【発明の属する技術分野】
本発明は、皮膚貼付薬用基材シートとこれを用いてなる皮膚貼付薬に関する。詳しくは、本発明は、粘着剤層中の薬剤ほか、通常、粘着剤層に含まれている流動パラフィンや粘着付与剤等の添加剤の基材シートへの移行がないと共に、柔軟で、貼着した皮膚の伸縮に対してよく追従する皮膚貼付薬用基材シートと、このような基材シートを用いてなる使い勝手のよい皮膚貼付薬に関する。更に、本発明は、上記皮膚貼付薬用基材シートの製造方法に関する。

【従来の技術】
従来、例えば、消炎鎮痛剤やTTS製剤等の薬剤を含有する粘着剤を基材シート上に積層してなる粘着性皮膚貼付薬が経皮投与薬として医療用途に広く用いられている。このような皮膚貼付薬のための基材シートとしては、従来、柔軟で皮膚になじみよい塩化ビニル樹脂が広く用いられているが、反面、塩化ビニル樹脂からなる基材シートを用いる場合には、粘着剤層中の薬剤や、粘着剤層に通常、含まれる流動パラフィンや粘着付与剤等の添加剤が粘着剤層から基材シート中に移行し、基材シートを膨潤させ、変質させるのみならず、薬剤の経時的な投与を制御し難くなり、所期の治療効果を損なう問題がある。また、塩化ビニル樹脂シートに含まれる可塑剤が薬剤を含む粘着剤層中に移行して、粘着剤の性能を損なう問題もある。

そこで、このような問題を解決するために、例えば、高分子量ポリエステル可塑剤を配合した軟質塩化ビニル樹脂フィルムを耐薬品性や耐溶剤性にすぐれるポリエチレンテレフタレートフィルムの表面に積層接着し、裏面に薬剤を含む粘着剤層を設けて、ポリエチレンテレフタレートフィルムをバリヤ層として機能させた皮膚貼付薬が提案されている(例えば、特許文献1参照)。このような皮膚貼付薬は、粘着剤層中の薬剤や添加剤が基材シ−トに移行することがなく、また、基材シート中の可塑剤も粘着剤層中に移行することがなく、しかも、柔軟であり、皮膚によく追随するので、使い勝手がよいものである。

しかし、近年、塩化ビニル樹脂は、焼却時に有害なダイオキシンを発生するおそれがあることから、環境保護の観点から、皮膚貼付薬用基材シートについても、非塩素化材料への代替が求められている。

前述したように、ポリエチレンテレフタレートフィルムは、非塩素化材料のなかでも、耐薬品性や耐溶剤性にすぐれているが、ポリエチレンテレフタレートフィルム単独を基材シートとして適当な厚みとすれば、非常に硬く、柔軟性と皮膚への追従性がないので、そのようなポリエチレンテレフタレートフィルムを基材シートとする皮膚貼付薬シートは、使い勝手が悪く、また、皮膚に貼着したとき、シートの端部が浮き上がって、十分な治療効果に欠けたり、また、皮膚を損傷するおそれもある。

【発明が解決しようとする課題】
本発明は、従来の皮膚貼付薬とそのための基材シートにおける上述した種々の問題を解決するためになされたものであって、粘着剤層中の薬剤や添加剤が基材シートに移行して、基材シ−トを膨潤させるようなことがなく、しかも、柔軟で皮膚によく追随し、使い勝手のよい皮膚貼付薬用とそのための基材シートを提供することを目的とする。更に、本発明は、そのような皮膚貼付薬用基材シートの製造方法を提供することを目的とする。

【課題を解決するための手段】
本発明によれば、低密度ポリエチレン樹脂とオレフィン系熱可塑性エラストマーとから選ばれる少なくとも1種50〜100重量%とランダムポリプロピレン樹脂0〜30重量%とエチレン−ブチレンランダム共重合体0〜20重量%との樹脂組成物からなる厚み50〜200μmのポリオレフィン樹脂フィルムが厚み10μm以下のポリエチレンテレフタレートフィルムに接着剤にて接着されてなり、10%モジュラスが縦及び横共に35N/19mm幅以下である皮膚貼付薬用基材シートが提供される。

