ヒドロフロキノリンライブラリーを作製するためのコンビネーション法
本出願は米国仮特許出願第60/033665号(1996年12月18日出
願)の恩恵を請求するものである。
発明の分野
本発明はヒドロフロキノリン化合物の多様なライブラリー、そのようなライブ
ラリーの作製方法、および多様なヒドロフロキノリン化合物を容易に得ることが
できる供給源を保存し、提供する装置に関する。本コンビネーション(組合せ)
ライブラリーを収容する装置は薬剤開発のための化合物を同定するアッセイ系の
有用な構成部分である。
発明の背景
製薬業界において研究開発費は資本の大きな支出を占める。化合物の合成は費
用と時間がかかる研究開発の段階である。歴史的には、化学研究者は、医薬の手
がかり(リード)を開発するための生物学的スクリーニング用の高純度の化合物
を個々に合成し、分析した。そのような方法は新しい薬剤を市場にもたらすのに
成功したが、個々の合成と化合物の完全な特徴付けに限界があるため、新規医薬
活性化合物の発見はかなり遅れた。今日の競合的な製薬業界では、迅速で費用の
かからない薬剤開発の方法論がますます重要である。
近年、最新薬の発見には、一般に「ライブラリー」と呼ばれる数多くの(102
−106)の化合物を生成するコンビネーション化学が利用されてきた。コンビ
ネーション化学の重要な目的は医薬研究の手がかりとなる化合物を得るためにス
クリーニングすることができる数多くの新規化合物を生成することである。
理論的には、該ライブラリーを作製することができる化合物の総数は、該ライ
ブラリーの分子骨格(scaffold)上の置換位置に置換基を形成するのに利用できる
試薬の数によって決まる。コンビネーション法は、化合物の生成と生物学的スク
リーニング両方の自動化に適合させることにより、薬物発見の機会と効率が大い
に高まる。
コンビネーション化学は、化合物ライブラリーが混合物として作製され、陽性
のスクリーニング結果が得られる後まで個々の化合物の完全な同定が延期される
方法で行うことができよう。しかし、好ましいコンビネーション化学は、個々の
反応生成物が同時に合成されるが別々の容器で保持される「パラレルアレイ合成
」である。例えば、個々のライブラリー化合物はマイクロタイタープレートの別
々のウェル中で製造され、保存され、それぞれパラレルアレイの1メンバーを含
む該ウェル中でアッセイされる。標準化マイクロタイタープレートまたは同等の
装置はライブラリー合成時およびライブラリーのサンプリングまたはアッセイ時
のいずれでもプログラム化ロボットシステムに利用しやすいため、そのような装
置を用いることは有利である。
典型的には、コンビネーション化学のスキーム中の溶液相反応は、高収率の化
学反応を選択し、そして/またはかなり過剰の1試薬を用いることにより確実に
完結する。1試薬を過剰に用いる場合は、反応の完結により可溶性生成物と少な
くとも1つの可溶性非反応性試薬が生成する。
コンビネーション化学は薬剤開発の2つの異なる段階で用いることができよう
。発見段階では、リード化合物をみいだすために多様なライブラリーが作製され
る。第二の最適化段階では、最適な分子配置を得るため強力なリード化合物をよ
り狭い範囲で修飾する。
アニリンとアルデヒドの縮重により誘導されるイミンとジヒドロフランとの反
応による選ばれたヒドロフロキノリン化合物の製造法はJose Cabra1ら、Tetrahe
dron Letters,Vo1.29,547-550頁(1988),Jose Cabra1ら、Tetrahedron Letters
,Vol.30,7237-7238頁(1989)、およびKametaniら、Synthetic Communications,
vol.15,499-505頁(1985)に記載されている。該著者らは、環化反応の触媒にプロ
トン酸とルイス酸を用いること、およびベンジルイデンアニリンとジヒドロフラ
