【発明の属する技術分野】
本発明は、従来の手法を用いずに全血から血漿または血清を得ることが可能な方法及び該装置に関する。また全血から血漿または血清を得る方法として、血球成分の沈降を亢進する溶液を用いることを特徴とする分離方法及びそれを容易に行うよう工夫された装置に関する。

【従来の技術及び発明が解決しようとする課題】
現在血液検査は健康診断や疾患の治療経過を診断する手段として汎用され、検査用の分析装置の精度や稼動能力は日々向上しているものの、分析する血液検体を調整する作業はあまり改善されておらず煩雑で時間がかかる。第一に検体の採取となる採血は医者、看護婦や臨床検査技師といった有資格者のみが行える医療行為であり、検体採取の為に医療施設に出向かなければならない。次に、血液中の赤血球、白血球及び血小板が検査項目に影響することがあるため、生化学的検査の対象のほとんどが血漿や血清である。まず全血量で10mL近い血液が必要で、血清は血餅が生じるまでに時間を要し、更に遠心操作が必要になる。また抗凝固処理をした血漿は遠心操作のみであるが、遠心後血球成分が混在しないように注意深く血漿を分離する必要があるなど、血漿も血清もその調整過程は煩雑で時間を要する作業のため検査する側にとって大きな負担となる。
また作業上不可欠な遠心装置のない比較的小さな病院では検査を他の検査機関に依頼するため検査結果確認に時間を要するといった問題点もある。
このような方法では検査を受ける側行う側の両方の負担が大きく、更に検査を行う側には検体処理中の感染という危険性もある。
近年、分析装置の精度向上にともなって、微量の血漿又は血清で従来の血液検査を行うことが可能になり、更に全血から血漿又は血清を簡便に分離する手法が発明されてきた。
例えば多層の不織布フィルターを用いて全血から血漿を分離する手法が開発された(特開平11−295302)が、微量の血漿量を得るために3mL以上の全血が必要で検査を受ける側の負担は変わらず、またこのフィルターは特定の分析装置仕様であるため他の分析装置には使用できないため汎用には至っていない。
また指先をランセット等の針で傷つけて得られる微量の血液をペーパークロマトの原理で血球と血漿または血清に分離する方法(特開2001−188066)が発明されているが、分離後展開した血漿または血清の成分を界面活性剤等で抽出する必要があり、実際の測定に至るまでの工程が煩雑である上、抽出等で影響を受ける成分の測定は困難でやはり汎用には至っていない。
その他に中空糸を用いて全血から血漿を分離する方法が発明されているが、測定可能な量の血漿を得るためにはある程度の血液量が必要で、この方法でも受ける側に負担が生じる。
本発明者らは上述の課題を解決すべく鋭利検討を重ねた結果、少量の血液より血漿また血清成分を分離可能な方法を発明した。血球成分の内赤血球は電解質溶液と混合したとき、膜成分脂質のシアル酸が負に帯電する為、電離している陽イオンを血球周辺に引き寄せ、互いに反発することで血球成分同士の凝集を抑制していることが知られている(「輸血の副作用,合併症」輸血学第2版遠山博著1989,p333〜334)。そこで、血球同士の凝集を亢進させる為、電解質ではない糖等の水溶液に血液を添加し、血球成分を沈降させ血漿又は血清成分を分離させたところ、10分以内で血球の沈降が認められた。また糖以外のアミノ酸等でも同様の試験を実施したところ、同様の結果を得ることができた。この方法を利用することでμL単位の血液から血漿が分離可能となり、微量な検体でも測定可能な分析装置を用いることで基本的な生化学検査が十分に可能となった。
また、沈降させた血球成分は崩壊していない為、緩衝液に再溶解することで、血球を試験対象とした形態・細胞性免疫検査・遺伝子関連検査等の検査も可能である。

