作成し、効率的で安定したゴム生産が可能である。

Therefore, the consumption of SBR may be replaced with natural rubber, but natural rubber is not always smoothly replaced due to its resource bias in the southern region.

Natural rubber is a cis-type isoprene rubber made from sap collected from a rubber tree, but other rubber resources are known to produce natural rubber, such as Tonaka, which can grow even in warm regions. . Prior to the invention of petroleum-based synthetic rubber, rubber materials were also collected from Tochu, and were widely used as covering materials for submarine electric wires, covering materials for golf balls, and dental materials. However, since synthetic rubbers have been synthesized at low cost, these have almost been replaced by synthetic rubbers.

Of course, the production of plant-based natural rubber requires growing plants. Thus, if the production of plant-based natural rubber is achieved anywhere in the world, this will have immense benefits to the rubber manufacturing industry. Usually, in order to obtain a plant-based natural rubber, a plant is cultivated, a rubber substance is biosynthesized in the plant body, and collected. However, it is known that a plant can biosynthesize a unique component (secondary metabolite) even in a plant culture such as callus without necessarily growing to a complete tree.

Example 1
Seeds collected from the Tonaka female strain of Ikunajima, Ehime Prefecture were aseptically seeded on a gellite medium containing 2% by weight of sucrose (gellite concentration: 0.24% by weight). Roots were cut into 2-3 cm from the seedlings 40 to 60 days after sowing, and cut pieces of the roots were collected.