ン、ジヒドロピラン、およびエチルビニルエーテルとの反応における立体異性体
比に対する種々の溶媒と触媒の影響について記載している。
本発明の方法は、新規リード化合物を同定するのに有用なヒドロフロキノリン
の多様な新規ライブラリーを作製することに基づく。該ライブラリーは、各リザ
ーバーが隣り合ったリザーバー中のものとは異なるあらかじめ決定されたライブ
ラリー化合物を含む、2次元配列のリザーバーを含む装置として作製され、保存
され、使用される。
発明の要約

さらに、本発明は一般的に下記の反応式1に従ってヒドロフロキノリンライブ
ラリーを製造する方法を提供する。
本発明の別の態様では、アッセイ物質、および2次元配列の一定のリザーバー
を提供するウェルプレート装置または同等の装置を含む、医薬的リードヒドロフ
ロキノリン化合物を同定するためのアッセイキットを提供する。ウェルプレート
装置は、各ウェル(リザーバー)がヒドロフロキノリンライブラリーの独特な反
応生成物を含む多様なコンビネーションライブラリーを提供する。ウェルプレー
ト装置はライブラリーを作製するための多反応ゾーンを提供し、ライブラリーを
保存し、ライブラリー化合物を容易に得ることができる供給源を提供するのに用
いられる。

発明の詳細な説明
本発明で用いている用語「アッセイキット」は、2つの協同的要素の集合物、
すなわち(1)ウェルプレート装置および(2)生物学的アッセイ物質を表す。
「生物学的アッセイ物質」とは、選択した疾病の状態に関連したスクリーニン
グにおいて、あらゆるライブラリー化合物の効果の生物学的評価を行うのに必要
な物質をいう。
「ライブラリー」は、コンビネーション化学法により作製した、共通骨格と1
またはそれ以上の変化し得る置換基を有する化合物のコレクションである。本発
明の該骨格はヒドロフロキノリンである。
「ライブラリー化合物」は、コンビネーションライブラリー中の個々の反応生
成物、単一化合物または異性体の混合物である。
「リード化合物」は、選ばれた疾病状態に関連する有意な活性がアッセイキッ
トにより証明された、選ばれたコンビネーションライブラリー中のライブラリー
化合物である。
「多様なライブラリー」は、コンビネーションライブラリーの骨格またはコア
構造上の置換基の構成原子、分子量、および構造が変化しやすく、全体的にみて
、密接に関連した同族体または類似体のコレクションではない(「方向性のある
ライブラリー」に比べて)ライブラリーを意味する。
「方向性のあるライブラリー」は各ライブラリー化合物が共通骨格を持ち、ラ
イブラリーが全体的にみて、リード化合物と密接に関連した同族体や類似体(「多
様なライブラリー」に比べて)のコレクションである、リード化合物の活性を最
適化することを目的とするコンビネーション化学法によって作製した化合物のコ
レクションである。
本発明で用いている用語「骨格(scaffold)」は、コンビネーションライブラリ
ーのメンバーである化合物の変化しない領域(本発明のヒドロフロキノリンコア
)をいう。
「置換基」は、コンビネーション合成法を通してヒドロフロキノリン骨格と結
合するか、または該骨格に組み込まれる化学基をいう。異なる官能基はライブラ
リー全体の分子の多様性を説明し、多様なライブラリーの場合は該骨格に生物活
性の多様性をもたらし、方向性のあるライブラリーの場合は特定の生物活性を最
適化するように選ばれる。
「試薬」は、ライブラリーの骨格上に置換基を配置するためにコンビネーショ
ン合成において用いられるあらゆる化合物、試薬を意味する。
「パラレルアレイ合成法」とは、個々のコンビネーションライブラリー化合物
が別々に製造され、保存される前後に意図的に混合されることなく保存される、
ライブラリーのコンビネーション化学合成を行う方法をいう。
「同時合成」とは、コンビネーション法の1製造サイクル中でライブラリー化