【課題を解決するための手段】
[1]本発明は、(1)血液(BL)と、非電解質または両性電解質または電荷し難い物質からなる群より選択される少なくとも一つの化学物質またはこれらの化学物質の組み合わせからなる化学物質(CS)を、水または電解質を全く或いはほとんど含まない溶媒(SL)より選択される少なくとも一つの溶媒(SL)またはこれらの溶媒(SL)の組み合わせからなる溶媒(SL)に溶かした液体とを混合するステップ、
(2)血液成分を分離するステップ、
少なくとも以上の2つのステップを含む血液成分の分離方法を提供する。
[2]本発明は、前記血液成分とは、主に赤血球または白血球または血小板からなる群より選択される少なくとも一つの成分またはこれらの成分の組み合わせからなる血球成分(BLC)と、主に血漿または血清からなる群より選択される少なくとも一つの液状成分(BLS)である[1]の血液成分の分離方法を提供する。
[3]本発明は、前記非電解質または両性電解質または電荷し難い化学物質(CS)を水または電解質を全く或いはほとんど含まない溶媒(SL)に溶解した水溶液(WSL)を調整し、当該水溶液(WSL)を血液成分のうち主に血球が含まれる成分を沈降させる手段として使用する[1]ないし[2]の血液成分の分離方法を提供する。[4]本発明は、前記水溶液(WSL)は血球表面への正電荷層の形成を抑制し、当該水溶液(WSL)中に血液を添加すると血球同士の作用によって主に血球成分(BLC)のみの沈降を亢進させる[1]ないし[3]の血液成分の分離方法を提供する。
[5]本発明は、前記化学物質(CS)が、非電解質または糖またはポリマーからなる群より選択される少なくとも一つの物質またはこれらの二種類以上の物質の組み合わせからなる物質である[1]ないし[4]の血液成分の分離方法を提供する。
[6]本発明は、前記化学物質(CS)が、単糖類、二糖類、多糖類、糖アルコール、緩衝剤、水溶性のポリマー類からなる群より選択される少なくとも一つの物質またはこれらの二種類以上の物質の組み合わせからなる物質である[1]ないし[5]の血液成分の分離方法を提供する。

【発明の実施の形態】
本発明の血液成分の分離方法は、(1)血液(BL)と、非電解質または両性電解質または電荷し難い物質からなる群より選択される少なくとも一つの化学物質またはこれらの化学物質の組み合わせからなる化学物質(CS)を、水または電解質を全く或いはほとんど含まない溶媒(SL)より選択される少なくとも一つの溶媒(SL)またはこれらの溶媒(SL)の組み合わせからなる溶媒(SL)に溶かした液体とを混合するステップと、(2)血液成分を分離するステップの少なくとも以上の2つのステップを含む。
本発明で血液(BL)とは、血球成分(BLC)と液状成分(BLS)の双方を含む血液である。
本発明で血液成分とは、主に赤血球または白血球または血小板からなる群より選択される少なくとも一つの成分またはこれらの成分の組み合わせからなる血球成分(BLC)と、主に血漿または血清からなる群より選択される少なくとも一つの液状成分(BLS)であり、血球成分(BLC)は主に赤血球、白血球及び血小板からなる血球成分を含み、液状成分(BLS)は主に血漿または血清からなる成分を含む。
本発明では、前記非電解質または両性電解質または電荷し難い化学物質(CS)を水または電解質を全く或いはほとんど含まない溶媒(SL)に溶解した水溶液(WSL)を調整し、当該水溶液(WSL)を血液成分のうち主に血球が含まれる成分を沈降させる手段として使用する。
前記水溶液(WSL)は血球表面に正電荷の層が形成し血球同士互いに反発しあうことを抑制する。さらに詳述すれば前記水溶液(WSL)は、前記ステップ(1)において、血球表面へ正電荷層が形成されるのを抑制し、当該水溶液(WSL)中に血液を添加すると血球同士の作用によって主に血球成分(BLC)のみの沈降を亢進させる。
前記化学物質(CS)と前記溶媒(SL)は、血球表面の正電荷層の形成を抑制し、血球の沈降を亢進させることのできる前記水溶液(WSL)を調整できるものであれば何でも良い。
例えば、前記血球沈降を亢進させる化学物質(CS)として、非電解質または糖またはポリマーからなる群より選択される少なくとも一つの物質またはこれらの二種類上の物質の組み合わせからなる物質を使用することができる。
また前記化学物質(CS)として、単糖類、二糖類、多糖類、糖アルコール、緩衝剤、水溶性のポリマー類からなる群より選択される少なくとも一つの物質またはこれらの二種類以上の物質の組み合わせからなる物質を使用することができる。

