【発明の属する技術分野】
本発明は、採取された血液から、血漿もしくは血清成分を分離し、分離された血漿または血清を容易に取り出すことを可能とする血液検査用容器に関し、より詳細には、血球成分の破壊により生じた血球内の液体による血漿または血清の汚染が生じ難い血液検査用容器に関する。

【従来の技術】
従来、血液から血球成分を除去し、臨床検査に必要な血漿もしくは血清を得るために様々な分離膜を有する血液検査用容器が提案されてきている。

下記の特許文献1には、直径0.05〜1μmの細孔を有し、外面開口率が40%以下、内面開口率が60%以上の中空繊維を用いて血液から血漿を採取する方法が提案されている。

また、下記の特許文献2には、平均直径が0.2〜5μm及び密度が0.1〜0.5g/cm3の繊維層により血漿または血清を分離する方法が提案されている。

しかしながら、特許文献1に記載の方法では、中空繊維を用いているため、ディスポーザブル製品としてはコストが高くなりすぎ、経済的でないという問題があった。また、特許文献2に記載の方法では、血漿または血清を分離し得るものの、ろ過速度が遅かった。圧力を加えてろ過速度を高めた場合には溶血が起こり、赤血球の破壊により血球内成分が漏洩するという問題があった。

他方、下記の特許文献3には、高分子極細繊維集合体または多孔質ポリマーを用い、血球成分と血漿または血清成分との移動速度差により分離を行う方法が開示されている。ここでは、繊維表面に親水性ポリマーが固定化されており、該親水性ポリマーが血漿または血清分離後に膨潤し、フィルタを閉塞することによりろ過が自動的に停止される。しかしながら、ヘマトクリットや粘度が異なる血液では、ろ過速度が変化するため、常に血漿または血清のろ過が終了した時点でろ過を停止させるように制御することができなかった。

【発明が解決しようとする課題】
上記のように、従来の血液から血漿または血清を分離する方法では、分離に際して赤血球などの血球成分の混入を確実に防止し、血漿または血清を簡便かつ確実に分離することができなかった。

本発明の目的は、上述した従来技術の現状に鑑み、赤血球の破壊に起因する血球内成分による血漿または血清の汚染が生じ難く、血液から血漿または血清を確実に分離することができる構造を備えた血液検査用容器を提供することにある。

【課題を解決するための手段】
本発明は、開口を有し、かつ採取された血液が収容される血液収容部と、前記血液から分離された血漿または血清を収納する血漿もしくは血清収納部と、前記開口を気密封止するように前記開口に取り付けられた栓体と、前記血液収容部と前記血漿または血清収納部との間に配置されており、血液から固形分と、血漿もしくは血清とを分離するフィルタとを備え、前記フィルタで分離された血漿もしくは血清がフィルタから前記血漿もしくは血清収納部に流下される血液検査用容器において、前記フィルタの前記血漿もしくは血清収納部側に、一定量の血漿もしくは血清を保持する血漿もしくは血清保持部が設けられており、該血漿もしくは血清保持部から、分離された血漿もしくは血清が前記血漿もしくは血清収納部に流下されることを特徴とする。

本願発明者らは、血液から血漿または血清成分と、血球成分とを分離するために、血球成分と血漿または血清成分との移動差を利用したフィルタ部材と、血球の混入を防止する血球停止膜とを用いれば、血液から血漿もしくは血清成分を簡便にかつ確実に取り出し得ることを見出した。従って、このようなフィルタ部材と、血球停止膜とを有する血液検査用容器を構成すれば、採取された血液から、血漿または血清成分と血球成分とを容易にかつ確実に分離することができる。

もっとも、上記分離が終了する時点は、赤血球が血球停止膜を閉塞した時点であり、この時、血液検査用容器内に減圧が残っている場合には、長時間保存すると、徐々に赤血球が破壊され、赤血球内の成分が液体成分として分離された血漿または血清成分に混入することがあった。

しかしながら、本発明では、上述した血漿もしくは血清保持部がフィルタの血漿もしくは血清収納部側に設けられているため、赤血球内の液体成分による血漿もしくは血清の汚染が効果的に抑制される。すなわち、血漿もしくは血清保持部には、一定量の血漿もしくは血清が保持される。他方、破壊された赤血球から赤血球内の液体成分がフィルタを通過して滲み出すには一定の時間を必要とする。従って、分離された血漿または血清成分が採取されるまでの間に赤血球の破壊による赤血球内の液体成分が、該血漿もしくは血清保持部に留まり、分離された血漿もしくは血清への分離が起こらず、従って、赤血球内の液体成分による汚染のない血漿または血清を確実に取り出すことができる。

【発明の実施の形態】
本発明では、血液から血漿または血清を分離するためにフィルタが用いられる。このフィルタについては、血液から血球と、血漿または血清とを分離することができる様々なフィルタ材料を用いることができ、特に限定されるものではない。もっとも、好ましくは、血球よりも血漿または血清を速く移動させるフィルタ部材と、フィルタ部材の下方に配置された血球停止膜とからなるフィルタが用いられる。

