アリールオキシおよびアリールチオ縮合ピリジン、
アリールオキシおよびアリールチオ縮合ピリミジン、およびそれらの誘導体
発明の分野
本発明は、新規化合物および新規医薬組成物に関し、ならびに、重度の鬱病(
major depression)、不安関連疾患、外傷後ストレス症、核上麻痺および摂食障
害を含めた精神障害および神経疾患の治療にこれらの化合物および医薬組成物を
利用する方法に関する。
発明の背景
41個のアミノ酸からなるペプチドである副腎皮質刺激ホルモン放出因子(本
明細書ではCRFと呼ぶ)は、下垂体前葉からのプロオピオメラノコルチン(P
OMC)由来ペプチド分泌の生理学的調節因子である[J.Riverらの、Proc.Nat.A
cad.Sci.(USA)80:4851(1983年)、W.Valeらの、Science 213:1394(1981年)]。
下垂体における内分泌の役割に加え、CRFは、このホルモンが中枢神経系にお
いて広く視床下部外に分布していること、また、脳内における神経伝達物質また
は神経調節物質の役割と一致して自律作用、電気生理学的作用および行為作用と
いった多様な作用を産み出していることが、その免疫組織化学的局在によって証
明されている[W.Valeらの、Rec.Prog.Horm.Res.39:245(1983年)、G.F.Koobの
、Persp.Behav.Med.2:39(1985年)、E.B.De Souzaらの、J.Neurosci.5:3189(1
985年)]。CRFが生理学的、心理学的および免疫学的各ストレッサーに対する
免疫系の応答の統合において重要な役割を果たしているという証拠もある[J.E.
Blalockの、生理学概説(Physiological Reviews)69:1(1989年)、J.E.Morley
の、Life Sci.41:527(1987年)].
CRFがうつ病、不安関連疾患および摂食障害を含む精神障害および神経疾患
において役割を果たしていることは、臨床データからも証拠づけられる。アルツ
ハイマー病、パーキンソン病、ハンチントン病、進行性核上麻痺および筋萎縮性
側索硬化症などが中枢神経系におけるCRFニューロン機能不全に関係している
ことから、それらの病因諭および病態生理学においてもCRFの役割が前提とさ
れている[E.B.De Souza、Hosp.Practice 23:59(1988年)を参照のこと]。
情動障害または重度の鬱病の場合、薬剤を服用していない個体の脳脊髄液(C
SF)ではCRF濃度が有意に高い[C.B.Nemeroffら、Science 226:1342(1984
年)、C.M.Bankiら、Am.J.Psychiatry 144:873(1987年)、R.D.Franceら、Biol
.Psychiatry 28:86(1988年)、M.Aratoら、Biol.Psychiatry 25:355(1989年)
]。さらに、自殺者の前頭皮質ではCRF受容体密度が有意に低下し、これはC
RF分泌過多と一致する[C.B.Nemeroffら、Arch.Gen.Psychiatry 45:577(1988
年)]。また、うつ病の患者では、CRF(静脈内投与)に対する副腎皮質刺激
ホルモン(ACTH)の応答が弱くなっていることが観察される[P.W.Goldら、
Am.J.Psychiatry 141:619(1984年)、F.Holsboerら、精神神経内分泌学(Psych
oneuroendocrinology)9:147(1984年)、P.W.Goldら、New Eng.J.Med.314:1129
(1986年)]。ラットおよびヒト以外の霊長動物における前臨床試験によっても
、CRF分泌過多がヒトのうつ病に見られる諸症状に関与しているという仮説が
支持される[R.M.Sapolskyの,Arch.Gen.Psychiatry 46:1047(1989年)]。三環
系抗うつ薬によってCRFレベルを変えることが可能であり、したがって脳内の
CRF受容体の数を調節することが可能であるという先行する証拠がある[Grig
oriadisら、神経精神薬理学(Neuropsychopharmacology)2:53(1989年)]。
また、不安関連疾患の病因諭でもCRFの役割が前提とされている。CRFは
動物内で不安発生作用を引き起こし、また、ベンゾジアゼピン/非ベンゾジアゼ
ピン抗不安薬とCRFとの間の相互作用については種々の行動不安モデルにおい
ても立証されている[D.R.Brittonら、Life Sci.31:363(1982年)、C.W.Berrid
geおよびA.J.Dunnの、Regul.Peptides 16:83(1986年)]。種々の行動パラダイ
ムにおいて推定上のCRF受容体拮抗物質α−らせん状ヒツジ由来CRF(9−
41)を使用した予試験によって、拮抗物質が質的にベンゾジアゼピンに類似の
「抗不安薬様」効果を引き起こすことが立証されている[C.W.BerridgeおよびA.