本発明の血液成分の分離装置は、少なくとも化学物質(CS)と血液(BL)とを混合し、静置する手段を有し、また静置後分離した液状成分(BLS)中に混在し得る夾雑細胞成分を捕獲するための分離手段を有することもある。
前記化学物質(CS)と血液(BL)を混合する手段として、少なくとも(1)血液成分(BL)と化学物質(CS)を含む水溶液(S)とを混合し、静置することが可能な手段を有し、少なくと(1)血液成分(BL)と化学物質(CS)を含む水溶液(S)とを混合する手段中に、前記血液成分中の血漿又は血清成分が希釈されて含まれる液状成分(BLS)を分離する為に血液成分中に主に血球成分を沈降させる化学物質(CS)を含む水溶液(WSL)を収納(装填或いは充填ともいう)している。
本発明の血液成分の分離装置は、少なくとも(1)血液成分(BL)と化学物質(CS)を含む水溶液(S)とを混合し静置する手段と、当該手段(1)に加えて、(2)静置後、沈降した血球成分(BLC)から上澄として分離されることにより、主に前記溶媒(SL)中で希釈された血漿または血清成分を含む液状成分(BLS)中に含まれる可能性がある血球成分(BLC)(夾雑細胞成分ともいう)を捕獲する分離手段、(3)前記の液状成分を分離手段へと導く手段として加圧や引圧を生じさせることが可能な或いは自然力にて稼動する駆動手段、(4)分離される前記液状成分(BLS)を収納する手段を有する。
前記液状成分(BLS)に残存する血球成分(BLC)を捕獲する為の分離手段は、次のいずれかの形態を採用することができる。前記化学物質(CS)と血液(BL)とを混合し静置する手段と連結及び/又は取り外しが可能な形態、または前記分離手段それ自体が前記分離される液状成分を収納する手段自体に固定した形態。
本発明で前記(1)血液成分(BL)と化学物質(CS)を含む水溶液とを混合し静置する手段とは、例えば、ポリプロピレンやポリエチレン等の高分子材料或いはガラス等の無機材料で成型された密閉性を有する容器3、13、23、33等であり、その形状は本発明の目的を達成することができるものであれば何でも採用することができる。但し、前記容器のうち23、33(例えば後述する図3、図4参照)は、その上部に分離手段22、32が接続可能なものである。
前記(2)静置後、沈降した血球成分(BLC)から上澄として分離されることにより、主に前記溶媒(SL)中で希釈された血漿または血清成分を含む液状成分(BLS)中に含まれる可能性がある血球成分(夾雑細胞成分ともいう)(BLC)を捕獲する分離手段3、13、23、33とは前記段落番号[0007]に記載の分離手段そのもの(例えば平膜或いはろ紙や中空糸やスポンジ状の樹脂発泡体等)、または同分離手段を血液の出口と入口を有するハウジング内に配置されるもの(例えば後述する図3、図4参照)、また容器(筒体ともいう)の下部(口部ともいう)に固定されたもの(例えば後述する図1、図2参照)で、分離手段そのものやハウジングや容器(筒体ともいう)の形状は本発明の目的を達成することが可能なものであれば何でも採用することができる。またハウジングや容器(筒体ともいう)内のフィルターの形状や配置は本発明の目的を達成することが可能なものであれば何でも採用することができる。
前記(3)液状成分を分離する手段として加圧や引圧を生じさせることが可能な駆動部(駆動手段ともいう)或いは自然力にて稼動する駆動部(駆動手段ともいう)5、15、27、37とは、例えば、シリンジのピストン27、37や前記容器に収まる容器(筒体)5、15等であり、ピストンは加圧する方向に、容器(筒体)は前記容器内に挿入する方向に押すことで、容器3、13、23、33内の液体を加圧する機能を有するものであれば何でも採用することができる。また前記(4)静置後分離される前記液状成分を収納する手段5、15、25、35(例えば後述する図1から4参照)とは液状成分内に希釈されて含まれる血漿または血清成分に影響しないもので分離される前記液状成分を採取することができる容器等である。例えば、ポリプロピレンやポリエチレン等の高分子材料或いはガラス等の無機材料で成型された密閉性を有する容器等であり、これらの形状は、本発明の目的を達成することができるものであれば何でも採用することができる。但し、収納手段5、15は前記(3)記載の駆動手段および前記(2)記載の分離手段としての両手段の機能を併せ持つ。
また収納手段(容器ともいう)15は前記液状成分を収納した後、栓体17にて下部を密閉することが可能である。

