【発明の詳細な説明】
新規微生物
本発明は免疫原性炭水化物莢膜の生産をコードするDNAで形質転換された新
規な非侵襲性又は非病原性のグラム陽性細菌宿主微生物、特にラクトコッカス・
ラクティス(Lactococcus lactis)微生物に関する。このような細菌宿主微生物
を含む組成物並びにワクチンにおけるその使用も提供される。
一つの重要な病原性微生物は、普通肺炎球菌と呼ばれるストレプトコッカス・
ニューモニエ(Streptococcus pneumoniae)である。発展途上国及び先進国にお
けるヒトの病気との関連でストレプトコッカス・ニューモニエ感染の継続的重要
性が権威ある総説により最近指摘された(1)。これは、地球規模で、この微生
物が急性呼吸器感染の最も普通にみられる原因細菌であると信じられ、そしてほ
とんどは発展途上国であるが、その結果毎年100万人の子供が死亡すると推定
されていると指摘している(2)。アメリカ合衆国では、肺炎球菌はなお細菌性
肺炎の最も普通の原因であり、そして病気の割合は若年の子供達、高齢者、そし
て無脾症、心臓、肺臓及び腎臓疾患、糖尿病、アルコール中毒症などの罹患し易
い状態を持つ患者、又は免疫抑制疾患、特にエイズを患う患者で特に高いことが
示唆されている(3)。これらのグループは肺炎球菌敗血症、従って髄膜炎のリ
スクが高くなり、従って肺炎球菌感染による死亡のリスクが大きくなる。肺炎球
菌は、先進国の子供達の支配的な感染であり続け、そしてかなりの出費を招いて
いる中耳炎や副鼻腔炎の主要な原因でもある。
肺炎球菌感染に対する有効な予防戦略の必要性がペニシリン耐性肺炎球菌の最
近の出現により注目を浴びている。米国の12の州の13の病院における肺炎球
菌単離菌の6.6%がペニシリンに耐性であることが見出され、そして単離菌の
一部は第3世代のシクロスポリンを含む他の抗生物質にも耐性であると最近報告
された(4)。ペニシリン耐性の割合は一部の病院ではもっと高くなる(20%
まで)(5)。肺炎球菌の中でのペニシリン耐性の出現はペニシリンが有効な治
療であった数十年の後に最近そして突然にやって来たので、これらの発見は警鐘
と考えられる。
従って、上記の理由で、肺炎球菌疾病の予防、制御、診断又は治療の諸手段の
改善を検討することについては説得力のある根拠がある。
全世界的な子供ワクチン接種プログラムに組み込まれうるであろう肺炎球菌ワ
クチンの開発に努力するとき、二つの主要な問題に遭遇することが良く知られて
いる。これらは、(a)肺炎球菌の抗原的多様性、少なくとも84種の免疫学的
に異なる血清型が知られており、そのそれぞれがワクチンとして使用されるとき
それ自身の血清型に対してのみ保護を誘発することができるだけであること、そ
して(b)ワクチンとして精製し使用するとき、莢膜多糖類(そのそれぞれが血
清型を決定し、そして主要な保護抗原である)が、侵襲性肺炎球菌感染及び髄膜
炎の最高の発症率を示す年代グループである2歳未満の子供達では信頼できる程
には保護的な抗体応答を誘発しないという事実(6)である。これらの問題は莢
膜抗原(例えば、グループBストレプトコッカス、ヘモフィルス・インフルエン
ゼ及びナイセリア・メニンゴコッカス)に基づく他のワクチン戦略にも当てはま
る。ただし、これらの種内では、莢膜の型不均質性の程度は、より限定的である
ようにみえる。
肺炎球菌感染の予防に関して米国で行われている現在の戦略は、最も普通の肺
炎球菌血清型の中の23種由来の莢膜多糖類抗原を含むワクチンを製剤化するこ
とである。これらの株を合わせると、米国及び欧州の子供達由来の侵襲的肺炎球
菌単離菌のほぼ90%を、そしてアジアにおける肺炎球菌感染の70%未満を占
める(7、8)。実用的には、本質的に様々な異種の莢膜を製造し、その品質を
コントロールし、そして適当な量で混合せねばならないので、これらのワクチン
を製造する費用は高くつく。
本発明は保護的免疫原をもっと単純な細菌醗酵(病原性の細菌であるストレプ
トコッカス・ニューモニエそれ自体ではなく、ラクトコッカス・ラクティスなど
の非病原性細菌の生育を含む)で生産することができる手段を提供する。本発明
は二つ以上の多糖類免疫原が同一細菌により生産され、それによって製造し、品
質を管理し、そして混合せねばならない異種の免疫原の数を減少させる手段をも
提供する。驚くべきことに、本発明者らは、病原性細菌の莢膜の合成に責任のあ
る酵素をコードするオペロンの全部又は一部を含むDNAを用いて、非侵襲性又
は非病原性のグラム陽性細菌宿主微生物を形質転換又はトランスフェクトすると
、病原性微生物の周囲に正常に見出される多糖類莢膜と免疫学的にそして構造的
に同一の多糖類莢膜を持つ宿主微生物を生ずることを発見した。全オペロンを使
用したときでさえもこの莢膜の正確な発現、分泌及び組立が生ずることは、これ
らのプロセスに伴う複雑さそしていわゆる「管理維持」遺伝子と呼ばれるもの、
例えばこのオペロンそれ自体の一部を形成するものではないがこの病原性微生物
由来の多糖類の移送に関与する遺伝子など、の関与の可能性を考慮すると、驚く
べきことである。
ラクトコッカス・ラクティスなどの非病原性グラム陽性細菌中で病原性細菌由
来の多糖類免疫原の発現及び/又は産生に関する記録は科学的文献には存在しな
い。従って、本発明はラクトコッカス・ラクティスなどの非病原性グラム陽性微
生物中で病原性細菌由来の多糖類免疫原の発現及び生産を達成する実用的方法を
