植物有害生物の抑制方法および固定化物
本発明は、架橋結合された担体構造体中にある、植物有害生物を死滅させる微
生物を、植物または植物の種子と接触させることにより植物有害生物を抑制する
ための方法に関する。本発明はさらに生物学的な有害生物抑制のための固定化物
(immobilizate)にも関する。
環境および海水の保護に関する関心が高まり、これまでの一般的な化学的植物
保護剤の使用を減少させるか、または禁止することが要求されるため、化学的植
物保護剤を生物学的な有害生物防除剤で代替するための試みが行われてきている
。そこで例えば、サトウダイコン線虫であるヘテロデラ・シャッヒチイ(Hetero
dera schachtii)をヒルステラ・ロッシリエンシス(Hirsutella rhossiliensis
)の如き線虫病原性真菌を用いて抑制することが知られている。
生物学的に活性な微生物の調節された投与を可能にし且つ土壌中でのそれらの
安定性を改良するために、微生物を架橋結合された構造体の中で固定化すること
がすでに提唱されている(1995 Annual Report of the Bundesforschungsanstal
t fur Landwirtschaft(FAL),page 90,section 1.1)。
細胞および酵素を含む微生物を固定化する方法はかなり昔から知られており、
非常に広範囲の高分子電解質系が固定化手段として提唱されていた。例えば、C
aイオンのような低分子量架橋結合用反応物は、アルギン酸塩の、完全に架橋結
合された球を形成するために使用でき、アル
ギン酸塩は微生物を架橋結合された構造体の中に固定化する。架橋結合された反
応物を含有しうる中空球、例えば液体芯を取り囲む中空球を形成することも知ら
れている。水中油滴型または油中水滴型エマルションの界面重合により膜を形成

面で膜を形成する。
米国特許第5,358,836号は固定化された真菌、細菌および線虫からなる
植物保護剤を開示している。アルギン酸塩および澱粉が固定化マトリックスとし
て挙げられている。固定化された活性物質が早期に乾燥することを防止するため
に、固定化物にエマルション−逆転油、例えばパラフィン油、を油用の吸収剤、
例えばシリカ、と共に加えて固定化物からの水の蒸発を遅らせることが提唱され
ている。
有効な生物学的に活性な微生物の植物上での固定化が所望する程度の効果を与
えなかったことが見いだされており、そのために実用的な応用は考えられなかっ
た。微生物と有害生物との間の相互作用が固定化により非常に大きく減じられる
。
本発明の基礎をなす問題は、存在する困難にもかかわらず生物学的な有害生物
防除剤の実用可能な適用方法を見いだすことである。
この問題に基づき、最初に挙げられたタイプの方法は、本発明によれば、架橋
結合された外側殻の内部に微生物が栄養培地と一緒に含まれていること並びに微
生物が、外側殼の内部で最初に成長しそして次にそこから脱出するように外側殻
が設計されていることにより特徴づけられる。
本発明によれば、微生物は中空構造体、好適には中空球、中に固定化され、こ
こで微生物用の栄養培地は微生物と共に中空構造体の内部に含
まれる。このようにして固定化物は微生物の増殖用の反応器として作用する。こ
の反応器の内部で、例えば微生物が増殖中のバイオマスにより生じた圧力により
外側殻を破壊することによりまたは好適にはそこを通って外側に成長することに
より、外側殻から脱出するほど多量のバイオマスを生成するまで微生物は安定な
生存可能な培養菌を生成しうる。
本発明に従う方法の好適な態様では、低い初期濃度の微生物だけが栄養培地と
共に含まれ、そして微生物が少なくとも100の係数(factor)だけ増殖するまで
外側殻からの脱出が起きないように外側殻が設計される。
この方法では比較的大規模な微生物の費用のかかる発酵を省略することができ
そして適用部位における固定化物内部での発酵により代替されるため、生物学的
な有害生物の抑制を効果的に且つ安価にすることができる。
本発明に従う方法は、従って、これまでは臨界的であり、重要であるとみなさ
れていた、有害生物抑制目的のための十分な量のバイオマスの適用を、適用部位
に相対的に高い濃度の微生物を適用することによってではなく、適用部位におい
てのみ固定化物により保護されている環境の中で必要量の微生物バイオマスを生

本発明に従う方法の一部として、架橋結合された担体構造体の多数の非常に小
さい球の中に固定された外側殻の内部に、微生物を含むことが
好適である。これは少なくとも1個の種子も外側殻の中に含まれている場合に特
に有利である。この場合に形成される系は有害生物の種子への侵入を防止する。

と、内部に微生物が栄養培地と共に含まれている架橋結合された外側殻からなっ
ている。
以上で説明したように、固定化物はさらに少なくとも1個の種子を含有するこ
とができる。
微生物は、架橋結合された担体構造体の多数の小球の中に固定された外側殻の
内部に含まれうる。この形態で球は種子に付着しうる。
外側殻を、微生物が付着している種子および付着している栄養培地上に直接形
成することができる。
外側殻は好適には少なくとも1種の高分子電解質から形成される。特に適する
高分子電解質は、穏やかな架橋結合条件の理由から、アルギン酸塩類であり、特
にCaイオンを有するもの、好適にはCaCl2の形態、またはセルロース類、
例えばカルボキシアルキルセルロース類、特にカルボキシメチルセルロース、ま
たはスルホアルキル基を含有するセ
ルロースエーテル類、特にスルホアルキルセルロース、好適にはスルホエチルセ
ルロースである。スルホエチルエーテル(SEC)とは別の適当なセルロースエ
ーテル類は特にスルホプロピルエーテル(SPC)並びにヒドロプロピルスルホ
アルキルセルロース類(例えばHPSEC、HPSPC)、ヒドロキシエチルア
ルキルセルロース類(例えばHESEC、HESPC)およびカルボキシメチル
スルホアルキルセルロース類(例えばCMSEC、CMSPC)のような混合セ
ルロース類である。これらのセルロース類はチトサンまたは好適にはポリジメチ

アルギン酸塩またはスルホエチルセルロース(SEC)の使用は少なくとも高
度の生分解性という利点を有する。
含まれる栄養培地は当然含まれる微生物に依存する。ヒルステラ・ロッシリエ
ンシス(Hirsutella rhossiliensis)のような真菌培養菌用には、トウモロコシ
粉末またはトウモロコシグルテンが特に適しており、好適には少割合の酵母濃縮
物が加えられる。

121℃で10分間にわたりオートクレーブ処理された120gの2.6%SE
C溶液をオートクレーブ処理されたトウモロコシグルテン(粒

この溶液をPDMDAACのオートクレーブ処理された2%溶液に1時間32

PDMDAAC溶液はSEC溶液中にある程度だけ透過することができ、それ