図1から4は本発明の血液成分分離装置の一例を示す概略図であり、以下これらに基づいて説明する。図1から4の血液成分分離装置はあくまでも例示であり、同様の作用効果を奏する血液成分分離装置は、全て本発明に含まれる。
図1の血液成分分離装置1は、血液成分と化学物質を含む水溶液Sとを混合する容器3と分離手段2、駆動部および分離後の液状成分を採取する容器5から構成される。
容器5は、人工的な加圧力で駆動或いは自然力による降下の何れによっても容器3内に挿入可能な構造(例えば容器5の外径を容器3の内径と同じかまたは若干小さく形成する)を有し、その下部(口部ともいう)に分離手段2が固定される構造(例えば分離手段2の装着溝を形成する)を有する。また、容器3内に駆動部および分離後の液状成分を採取する容器5が挿入されることで、容器3内の溶液が加圧され、分離手段2を通すことによって、主に希釈された血漿または血清成分を含む液状成分を分離することが可能な装置である。
容器3の上部、駆動部および分離後の液状成分を採取する容器5の上部と下部には密閉性を維持する為に、ポリプロピレンやポリエチレン等の高分子材料やガラス等の無機材料から成型されるキャップ6A、6B等が装着される。分離手段2とは例えば、平膜2M(或いはろ紙や中空糸)を駆動部および分離される液状成分を採取する容器5の下部に固定したものである。
図1の血液成分分離装置1は例えば次の順序で血液成分を分離することができる。
(1)化学物質が含まれる水溶液の入った容器3の上部キャップ6Bを外し、血液を同容器3内に添加する。
(2)再び容器3にキャップ6Bを装着し、転倒して血液と化学物質を含む水溶液とを混合し、静置する。
(3)静置することにより容器3内で、沈降した血球成分と、主に希釈された血漿または血清成分を含む液状成分とに分離される。
(4)容器5上部のキャップ6Aを外し、人工的に加圧し或いは自然的に降下させ、容器3内に挿入する。
(5)容器5に固定されている分離手段2によって主に希釈された血漿または血清成分を含む液状成分が膜2Mを介し、容器5内に採取される。
(6)容器5が容器3内に完全に挿入された後、同容器5上部に、再びキャップ6Aを装着し、容器内を密閉する。

図2の血液成分分離装置11は人工的な加圧或いは自然力による降下によって分離を行う装置である。図2(A)は装置1の概略図(断面図)で、図2(B)は、容器15の平面図である。血液成分分離装置11は図1の血液成分分離装置1と比較して、新たに分離される主に希釈された血漿または血清成分を含む液状成分と沈降した血球成分とを完全に分離する為に、キャップ16Aの下部に支持竿を装着し、該支持竿の先端に栓体17を装着したものである。これらの支持竿及び栓体17は、図3、図4のシリンジのピストン27、37と同様の役割を果たす。
キャップ16Aを容器15上部に装着することによって、栓体17は、駆動部および分離された液状成分を採取する容器15下部を塞ぎ、容器13内で沈降している血球成分と液状成分とを完全に分離することが可能となる。また容器15は、キャップ16Aと栓体17によって密閉性を有し、容器13から取り外すことが可能となる。
また図1と比較して、新たに容器13は内部に、栓体17を挿入する時、分離された液状成分を栓体17上部に通液するための通液溝18と、栓体17で容器13内を密閉するための栓体止め19を有する。分離手段12とは例えば、平膜12M(或いはろ紙や中空糸等)を駆動部および分離される液状成分を採取する容器15の下部に固定したものである。
図2の血液成分分離装置11は、例えば次の順序で血液成分を分離することができる。
(1)化学物質が含まれる水溶液が入った容器13上部のキャップ16Bを外し、血液を同容器13に添加する。

図2の血液成分分離装置11と比較して、新たに化学物質と血液とを混合する容器23内に上下に稼動するピストン等の駆動部27を有する。分離手段22とは、例えば、液状成分の入口と出口を有するハウジング23H内に平膜23M(或いはろ紙や中空糸等)を配置したものである。またハウジング23Hは、容器23と容器25のそれぞれの口部に装着および取り外しが可能な構造を有している。
図3の血液成分分離装置21は、例えば次の順序で血液成分の分離ができる。
(1)化学物質が含まれる水溶液の入った容器23のキャップ26Bを外し、血液を同容器26内に添加する。
(2)再び容器23にキャップ26Bを装着し、転倒して血液と化学物質を含む水溶液とを混合し、静置する。
(3)静置することにより容器26内で、沈降した血球成分と、主に希釈された血漿または血清成分を含む液状成分とに分画される。
(4)容器25上部のキャップ26Aを外す。
(5)分離手段22の接続部22Aと、容器23の上部および容器25の下部と装着する。
(6)駆動部27を押して容器23内を加圧し、分離手段22内によって、主に希釈された血漿または血清成分が膜22Mを介して容器25内に採取される。

