本発明は、一般的に、好酸球レベルを枯渇する方法、およびそのために使用さ
れる組成物に関し、そしてより詳細には、好酸球の増加した集団に関連する疾患
または状態を処置する方法に関する。
発明の背景
好酸球は、顆粒球系統の白血球細胞である。それらの正常な機能として、寄生
的な(parasitic)感染、特に、寄生虫様の感染の防止が挙げられる。例えば、J

現在のグルココルチコイドステロイドは、アレルギー性疾患(例えば、喘息)
の急激な影響を処置するための好ましい治療用薬物である。しかし、延長された
ステロイド処置は、多くの有害な副作用を付随する。例えば、GoodmanおよびGil

より最近は、特定の免疫障害に関連する好酸球増加症が、インターロイキン-5
(IL-5)に対するアンタゴニストの投与によって処置され得ることが示されてい

特許第5,096,704号を参照のこと。IL-5は、強力な好酸球分化因子であるが、こ
れはまた、他の免疫細胞(例えば、B細胞)に対しても効果を発揮する。しかし

好酸球増加症に関連する障害を処置するための代替的なまたは相補的なアプロ
ーチの利用可能性は、重要な臨床的な有用性を有する。関連する組織における好
酸球の産生または蓄積の予防は、これらの障害または疾患の基礎となる原因をブ
ロックし得る。
発明の要旨
本発明は、好酸球ケモカインであるエオタキシン(eotaxin)のレセプターに
対するアンタゴニストの有効量を投与することによって、好酸球を枯渇する方法
を提供する。発明者らは、これによって好酸球の数の減少を導くことを観察した
。CCR3と命名されているレセプターは、好酸球上で主に発現される。好ましくは
、CCR3のアンタゴニストはモノクローナル抗体であるか、または標準的な技術に
よりそれらから誘導される結合組成物である。
本発明は、CCR3レセプターに特異的に結合する抗体または結合組成物を提供し
、サンプル中の好酸球増加症のレベルを低下させ、そしてリガンドの結合をブロ
ックしない。好ましい実施態様において、抗体はモノクローナル抗体である。抗
体または結合組成物、および薬学的に受容可能なキャリアを含む滅菌組成物もま
た、提供される。
本発明は、有効量の抗体または結合組成物を、単独で、またはインターロイキ
ン-5(IL-5)もしくはステロイドに対するアンタゴニストと組み合わせてのいず
れかで投与することによる、個体における好酸球に関連する疾患または障害を寛
解させる方法を含む。好ましい実施態様において、抗体はモノクローナル抗体で

過好酸球増加症であり得る。IL-5に対するアンタゴニストは、インタクトなモノ

有効量のIL-5モノクローナル抗体、その結合フラグメント、または可溶性のレ

本発明はさらに、サンプル中の好酸球の存在について検出する方法を提供する

抗体または結合組成物と混合物とを接触させる工程、および細胞の集団を単離
する工程による、細胞の混合物からCCR3レセプターを発現する細胞の集団を単離

I.概論
本発明は、好酸球特異的ケモカインのレセプターであるエオタキシンが、好酸
球上に主に発現されるという知見に、一部基づく。約30個のモノクローナル抗体
(mAb)のパネル全体からは、驚くべきことに、2つのmAbは、寄生虫に寄生され
たマウス中の好酸球のレベルを有意に低下し得たが、レセプターへのリガンド結
合をブロックしなかった。低下したレベルは、任意のいくつかの機構(例えば、
補体結合/枯渇、またはマクロファージによるオプソニン作用)に起因し得る。

ケモカインは、特異的なGタンパク質に連結されている7回の膜貫通レセプタ
ーに結合することによって、古典的に白血球の通行を媒介する、化学誘引物質サ
イトカインのサブファミリーである。ケモカインは、最初の2つのシステインの
一次配列に基づいて4つの群に分けられる:CXC、CC、C、および新たに発見され

II.抗体;結合組成物の作製
本発明は、CCR3(好ましくは、哺乳動物、例えば、霊長類、ヒト、猫、イヌ、
ラット、またはマウス))に特異的に結合する抗体または結合組成物の使用を提
供する。抗体は、それらの天然に存在する(全長)形態またはそれらの組換え形
態の両方で、個体、多型、対立遺伝子、株、または種々の変異体、およびそれら
のフラグメントを含む、種々のCCR3タンパク質に対して惹起され得る。特に、興

ポリクローナル抗体を産生する方法は、当業者に周知である。代表的には、免
疫原(好ましくは、精製されたタンパク質)がアジュバントと混合され、そして
動物はその混合物で免疫される。免疫原調製物に対する動物の免疫応答は、試験
採血を行うこと、および目的のCCR3タンパク質に対する反応性の力価を決定する

抗体は、特異的結合組成物の単なる1つの形態である。しばしば類似の用途を

CCR3タンパク質またはペプチドの測定のためのイムノアッセイは、当業者に公
知の種々の方法によって行われ得る。簡潔には、タンパク質を測定するためのイ
ムノアッセイは、競合または非競合結合アッセイのいずれかであり得る。競合結
合アッセイにおいて、分析されるべきサンプルは、固体表面に結合された捕捉試

ク質からの結合した標識タンパク質の分離を達成するために、固体に表面に結合

あるいは、分離工程が必要ではない同種のイムノアッセイが、行われ得る。こ

CCR3タンパク質はまた、種々の非競合イムノアッセイ方法によっても決定され

ウェスタンブロット分析が、サンプル中のCCR3タンパク質の存在を決定するた
めに使用され得る。電気泳動が、例えば、タンパク質を含有すると疑われる組織
サンプルについて行われる。タンパク質を分離するための電気泳動、および適切
な固体支持体(例えば、ニトロセルロースフェルター)へのタンパク質の転移後
、固体支持体は、タンパク質に反応性である抗体とともにインキュベートされる

くインキュベーションによって検出され得る。

mAbおよび結合組成物の結合および活性を評価するためのスクリーニングは、

成物は、循環する好酸球のレベルを有意に低下させる。
抗体の評価は、他の動物(例えば、人)において種々の方法を使用して行われ
得る。例えば、血液サンプルが、好酸球関連疾患または障害に罹患している患者
から、候補のmAbでの処置の前および後に採取され、そして好酸球の計数が行わ
れる。抗体は、例えば、FACS、組織染色によって、インビトロ培養物中で、好酸
球増加症の程度を評価するために、診断的な状況で使用され得る。
IV.用途
本発明は、好酸球増加症に関連する疾患(例えば、肺性の炎症、皮膚炎など)
