アセトアミノフェン塩
発明の分野
本発明はアセトアミノフェンの塩に関し、特にアセトアミノフェンのアルカリ
金属塩及びアルカリ土類金属塩に関する。
背景技術
アセトアミノフェン(APAP)は、鎮痛活性と解熱活性の両方を有する治療
薬として十分に確立されている。しかし、アセトアミノフェンの水への溶解性が
低いことと苦味により、患者にとって許容できる経口投与形態に製剤することが
困難になっている。大部分の市販アセトアミノフェン経口投与形態は、アセトア
ミノフェンの粒に良い味のマスクコーティングするか、薬剤の苦味を隠すために
香料と甘味料を用いている。
アセトアミノフェンの溶解性と味を処理する他の方法には、アセトアミノフェ
ンのアミノ酸エステルの形成がある。I.M.Kovachは、Diss.Abstr.Int.B 1
975年36(2)、第734頁乃至第735頁に、p−アセトアミドフェニルグリ
シネート(APG)、α−p−アセトアミドフェニルアスパルテート(AAPA
)及びβ−p−アセトアミドフェニルアスパルテート(BAPA)の合成を示し
ている。これらのエステルはアセトアミノフェンより苦味が少ないと報告されて
いる。APG−HBrはアセトアミノフェンの5倍水に溶けるが、BAPA−H
ClはAPAPの4倍水に溶けにくい。
また、脂肪親和性カルボン酸等の疎水性化合物は、通常適当な塩を形成してそ
の化合物の水溶性を改善することが知られている。ナトリウムイブプロフェンと
ナトリウムナプロキセンは、塩形態で水溶性が改善された薬学的に活性な脂肪親
和性カルボン酸の例である。これらの塩は一般にカルボン酸を水酸化ナトリウム
又は水酸化カリウム等の強塩基と反応させて製造する。
1978年9月11日発行のUSSR発明者証第629,209号は、4−ア
セチルアミノフェノールをジメチルホルムアミド等の有機溶媒の溶液で水酸化カ
リウム等のアルカリ剤と反応させ、その後得られたカリウムフェノラート溶液を
クロレックスと反応混合物の沸点で反応させて、ビス−[β−(4−アセチルア
ミノフェニルオキシ)エチル]エーテルを製造する方法を示している。得られた
エーテルは蠕虫疾患を患う動物の治療に有効であると報告されている。
1993年3月23日発行のUSSR発明者証第1,803,833号は、蛍
光強度測定用のアセトアミノフェンの製造方法を示している。アセトアミノフェ
ンサンプルは、初めにイソプロピルアルコールに溶解し、その後、KOH:クロ
ロホルム体積比が3−4である水酸化カリウム溶液とクロロホルムの8%溶液で
処理して、製造する。その後、15分間乃至20分間70℃乃至80℃で加熱し
て、サンプルの蛍光強度を測定する。
上記のUSSR発明者証は共にアセトアミノフェンを水酸化カリウムで処理す
ることを報告しているが、いずれの文献もアセトアミノフェンのカリウム塩の単
離は報告していない。
米国特許第5,360,615号のM.S.Yu等は、部分的にイオン化して酸性
、塩基性又は両性の薬剤の溶解性を高め、ソフトゲルの充填、2ピースカプセル
化又は錠剤の製造に適した高濃度の主に非水溶性溶液を製造する薬学的担体シス
テムを論じている。アセトアミノフェン溶液はポリエチレングリコールに、25
%(wt.)乃至40%(wt.)のアセトアミノフェン、アセトアミノフェン
1モル当り0.4モル乃至1.0モルの水酸化物イオン及び1%(wt.)乃至
20%(wt.)の水を含んでいた。ソフトゲル充填として使用するのに適した
アセトアミノフェンの濃縮溶液の一例は、1等量APAP(35重量%)、1等
量水酸化カリウム及び残りはポリエチレングリコール600を含んでいた。
D.L.Simmonsの米国特許第5,273,759号は、固体のMg(OH)2を
、APAP含有錠剤に添加することを示している。
しかしながら、Yu等もSimmonsもアセトアミノフェンの個々の塩の単離は報告
していない。
アセトアミノフェンの従来の形態に比べて、水溶性と味が改善されたアセトア
ミノフェンの単離した塩が必要とされている。
発明の開示
本発明は単離したアセトアミノフェン塩を提供する。好ましくは、単離した塩
はアセトアミノフェンのアルカリ金属塩及びアルカリ土類金属塩である。
本発明の他の態様では、単離した塩は以下の式を有する。
ここで、nは1又は2であり、nが1のときMはアルカリ金属であり、nが2の
ときMはアルカリ土類金属であり、Xは0乃至約10である。これらの塩はアセ
トアミノフェンの遊離酸形態より水溶性が改善され苦味が減っていた。また、本
発明はこれら塩の製造方法も含む。
本発明の他の態様は、鎮痛及び/又は解熱治療薬が必要な哺乳類にこれらの塩
を投与する方法に関する。さらに、本発明はアセトアミノフェン塩を含む経口投
与形態に関する。