図4の血液成分分離装置31は、加圧力により、容器33内の主に希釈された血漿または血清成分を含む液状成分を分離することが可能な装置である。
図3の血液成分分離装置21と比較して、分離された液状成分を採取する容器35上部にキャップ36Aが装着された連結針38Aを有する連結部38を配置したものである。
分離手段32は、図3に例示した分離手段21の膜22Mの平膜やろ紙等に代えて、中空糸膜32Mを容器33内で分離された主に液状成分の入口と分離膜32Mを介して分離される液状成分の出口を有するハウジング内32Hに配置した実施例である。
図4の例示に従って詳細に説明すると、ハウジング32Hの下部には、容33内で分離される内溶液の入口(液体通路を有し液体を中空糸膜32M内側に導入する連結部32A)、右側部には中空糸32Mにて分離された液状成分を排出する為の出口(液体通路を有し液体を容器35に排出する連結部32D)がそれぞれ装着されている。また左側部と上部にはそれぞれ分離手段32に通液する容器33の内溶液或いは分離手段32外に排出される液状成分を透過しない材質・構造を有する通気部32B、32Cが装着されている。さらに同通気部32B、32Cにはそれぞれ、シート状等の封止部材34A、34Bが装着され、ハウジング32H内を外気と通気するまでは同通気部32Bと32Cは封止材34A、34Bによって密閉された状態である。
以上説明したように、図1から図8に例示した血液成分分離装置1、11、21において、各分離手段2、12、22ないしハウジング22H(内部には平膜やろ紙等が配置される)に代えて、本実施例のハウジング32H(中空糸膜32Mが配置される形態)を使用することも可能である。逆に、本実施例の血液成分分離装置31において、分離手段32のハウジング32H(中空糸膜32Mが配置される形態)に代えて、図1から図3に例示した血液成分分離装置1、11、21ないし前記ハウジング22H(内部には平膜やろ紙等が配置される)を使用することも可能である。
図4の血液成分分離装置31は例えば次の順序で血清成分を分離することができる。
(1)化学物質が含まれる水溶液の入った容器33の上部キャップ36Bを外し、血液を同容器33内に添加する。
(2)再び容器33にキャップ36Bを装着し、転倒して血液と化学物質を含む水溶液とを混合し、静置する。
(3)静置することにより、容器33内で、沈降した血球成分と、主に希釈された血漿または血清成分を含む液状成分とに分画される。
(4)容器33に分離手段32を装着する。
(5)分離手段32上部の通気部32Bの封止材34Aを外して、通気可能な状態にする。
(6)駆動部37を押して容器33内の溶液を加圧し、分離手段32に通液する。
(7)分離手段32内によって、主に希釈された血漿または血清成分が膜32Mを介して容器35内に採取される。
(8)分離手段32上部の通気部32Bを封止材34Bで再び密閉し、隣接した通気部32Cの封止材34Aを外す。
(9)容器35上部のキャップ36Aを外し、同容器35の連結部38と分離手段32の連結部32Dを装着し、分離手段32内で分離された液状成分を同容器35内に採取する。
(10)分離手段32と容器35とを取り外し、容器35を同容器35から外してキャップ36Aで同容器35を再び密閉する。

実施例1.
本実施例は、数種類の化学物質を用いて水溶液を調整し、血液を添加した後血球成分の沈降が亢進し約10分程度で、主に血球を含む成分と主に血漿又は血清である液状成分とに分離可能か否かの検討を行う為に実施した。
本実施例では、以下の材料を使用した。
人血液を用い、化学物質を含む水溶液として10%マルトース水溶液,5%及び20%マンニトール水溶液,2.25%グリシン水溶液,3.6%トリスアミノメタン水溶液,20%ポリビニルピロリドン水溶液,3%ポリビニルピロリドン/5%ソルビトール水溶液,2.45%ポリビニルピロリドン/4.25%ソルビトール水溶液,0.1%メチルセルロース/5%マンニトール水溶液,0.1%メチルセルロース/5%HEPES(2−〔4−(2−ヒドロキシエチル)−1−ピペラジニル〕エタンスリルホン酸)水溶液,5%ACES(N−(2−アセトアミド)−2−アミノエタンスルホン酸)緩衝液,5%MOPS(3−(N−モルホリノ)プロパンスルホン酸)緩衝液,6%BES緩衝液((N,N−ビス(2−ヒドロキシエチル)−2−アミノエタンスルホン酸),7%HEPES(2−〔4−2(ヒドロキシエチル)−1−ピペラジニル〕エタンスルホン酸)緩衝液,10%デキストラン10%マルトース/リン酸緩衝液,5%及び20%マンニトール/リン酸緩衝液を調整して使用した。また市販の生理的リン酸緩衝食塩水(PBS(−)),生理食塩液,10%デキストラン注射液,赤血球保存液MAP液,全血用液保存液ACD液,CPD液又はCPDA−1液をそのまま使用した。また必要に応じ、化学物資を含む水溶液と血液を混合する容器としてプラスチック製の容器(栄研チューブ1号/栄研機材)、混合した溶液を放置するものとして1mLのシリンジ(テルモ社製)を用いた。
血液を様々な水溶液中に添加したとき、血球成分に加わる重力によって放置することでどのような水溶液を用いた場合でも血球成分と血漿または血清成分に分離する。しかし時間が経つにつれ血漿或いは血清中の成分は影響を受ける為、短時間での分離が必要となる。
従って血液と化学物質を含む溶液を混合したときから約10分内で血球成分が沈降する条件を検討し、分離可能であったと判断した。10分以上かかった物は分離不可能と判断した。