上記血球停止膜は、上記フィルタ部材を通過した血液中の血球成分を捕捉し、分離後の血漿または血清に血球成分が混入することを防止する作用を有する。
上記血球よりも血漿を速く移動させる性質のフィルタ部材については、特に限定されないが、例えば細い繊維径を有する合成高分子もしくはガラスからなる繊維または多孔性高分子などを用いることができる。もっとも、血液中の測定成分を吸着する場合には、吸着を抑制するためにフィルタ部材を構成する材料を表面処理しておくことが望ましい。表面処理剤としては、特に限定されないが、ポリエーテル系もしくはシリコーン系などの潤滑剤、ポリビニルアルコールやポリビニルピロリドンなどの親水性高分子類もしくは天然の親水性高分子類、または高分子界面活性剤などを用いることができる。

上記フィルタ部材が合成高分子またはガラスからなる繊維の場合、平均繊維径は0.2〜5.0μmの範囲が好ましい。0.2μm未満では溶血が生じやすくなり、5.0μmを超えると、血球と血漿もしくは血清とを分離するために繊維を高密度に充填する必要があり、またフィルタ部材の量が多くなるため、コストが高くなることがある。より好ましくは、繊維径は0.5〜3.0μmの範囲とされる。

血球停止膜は、孔径が0.05〜2.0μmの複数の孔、好ましくは多数の孔を有し、かつ空隙率が30%以下、好ましくは25%以下であることを特徴とする。孔径が0.05μm未満では、血漿もしくは血清中のタンパク質や脂質が目詰まりを起こしやすくなり、2.0μmより大きい場合には、赤血球が変形し、容易に膜を通過するおそれがある。また、空隙率が30%を超えると、赤血球に負荷がかかりやすくなり、溶血を引き起こしやすくなる。

上記血球停止膜における孔の形状については特に限定されないが、孔の内周面が滑らかな形状であるものが望ましい。例えば、孔が膜の一方の面から他方の面に直線状に貫通している形状のものが、赤血球へのダメージが少なく、特に溶血も引き起こし難いため、望ましい。

貫通孔の開口部平面形状及び貫通孔の横断面形状は特に限定されないが、鋭い角を有する形状は好ましくない。従って、貫通孔の開口部平面形状及び横断面形状は円または楕円などの曲線状の形状が好ましい。

血球停止膜の材質は特に限定されず、合成高分子及び天然高分子のいずれをも用いることができる。血球停止膜を構成する材料としては、例えば、セルロース混合エステル、ポリビニリデンジフルオライド、ポリテトラフルオロエチレン、ポリカーボネート、ポリプロピレン、ポリエステル、ナイロン、ガラス、アルミナなどを挙げることができる。

本発明において分離作業の対象となる血液は全血でもよく、希釈された血液試料であってもよい。また、血液は人の血液に限定されず、動物の血液であってもよい。さらに、血液は、新鮮血であってもよく、ヘパリン、エチレンジアミン四酢酸塩またはクエン酸などの抗凝固剤が添加された血液であってもよい。

上記血球停止膜を用いて、血液から、血球成分と、血漿もしくは血清とを分離するに際しては、膜の一方面に血液が供給され、ろ過により上記分離が果たされる。

上記分離作業に際しては、赤血球が目詰まりすることにより、ろ過が終了する。この場合、過大な圧力を加えると、溶血を引き起こすおそれがある。しかしながら、通常の孔を有する分離膜では、僅かな圧力を加えた場合でも溶血が生じるのに対し、本発明に係る血球停止膜を用いた場合には、より大きな圧力を加えた場合であっても溶血が生じ難い。すなわち、60kPa以下の圧力を加えてろ過したとしても、溶血が生じ難い。これは、空隙率を30%以下にすることで血球停止膜の表面形状が滑らかとなり血球へのダメージを少なくできるからである。

60kPaよりも大きな圧力を加えた場合には、赤血球が徐々に破壊されることがある。従って、好ましくは、60kPa以下の圧力を加えることが望ましく、それによって得られた血漿もしくは血清を用いて正確な検査値を得ることができる。

上記フィルタでは、上記血球停止膜の前段に、血球よりも血漿を速く移動させるフィルタ部材が直列に接続されている。血液が供給されると、まず血液が該フィルタ部材を通過するが、その際に血漿が速やかに血球停止膜側に向かって移動し、血球停止膜において血漿が速やかに血球停止膜を通過する。従って、血球と血漿とを効率良く分離することができる。このフィルタにおいては、血漿がろ過された後、赤血球が血球停止膜の孔を閉鎖した時点でろ過が終了される。

本発明に係る血液検査用容器は、上記フィルタを内部に収納した構造を有し、上述したフィルタ部材及び血球停止膜からなるフィルタが内部に収納された検査用容器が好ましい。次に、この血液検査用容器の構造について説明する。