更に、本発明によれば、上記基材シートとこの基材シートのポリエチレンテレフタレートフィルムの裏面に設けられた薬剤を含む粘着剤層とからなる皮膚貼付薬が提供される。

【発明の実施の形態】
本発明による皮膚貼付薬用基材シートにおいて、ポリエチレンテレフタレートフィルムは、その厚みが10μm以下であることが必要である。ポリエチレンテレフタレートフィルムの厚みが10μmを越えるときは、得られる基材シートが皮膚貼付薬用基材シートとして用いるには硬すぎて、皮膚への追従性が悪い。本発明によれば、ポリエチレンテレフタレートフィルムの厚みは、得られる基材シートが柔軟性を有するように、通常、7.5μm以下であることが好ましく、特に、1.8〜5μmの範囲にあることが好ましい。

従って、本発明によれば、例えば、厚み3.5μm、破断強度29.4kgf/mm2 、2%(縦方向)モジュラスが8.6kgf/mm2 であるポリエチレンテレフタレートフィルムを好適に用いることができる。

本発明による皮膚貼付薬用基材シートは、このようなポリエチレンテレフタレートフィルムにポリオレフィン樹脂フィルムが接着剤にて接着されてなるものであり、ここに、得られる基材シートが柔軟で皮膚への追従性にすぐれると共に、所要の強度を有するように、このポリオレフィン樹脂フィルムは、その基材樹脂として、低密度ポリエチレン樹脂とオレフィン系熱可塑性エラストマーとから選ばれる少なくとも1種50〜100重量%を有することが必要であり、更に、その用途からの要求特性として、厚みが50〜200μmの範囲にあることが必要である。

本発明において、低密度ポリエチレン樹脂は、特に限定されるものではないが、所謂メタロセン触媒による直鎖低密度ポリエチレン樹脂(LLDPE)と呼ばれるものが好ましく用いられる。また、上記TPOとしては、その一つとして、ポリプロピレンと(場合によっては、ポリブテンと)エチレン−プロピレン共重合体成分とを有するブロック又はグラフト共重合体からなる所謂リアクターTPOがよく知られており、本発明によれば、このようなリアクターTPOが好ましく用いられる。このようなリアクターTPOの具体例としては、例えば、モンテル社の「キャタロイ」、(株)トクヤマの「PER」、チッソ(株)の「ニューコン」等が市販品として知られており、本発明においては、このような市販品を好適に用いることができる。このような低密度ポリエチレン樹脂とオレフィン系熱可塑性エラストマーとは、単独で、又は混合物として用いられる。

本発明によれば、必要に応じて、上記基材樹脂と共にランダムポリプロピレン樹脂やエチレン−ブチレンランダム共重合体を用いてポリオレフィン樹脂フィルムとすることもできる。ランダムポリプロピレン樹脂は、得られるポリオレフィン樹脂フィルムに適度の硬さを付与するのに有用であり、樹脂組成物の0〜30重量%の範囲、好ましくは、3〜25重量%の範囲で用いられる。このようなランダムポリプロピレン樹脂としては、例えば、日本ポリオレフィン(株)製のジェイアロマーPM620が市販品として知られている。本発明においては、このような市販品を好適に用いることができる。

他方、エチレン−ブチレンランダム共重合体は、得られるポリオレフィン樹脂フィルムに柔軟性を付与するのに有用であり、樹脂組成物の0〜20重量%の範囲、好ましくは、3〜15重量%の範囲で用いられる。エチレン−ブチレンランダム共重合体としては、例えば、JSR(株)製のEMB2041Pが市販品として知られている。本発明においては、このような市販品を好適に用いることができる。

更に、本発明によれば、このように、ポリエチレンテレフタレートフィルムにポリオレフィン樹脂フィルムが接着剤にて接着されてなる基材シートは、柔軟で、しかも、腰を有して、使い勝手のよい皮膚貼付薬を与えるように、10%モジュラス(試料19mm幅短冊状、チャック幅100mm、標線間50mm、引張速度50mm/分)が縦及び横共に35N/19mm幅以下であることが必要であり、特に、5〜35N/19mm幅の範囲にあることが好ましい。

本発明によれば、ポリエチレンテレフタレートフィルムとポリオレフィン樹脂フィルムは、接着剤にて接着されている。この接着剤としては、特に、一液型又は二液型ポリウレタン接着剤が好ましく用いられ、これらの一液型又は二液型ポリウレタン接着剤としては、なかでも、ポリエステルポリウレタン接着剤が好ましく用いられる。