具体的な血液分離方法は以下のとおりである。
1.検討する化学物質を用いそれぞれの水溶液を調整した。
2.1.で調整した水溶液を栄研チューブに分注した。
3.2.の栄研チューブ内に、水溶液と血液とが1:5の比率となるように血液を添加した。
4.1mLのシリンジを用い3.で混合した溶液を採取した。
5.シリンジを立てて静置し、分離の可不可を判断した。
結果を表1に示す。(分離可能を○、分離不可を×とした。)
13種類の水溶液で血球成分沈降が確認され10分内で血球成分と血漿成分に分離した。但し同じ物質をリン酸緩衝液に溶かすと緩衝液中の電解質による為か分離を確認することができなかった。また市販の水溶液も同様でも電解質を含むためか、分離されなかった。

実施例2.
本実施例では血球沈降を促す補助物質を、血球沈降させる化学物質と混合して水溶液とし、補助物質の添加未添加によってその沈降に差異が見られるか否かの検討を行う為に実施した。
本実施例では以下の材料を使用した。
人血液を用い、化学物質として5%マンニトール水溶液に、補助物質として塩化マグネシウムをマグネシウムイオン濃度で0.5,1,2,5,7.5,10meq/Lとなるように添加した。
また必要に応じ化学物質及び補助物質と血液を混合し、静置する容器としてプラスチック製の容器(栄研チューブ1号/栄研機材)を用いた。
補助物質の未添加時の沈降時間と比較し、それよりも早かった場合、沈降時間を早める補助物質として効果があると判断した。
具体的な血液分離方法は以下のとおりである。
1.血球成分を沈降させる化学物質のマンニトールと補助物質の塩化マグネシウムを混合し、マンニトール終濃度は5%で固定し、塩化マグネシウムをマグネシウムイオン濃度で0.5,1,2,5,7.5,10meq/Lとなるような水溶液をそれぞれ調整した。また対照として5%マンニトールのみの水溶液を調整した。
2.1.で調整した水溶液を栄研チューブに分注した。
3.2.の栄研チューブ内に、水溶液と血液とが1:5の比率となるように血液を添加した。
4.そのまま栄研チューブを静置し、塩化マグネシウム未添加のものと沈降速度との比較をした。
結果は表2に示す。
塩化マグネシウムを添加した全ての水溶液では血球成分の沈降が、塩化マグネシウム未添加のものよりも早いことが確認された。

実施例3.
本実施例は、血液と化学物質を含む水溶液とを混合し、静置することによって主に血球成分を沈降し、上清には主に希釈された血漿または血清成分を含む液状成分と血球成分が若干残存しており、前記液状成分のみを最終的に分離する為、この残存する血球成分を幾つかの分離手段で捕獲可能か否かを検討したものである。
本実施例では以下の材料を使用した。
人血液を用い、分離手段として多孔質体ポリマーの平膜(親水性ポリビニリデンジフロライド、ニトロセルロース、親水性ポリテトラフロロエチレン):直径13mm,孔径0.65〜2.0μmおよび無機材質のガラスろ紙:直径13mm,保留粒子径0.4〜0.8μm,血球成分を沈降させるための化学物質として5%マンニトール水溶液、駆動手段としてシリンジ(2.5mL)、血液と化学物質を含む水溶液とを混合する手段として先端口部を密閉したシリンジ(5mL)、分離体は駆動手段であるシリンジ(2.5mL)にシリコンゴムを用いて固定した。
具体的な血液成分の分離方法は以下の通りである。
1.5mLのシリンジのピストンを抜き、先端口部を下に向けてセットした。
2.5mLのシリンジ内に5%マンニトール水溶液を入れ、血液を添加した。
3.そのまま5分間静置し、血球成分を沈降させた。
4.分離体を固定した2.5mLのシリンジを、5mLのシリンジ内に押し入れ、分離体を介し主に血漿または血清成分を含む液状成分を2.5mLのシリンジ内に採取した。
5.分離前の液状成分と採取した液状成分の血球計測を行って、分離の可否を確認した。
結果は表3に示す。