初めて開示するものである。さらに、本発明はこのような微生物の多糖類発現株
により多糖類莢膜への免疫応答が引き出され得ることを開示する。
本発明が多くの異なる種類の用途に用いることが可能であることを熟練者は認
めるであろうが、抗莢膜に基づくワクチンそして殊に肺炎球菌用ワクチンの分野
に対するその貢献は特に重要である。本発明によって開かれた別の重要な可能性
は、免疫原性多糖類莢膜に対する、粘膜経由、例えば経鼻又は経口で送達できる
、ワクチンの提供である。
こうして、第1の側面で本発明は病原性細菌由来の多糖類免疫原の生産に責任
のある一つ以上の酵素をコードするDNAで形質転換又はトランスフェクトされ
た非侵襲性又は非病原性のグラム陽性細菌を提供する。
使用することができるグラム陽性細菌の例としては、リステリア・イノキュア
(Listeria innocua)、スタフィロコッカス・キシローサス(Staphylococcus xyl
osus)、スタフィロコッカス・カルノーサス(Staphylococcus carnosus)、スト
レプトコッカス・ゴルドニイ(Streptococcus gordonii)、ラクトコッカス・ス
ピーシーズ(Lactococcus species)又はラクトバシラス・スピーシーズ(Lactobac
illus species)が挙げられる。本発明の一つの好ましい態様はラクトコッカス・
ラクティスを使用するものである。
また、本発明はグラム陽性病原性細菌の弱毒株、例えば、リステリア、例えば
、リステリア・モノサイトトゲネス(Listeria monocytogenes)のワクチン株を
利用することができる。
好ましい態様では、多糖類免疫原は多糖類莢膜を形成する。このような莢膜の
一つの実例はストレプトコッカス・ニューモニエ由来のものである。好ましい莢
膜血清型の選択は場所等によって決定されると理解されよう。こうして、一般に
、上記の23個の最も普通の多糖類血清型の一つ以上を発現させたいと望むこと
になる。しかしながら、このグループ内でさえ、ある領域の許容可能なレベルの
保護を達成するために、より少ない数の発現を提供する必要があるにすぎない。
本発明は、おそらく、発現される莢膜多糖類の「ライブラリー」を構築し、つい
で特定領域又は特定集団において最も普通の血清型用に特注されたワクチンを生
産させるためそれを適当に混合することにより、エス.ニューモニエ由来の84
種の莢膜血清型の他のものを発現させるためにも勿論使用することができる。
本明細書に記載した実施例はエス.ニューモニエ由来の多糖類免疫原を生産し
及び/又は送達するためのエル.ラクティスの使用に関するけれども、本発明は
いかなる病原性細菌由来の天然又は組換え多糖類免疫原の生産及び/又は送達に
も適用することができる。これらの多糖類免疫原は、例えば、グループBストレ
プトコッカス由来の多糖類免疫原、動物やヒトに病気を惹起する他の種のストレ
プトコッカス由来の多糖類免疫原、莢膜に包まれたスタフィロコッカス及び他の
グラム陽性病原体由来の多糖類免疫原、又はたとえそれがグラム陽性であろうと
グラム陰性であろうと(イー.コリ、ヘモフィルス・インフルエンゼ(Haemophi
lus influenzae)、ナイセリア・メニンジティディス(Neisseria meningitidis
)の侵襲性莢膜化株など)、その多糖類莢膜生合成遺伝子がエル.ラクティスな
どの微生物中で発現することができる如何なる他の微生物由来の多糖類免疫原な
どであってもよく、無害な宿主中で実用的に発現させることができるものである
。
グループBストレプトコッカス(GBS)の莢膜多糖類はエル.ラクティスな
どの無害な微生物を用いて生産及び/又は送達されうる莢膜の例である。ヒトの
疾病と関連するGBSは一般に莢膜を有し、9種の血清学的に異なる莢膜多糖類
の中の一つを含んでいる。小さな例外はあるが、これらの多糖類はグルコース、
ガラクトース、N−アセチルグルコサミン及びN−アセチルノイラミン酸又はシ
アル酸を含んでいる。単糖類の反復ユニット中への具体的配置が各莢膜の免疫学
的特異性を決定する。タイプIII GBSオペロンの染色体領域の地図が作成され
、DNAプローブを用いるサザン・ハイブリダイゼーション研究により、テスト
した血清型の中ではその全体領域が高度に保存されていることが明らかにされた
(9、10)。さらに、この領域の両側の遺伝子の配列は多糖類の合成に関与す
る酵素群をコードする他の細菌の遺伝子と著しい相同性を共有することが発見さ
れた。グループBストレプトコッカスの莢膜生合成オペロンの両側には、こうし
てcpsF及びcpsE(それぞれ、CMP−シアル酸シンテターゼ及びアセチ
ルトランスフェラーゼをコードすると考えられる)と命名された遺伝子が一方の
端に、そしてこのcpsサブユニットの合成及び移送/重合に関与すると考えら
れる遺伝子が他方の端に位置するようにみえる。従って、他のオペロンの生合成
遺伝子はタイプIII cpsオペロン中で同定された保存された遺伝子の公表され
た配列に基づくプライマーを用いてPCR増幅により容易に得ることができる。
GBScps生合成遺伝子が容易に得ることができるとなれば、例えば、これら
の酵素の不可欠な組合せを発現させそしてGBS莢膜多糖類を生産するエル.ラ
クティスの株を構築することが可能となりうる。