J.Dunnの、Horm.Behav.21:393(1987年)、脳の研究概説(Brain Research Revi
ews)15:71(1990年)]。神経化学的研究、内分泌の研究および
受容体結合の研究のいずれによっても、これらの疾患におけるCRFの関わりに
ついてのさらなる証拠が付け加えられ、CRFとベンゾジアゼピン抗不安薬との
間の相互作用が立証されている。ラットでのコンフリクトテスト[K.T.Britton
ら、精神薬理学(Psychopharmacology)86:170(1985年)K.T.Brittonら、
Psychopharmacology 94:306(1988年)]および聴覚刺激テスト(acoustic star
tle test)[N.R.Swerdlowら、Psychopharmacology 88:147(1986年)]の、い
ずれにおいても、クロルジアゼポキシドによってCRFの「不安発生」作用は弱
まっている。ベンゾジアゼピン逆作用物質(FG7142)がCRFの作用を増
強したのに対し、ベンゾジアゼピン受容体拮抗物質(Ro15−1788)は、
オペラントコンフリクトテストにおいて行動活性のみ示さなかったが、用量依存
的にCRFの作用を逆転させている[K.T.Brittonら、精神薬理学94:306(1988年)
]。
標準的な抗不安薬および抗うつ薬の治療効果が引き起こされるもととなる作用
機序および作用部位は、いまだ解明されていない。しかし、それらが、不安関連
疾患において観察されるCRF分泌過多の抑制に関係しているとの仮説が立てら
れている。種々の行動パラダイムでCRF受容体拮抗物質(α−らせん状CRF9-41
)の効果を調べる予備的研究によって、CRF拮抗物質が質的にベンゾジア
ゼピンに類似の「抗不安薬様」効果を引き起こすことが立証されたことは特に興
味深い[G.F.KoobおよびI.T.Britton、副腎皮質刺激ホルモン放出因子:神経ペ
プチドの基礎および臨床研究(Corticotropin-Releasing Factor:Basic and Cli
nical Studies of a Neuropeptide)、E.B.De SouzaおよびC.B.Nemeroff編、CRC
Press p221(1990年)を参照のこと]。
デュポン・メルク社のPCT出願US94/11050には、下式の副腎皮質
刺激ホルモン放出因子拮抗化合物、ならびに精神障害および神経疾患の治療のためのそれらの使用法が記載されてい
る。その記載には、下式の縮合ピリジンと縮合ピリミジンが含まれている。
上式で、ZはCR2またはN、AはCR30またはN、DはCR28またはNであり
、R3はアリールオキシまたはアリールチオであることができる。
ファイザー社のWO95/33750には、CNSおよびストレス異常症の治
療に有用な副腎皮質刺激ホルモン放出因子拮抗化合物が記載されている。その記
載には下式の化合物が含まれている。上式で、AはCR7またはNであり、BはOCHR1R2またはSCHR1R2であ
り、R1は置換または非置換アルキルであり、R2は置換もしくは非置換アルキル
またはアリールまたはヘテロアリールであり、R3はメチル、ハロ、シアノ、メ
トキシ等であり、R4はH、置換または非置換アルキル、ハロ、アミノ、
ニトロ等であり、R5は置換もしくは非置換アリールまたはヘテロアリールであ
り、R6はHまたは置換もしくは非置換アルキルであり、R7はH、メチル、ハロ
、シアノ等であり、R16およびR17は一緒になってオキソ(=O)基を形成して
おり、Gは=O、=S、=NH、=NCH3、水素、メチル、メトキシ等である
。
ファイザー社のWO95/34563には、下式の化合物を含む副腎皮質刺激
ホルモン放出因子拮抗化合物が記載されている。
上式で、A、BおよびR基はWO95/33750で定義したと同様に定義され
る。
ファイザー社のWO95/33727には、下式の副腎皮質刺激ホルモン放出
因子拮抗化合物が記載されている。
上式で、AはCH2であり、Zはヘテロアリール部分であることができる。
