説明する。本発明は次の順序に従って行うものである。 (1)患者血清の分離 患者静脈より約50mlの血液を採取し、遠心して血清

する組織の大きさは約1cm2であり、組織検査に用い
るものとサイズは同様である。この場合治癒後の傷跡も
殆ど目立たない。 (3)毛包の分離 採取した組織を手術用顕微鏡下に毛包部を傷つけないよ
うに単離する。毛包最下端のバルブと呼ばれる部分を微
小手術用メスで切断し、結合織性部分を翻転し、毛乳頭
を露出する。ここで毛乳頭をメスを用いて切離し、予め
用意しておいた別の培養皿に移動する。 (4)毛乳頭の培養 直径3.5cmの培養皿に約10〜20個の毛乳頭を集
め、培養皿と接着するように静かに沈める。この時の培
養液は患者血清10%を含むDMEMである。更に非必
須アミノ酸とストレプトマイシン、ぺニシリン、ファン
ギゾンを加える。これらの培養皿を37℃、5%CO2
の環境下で約2週間静置する。2週間後に倒立顕微鏡下
で細胞の増殖が確認できたならば、培養液を交換する。
この時の培養液の交換頻度は細胞の数に応じ適宜調整し
てよい。通常は3日に一度程度である。観察中に充分に
細胞増殖したところで初めて0.05%トリプシンを用
いて細胞を培養皿より剥離浮遊させ、細胞懸濁液を作成
し、継代培養を行う。これらの細胞は36〜40時間の
ダブリングタイムで細胞分裂が見られ増殖するので、細
胞密度が過密になればその都度継代を行う。 (5)液体窒素による細胞の保存 3〜4回の継代を行ったところで全ての細胞を回収し、
10%グリセロールを含む10%患者血清入りの培養液
にて懸濁した後、約1×106個づつに分注し、液体窒
素にて保存する。尚、これら一連の過程で患者血清を添
加物として用いているが、これは患者本人の血清である
ため、あらゆる血清を介する感染症(ウイルス、細菌
等)の危険性がなく、また術前検査と同時に血液採取を
行えば、手技的な煩雑さも回避できるという利点がある
からである。