宿主微生物を形質転換又はトランスフェクトするために使用するDNAは多糖
類莢膜の合成に必要な酵素類をコードするオペロンの全部か又は一部を含むこと
ができる。本質的な要件は、もちろん、外因性DNAによりコードされる酵素が
宿主微生物中に存在する一つ以上の酵素と一緒になって莢膜のための完全な合成
経路を提供することである。
以下に例示する一つの態様では、本発明者らは肺炎球菌タイプ3cps生合成
オペロン由来のDNA断片をエル.ラクティス中のプラスミドベクター上にクロ
ーニングすることができることを示す。さらに、エル.ラクティスを肺炎球菌生
合成オペロンの断片を保持するプラスミドで形質転換すると、エル.ラクティス
中で多糖類莢膜の生産が起こり、そしてこの多糖類莢膜は肺炎球菌中で生産され
る莢膜に抗原的にそして構造的に同一である。下記の実施例1では、エル.ラク
ティスで発現されるこの莢膜遺伝子がそれ自身のプロモーターと翻訳シグナルを
保持している。しかしながら、熟練者は他のプラスミド、プロモーター及び/又
は翻訳シグナル、例えば宿主微生物由来のものを用いて本発明を実施することが
可能であることを認めるであろう。実際、最近文献(11、12)に記載された
構成的又は誘導的ラクトコッカスプロモーターを使用することにより、莢膜合成
のレベルを変化させ、そしてエル.ラクティスにより生産される莢膜のレベルを
高めることができる可能性がある。同様な結果がこのような「注文仕立ての」プ
ロモーター等を用いて他の微生物でも得ることができるはずである。
さらに、高レベルの多糖類発現がウエルズら(13及びGB−B−22783
58号)により開発された誘導可能なT7発現システムを用いエル.ラクティス
で達成することができるであろうことが予想されるであろう。
二つ以上の異種莢膜多糖類の合成に必要な遺伝子を保持する細菌の株を構築す
ることによるか(異種のcpsオペロンが染色体の異なる遺伝子座に組み込まれ
たときは肺炎球菌中で可能であることが示された(14))、又は二つ以上の血
清型由来の保護的エピトープが同一多糖類内に存在するような組換え多糖類を生
産する、例えばエル.ラクティスの株を構築することにより、ラクトコッカス・
ラクティスなどのグラム陽性細菌で二つ以上の抗原を同時に生産することができ
るであろう。二つ以上の異種莢膜エピトープを含むように莢膜多糖類を工学処理
することは、病原性細菌由来の莢膜生合成酵素類の不可欠な組合せを発現する例
えばエル.ラクティスの株を構築することにより可能となりうる。
肺炎球菌多糖類抗原などの多糖類抗原を生産するために、エル.ラクティスな
どの非病原性GRAS微生物を使用することは、ワクチンとして多糖類を生産し
そして製剤化する費用を大きく減少させる手段を提供する。エル.ラクティスの
高生産株は多糖類(この多糖類はついで精製しそして選ばれた担体タンパク質に
結合させられる)を生産するために使用することができるし、あるいは多糖類を
生産するエル.ラクティスの細胞はワクチンとして直接使用することもできよう
。
こうして、第2の側面で、本発明は病原性細菌の多糖類免疫原を生産する方法
であって、その多糖類免疫原の生産に責任のある一つ以上の酵素をコードするD
NAを用いて非侵襲性又は非病原性のグラム陽性細菌を形質転換する工程及び/
又はその細菌を培養する工程を含む方法を提供する。
上に論じたように、本発明のグラム陽性細菌は多糖類免疫原を送達するのに使
用することができよう。こうして、第3の側面では本発明は医薬における使用の
ために本発明の微生物を提供する。
特に、本発明は莢膜多糖類抗原に対するワクチンを提供する一つの代替手段を
提供する。こうして、第4の側面では、本発明は本発明の非侵襲性又は非病原性
グラム陽性細菌を含むワクチンを提供する。このようなワクチンは、多糖類を発
現する微生物、例えばエル.ラクティスの生きた調製物か又は殺菌された調製物
を含むことができよう。特に、粘膜経路を経て投与されるのに適するワクチンが
提供される。このような粘膜経路の例としては、鼻、口、経気管支組織、眼、耳
、直腸、膣、尿道、又は舌下が挙げられる。粘膜経路としては経鼻及び経口が特
に好ましい。本発明のワクチンはヒトに使用するために特に提供される。もっと
も、このようなワクチンは動物へのワクチン接種プログラムに使用するために製
剤化することもできるであろう。
本発明の微生物を製剤化するときは、熟練者はある範囲の遅延放出、時間を定
めた放出、又は適当に保護された製剤、例えばワクチンがそれが最も有効となる
ような部位、例えばパイアー斑に送達されることを確保するための腸溶被覆カプ
セルを勿論利用することができる。さらに、本発明のワクチンは従来からのアジ
ュバントを含んでもよい。
このようなワクチンは粘膜経路の免疫処置を利用するであろう。そして発現さ
れた多糖類(単数又は複数)に対する粘膜の免疫応答及び全身的免疫応答の両方
を引き出すことが予想されるであろう。エル.ラクティスなどのグラム陽性微生
物はタンパク質抗原に対する有効な送達ビークルとして使用することもできるこ
とが示された(国際特許出願第PCT/GB96/02580)。これらの研究
では、抗原である破傷風毒素断片Cを発現する組換え体エル.ラクティスを用い
てマウスを経口及び経鼻で免疫処置したところ、発現した抗原に対する高い保護
レベルの血清抗体応答が誘導された。
従って、第5の側面で、本発明は多糖類莢膜を有する病原性細菌に対するワク