Gangulyらの米国特許第4,076,711号には、下式のトリアゾロ[4,
5−d]ピリミジンが記載されている。
上式で、Xはハロ、−NR1Rまたはアルコキシであって、但しR1とRは各々H
またはアルキルであり、Yはアルキル、シクロアルキル、ヒドロキシシクロアル
キル、フェニル、ビシクロアルキルまたはフェニルアルキルまたはビシクロアル
キルアルキルであり、QはHまたはYである。この特許では、これら化合物が乾
癬の治療に有用であることが述べられている。
TanjiらのChem.Pharm.Bull.39(11)3037-3040(1991年)には、下式のトリア
ゾロ[4,5−d]ピリミジンが記載されている。
上式で、ハロはI、BrまたはClであり、Phはフェニル、Meはメチルであ
る。これら化合物の有用性については一切記載されていない。
SettimoらのIl Farmaco,Ed.Sc.,35(4),308-323(1980年)には、下式の8−ア
ザアデニン(トリアゾロ[4,5−d]ピリミジン)が記載されている。
上式で、R1はHまたはベンジルであり、R2はp−メチルフェニルである。
BiagiらのIl Farmaco,49(3),183-186(1994年)には、下式のN(6)−置換
2−n−ブチル−9−ベンジル−8−アザアデニンが記載されている。上式で、R2はアルキル、フェニルまたはベンジルであることができる。この論
文では、これら化合物がアデノシン受容体に対して親和性を有することが述べら
れている。
ThompsonらのJ.Med.Chem.1991,34,2877-2882には、下式のN6,9−二置換ア
デニンが記載されている。
上式で、Phはフェニルまたは(C−2が非置換の場合は)2−フルオロフェニ
ルである。この論文では、これら化合物がA1アデノシン受容体に対して選択的
親和性を有することが述べられている。
KelleyらのJ.Med.Chem.1990,31,606-612には、下式の化合物が記載されている
。
上式で、R6はNHC6H5でありR9はCH2C6H5である。この諭文は、この化
合物が抗痙攣活性テストで不活性であったと報告している。またこの諭文では、
前記化合物の種々の6−(アルキルアミノ)−9−ベンジル−9H−プリン類似
化合物が抗痙攣活性を示したことが報告されている。
KelleyらのJ.Med.Chem.1990,33,1360-1363には、下式の6−アニリノ−9−ベ
ンジル−2−クロロ−9H−プリンが記載されている。
上式で、Bzはベンジルまたは(R4がの場合は)p−メチルベンジルであり、
R4はHまたはアルキル、アルコキシ、ハロ、シアノ、ニトロ等である。これら
化合物の抗ライノウイルス活性テストについても報告されている。
KelleyらのJ.Heterocyclic Chem.,28,1099(1991年)には、下式の6−置換−
9−(3−ホルムアミドベンジル)−9H−プリンが記載されている。
上式で、R1はNH2またはNHCHOである。R1がNHCHOの化合物につい
てベンゾジアゼピン受容体結合をテストしたところ、種々の類似化合物が活性で
あったにもかかわらず、不活性であった。
KhairyらのJ.Heterocyclic Chem.,22,853(1985年)には、下式で表される幾
つかの9−アリール−9H−プリン−6−アミンの合成について記載されている
。上式で、R基はH、メチル、エチル、イソプロピル、クロロまたはフルオロであ
る。
HoechstのEP第298467号(1989年)には、下記の構造を有する化
合物を含め、アザプリンについて記載されている。
上式で、QはO、S、SO、SO2またはNH2であり、XはO、S、SOまたは
SO2であり、ZはH、ハロゲン、CF3、1−3Cアルコキシまたはアルキルチ
オであり、R2はアルキルまたはアルコキシであり、R3はOR2である。これら
の化合物は、ウイルス性疾患、自己免疫疾患および癌の治療に有用であるとされ
ている。
エスエス製薬(株)の日本国特開昭59−062595号(1984年)およ
び日本国特開昭56−131587号(1981年)には、下記の構造を有する
