これまでゼリー食品に使用される材料として、古くはゼラチンや寒天から始まり、カラギーナン、キサンタンガム、グァーガム、アルギン酸、ペクチン、ローカストビーンガム、マンナンなどが使用されている。これらの材料は、製造システムの高度化や、流通事情、あるいは嗜好性の変化から、1種類の使用で消費者の多様な要求を満たすことが難しく、通常、2種あるいはそれ以上の多糖を混合使用される。例えば、カラギーナンとローカストビーンガム、キサンタンガムとグァーガム、アルギン酸とペクチンなど、好適な組み合わせが広く知られ、目的に応じて様々な様態の商品となり、広く流通している。

このように商品化されたゼリー食品の物性の良さに着目し、咀嚼や嚥下といった食物摂取機能の低下した高齢者や乳幼児に多い、誤嚥防止に、医薬品ゼリー製剤(特開平9−187233、特開平9−194346)が提案された。 即ち、上述のように、すでに汎用されているカラギーナンにローカストビーンガムの組み合わせに(Rees,D.A., Bioch.J., 126:257−273,1972)、ポリアクリル酸化合物を添加することで、保存安定性に優れたゼリー製剤を完成させた。 特に、ゼリー製剤は、製造後、室温と言っても、地域や季節により温度変化の激しい条件で長期間保存されると、離漿が見られるようになり、形状を維持できないばかりか、このゼリー組成物中に含まれる肝心の薬効成分そのものも維持できなくなるので、この点で、吸水性の高いポリアクリル酸化合物の添加は、離漿を防ぎ製剤の安定化を図る上で好ましい工夫と云える。

一般に、ゼリー製剤は、従来服用された錠剤やカプセル剤などの固形製剤に比べ、液剤やシロップ剤と同様、比較的飲みやすいと云える。 しかも、液剤などに比べ、誤嚥もなく、また、薬効成分の持つ苦みやえぐみなども、避けることができる点で優れている。 更に、その製剤化の面でも、明らかに薬効成分が不安定なものを除けば、可溶性のものはもちろんのこと、特に、不溶性や難溶性のものでも、いずれも均一に分散してゼリー製剤を作製することができる点で、液剤やシロップに勝る。

ところが、このように優れた特性を持つゼリー製剤も、薬効成分の1回あたりの服用量が多いものについては、問題が残る。 というのは、1回投与量の多い薬剤は、通常の錠剤やカプセル剤などでは、大きな形状にするか、あるいは、複数個服用することで解決を図るが、そのどちらを選んでも、服用しずらいことに変わりがない。 従って、このような多量の薬効成分を含有する経口製剤を作製する上で、服用しやすいゼリー製剤化が望まれる。 この典型的な例として、血液及び尿のアルカリ化によるアシドーシスや、酸性尿の改善作用を示す、クエン酸アルカリ塩を含有する製剤が上げられる。

現在、これらの製剤としては、用時溶解性の粉末粒「ウラリット(登録商標)−U」及び錠剤「ウラリット(登録商標)錠」が臨床使用されているが、前者の粉末粒では、かなり多量の水で希釈しなくては飲みづらいこと、一方、錠剤では、比較的飲みやすくなってはいるものの、服用後、胃内に局所的にアルカリ塩が残るため、胃に不快感が表れる。