チンを主体に接種する方法であって、本発明のグラム陽性細菌の有効量を主体に
投与する工程を含む方法を提供する。
さらなる側面では、本発明は多糖類莢膜を有する病原性細菌に対する粘膜免疫
を誘導する方法であって、本発明のグラム陽性細菌の有効量を主体に投与する工
程を含む方法を提供する。
粘膜経路の免疫処置の主要な利点の一つは、それが粘膜免疫システムを刺激す
る能力を有し、こうして身体の粘膜表面で抗原特異的抗体の分泌を誘発すること
である。粘膜表面のコロニー形成は莢膜を有する細菌(例えば、肺炎球菌、ヘモ
フィルス・インフルエンゼ、ナイセリア・メニンジティディス及びグループBス
トレプトコッカス・アガラクティエ(Streptococcus agalactiae))を含め多く
の病原体による感染の前提条件であるから、粘膜表面に抗莢膜抗体を誘発するワ
クチンは感染予防に、そして集団でのこの微生物の無症候の移送の予防にもより
有効であるかも知れない。
他の側面では、本発明は下記のものを提供する、
i) 病原性細菌由来の多糖類免疫原の生産に責任のある一つ以上の酵素をコー
ドするDNAを含むDNA構築物、
ii) i)で規定された構築物を含むベクター、例えば、プラスミドベクター、
iii) 非侵襲性又は非病原性グラム陽性細菌の形質転換又はトランスフェクショ
ンにおけるi)で規定された構築物の使用、そして
iv) 多糖類莢膜を有する病原性細菌に対するワクチンの調製における本発明の
細菌の使用。
本発明の各側面の好ましい性質は必要な変更を加えて他の側面のそれぞれにも
同様に適用可能である。
本発明は下記の実施例によりここに記述されるが、これらの実施例は如何なる
意味でも制限として理解すべきでない。これらの実施例は下記の図面を参照する
。
図1はタイプ3莢膜多糖類合成に関与する酵素をコードする領域のDNA配列
上のプライマーS1、A1及びA2の位置を示す(ゲンバンク、受託番号U15
171(19)。
図2はpTREPにおけるpTREPカセットの配列及び不可欠な性質を示す

図3はストレプトコッカス・ニューモニエにおけるタイプ3莢膜多糖類(cp
s)の生合成経路を示す。グルコース−6−リン酸をエス.ニューモニエで見出
されたタイプ3莢膜多糖類構造に変換するには4個の酵素が必要である。莢膜の
移送及び結合のためには、さらなる機能が必要である。
図4はエス.ニューモニエ中のタイプ3cps遺伝子座の遺伝的構成を示す。
二つのプロモーターが存在する(Pと示す)。一方のプロモーターはcps3D
遺伝子及びcps3S遺伝子を複製し、そして他方はcps3U遺伝子及びcp
s3M遺伝子を複製する。DNA断片CPS1及びCPS2は、鋳型としてタイ
プ3肺炎球菌染色体DNAを用い、そしてプライマーS1をプライマーA2及び
プライマーA3とそれぞれ組み合わせて用いるPCRにより作成した。これらの
プライマーはpTREP中にこの断片をクローニングし易いようにその5’末端
にBamHI制限エンドヌクレアーゼ部位を含むように設計した。
図5は、プラスミドpTREP、pTREP−CPS1及びpTREP−CP
S2の図式的表示を示す。MLSはマクロライド、リンコスアミド及びストレプ
トグラミンB耐性決定因子を表し、Tは下流のターミネーターを表す。不可欠な
制限部位の位置が示してあり、クローニングされた肺炎球菌のDNA断片である
CPS1及びCPS2(上記の図1B)は太い線で示してある。
図6は、実施例に記述されたようなタイプ3特異的抗血清を用いる、エル.ラ
クティスによるタイプ3cps生産の免疫検出を示す。(1)pTREPを含む
エル.ラクティスの非発現株、(2)エル.ラクティスのcps発現株UCP1
619、及び(3)エル.ラクティスのcps発現株UCP1618。
図7は、(1)ラクトコッカス・ラクティスの発現株UCP1619及び(2
)タイプ3ストレプトコッカス・ニューモニエの精製された多糖類の類似のプロ
トンNMRスペクトルを示す。4個の水素ピーク(A、B、C及びD)の積分は
莢膜多糖類の二つの試料について類似の割合を与えた(1では、A:B:C:D
=10.2:28.3:53.8:7.7であり、2では、A:B:C:D=1
3.8:31.1:46.2:8.9である)。これらの結果はエル.ラクティ
スにより生産されるcpsの構造はタイプ3エス.ニューモニエで天然に生ずる
ものと同じであるという証拠となる。
図8は、タイプ3cpsを発現するエル.ラクティスの組換え体株で免疫処置
したマウスにおける抗タイプ3cps−IgG血清抗体応答を示す。免疫処置後
21日目と35日目におけるcps特異的抗体の平均タイターは未処置群マウス
よりも有意に(p=0.004)高かった。誤差のバーは平均値からの標準偏差
を表す。これらの結果はcpsを発現するエル.ラクティスでの免疫処置がマウ
スで特異的な抗cps血清抗体応答を誘発することを示す。
図9は、(1)タイプ3エス.ニューモニエ(335Kで60時間)及び(2
)エル.ラクティス株1619(320Kで44時間)由来の精製cpsの13C
NMRスペクトルを示す。両スペクトル共600Mhz装置で得られた。両ス
ペクトルで、炭素ピークの位置は同一である。
図10は、生きたエス.ニューモニエでチャレンジした後4時間目における、
実施例6に記述したようにエル.ラクティス1619で免疫処置したマウス又は