発明を実施するための最良の形態
本発明の前には、APAPのいかなる個々の塩(フェノラート)の単離は報告
されていなかった。さらに、いかなる脱プロトン化APAP種(deprotonated AP
AP species)スピーシーズのインシトゥ(in situ)溶液特性も報告されてなかった
。本発明で使用する、アセトアミノフェンの「遊離酸(free acid)」とは、AP
APのプロトン(protonated)化フェノール形態を意味する。
科学文献でAPAP塩についての検討がない一因は、APAPのアニオン形態
が水性溶液中(pH>11)では短期間(<24時間)でしか安定でない事実に
ある。塩が形成した後すぐに単離しなければ、p−アミノフェノラート(PAP
)が形成して生成物が変色する可能性がある。
本発明で使用する、アセトアミノフェンの単離した塩とは、アセトアミノフェ
ンのフェノール性プロトンの脱プロトン化により生成されるp−ヒドロキシアセ
トアニリドの塩を示す。好ましくは、単離した塩はアセトアミノフェンのアルカ
リ金属塩及びアルカリ土類金属塩である。本発明の他の態様では、単離した塩は
以下の式を有する。

APAPの塩は、APAPと、所望の1価又は2価の金属水酸化物との一工程
水性反応により製造される。適当な1価又は2価の金属水酸化物の例には、水酸
化ナトリウム、水酸化カルシウム、水酸化リチウム、水酸化カリウム、水酸化マ
グネシウム及び水酸化セシウムがある。水酸化物のアセトアミノフェンに対する

PAPと金属水酸化物を水に、又は水と、アセトニトリル、メタノール、イソプ
ロパノール、エタノール又はテトラヒドロフラン等の水混和性有機溶媒との混合
物に溶解する。その後、より極性の低い水混和性反応混合物を添加して、沈殿に
より、粗反応生成物を回収又は単離する。一般に、PAPの形成により生成物が
変色する可能性を減らすために、反応生成物が形成したらできる限り速く回収又
は単離すべきである。その後、最終生成物を真空乾燥してもよい。
本発明のAPAP塩はまたカチオン交換反応しやすい。例えば、アセトアミノ
フェンの1価金属塩の水性スラリー又は溶液が2価金属カチオンと接触すると、
カチオン交換反応によりアセトアミノフェンの無水2価金属塩が形成される。そ
の後、塩は速やかに回収される。特に、C16H16N2O4Caは、C8H8NO2N
aの水性溶液を0.5等量の塩化カルシウム(CaCl2)と反応させて製造で
きる。室温より高い温度で真空乾燥した後、得られたC16H16N2O4Caは無水
物であった。
無水形態の他に、APAP塩の様々な水和形態が反応条件によって製造できる
。好ましくは、これらの水和塩はAPAP塩1モル当り、水は10モルより少な
く、例えば、アセトアミノフェンナトリウム5水和物、アセトアミノフェンナト
リウム6水和物、アセトアミノフェンナトリウム7水和物、アセトアミノフェン
カルシウム2水和物及びアセトアミノフェンリチウム6水和物がある。
本発明のAPAP塩の22℃での水溶性は、ナトリウム塩が490mg/mL
乃至540mg/mL、リチウム塩が、450mg/mL乃至470mg/mL
、カルシウム塩が13mg/mLである。従って、ナトリウム塩、リチウム塩及
びカルシウム塩は、それぞれAPAP遊離酸の約260mg/mL乃至280m
g/mL、250mg/mL乃至270mg/mL及び10mg/mLと等価の
溶解性を有する。
APAP塩は、アセトアミノフェンの従来の遊離酸形態と比べると、溶解速度
がかなり上昇している。USP溶解装置2(パドルスピード:50rpm)を用
いた37℃の0.1Nの塩酸において、30秒後のアセトアミノフェンの濃度は
以下の通りであった。