合物を生成することをいう(時間的に同じ瞬間にすべてのライブラリー化合物が
生成されるわけではない)。
「反応ゾーン」とは、本発明のコンビネーション化学ライブラリー化合物製造
法が実施され、個々のライブラリー化合物が合成される個々の容器の位置をいう
。適切な反応ゾーンはウェルプレート装置の個々のウェルである。
「ウェルプレート装置」とは、次元的に固定された一定の位置に多数のライブ
ラリー化合物を保持することができる構造をいう。
「非干渉置換基」は本発明の方法を有意に妨げることのない、安定なヒドロフ
ロキノリンライブラリー化合物を生成する基である。
「アリール」は1またはそれ以上の芳香族環(環炭素原子各5〜6個)を意味
し、1またはそれ以上の非干渉置換基を有する置換アリールを含む。多アリール
環はナフチルのように融合するか、ビフェニルのように融合していなくてよい。
「アルキル」は炭素数1〜20の直鎖または分岐鎖、または環状炭化水素を表
す。
「置換(された)アルキル」は1またはそれ以上の非干渉置換基を有するアル
キルである。
「ハロ」はクロロ、フルオロ、ヨード、またはブロモを意味する。
「複素環」または「複素環基」は、非飽和または芳香族特性を持つかまたは持
たず、所望により置換された、少なくとも一つの環原子が炭素でない、1または
それ以上の、5、6または7原子の環を意味する。好ましい異種原子には、硫黄
、酸素、および窒素が含まれる。多環は、キノリンやベンゾフランのように融合
しても、4−フェニルピリジンのように融合していなくてもよい。
「置換(された)複素環」または「置換(された)複素環基」は、1またはそれ以
上の非干渉置換基を有する複素環である。複素環構造の置換に適した基には、限
定されるものではないが、ハロ、C1−C10アルキル、C2−C10アルケニル、C2
−C10アルキニル、C1−C10アルコキシ、C7−C12アラルキル、C7−C12ア
ルカリル、C1−C10アルキルチオ、アリールチオ、アリールオキシ、アリール
アミノ、C3−C10シクロアルキル、C3−C10シクロアルケニル、ジ(C1−C10
)−アルキルアミノ、C2−C12アルコキシアルキル、C1−C6アルキルスルフィ

「有機部分」は少なくとも1個の炭素原子を介して共有結合した非干渉置換基
を含む置換基を意味する。結合炭素原子上で置換するのに適した基には、限定さ
れるものではないが、水素、ハロ、C1−C10アルキル、C2−C10アルケニル、
C2−C10アルキニル、C1−C10アルコキシ、C7−C12アラルキル、C7−C12
アルカリル、C1−C10アルキルチオ、アリールチオ、アリールオキシ、アリー
ルアミノ、C3−C10シクロアルキル、C3−C10シクロアルケニル、ジ(C1−C10

ヒドロフロキノリンの多様なライブラリーは本発明に従って提供される。本発
明の装置として具体化されるヒドロフロキノリンライブラリーは、医薬的および
農業的候補のスクリーニングアッセイを介して新規の生物学的に活性なヒドロフ
ロキノリン化合物を同定するのに用い、構造と活性の関係を定義する研究に用い
、そして/または臨床研究に用いるための多様なヒドロフロキノリン化合物を容
易に得ることができる供給源として役立つ。
本発明により提供されるライブラリーには式(I):

ーが提供される。
本発明の別の態様では、R1およびR1'が水素および非干渉置換基から独立し
て選ばれ、R2がC1−C10アルキル、置換(C1−C10アルキル)、またはアリ
ールであり、R3が水素である上記式Iの化合物のライブラリーが提供される。
本発明のさらに別の態様では、R1およびR1’が独立して水素、またはハロ、
C1−C10アルキル、C2−C10アルケニル、C2−C10アルキニル、C1−C10ア
ルコキシ、C7−C12アラルキル、C7−C12アルカリル、C1−C10アルキルチ