対照マウスからのBAL中に存在するエス.ニューモニエの、チャレンジした接
種物に対する%を示す。
図11は、生きたエス.ニューモニエでチャレンジした後4時間目における、
実施例6に記述したようにエル.ラクティス1619で免疫処置したマウス又は
対照マウスからのBAL中のエス.ニューモニエの細菌回収を示す。
図12は、a)長いDNA断片を合成するために使用したオリゴヌクレオチド
プライマーであって、グループBストレプトコッカスの9種の血清型全てからの
莢膜生合成遺伝子を含むプライマー、及びb)GBSの6種の異なる血清型から
得られたPCR産物の制限エンドヌクレアーゼ消化物を示す。実施例1 エス.ニューモニエのタイプ3cps生合成経路酵素のエル.ラクティスでのク ローニング及び発現
i)生合成経路酵素をコードするDNA断片の調製
タイプ3多糖類莢膜を生産するストレプトコッカス・ニューモニエ株WU2の
ゲノムDNAを、グルコースを含むBTS培地(ディフコ)で生育させた細胞か
ら標準的方法により調製した。この細菌はジュアン遠心分離機(モデルCR31
2)中3000gで10分間の遠心分離により生育培地から回収し、そして10
0μlのDNAプレプ緩衝液(10mMトリス−塩酸、pH8.0、1mMのE
DTA、25%w/vスクロース)中に再懸濁し、これに10μlの10%w/
vドデシル硫酸ナトリウム(SDS)及びプロテイナーゼK(シグマ)の20m
g/ml溶液0.6μlを添加した。この溶液を37℃で一晩インキュベートし
、ついでDNAをクロロホルム、フェノール及びイソアミルアルコール(IAA
)(24:24:2)の混合液で2回そしてクロロホルム:IAA(98:2)
で2回抽出した。3M酢酸ナトリウム0.1倍容と絶対エタノール2倍容を添加
することによりこのゲノムDNAを20分間−70℃で沈澱させた。DNAのペ
レットを遠心分離により回収し、100μlのTE(トリス−塩酸pH8.0、
1mMのEDTA)で再懸濁し、4M塩化リチウムの20μl及び2.5倍容の
エタノールを添加することにより第2回目の沈澱を行った。精製したゲノムDN
Aは最後にTEに再懸濁し、4℃に貯蔵した。
タイプ3莢膜生合成に必要な遺伝子をコードするDNA断片は、次に鋳型とし
てエス.ニューモニエのゲノムDNAを用い、タイプ3莢膜遺伝子座の公開され
た配列に基づくオリゴヌクレオチドプライマー(センスプライマーS1とアンチ
センスプライマーA2又はA3)を用いるPCRにより取得した(cpsオペロ
ンおよびプライマーの配列については図1を見よ)。このPCRのセンスおよび
アンチセンスプライマーは続くクローニング工程を容易にするためそれらの5’
末端にBamHI制限エンドヌクレアーゼ部位を含むように設計した。
このPCR反応混合液は、製造者(ストラタジーン)により供給されたTaq
プラス反応緩衝液中に、約15ngのWU2株由来の染色体DNA、20pmo
lの各プライマー、250μMの各dNTP、2ユニットのTaqポリメラーゼ
及び1μlのTaqエクステンダー(ストラタジーン)を含んでいた。変性工程
(95℃で30秒間)、アニーリング工程(60℃で60秒間)及びポリメラー
ゼ伸長工程(72℃で180から240秒間)を含む35サイクルの後、プライ
マーS1+A2及びS1+A3で得られたPCR産物を精製しそして標準的方法
を用いるアガロース・ゲル電気泳動により予想されたサイズのもの(それぞれ約
3及び44kb)であることを測定した。
ii) pTREX及びpTREPの構築
推定的RNS安定化配列、翻訳開始領域及び標的遺伝子のための複数のクロー
ニング部位をコードする合成オリゴヌクレオチド類は、150mMのNaClを
含む1×TBEの200μl中の各オリゴヌクレオチド200μg中の各オリゴ
ヌクレオチド20μgをボイルし、室温で冷却させることによりアニールした。
アニールしたオリゴヌクレオチドは250μMのデオキシヌクレオチド三リン酸
及び1.5mMのMgCl2を含む1×TF1緩衝液中、Tf1DNAポリメラ
ーゼを用い、35℃、45℃、55℃及び65℃でそれぞれ1分間、その後72
℃で10分間伸長させた。センス及びアンチセンス・オリゴヌクレオチドには、
さらなるクローニングをし易くするためその5’末端にそれぞれNheI及びB
amHIに対する認識部位を含めた。
得られた2本鎖DNAをNheI及びBamHIで切断しpUC19の誘導体
であるpUC19NT7中のXbaI部位とBamHI部位の間にクローニング
した。このpUC19NT7はEcoRI部位とHindIII部位の間にクロー
ニングされたpLET1由来のT7発現カセット(17)を含んでいた。得られ
た構築物はpUCLEXと命名した。
次いで、pUCLEX中の完全な発現カセットをHindIIIで切断しそして
平滑化することにより取り出し、その後EcoRIで切断した後pIL253の
EcoRI部位及びSacI(平滑化)部位中にクローニングしてベクターpT
REXを作成した。
プラスミドpTREPは次のようにして構築した。すなわち、ビー.リヘニフ
ォルミスのペニシリナーゼ遺伝子の3’末端由来のrho独立ターミネーター、
PUC逆配列プライマー、プロモーター挿入のための複数のクローニング部位及
び普遍的翻訳停止配列(すなわち、3リーディング・フレームすべての停止コド
ン)をコードする合成オリゴヌクレオチド類を上述のようにアニールした。アニ