実施例4.
本実施例は5%マンニトール水溶液及び10%マルトース水溶液を用いて血液から血漿成分を分離し、分離された血漿成分(TP,ALB,GOT,GPT,γ−GTP,T−CHO,TG,HDL−C,BUN,CRNN,UA,GLU)の分析を行った。
本実施例では以下の材料を使用した。
血液成分として人全血を用いた。血液と化学物質を混合する溶液として栄研チューブ(栄研機材社製)、混合後放置する容器として1mLシリンジ(テルモ社製)、分離される血漿中に若干残存する血球成分を捕獲する分離手段として孔サイズ0.25μmのPVDF性メンブランフィルター(Milipore社製)を用いて試験を行った。
また血漿成分の測定装置として、日本電子社製自動分析装置のBM−2000Cを使用した。

具体的な血液成分の分離方法は以下のとおりである。
1.各水溶液を調整し、栄研チューブ内に分注した。
2.血液と化学物質を含む水溶液との混合比が1:5となる様、栄研チューブに血液を添加した。
3.1mLのシリンジを用い、2.で混合した溶液を採取し、立てた状態で静置した。
4.血球成分が沈降したらシリンジの先端にフィルターを接続した。
5.シリンジを加圧し、分離された血漿をフィルターに通しながら、分析装置用カップに分注した。
6.血漿成分12項目について自動分析装置BM−2000Cにて測定を行った。
7.化学物質の変わりに生食を用いて同比率で血液を希釈したものを対照とし、その値との一致率を求めた。
結果は表4に示す。
マンニトールで分離した血漿成分は全て対照の測定値との一致率が8割以上近く見られ、分離方法或いは操作による血漿成分の影響は無いと判断した。
一方、マルトースで分離した血漿成分は、TP及びグルコース以外の項目は全て対照の測定との一致率が9割近いが、TP及びグルコースの測定値が対照測定値のTPは130%以上,グルコースは190%以上で測定系に何らかの影響があったことが確認された。マルトースはグルコース2分子が結合した構造を持つ為、グルコース濃度に影響があったと考える。従って実際の仕様ではマンニトールを用いることが望ましいと考えられる。

【発明の作用効果】
(1)本発明を用いることにより、従来の煩雑で検査を受ける側行う側両方に大きな負担となっていた血清分離法や血漿分離法を用いなくとも、基本的な生化学的検査を行うことができるようになった。
(2)例えば従来検査を行う為に必要であった10mL近い血液がμL単位の血液量で検査対照となる血漿や血清が得られ、また微量であるため医療機関に出向く必要も無く、場所・時間を選ばず誰もが検査を受けることが可能になるため被験者の負担が大変軽くなる。
(3)またこれまでのように煩雑な手段でなく大変容易な方法であること、そのため検体処理を有資格者が行う必要も無く被験者自身でも可能となる等検査側の負担もかなり軽くなる。
(4)特に被験者自身が血漿または血清の分離を行った場合、検査者は検体を分析装置のオートサンプラー等に並べるだけとなり、検査費用の低下をも可能とする。
(5)また用いる物質も特殊で無く血球や血漿成分への影響も見られ無い為、従来の検査結果に劣らない信憑性の高い結果を得ることができるようになった。
(6)これまでに発明されたペーパークロマトグラフを用いた在宅用検査用具の方法とは異なり、本方法は前記方法において必要であった検査までの煩雑な手法が全く無く、前述した通り誰もが可能な安易な方法である。
(7)また微量の血漿で分析可能な装置が必要となるものの、今日の分析装置機能向上からすれば将来的に問題とはならず、どのような機械でも測定可能な利点を有している。

Secondly, most of biochemical tests are performed on plasma and serum because red blood cells, white blood cells, and platelets in blood may affect test items. First, a blood volume of about 10 mL is required, and serum requires a long time to form a blood clot, and further requires centrifugation. In addition, anticoagulated plasma is only centrifuged, but it is necessary to carefully separate plasma after centrifugation so that blood cell components do not mix. This places a heavy burden on the inspector.
Also, in a relatively small hospital without a centrifugal device that is indispensable for work, there is also a problem that it takes time to confirm the test result because the test is requested to another test organization.
In such a method, the burden on both the side receiving the test and the side performing the test is large, and the side performing the test also has a risk of infection during sample processing.
In recent years, as the accuracy of the analyzer has been improved, a conventional blood test can be performed with a small amount of plasma or serum, and a technique for easily separating plasma or serum from whole blood has been invented.
For example, a method of separating plasma from whole blood using a multi-layer nonwoven fabric filter has been developed (JP-A-11-295302). The load does not change, and this filter cannot be used for other analyzers because it has a specific analyzer specification, so that it has not been widely used.
In addition, a method of separating a small amount of blood obtained by injuring a fingertip with a needle such as a lancet into blood cells and plasma or serum based on the principle of paper chromatography (Japanese Patent Application Laid-Open No. 2001-188066) has been invented. It is necessary to extract serum components with a surfactant or the like, and the process up to the actual measurement is complicated, and it is difficult to measure the components affected by extraction or the like, which is not yet widely used.
In addition, a method of separating plasma from whole blood using hollow fibers has been invented, but a certain amount of blood is required to obtain a measurable amount of plasma, and this method also burdens the recipient. .
The present inventors have conducted intensive studies to solve the above-described problems, and as a result, have invented a method capable of separating plasma or serum components from a small amount of blood. When red blood cells of the blood cell component mix with the electrolyte solution, the sialic acid of the membrane component lipid is negatively charged, so attracting ionized cations around the blood cell and repelling each other suppresses aggregation of blood cell components. ("Side effects and complications of blood transfusion", Transfusion Science 2nd Edition, Hiroshi Toyama, 1989, pp. 333-334). Then, in order to enhance the aggregation of blood cells, blood was added to an aqueous solution of sugar or the like, which was not an electrolyte, and blood cell components were sedimented to separate plasma or serum components. By using this method, plasma can be separated from μL of blood, and a basic biochemical test can be sufficiently performed by using an analyzer capable of measuring even a small amount of sample.