図1は、本発明の塩の錠剤溶解速度を示している図である。APAPのナトリ
ウム塩、リチウム塩及びカルシウム塩と、従来の形態のAPAPを、それぞれ錠
剤に圧縮し、上記の条件により溶解速度を測定した。溶解媒体を、遊離酸形態の
アセトアミノフェンについて、アッセイした。図1では本発明の塩が従来の遊離
酸より、アセトアミノフェン溶解速度がかなり速いことを示している。
アセトアミノフェンのカルシウム塩とナトリウム塩は、従来の遊離酸形態のア
セトアミノフェンの苦味を有してないことが観察された。カルシウム塩はほとん
ど味がなく、ナトリウム塩は少し塩辛かった。本発明の塩の味が改善されている
ので、改善された味を有するアセトアミノフェンの経口投与形態の製剤が可能と
なる。
遊離酸形態に対し、本発明の単離した塩では、アセトアミノフェンの作用の開
始が早まると考えられる。本発明の塩の溶解性が高いことにより、アセトアミノ
フェンの血漿濃度のピークが速くなる。この特性により、アセトアミノフェンの
鎮痛活性及び/又は解熱活性の作用の開始が潜在的に早まる。
本発明のアセトアミノフェン塩は、治療有効量を哺乳類に投与できる。治療有
効量とは経口投与で所望の治療反応を引き起こせる量であり、当業者は容易に決
定できる。かかる量の決定の際には投与する特定の化合物、化合物のバイオアベ
イラビリティ、投与方式、患者の年齢と体重及び他の因子を考慮しなくてはなら
ない。典型的なヒトへの経口投与量ユニットは約80mg乃至1000mg(A
PAP遊離酸ベース)の範囲である。
一般に、本発明のアセトアミノフェン塩は固体形態で経口投与する。適当な固
体製剤の例として、飲み込むことのできる、噛むことのできる、又は速く溶ける
錠剤、ピル、カプセル、カプレット(caplet)、粉末、ウェハース、サシェイ(sac
hets)、ゼラチンコート錠剤及び粒体がある。固体形態を製造するときアセトア
ミノフェン塩は、コーンスターチ、タルク、リン酸カルシウム、硫酸カルシウム
、ステアリン酸カルシウム、ステアリン酸マグネシウム、ステアリン酸、ソルビ
トール、微結晶セルロース、マンニトール、ゼラチン、天然ゴム又は合成ゴム等
(カルボキシメチルセルロース、メチルセルロース、アルギネート、デキストラ
ン、アカシアゴム、カラヤゴム、イナゴマメゴム等)の従来の固形充填剤又は担
体及び他の従来の担体と混ぜ合わすことができる。さらに、希釈剤、バインダー
、潤滑剤、崩壊剤、着色剤及び香味料等の他の成分も使用できる。投与形態はフ
ィルムコーティングされていてもよい。
従来の方法を本発明の固形投与形態の製造に使用できる。適当な技術が、Remi
ngton's Pharmaceutical Sciences,第18版、第89章(1990年)に記載さ
れており、この文献は参考としてここに取り込む。
以下の実施例は本発明の特定の実施形態を示す。しかし、本発明はこの実施例
の特定事項に限定されずに、請求の範囲に限定される。特に示さない限り、以下
の比率と割合は重量によるものである。実施例I
この実施例はアセトアミノフェンナトリウム(C8H8NO2Na・6H2O)の
製法を示す。
30mLの1N NaOH溶液(0.030モル)を、25mLの水中の4.
53g(0.033モル)アセトアミノフェンの攪拌懸濁液に加えた。全ての固
形を溶解した後、溶液を素早く攪拌しながら、200mLのアセトニトリルを加
えた。得られた白色沈殿(9.15g、6水和物として収率99%)をフリット
に集め、テトラヒドロフラン(THF)で洗い、室温で乾燥した。1H NMR

実施例II
この実施例はアセトアミノフェンナトリウム(C8H8NO2Na・7H2O)の
製法を示す。
80g(2.00モル)のNaOHを400mLの水に溶かし、302g(2
.00モル)のAPAPが2100mLのイソプロパノールに溶けて入っている
フラスコに、50℃で攪拌しながら滴下した。溶液を室温に冷やすと、オフホワ
イト(黄白色)の沈殿が形成された。固体を濾取し、200mLのイソプロパノ
ールで3回洗い、真空乾燥した(500g、7水和物として84%)。1H N
MRとIRスペクトルはC8H8NO2Na・6H2Oと同一であった。燃焼分析C8
H8NO2Na・7H2Oの計算値:C32.11、H7.41、N4.68;測
定値:C31.99、H7.38、N4.31;水含有量C8H8NO2Na・7
H2Oの計算値:42.1%;測定値:42.7%(カールフィッシャ)。22
℃での水溶性は541mg/mLであった。
実施例III
この実施例はアセトアミノフェンカルシウム(C16H16N2O4Ca・2H2O
)の製法を示す。
5g(0.033モル)のAPAPと1.22g(0.016モル)Ca(O
H)2を200mLの水に懸濁し、混合物を4時間攪拌すると、全ての固体は溶
液に溶けた。溶液を液体窒素浴で凍結し、凍結乾燥すると、明るい微結晶オフホ
ワイト固体(5.44g、2水和物ベースで100%粗収率)が残った。1HN