ールしたDNA断片をEcoRV及びBamHIで切断し、EcoRIで切断し
ておいたpTREX中にクローニングし、平滑化し、次いでBamIで切断した
。生ずるプラスミドをpTREPと命名し、以下に述べるようにエス.ニューモ
ニエ由来のcpsオペロン遺伝子のクローニングに使用した。pTREPの詳細
は図2及び図5に示す。
iii) エス.ニューモニエ由来のcpsオペロン遺伝子のpTREPへのクロ
ーニング
精製したDNA断片(cps1及びcps2は図5に示す)を標準的条件を用
いBamHIで消化し、ライゲーションキット(アマシャム)及び製造者により
推奨された条件を用いてBglII及びBamHIで切断し脱リン酸化し精製した
プラスミドpTREPに連結した。この連結されたDNAを先に記述された(1
6)ようにコンピテントな(受容能力のある)エル.ラクティス中にエレクトロ
ポレート(電気穿孔)し、プラスミドを保持する形質転換体を選択するため形質
転換体を5μg/mlのエリスロマイシンを含むGM17寒天上で回収した。得
られた、エス.ニューモニエタイプ3cps遺伝子座の3及び4kbDNA断片
を保持するプラスミドをそれぞれpTREP−CPS1及びpTREP−CPS
2と命名した。これらを図5に示す。pTREP−CPS1にクローニングされ
た3kbの肺炎球菌DNA断片は、UDPグルコースデヒドロゲナーゼ及びタイ
プ3莢膜多糖類シンテターゼ及びそれらの関連するプロモーターをコードするc
ps3D及びcps3S遺伝子を含んでいる(図5)。pTREP−CPS2中
にクローニングされた肺炎球菌DNA断片はほぼ1kb長くそしてcps3D及
びcps3Sに加えてグルコース−1−リン酸・ウリジル・トランスフェラーゼ
に対するcps3U遺伝子を含んでいる(図3)。cpsM遺伝子はクローニン
グしなかった。これはこの酵素がエル.ラクティス中に天然に存在すると予想さ
れたからである。以下の実施例2に示すように、この予想は正しかった。
プラスミドpTREP−CPS1及びpTREP−CPS2を保持するエル.
ラクティス株はそれぞれUCP1618及びUCP1619と命名した。これら
の株による多糖類莢膜の生産はスライドガラス上の細菌の薄い乾燥スメアをグラ
ム及びニグロシンで染色した後光学顕微鏡により肉眼で見ることができる。実施例2 エル.ラクティスにより生産される莢膜多糖類は肺炎球菌cpsタイプ3に特異 的な抗血清により認識される。
エル.ラクティス株、UCP1618及びUCP1619を新鮮な一晩培養か
ら0.5%w/vグルコース(GM17)及びエリスロマイシン(5μg/ml
)を含むM17ブロス中に100倍に希釈し、30℃で3時間生育させた。次い
で、この細菌培養を遠心分離によりほぼ10倍に濃縮し、そして5μlをニトロ
セルロース片上にスポットし乾燥させた。負の対照としてベクターpTREPを
保持するエル.ラクティス株を同様に処理し、そしてニトロセルロース濾紙に適
用した。既に記述されたように(12)、タイプ3cps抗血清を用いてイミュ
ノブロッティングを行った。ただし、ブロッキング剤として牛血清アルブミンの
代わりに乾燥ミルク(2%w/v)を使用した。その結果、タイプ3cps抗血
清はエル.ラクティス株UCP1618及びUCP1619がその上にスポット

とが可能となりうる。実施例3 組換えエル.ラクティスにより生産されるタイプ3多糖類莢膜のプロトン核磁気 共鳴(NMR)分析
エル.ラクティスにより生産される莢膜多糖類は、MOPS緩衝化培地(MO
PS,アレクシス・コウ,6.7g/リットル、pH7.2、イースト・ナイト
ロジェン・ベース,ディフコ,25.11g/リットル、カゼイン酵素水解物,
シグマ,2g/リットル、グルコース0.5%w/v及びエリスロマイシン5μ
g/ml)に一晩生育させたエル.ラクティス株UCP1619から精製した。
100%トリクロロ酢酸0.25倍容を添加し、ジュアン遠心分離機(モデルC
R312)中、2000rpmで10分間遠心分離することにより培養液からタ
ンパク質を除去した。cpsは、等容量のアセトンで沈殿させ、ジュアン遠心分
離機(モデルCR312)中、3000rpmで10分間遠心分離することによ
り回収した。次いで、エル.ラクティスからのcpsを5%(w/v)酢酸ナト
リウム中に再溶解し、残りのタンパク質をクロロホルムとブタノールの混合液(
5:1)で抽出することにより除去した。このcpsを等容量のエタノールで再
度沈殿させ、エタノールとアセトンの両方で1回洗浄し、そして脱イオン水中に
再溶解した。ついで、このcpsを長期間貯蔵のため凍結乾燥した。
エル.ラクティス株UCP1619により生産された精製cps及びエス.ニ
ューモニエからの高度に精製したタイプ3cpsの市販品(メルク・シャープ・
ドーム)を約3.3mg/mlの濃度で重水D2O中に再懸濁し、そしてDRX
500NMRスペクトル分光器(ブルーカー・コウ.ドイツ)中、320°Kで
プロトンNMRスペクトルを分析した。
天然の肺炎球菌cps調製物及びラクトコッカス誘導cps調製物のプロトン
NMRスペクトルは極めて類似している。各水素グループと関連する小さなピー
クの相対強度における僅かな変動は試料の粘度の差に帰するのが最も妥当である
(図7)。同様に、これらの試料の13C−NMRスペクトルは同一の位置にピー
クを示す。これらの結果はこの二つの多糖類試料の反復ユニットの分子構造が同