このような一液型又は二液型ポリウレタン接着剤を用いて、ポリエチレンテレフタレートフィルムとポリオレフィン樹脂フィルムを接着する手段は、特に限定されるものではないが、しかし、ポリオレフィン樹脂フィルムに上記一液型又は二液型ポリウレタン接着剤を塗布し、乾燥させ、必要に応じて加熱下に、ポリエチレンテレフタレートフィルムと貼り合わせるドライ・ラミネーションによることが生産効率上、有利であり、しかも、このようにして得られる基材シートにおいては、ポリエチレンテレフタレートフィルムとポリオレフィン樹脂フィルムとの間に強い接着を得ることができる。

本発明による皮膚貼付薬は、このような基材シートのポリエチレンテレフタレートフィルム上に薬剤を含む粘着剤層を積層してなるものである。ここに、粘着剤は、特に限定されるものではないが、皮膚貼付薬を皮膚に貼付した際のすぐれた粘着性、肌へのなじみ性、適度の弾性等を有するのみならず、剥離時に皮膚に刺激を与えず、容易に剥離し得るところから、スチレン−ジエン−スチレンブロック共重合体やアクリル酸アルキルエステル共重合体等が好ましく用いられる。

このようなスチレン−ジエン−スチレンブロック共重合体の具体例としては、例えば、スチレン−ブタジエン−スチレンブロック共重合体やスチレン−イソプレン−スチレンブロック共重合体等を挙げることができる。これらスチレン−ジエン−スチレンブロック共重合体は、粘着剤として単独又は混合物として用いてもよい。更に、粘着剤は、前述したように、流動パラフィンや粘着付与剤のほか、必要に応じて、ポリイソブチレン、ロジン変性樹脂、水添ロジンエステル、可塑剤、充填剤、老化防止剤等、種々の添加剤を含有していてもよい。

本発明による皮膚貼付薬は、種々の薬剤を上記粘着剤に混合し、これを基材シートのポリエチレンテレフタレートフィルム上に積層してなるものである。従って、上記薬剤は、経皮吸収性薬剤であれば、特に、限定されるものではなく、例えば、局所刺激剤、抗炎症性剤及び鎮痛剤、中枢神経作用剤(睡眠鎮静剤、抗てんかん剤、精神神経用剤)、利尿剤、血圧降下剤、冠血管拡張剤、鎮咳去痰剤、抗ヒスタミン剤、不整脈用剤、強心剤、性ホルモン剤、副腎皮質ホルモン剤、局所麻酔剤、抗真菌剤等を挙げることができる。これらの薬剤は、単独で、又は2種以上が配合して用いられる。より具体的には、例えば、局所刺激剤、抗炎症性剤及び鎮痛剤の例として、例えば、サリチル酸、サリチル酸メチル、サリチル酸グリコール、L−メントール、ハッカ油、チモール等を挙げることができる。

粘着剤層におけるこのような薬剤の割合は、薬剤にもより、また、何ら限定されるものではないが、通常、1〜25重量%、好ましくは、5〜20重量%の範囲である。

【発明の効果】
以上のように、本発明による皮膚貼付薬用基材シートによれば、柔軟で強度を有するポリエチレンテレフタレートフィルムに柔軟で弾力性を有するポリオレフィン樹脂フィルムを接着してなり、基材シートとしての強度と柔軟性を有しながら、適度の腰と皮膚の伸長によく追従する弾力性とを有するものである。

The pressure-sensitive adhesive sheet according to Comparative Example 1 is a sheet in which only a film made of a polyolefin resin is used as a base sheet, and the pressure-sensitive adhesive is directly laminated on the base sheet. Bleeding of paraffin and tackifier is also remarkable. The pressure-sensitive adhesive sheets according to Comparative Examples 2 and 4 are not practical as a skin patch because the polyethylene terephthalate film in the base sheet is too thick, hard, and lacks flexibility. The pressure-sensitive adhesive sheets according to Comparative Examples 3 and 5 each have a base sheet obtained by bonding a polyethylene terephthalate film to a random polypropylene resin film, and use a low-density polyethylene resin or a reactor TPO as a polyolefin resin component. It is hard to use and lacks flexibility, so it is not practical as a skin patch. In contrast, any of the pressure-sensitive adhesive sheets according to the present invention can be suitably used as a skin patch.