化合物を含む、トリアゾロピリミジン誘導体について記載されている。
下式で、QはO、S、SO2であり、Rはアミノまたは置換アミノ、アルコキシ
、ベンジルオキシ、ハロゲン、あるいはフェニルヒドラジノである。これらの化
合物は、抗癌剤として有用であるとされている。 富士写真フィルム(株)の日本国特開昭60−194443号(1985年)
には、下記の構造を有する化合物を含め、アザインデンについて記載されている
。
上式で、QはO、Sであり、R1、R2およびR3はH、アルキル、アリール、ア
ラルキル、アミノ、ヒドロキシル、アルコキシ、カルバモイル、アリールオキシ
、アルコキシカルボニル、シアノ、ハロゲン、アルキルチオ、アリールチオ、カ
ルボキシルまたはメルカプトであるが、但し置換基の少なくとも1個はメルカプ
トである。これらの化合物は、高い写真感度およびハロゲン化銀エマルシジョン
に対するコントラストを可能にする感光剤として有用であるとされている。
発明の概要
本発明は、CRF受容体拮抗物質であって下式Iまたは下式IIで表すことが
できるような新規化合物類、または薬剤学的に許容可能なそれらの塩またはプロドラッグ体であり、これら化
合物において、
XはNまたはCR1であり、
YはNまたはCR2であり、
ZはNR3、OまたはS(O)nであり、
GはOまたはSであり、
QはOまたはS(O)nであり、
Arはフェニル、ナフチル、ピリジル、ピリミジニル、トリアジニル、フラニル
、キノリニル、イソキノリニル、チエニル、イミダゾリル、チアゾリル、インド
リ
ル、ピロリル、オキサゾリル、ベンゾフラニル、ベンゾチエニル、ベンゾチアゾ
リル、イソキサゾリルまたはピラゾリルであって、各々は任意に1〜4個のR5
基で置換され、
R1はいずれの場合も独立にH、C1−C4アルキル、C2−C4アルケニル、C2−

ソキサゾリルまたはピラゾリルであって、それらは、C1−C6アルキル、C3−
C6シクロアルキル、ハロ、C1−C4ハロアルキル、シアノ、−OR7、SH、−
S(O)nR13、−COR7、−CO2R7、−OC(O)R13、−NR8COR7、

に選択される1〜3個の置換基で任意に置換され、
nはいずれの場合も独立に0、1または2であり、
但し、式Iでは、XおよびZがそれぞれNであってYがCR2の場合、R1および
R2はメルカプト基ではあり得ない。
本発明には、治療上有効な量の式Iまたは式IIの化合物を哺乳動物に投与す
ることを含む、哺乳動物における情動障害、不安、うつ病、過敏性腸症候群、外
傷後ストレス症、核上麻痺、免疫抑制、アルツハイマー病、胃腸疾患、拒食症も
しくはその他の摂食障害、薬物もしくはアルコール禁断症状、薬物嗜癖、炎症性
疾患または受胎力問題などの治療方法が含まれる。
また本発明には、薬剤学的に許容可能な担体および治療上有効量の前記いずれ
かの化合物1つを含む医薬組成物も含まれる。
本発明が提供する化合物(特に本発明の標識化合物)は、潜在的に可能な医薬
品のCRF受容体結合能力を測定する際の標準および試薬としても有用である。発明の詳細な説明
本発明の多くの化合物は、1つまたはそれ以上の不斉中心または不斉面を有す
る。特段の指示がない限り、すべてのキラル体(鏡像異性体およびジアステレオ
異性体)およびラセミ体が本発明に含まれる。オレフィン、C=N二重結合など
の多くの幾何異性体もこれらの化合物中に存在するが、本発明においてこのよう
な安定な異性体のすべてについても企図するものである。これら化合物は光学活
性体またはラセミ体に単離できる。ラセミ体を分離するかまたは光学活性出発材
料から合成するなど、光学活性体を生成する方法は当業界では公知である。特定
の立体化学または異性体が特に指示されていない限り、一構造体のキラル体(鏡
像異性体およびジアステレオ異性体)およびラセミ体のすべて、また、幾何異性
体のすべてを意図するものである。
「アルキル」という用語は、指定数の炭素原子を有する、分枝鎖および直鎖の
両アルキルが含まれる。「アルケニル」には、エテニル、プロペニル等の、直鎖
状または分枝鎖状であって、その鎖上の安定な位置に1つまたはそれ以上の不飽