じであるという証拠を提供する(図9)。実施例4 単離された莢膜多糖類及び莢膜を有するエル.ラクティスに対する免疫応答の研 究
cpsを発現する組換えエル.ラクティス及びエス.ニューモニエ由来の精製
cpsに対する免疫応答をBALB/cマウスの同系交配株の腹腔内(i/p)
接種により比較した。5匹1群のマウスに、アジュバントなしで、cpsを発現
する組換えエル.ラクティス(株UCP1619)の種々の投与量又はエス.ニ
ューモニエ由来の精製したタイプ3cps抗原の当量を用いて皮下に免疫処置し
た。免疫処置後7、21、35及び49日目に血液試料を集め、その抗−cps
抗体レベルをELISAで測定した。
ELISAでは、ミクロタイター・プレート(ヌンク、マクシソーブ)を、各
ウエルにエス.ニューモニエ由来のcpsの50μg/ml溶液を添加しそして
37℃で5時間っいで4℃で一晩インキュベートすることにより、エス.ニュー
モニエ由来の精製cpsで被覆した。非特異的タンパク結合部分は各ウエルにP
BS中の1%w/vBSAの100μlを添加しそして室温で1時間そのプレー
トをインキュベートすることによりブロックした。
このプレートを洗浄緩衝液(WB、PBS、0.05%(w/v)トゥイーン
20)中で3回洗浄した後、PBS中の1%w/vBSAで希釈した血清試料を
このプレートに添加し(50μl/ウエル)、室温で2時間インキュベートした
。ついで、このプレートをWBで5回洗浄しそして1%BSA(50μl/ウエ
ル)を含むWBで希釈した第2抗体(抗マウスIgG−アルカリホスファターゼ
複合体)と共に室温で1.5時間インキュベートした。最後にこのプレートをW
Bで5回洗浄しそしてジエタノールアミン緩衝液中の基質ニトロフェニルホスフ
ェートと共にインキュベートし、そしてELISAリーダーを用い405nmの
波長で検定した。
各血清試料について、予め免疫化したマウスのプールした血清の50倍希釈液
と同じ吸光度(405nm)の値を示す終点タイターをELISAデータを用い
て計算した。免疫処置後7、21、35及び49日目の異なる実験グループのマ
ウスに対する特異的抗cps抗体レベルの平均終点タイターを示す棒グラフを図
8に示す。
これらの結果から、精製cps又は当量のcpsを発現するエル.ラクティス
の1回投与でマウスを免疫処置すると有意のcps特異的IgG血清抗体応答が
引き出されることが明らかである。免疫処置後21及び35日目に、免疫処置グ
ループのマウス全ての平均のcps特異的抗体タイターは無処置のものよりも有
意に高かった(p=0.004、すべての統計分析は無パラメータ・マン・ホイ
ットニーUテストを用いて計算された)。21日目には、cpsの当量を発現す
るエル.ラクティスの精製cpsで免疫処置したマウスグループのIgG抗体タ
イターの間には有意の差は存在しなかった(p>0.05)。35日目には、5
μgのcps(精製したものであろうとエル.ラクティスにより発現したもので
あろうと)を与えたマウスグループのcps抗体タイターは、cps低投与量で
免疫処置されたグループよりも有意に高かった(p=0.048)。免疫処置後
49日目までに、免疫処置グループマウスのcps特異的血清IgG抗体タイタ
ーは、未処置グループと有意に異ならないレベルまで減少し始めていた。実施例5 エル.ラクティスで発現させるための他の肺炎球菌莢膜オペロン遺伝子のクロー ニング
他の血清型のタイプ特異的cps生合成オペロンの上流に位置するORF(オ
ープン・リーディング・フレーム)内の肺炎球菌DNA配列の間には高い相同性
がある。これはタイプ3肺炎球菌単離菌の莢膜遺伝子座由来の公表されたDNA
配列(18、19)と、タイプ14(ヌイテンら,ゲンバンク受託番号X857
85)及び19F(20)などの他の血清型との比較から明らかである。このD
NA配列の相同性はその配列が未知である肺炎球菌の他の血清型由来のcps生
合成オペロンをコードするDNA断片の逆PCR増幅のためのプライマーを設計
するために使用することができる(以下を見よ)。
さらに、上に参照した保存されたDNA配列は他のグラム陽性細菌の莢膜生合
成オペロンに隣接して発見されてもいる(21及びその中の引用文献)。このこ
とはこの方法が広い範囲の莢膜を有する細菌に適用可能であることを示唆する。
ここに示す実施例では、肺炎球菌血清型4のcps生合成オペロンを含むと予
想される一つの断片を逆PCRにより増幅した。肺炎球菌血清型4由来のゲノム
DNAは実施例1に記述した方法で調製した。プライマーS2及びA4は上に引
用した公表された保存配列を用いて設計し、そして公表された方法(22)を用
いて逆PCRのために使用した。約13kbのDNA断片をPCR反応で増幅し
た(変性94℃で30秒間、アニーリング60℃で60秒間、伸長72℃で8分
間の30サイクル、そして最終伸長期間72℃で10分間のインキュベーション
)。この断片のDNA配列はタイプ4オペロンのcps生合成酵素をコードする
ことが予想される。実施例6
1.免疫処置のためのエル.ラクティスの調製
エル.ラクティス1601(プラスミドを有する)及びエル.ラクティス16
19(莢膜を有する)の生育のために、粉末状のM17ブロスとグルコースをd
dH2O中に溶解し、オートクレーブし、ついで冷却する。エリスロマイシン(
5μg/ml)をこの培地に添加する。