本発明は、以下の反応式に示すパラレルアレイ合成技術においてコンビネーシ
ョン化学を用いる式Iのヒドロフロキノリン化合物のライブラリーの製造方法も
提供する。
反応式1 この方法には、一連の所望により置換されたアニリンと一連の所望により置換
されたアルデヒドを反応させることにより置換イミン中間体を生成し、次いで、
さらにルイス酸の存在下で各中間体と所望により置換されたジヒドロフランを反
応させることにより、それぞれアニリン試薬、アルデヒド試薬、およびジヒドロ
フラン試薬から誘導されたR1、R1’、R2、およびR3の4つの部位の多様性を
有するヒドロフロキノリン化合物のライブラリーを作製する工程が含まれる。各
化合物は別の反応ゾーンで作製され(すなわち、パラレルアレイ合成)、あらか
じめ決定された生成物である化合物がプレートおよび反応ウェル番号により確定
される。
アニリン、アルデヒド、およびジヒドロフラン試薬は市販されているか、市販
されている出発物質から製造される。本発明に用いるアニリンは、式:
[式中、R1およびR1’は非干渉基、すなわち、アルデヒドおよびシクロペンタ
ジエンとの反応に干渉しない置換基である]
で示される化合物である。典型的にはアニリン試薬の分子量は約100〜約60
0である。
本発明のヒドロフロキノリンライブラリーの作製に用いる適切なアニリンの例

本発明のヒドロフロキノリンライブラリーの作製においてイミン中間体を形成
するのに有用な特に適したアルデヒドは、さらに下記式(ここでLは−CHOで
ある)で示される。
所望により置換されたジヒドロフラン反応体も市販品を用いるか、市販の出発
物質を用いて合成することができる。好ましくは置換基R3は水素、アルキル、
または置換アルキルである。最も典型的には、R3は水素、C1−C10アルキル、
または置換C1−C10アルキル(ここで、該置換基は非干渉基である)である。
水素以外のR3基の例にはメチル、エチル、プロピル、ブチル、イソプロピル、
2−メトキシエチル、2−クロロプロピル、ベンジル、および4−ブロモベンジ
ルがある。
上記式Iのヒドロフロキノリンライブラリー化合物の製造には、イミン中間体
を最初に形成し、次いで酸、典型的にはプロトン酸および/またはルイス酸、例
えばトリフルオロ酢酸および塩化第二鉄の存在下で、所望により置換されたジヒ
ドロフランと反応させる、二段階の工程が含まれる。反応の進行/完結は薄層ク
ロマトグラフィ(TLC)を含む多くの通常の技術により測定することができる
。
本ライブラリー化合物の生成方法の第1段階では、それぞれ典型的には適切な
有機溶媒中の溶液中の、当量の所望により置換されたアニリンおよびアルデヒド
を反応ゾーン中で混合し、好ましくは周囲温度で一夜、次いで〜50℃で約4時
間反応させた。適切な有機溶媒は、各反応体が可溶性であり、イミン形成縮重反
応と干渉しないものである。適切な溶媒には、メタノールまたはエタノールのよ
うなアルコール、ジエチルエーテルのようなエーテル、酢酸エチルのようなエス
テル、および塩化メチレン、クロロホルムもしくは1,2−ジクロロエタンのよ
うなハロゲン化炭化水素溶媒が含まれる。反応温度は重要ではないが、約25℃
〜約50℃で反応を行うことにより良い結果が得られた。イミン形成反応の進行
は、例えば薄層クロマトグラフィによりモニターすることができる。
イミン中間体形成段階が完結した後、反応混合物を減圧下で蒸発させ、各反応
ゾーン中に中間体イミンを含む中間体生成物を得る。次に、各反応ゾーン中のイ
ミン中間体をハロゲン化炭化水素溶媒、好ましくは塩化メチレンに溶解し、触媒
量のトリフルオロ酢酸(TFA)および塩化第二鉄(FeCl3)の存在下で、
典型的にはイミン中間体に対して10〜30%モル過剰の、溶液中の2,3−ジ
ヒドロフランと反応させる。TFAおよびFeCl3は、典型的にはイミン中間