和な炭素−炭素結合が存在する炭化水素鎖が含まれる。「アルキニル」には、エ
チニル、プロピニル等の、直鎖状または分枝鎖状であって、その鎖上の安定な位
置に1つまたはそれ以上の三重の炭素−炭素結合が存在する炭化水素鎖が含まれ
る。「ハロアルキル」には、指定数の炭素原子を有し、1つまたはそれ以上のハ
ロゲンで置換されている分枝鎖または直鎖の両アルキルを含むことを意図する。
「アルコキシ」は、酸素によって橋かけ結合している指定数の炭素原子を有する
アルキル基を表す。「シクロアルキル」には、シクロプロピル、シクロブチル、
シクロペンチル、シクロヘキシル等の、単環式、二環式または多環式環状化合物
の系を含む飽和環基を含むことを意図する。「ハロ」または「ハロゲン」には、
フルオロ、クロロ、ブロモおよびヨードが含まれる。
本明細書で用いる「置換」という用語は、指定原子上の1つまたはそれ以上の
水素が、指示された群から選択された原子によって置き換わることを意味する。
但し、指定の原子の正常な原子価を超えて置き換わることはなく、また、置換に
よって安定な化合物がもたらされるものとする。置換基がケト(すなわち=O)
であれば、指定原子上の2つの水素が置き換わることになる。
置換基および/または変数の組み合せは、このような組み合せによって安定な
化合物がもたらされる場合にのみ許容される。「安定な化合物」または「安定な
構造」とは、反応混合物から使用可能な純度まで分離でき、ならびに有効な治療
薬への調剤に耐え得るほど十分強固な化合物であることを意味する。
「薬剤学的に許容可能な塩」には、式(I)および式(II)の化合物である
酸性塩または塩基性塩が含まれる。薬剤学的に許容可能な塩の例には、アミンな
どの塩基性残基の鉱酸塩または有機酸塩、カルボン酸などの酸性残基のアルカリ
塩または有機塩、その他が含まれるが、これらに限定されるものではない。
本発明の化合物である薬剤学的に許容可能な塩は、遊離酸または遊離塩基の状
態でのこれら化合物を、化学量論的量の適切な塩基または酸と、水または有機溶
媒、あるいは水と有機溶媒の混合溶媒中で反応させることによって生成すること
ができ、一般に、エーテル、酢酸エチル、エタノール、イソプロパノールまたは
アセトニトリルなどの非水性媒体が好ましい。好適な塩の一覧は、レミントンの
医薬品科学(Remington's Pharmaceutical Sciences),第17版,Mack Publishi
ng Company社,Easton,PA,1985,p.1418に見いだせるが、その開示を引用によって
本明細書に組み込むものとする。
「プロドラッグ」とは、このようなプロドラッグを哺乳動物である被検者に投
与すると、生体内で式(I)または式(II)の活性な親薬物(parent drug)
を放出するような共有結合性担体であるとみなされる。式(I)および式(II
)の化合物のプロドラッグは、これら化合物に存在する官能基を、型どおりの操
作においてまたは生体内でその修飾が開裂し、親化合物になるような方法で、修
飾することによって調製する。プロドラッグは、ヒドロキシ基、アミン基または
スルフヒドリル基が、哺乳動物である被検者に投与されると開裂して遊離のヒド
ロキシル、アミノまたはスルフヒドリルの各基を形成するような基のいずれかに
結合しているような化合物を含む。プロドラッグの例には、式(I)および式(
II)の化合物のアルコール官能基およびアミン官能基の酢酸の、ギ酸のおよび
安息香酸の各誘導体等を含むが、これらに限定されるものではない。
本発明の化合物の「治療上有効な量」という用語は、異常レベルのCRFに拮
抗するのに有効な量、またはホストの情動障害、不安またはうつ病の症状を治療
するのに有効な量を意味する。
合成
本発明の新規の置換縮合ピリミジンを、以下に略記した一般的スキームの1つ
によって調製する。このスキームにおいて、Ar、Q、G、X、Y、Z、R1、
R2、R3、R13は前記の通りであり、また、Lはハロ、メタンスルホネート、p
−トルエンスルホネートまたはトリフラートなどの好適な離脱基を表す。