冷凍ストックからの細菌を使用して培地100ml容量に接種する。振盪イン
キュベータ中、30℃で一晩培養する。この細菌を10,000×gで4℃で1
0分間の遠心分離により培地から分離する。この細胞をPBSに再懸濁し、PB
S中で遠心分離により2回洗浄する。最後の懸濁液を405nmで光学密度を測
定することによりコロニー形成ユニットを評価する。細胞の濃度を所望の濃度に
調整する。
2.免疫処置手順
ラクトコッカス・ラクティス1619を5×109CFU/mlの濃度及び0
.1mlの総容量(5×108CFU)に調整し、6BALB/c雄のSPFマ
ウス(7週齢)のそれぞれに26ゲージ針を用い皮下注射により投与した。マウ
スを生きている細菌の同濃度で0、14、及び28日に免疫処置した。対照のマ
ウスは6匹の未処置の動物であった。
3.マウスの気管内細菌チャレンジ
ストレプトコッカス・ニューモニエNTCC12695を用いる細菌チャレン
ジは42日目に行った。マウス(BALB/c)を静脈内サファン(saffan)麻
酔法(0.15ml、kg体重当たりPBS中20mgのアルファドン、ピトマ
ン−ムア、Nth Ryde,NSW,オーストラリア)により沈静化した。気管を口から露
出させ、プラスチック中に引っ込められる針を持つ22.5Gのカテーテル(テ
ルモ、東京、日本)を気管開口部に0.5〜0.75cmの距離まで挿入した。
カテーテルから針を取り除き、カテーテルのハブ中にPBS中のエス.ニューモ
ニエの20μl容(5×105コロニー形成ユニットを含む)を入れた。この種
菌をカテーテル中のハブの中に1mlのシリンジを封じこめることにより生ずる
0.3ml容の空気2回で散布した。
4.細菌チャレンジの後の試料の回収
腹腔内注射によりペントバルビタールナトリウムを過剰投与することによりマ
ウスを殺した。希釈していないペントバルビタールナトリウムの0.2mlを1
mlシリンジ上の26ゲージ針を用いて注射した。1.5mlのエッペンドルフ
内に置かれた1mlのシリンジと26ゲージの針を使って心臓穿刺により血液を
採取し、血清を集めるために凝血させた。頸を経て気管を露出させ、背面の結合
組織を切断することにより切り離し、喉頭の近くで切断し、そして気管の中に挿
入された1mlのシリンジが付いた20ゲージのカニューレを用い、肺の中に0
.5mlのPBSを注入しそして回収することにより気管支肺胞洗浄(BAL)
を取得した。回収されたこのBALを1.5mlのエッペンドルフの中に入れた
。このBALについて、CFU測定用ブラッド・アガー・プレート(血液寒天平
板)上にその10倍逐次希釈液を塗布することにより細菌の存在の有無を評価し
た。
4℃、450×gで10分間の遠心分離(ジュアンBR3.11、セント ナ
ザール、フランス)により血清を分離し、そして必要になるまで−80℃で貯蔵
した。100μlのBALを遠心分離機の中に置きそしてスライドガラス上に1
0分間遠心分離した。スライドをディフ・クイックで染色し、そして白血球の別
の集団をスライド当たり3視野からカウントし、動物当たりの平均%カウント、
ついでグループ当たりの平均%カウントを求めた。BALを4℃、1000rp
mで10分間遠心分離(ジュアンBR3.11、セント ナザール、フランス)
した。その上清を取得し必要になるまで−80℃で貯蔵する。そのペレットを5
0μlPBS及び50μlメチレンブルー中に再懸濁した。その白血球を血球計
でカウントし、そして上述の分析のため計算はBALの除去に対して調整された

エス.ニューモニエNTCC12695と相同な多糖類莢膜を発現するエル.
ラクティス1619の皮下注射により免疫処置された動物では、肺の気管支肺胞

肺に感染させるために使用したエス.ニューモニエ株と相同な多糖類莢膜を発
現するエル.ラクティス1619の皮下注射により免疫処置された動物のBAL
中では、回収された総細菌数は少なくなっていた。実施例7
タイプIII GBSオペロンの染色体領域の公表された地図(1)に基づいて、
本発明者らはグループBストレプトコッカスの9種の血清型すべてからの莢膜生
合成遺伝子を含む長いDNA断片を合成する(PCRにより)ために使用された
オリゴヌクレオチドプライマー(図12a)を設計した。PCRプライマーはc
psF遺伝子に対して公表された配列(EMBLデータベース受託番号SA19
899)及びcpsC遺伝子(EMBLデータベース受託番号SACPSABD
)を用いて設計された。GBSの8種の異なる莢膜血清型由来のゲノムDNAを
用いて、13kbから16kbまでにわたるサイズのDNA断片がPCR反応に
よって増幅された。PCR反応はタックプラスシステム(ストラタジーン・エル
ティーディー)、鋳型としてゲノムDNAの約150ng及び製造者により推奨
された緩衝液中の各プライマー40pmolを用いて行われた。この反応はゲノ
ムDNAを変性するため95℃で5分間インキュベートし、その後95℃で30
秒間の変性、60℃で1分間のアニーリング、72℃で15分間の伸長の5サイ
クル、次いで95℃で30秒間の変性、55℃で1分間のアニーリング、72℃
で15分間の伸長の5サイクル、次いで95℃で30秒間の変性、65℃で1分
間のアニーリング、72℃で15分間の伸長の25サイクル、そして72℃で1
0分間の最終の伸長期間、インキュベートした。
GBSの6種の異なる血清型から得られたPCR産物の制限エンドヌクレアー
