Abstract

PROBLEM TO BE SOLVED: To achieve the easy application of a hyaluronic acid solution to a wide range of medical applications, e.g. medical care, beauty, skin care, supplement and health care goods by safely and stably retaining a high concentration of an aqueous solution composition of powdery hyaluronic acid rich in collagen extracted from cockscombs for a long period of time.
SOLUTION: The slightly water-soluble hyaluronic acid that is extracted from cockscombs and is rich in collagen is dispersed and dissolved in water via a mixed solvent of ethanol and sorbitol to form the aqueous solution composition of at least 4-5% concentration. As powdery hyaluronic acid is converted into the aqueous solution, complicated and troublesome operations for powdery substance can be avoided, particularly the application of powdery and slightly water-soluble hyaluronic acid to medicines, cosmetics, supplements and the like becomes possible and also can be used as a substitute for bovine collagen.
COPYRIGHT: (C)2003,JPO

Description: BACKGROUND OF THE INVENTION 1. Field of the Invention The present invention relates to a health food which has been attracting attention as an effective and stable aqueous solution of a hyaluronic acid powder containing a large amount of collagen present in cockscomb, The present invention relates to a hyaluronic acid compound aqueous solution composition for easily and conveniently utilizing an application method for cosmetics, nutritional supplements, and the like. [0002] The hyaluronic acid compound has a large molecular weight and is originally water-soluble, but is a substance which is hardly soluble in water and has poor long-term stability as an aqueous solution. Or, it is usually used for cosmetics, and in each manufacturing process it was dispersed and dissolved by a unique method. However, hyaluronic acid compounds
As its features began to attract attention, those that could be provided in liquid form were more advantageous and needed to be added as appropriate. For this purpose, for example, sodium hyaluronate has been put into practical use as an aqueous solution of about 1 to 2%, but the hyaluronic acid compound is a complex polymer blend containing amino acids, and it is extremely difficult to make an aqueous solution.
%, The absolute content of the aqueous solution is small, and more water is added than necessary to satisfy the required amount, and a high-concentration solution of 5% or more is desired as a commercial value if possible.
As food solubilizers were limited by food, there were no restrictions on physical and chemical safety and there was no choice but to provide powdered products. Also, the inventor
Filed on October 10, 2001 (Japanese Patent Application No. 2001-2001)
346743) is stable under normal conditions, but contains a moderate amount of organic acid such as ascorbic acid, citric acid, malic acid and acetic acid. It was found that this kind of beverage was not suitable for health-conscious beverages. This is considered to be due to salt breakage in the acidic region where the pH of the sucrose fatty acid ester itself is low, and this disturbs the overall aqueous balance. SUMMARY OF THE INVENTION The present invention provides a low water-soluble low-concentration hyaluronic acid compound of 3 to 7%.
It is an object of the present invention to provide a composition which is preferably an aqueous solution of about 5% and has a pH which is physically and chemically stable even in a wide range of about 3 to 9 and hardly changes biochemically. Furthermore, the viscosity itself is high at room temperature (800 c
In the case of p ~), coagulation and phase separation were formed when mixed with other liquid preparations, and they did not match well. In particular, they became viscous at 10 ° C or less, and were sometimes concerned about homeostasis as commercial products. Also, if you transport or stock more than 98% of water, you will need
It was unnecessarily increased, and it was economically irrational. SUMMARY OF THE INVENTION The present invention provides
As a means for forming a stable aqueous solution of a poorly water-soluble hyaluronic acid compound at a high concentration (2 W% or more, preferably 3 to 7 W%), sorbitol and ethyl alcohol are used.
It has been found that combinations of polyhydric and polyhydric alcohols are effective. Both alcohols are extremely safe as food additives, and if the quantitative ratio is devised,
Can be used for a wide range of applications. The standard composition of the present invention An aqueous solution composition that is stable via water is obtained.
The composition may comprise propylene glycol, A water-soluble solvent such as, for example, or a polysaccharide can be added as appropriate. Sorbitol is commercially available as a sweetener for food additives, usually in a 60-70% aqueous solution. Edible (not denatured) ethyl alcohol can be used. In the composition of the present invention, the hyaluronic acid compound powder is dispersed in ethyl alcohol in advance, and sorbitol is added thereto and mixed well to prevent agglomeration of the hyaluronic acid compound. Stir vigorously while adding hot water. This can be left standing for half a day to give a brown-amber appearance. In the present invention, the content of ethyl alcohol used as a dispersing aid in advance in the hyaluronic acid compound is generally 3 to 10 W%, but is preferably suppressed to 5% or less. Can be reduced. The main component of the hyaluronic acid compound used in the present invention is a powdery substance obtained from chicken crown, so-called tosaka, which is extracted and purified after pulverization, blood removal, degreasing, heating, water addition, dehydration, and 18 kinds (eg, lysine, histidine) , Phenylalanine, tyrosine, leucine, valine, glycine, etc.) 80% collagen protein, hyaluronic acid 5
% Is the main component, and the molecular distribution with an average MW of 10,000 to 50,000 is 3,000 to 30,000, which is a dispersion type polymer compound centered at about 50%. It is heavily used. [0008] The aqueous solution of the hyaluronic acid compound of the present invention is used as a health care product derived from hyaluronic acid and collagen widely in soft drinks, supplements, dietary supplements, beauty foods, etc., at a usage concentration of about 3 to 10%. be able to. Further, the aqueous solution of the hyaluronic acid compound of the present invention may contain various additives as food emulsifiers (egg yolk lecithin, sorbitan fatty acid ester, propylene glycol, fatty acid ester, glycerin fatty acid ester), parabens, polylysine, benzoate, sorbic acid. Salt, vitamin C, vitamin E, retinol, riboflavin, calciferol, BHT, BHA, natural pigments, flavors, and plant extracts can be included in a range that does not impair the original stability. Hereinafter, the present invention will be described with reference to examples. Embodiment 1 The "stability of the aqueous solution" was examined under several conditions using the prototype. Leave at 40 ° C. for 60 days −10 ° C. Leave for 45 days 10% aq (pH: 6 to 7) 10% aq (pH: 2.7 to 3.5) 10% aq (pH: 7.8 to 8.8) The results of the evaluation are shown in Table 2. Example 2 Hyaluronic acid compound (Example 1)
Edible ethyl alcohol (C 2 H 5 OH) 5ml , sorbitol 15 g, the pH was adjusted to 100ml of sodium benzoate 0.2g of purified water with citric acid from 2.7 to 3.2
(25 ° C) is prototyped. The liquid was centrifuged at 1200 rpm and 300 SEC, and no change was observed as a result of observation of the liquid after the centrifugal separator. Among them, those excluding ethyl alcohol and those excluding sorbitol were separated into two layers to form cloudy precipitates. Using the product of the present invention prototyped by the above method (within 5%) Jasmine herb tea Isotonic beverage Green tea Soda water Lactic acid bacteria drink The plum juice was directly used for soft drinks. Also, skin care lotion, skin care cream (emulsion), lotion, cleansing cream, weak acid treatment, hair care products (shampoo, rinse,
It can be widely used arbitrarily for cosmetics such as hair dyes and dressers) weak acid soaps and quasi-drugs. According to the present invention, various drinks, foods, and the like of hyaluronic acid compounds that are more effective for human beauty and health in place of bovine collagen and hyaluronic acid, which are increasingly attracting attention in the future, will be described.
Supplements, health food and drinks, skin care, hair care,
Pharmaceutical cosmetics, toiletry goods (Champulins,
It can be widely and easily used for hair dressers, hair dyes, and hair growth products.