体の量に基づいて約1〜約20%のモル当量に相当する量で加える。他の強プロ
トン酸および/またはルイス酸、例えば、トリフロメタンスルホン酸、ポリリン
酸、クロロスルホン酸、塩化アルミニウムおよび3フッ化ホウ素を用いることが
できる。典型的には、該反応は室温で約24時間振盪しながら行う。反応の進行
は薄層クロマトグラフィまたは他の適切な当該分野で認められた分析技術により
追跡することができる。
典型的には、該反応は室温で約24時間後に完結する。反応が完結した後、さ
らなる溶媒を含む各反応ゾーンに中性アルミナ/SiO2の3:1混合物約90
mgを加え、約24〜40時間後、各反応混合物を濾過してきれいなリザーバー
に入れる。反応ゾーンと濾過したアルミナ/SiO2を塩化メチレンですすぎ、
各濾過物に洗浄液を加える。混合濾過物および洗浄液を減圧下で蒸発させ、各リ
ザーバー中にライブラリー化合物を得る。各ライブラリー化合物の試料をクロマ
トグラフィ、好ましくは複合クロマトグラフィ/質量分析により分析する。
上記式Iのヒドロフロキノリンライブラリーを作製するのに用いる本発明の方
法は液体反応媒質を保持することができるあらゆる容器中で行うことができよう
。ある態様では、本発明の方法はパラレルアレイ合成に適合した容器中で行われ
る。
特に、本発明のヒドロフロキノリンライブラリーは、図1および2に示す96ウ
ェルプレート中で形成することができる。該装置は、最も典型的には、本発明の
ヒドロフロキノリンライブラリーの1メンバーが各リザーバー中で製造される2
次元配列の一定のリザーバー中の多反応ゾーンを提供する。このように本発明の
多様なヒドロフロキノリンライブラリーは、各リザーバーまたはウェルがヒドロ
フロキノリンライブラリーのライブラリー化合物を含む、多数のリザーバー配列
(例えば、ウェルプレート)を含む。したがって、該ライブラリー化合物は典型
的にはそのウェルプレート番号とX欄とY列のウェルプレートの座標を参考にし
て確定される。
リザーバー配列中のライブラリーメンバーの化合物を同時に製造した後、化合
物のすべてまたは一部を他のリザーバー配列(例えば、ウェルプレート)に移す
ことにより、ライブラリー装置の多数のコピーを製造するか、またはライブラリ
ーをさらなる反応条件に置く。ライブラリー装置のコピー(各リザーバーがライ
ブラリーのあらかじめ決定されたメンバーを含む、2次元配列の一定のリザーバ
ーをそれぞれ含む娘ウェルプレート)は自動化アッセイ機の取り替え自由な要素
として有用である。本発明の装置により、多様な構造的に関連したヒドロフロキ
ノリン化合物を好都合に得ることができる。本発明を構成および使用するのに用
いるある好ましいリザーバー配列はマルチウェルタイタープレート、典型的には
96ウェルマイクロタイタープレートである。
図1は、本発明のウェルプレート装置の上端表面を示す。ウェルプレート(1
)はパラレルアレイ合成のための液体を保持できる96ウェル(窪み)を有する
プラスチックプレートである。個々の反応生成物を各ウェル中に作製し、ウェル
プレートの座標により標識する。例えば、位置(2)のライブラリー化合物は英
数字座標「A6」により確定される。
図2は本発明のライブラリーの製造に用いる改良ウェルプレート装置の側面図
を示す。ウェルプレート(3)はウェル(4)とフィルター(5)、保持フリッ
ト(6)、および該方法を実施するのに用いる液体反応媒質(7)を含む。ウェ
ルの底には、留め金(11)で所定の位置に維持した上部カバー(9)と底カバ
ー(10)により適切な位置に保持されたガスケット(8)によりシールされた
流出口がある。
そのようなウェルプレートは典型的には標準的96ウェルプレートを用いて製
造される。穴はプレートの各ウェルの底にドリルで開けられ、各ウェルの底に多
孔フリット(frit)を置く。次に、各プレートを留め金部品中に置き、ウェルの底
をシールする。