In this example, a plasma component was separated from blood using a 5% mannitol aqueous solution and a 10% maltose aqueous solution, and the separated plasma components (TP, ALB, GOT, GPT, γ-GTP, T-CHO, TG, HDL- C, BUN, CRNN, UA, GLU).
In this example, the following materials were used.
Human whole blood was used as a blood component. Eiken tube (manufactured by Eiken Kikai Co., Ltd.) as a solution for mixing blood and chemical substances, 1 mL syringe (manufactured by Terumo) as a container to be left after mixing, separation means for capturing blood cell components slightly remaining in plasma to be separated Using a PVDF membrane filter (manufactured by Millipore) having a pore size of 0.25 μm.
In addition, an automatic analyzer BM-2000C manufactured by JEOL Ltd. was used as a measuring device for plasma components.

The specific method of separating blood components is as follows.
1. Each aqueous solution was prepared and dispensed into an Eiken tube.
2. Blood was added to the Eiken tube so that the mixing ratio of the blood and the aqueous solution containing a chemical substance was 1: 5.
3. Use a 1 mL syringe. The solution mixed in was collected and left standing in an upright state.
4. When the blood cell component settled, a filter was connected to the tip of the syringe.
5. The syringe was pressurized, and the separated plasma was dispensed into an analyzer cup while passing through a filter.
6. Twelve items of plasma components were measured by the automatic analyzer BM-2000C.
7. The blood was diluted at the same ratio using a raw food instead of a chemical substance as a control, and the rate of agreement with the value was determined.
The results are shown in Table 4.
All of the plasma components separated by mannitol showed nearly 80% or more agreement with the measured values of the control, and it was judged that there was no influence of the plasma components by the separation method or operation.
On the other hand, in the plasma components separated by maltose, all items other than TP and glucose have a concordance rate of nearly 90% with the control measurement. It was confirmed that the measurement system had some influence at 190% or more. Maltose has a structure in which two glucose molecules are bonded, and thus it is considered that glucose concentration was affected. Therefore, it is considered preferable to use mannitol in actual specifications.

Operation and Effect of the Invention
(1) By using the present invention, it is possible to perform a basic biochemical test without using a serum separation method or a plasma separation method, which has conventionally been a burden on both the testee and the tester. Is now available.
(2) For example, plasma or serum serving as a test control can be obtained with a blood volume in the order of μL from blood of about 10 mL, which was conventionally required for performing a test, and since it is a very small amount, there is no need to go to a medical institution. Since the test can be performed by anyone at any time, the burden on the subject is greatly reduced.
(3) In addition, the method is not a complicated means as in the past, but a very easy method. Therefore, it is not necessary for a qualified person to perform sample processing, and the test can be performed by the subject himself.
(4) In particular, when the subject himself / herself separates the plasma or serum, the examiner only has to arrange the samples on an autosampler or the like of the analyzer, thereby making it possible to reduce the examination cost.
(5) Since the substance used is not special and has no effect on blood cells and plasma components, it is possible to obtain highly reliable results which are not inferior to conventional test results.
(6) Unlike the method of in-home inspection tool using a paper chromatograph invented so far, this method has no complicated method up to the inspection required in the above method, and as described above, Is an easy way.
(7) Although an apparatus capable of analyzing a small amount of plasma is required, it will not be a problem in the future in view of the improvement in the function of the analyzer today, and has an advantage that measurement can be performed with any machine. .