本発明における血液検査用容器の物理的な構造については、開口を有し、かつ採取された血液が収容される血液収容部と、分離された血漿または血清を収納する血漿もしくは血清収納部と、上記開口を気密封止するように開口に取り付けられた栓体と、血液収容部と血漿または血清収納部との間に配置された上記フィルタとを備える限り特に限定されるものではない。好ましくは、血液検査用容器は、気密容器で構成され、内部が予め減圧されている。従って、内部の減圧を利用して、例えば栓体に真空採血針を刺通させることにより、血液を血液収容部に容易に採取することができる。

本実施形態では、上記血漿または血清保持部8を構成するために、筒状部材3は、血漿または血清保持部8の上面において分割され得るように構成されている。すなわち、筒状部材本体3Aと、突出部3bとが、図1の矢印Aで示す部分で分割されており、両者はねじ止めまたは嵌合等により着脱自在に固定されるように構成されている。そして、突出部3bを筒状部材本体3Aから取り外すことにより、血漿または血清保持部8が露出されるように構成されている。従って、血漿または血清保持部8に、適宜の充填剤を充填し、しかる後突出部3bを筒状部材本体3Aに取り付けることにより、充填剤の量に応じて血漿または血清保持部8の体積が調整され得るように構成されている。

そして、ろ過が終了した時点では、血漿または血清保持部8にも、分離された血漿または血清の一部が貯留されている。この状態において、ある程度の時間保存したとしても、また保存に際し、血球停止膜7に捕捉された赤血球が破壊し、中の液体成分が下方に漏洩したとしても、漏洩した液体成分は血漿または血清保持部8において一旦留まる。そのため、ある程度の時間経過した場合であっても、赤血球の破壊により漏洩した赤血球内の液体成分は、血漿または血清保持部8に留まるため、血漿または血清収納部9に貯留されている、分離後の血漿または血清への上記液体成分の混入を確実に防止することができる。

上記血漿または血清保持部8の空間の大きさは、上記のような作用を考慮すると、ある程度大きいことが好ましい。もっとも、血漿または血清保持部8の体積が大きすぎると、血漿または血清収納部9に貯留される分離済みの血漿または血清の量が少なくなるおそれがある。また、血漿または血清保持部8の体積は、5〜100μL程度とすることが望ましい。

合成樹脂からなる外管2及び筒状部材3を有する血液検査用容器1を用いた。ここでは、シリコーン製のシーリング剤を充填剤として血漿または血清保持部8に適宜の量を充填することにより、血漿または血清保持部8の体積を調整した。なお、充填剤を入れない場合の血漿または血清保持部8の体積は100μLである。

上記実施例1〜6及び比較例の各血液検査用容器を用い、健常人のボランティアからシリンジにより採血した血液2mLを栓体4から注入した。血液注入後、真空採血針を栓体4に刺通し、栓体4の内部と外部とを連通させ、血液検査用容器1の内部と外部との圧力差により血液の分離を開始した。まず、血漿または血清収納部にフィルタ4を通過した血清が徐々に流下され、しかる後血球が血球停止膜に到達した時点でろ過速度が大きく低下した。この時点を血液分離の終了時とした。血液分離終了時からしばらく放置した場合、実施例1〜6では赤血球が徐々に破壊され、血漿または血清保持部8内の液体が徐々に赤みを帯びてきた。すなわち、溶血した血清が血漿または血清保持部8に混入してきた。この溶血した血漿または血清が、下方の血漿または血清収納部9に混入するまでの時間を測定した。また、この時間に達した点における血清量を測定した。すなわち、血漿または血清収納部9に貯留されている分離後の血清量を測定した。また、比較例では、血液分離の終了時から、血漿または血清収納部に貯留されている血清が赤みを帯びてきたまでの時間を求め、かつ血漿または血清収納部に貯留されている分離後の血清量を測定した。結果を下記の表1に示す。

表1から明らかなように、実施例1〜6では、分離後溶血した血清の血漿または血清収納部9に混入するまでの時間が1分以上であったのに対し、比較例では、分離終了後1分経過する前に溶血した血清の混入が見られた。

また、血漿または血清保持部8の体積が5〜100μLである実施例1〜4では、溶血した血清の混入までの時間を2分以上とすることができ、かつ汚染されていない血清回収量も160μL以上と多かった。

【発明の効果】
本発明に係る血液検査用容器では、フィルタの血漿もしくは血清収納部側に、一定量の血漿もしくは血清を保持する血漿もしくは血清保持部が設けられているため、フィルタにおける分離が終了した後に赤血球の破壊により血球内の液体が漏洩したとしても、漏洩した液体が一定時間血漿もしくは血清保持部に貯留される。従って、分離された血漿もしくは血清への上記液体成分の混入を一定の時間防止することができる。従って、分離終了後に容器内の残圧を開放することなく、溶血による液体成分の汚染が生じ難い血漿もしくは血清を確実にかつ簡便に取り出すことができる。