実施例IV
この実施例はアセトアミノフェンリチウム(C8H8NO2Li・6H2O)の製
法を示す。
5g(0.033モル)のAPAPを30mLのイソプロパノール/THF(
1:3、アルゴンで脱気)に溶解した。この溶液を、20mLの水(アルゴン脱
気)に溶解した1.38g(0.033モル)のLiOHが入っているフラスコ
に素早く加えた。無色の溶液を0℃で16時間貯蔵すると、白色結晶が形成した
。この結晶をアルゴン下で濾過し、THFで洗い、16時間真空乾燥した(4.

この実施例はアセトアミノフェンリチウム(C8H8NO2Li・6H2O)の他
の製法を示す。
アセトアミノフェン(15.1g;0.1モル)、水90mL及び水酸化リチ
ウム1N(100mL、0.1モル)を2Lビーカーに入れた。溶液が透明にな
ったら、アセトニトリル(1500mL)を却えた。得られた白色固体を濾過し
、THF(約500mL)で洗い、大気中で乾燥して、乾燥白色固体を得た(2

実施例VI
この実施例は無水アセトアミノフェンカルシウム(C16H16N2O4Ca)の製
法を示す。
アセトアミノフェン(90.6g、0.60モル)を135mLの水に懸濁し
、水酸化ナトリウム(24.0g、0.6モル)と36mLの水を含む溶液を1
8℃乃至26℃で30分かけて加えた。得られたNaAPAP−スラリーに、塩
化カルシウム(CaCl2)(44.1g、0.3モル)と54mLの水を含む
溶液を20℃乃至25℃の室温で30分間かけて加えた。その後、反応混合物を
60分間以内で60℃まで加熱した。60℃に達したら直ぐに、スラリーを60
分間以内で20℃に冷やし、20℃で30分間攪拌した。得られたC16H16N2
O4Ca(79g、78%)を濾取し、イソプロピルアルコール(75mL)で
洗い、80℃で真空下一晩乾燥した。1H NMR(D2O)δ7.01(d,8
,4H)、6.57(d,8,4H)、2.06(s,6H,CO−CH3);

実施例VII
アセトアミノフェンナトリウムのバイオアベラビリティを測定するために、犬
で実験した。対照として、アセトアミノフェンの遊離酸形態を用いた。以下の組
成の圧縮円筒形ペレットを作製した。
アセトアミノフェンナトリウム − そのまま圧縮(賦形剤無し)
対照 − 150mgのAPAP、30mgの微結晶セルロース及び
30mgのデキストレート
最初の投与時に体重が約9kg乃至14kgの8匹の雄純潔種ビーグル犬を実
験に用いた。犬に、PMI(登録商標)保証Canine Diet Meal No.5007と水を両
方とも自由に摂取させた。犬は投与前一晩約12時間断食させ、投与後4時間食
物を戻した。
犬を2つのグループに分け、各グループにアセトアミノフェンナトリウム又は
対照(遊離酸APAP)ペレットのいずれかを投与した。300mgのアセトア
ミノフェン遊離酸と同等な一回投与量を、胃チューブを用いて経口胃管栄養法に
より投与した。各投与後に20mLの水を与えた。一週間後、各グループにアセ
トアミノフェンの他の形態(APAPナトリウム)を再び投与した。12血液サ
ンプルを各犬から各投与日(投与前1日からその後11日まで)に採取した。血
漿を分離してアセトアミノフェンについてテストした。

の図では、本発明のアセトアミノフェン塩が遊離酸アセトアミノフェン対照より
速く吸収されるのを示している。アセトアミノフェン塩のTmaxがより速いこと
は、遊離酸対照と比べて、鎮痛及び解熱活性の作用の開始がより速くなることを
示している。
上記の実験から、本発明の精神と範囲から離れることなく、様々の変更が可能