スキーム1 タイプIII(スキーム1)の化合物は、タイプIの化合物(但し、Q=Oま
たはS)であるアルカリ金属塩を[AshleyおよびHarrisの、J Chem Soc.,677(1
944年)、Albertらの、J Chem Soc.,3832(1954年)]の文献による方法に従っ
て作ったタイプIIの化合物と、アセトニトリルなどの溶媒中で温度0℃および
50℃の間の温度で縮合することによって調製される。
タイプIVの化合物は、タイプIIIの化合物を、ジオキサンなどの溶媒中で
温度25℃および100℃の間の温度で、4−アミノヘプタンなどの第一級アミ
ンで処理することによって得られる。これらのアミノ付加物は、炭素に分散させ
た白金などの触媒の存在下、大気圧または高圧にて水素で還元することによって
、あるいは亜ジチオン酸ナトリウム、または酢酸中の鉄、などの還元剤によって
、タイプVの化合物に転化される。
タイプVIの化合物は、環状エーテルまたは芳香族炭化水素などの有機助溶剤
を含むかまたは含まない、酸の水溶液の存在下、タイプVの化合物をアルカリ金
属亜硝酸塩でジアゾ化および環化することによって調製される。
タイプVIIの化合物は、タイプVの化合物を、高沸点エーテルまたは芳香族
炭化水素などの溶媒の存在下または非存在下、100℃〜200℃の間の温度で
、ホスゲン、チオホスゲン、カルボニルジイミダゾール、チオカルボニルジイミ
ダゾール、尿素、チオ尿素、グアニジン等と縮合させることによって調製される
。
スキーム2
タイプVIIIの化合物は、タイプVの化合物を、エーテルまたは芳香族炭化
水素などの溶媒の存在下または非存在下、0℃から200℃の間の温度で、酸、
酸塩化物、無水物、アミドまたはオルトエステルなどの試薬と縮合させることに
よって調製される。
タイプVIIの化合物(この場合R13=H)を、水酸化ナトリウムなどの塩基
の存在下または非存在下、テトラヒドロフランまたはDMFなどの溶媒中、0℃
から100℃の間の温度で、ハロゲン化アルキル等の試薬でアルキル化すると、
タイプVIIIの化合物が生じる。
別法として、タイプVIIの化合物はタイプX(スキーム2)の化合物から調
製される。これらのジアミノピリミジン、すなわちXは、タイプIIの化合物を
、亜ジチオン酸ナトリウム、酸の存在下での鉄または亜鉛(これらに限定される
わけではない)などの還元剤で処理するか、または接触水素化することにより合
成されるタイプIXのジクロロアミノピリミジンから作られる(LaRock、Compre
hensive Organic Transformations,VCH Publishers,NY,1989,411を参照のこと
)。ジアミノ化合物すなわちXを、タイプVIIの化合物をタイプVの化合物か
ら調製するために記述したと同様な方法を用いて、タイプXIIの化合物に
転化し、次いでタイプXIIの化合物を、DMFまたは2−エトキシエタノール
などの溶媒中、25℃および200℃の間の温度で、タイプIの化合物の塩と縮
合させる。
別法として、タイプVIIIの化合物は、先ず、タイプVの化合物からタイプ
VIIIの化合物を調製するために記述したと同様な方法を用いて、タイプXの
化合物をタイプXIIIの化合物に転化し、次いでこのようにして得られたタイ
プXIIIの化合物を、タイプIの化合物をタイプIIの化合物に転化するため
に記述した条件下でタイプIの化合物の塩と縮合させることによって、タイプX
の化合物から調製される。
別法として、タイプVIの化合物は、タイプVの化合物をタイプVIの化合物
にするために先に記述したように、ジアゾ化および環化によってタイプXの化合
物から調製されて、タイプXIの化合物を生じる。続いて、先に記述したように
タイプXIの化合物とともにタイプIの化合物の塩で処理すると、タイプVIの
チアゾロ付加物が得られる。
本発明の化合物およびその合成を、以下の実施例および調製法によってさらに
詳しく説明する。
実施例1 3−ブロモ−4−ヒドロキシ−5−メトキシアセトフェノン
0〜5℃に保たれたアセトバニロン(10.0g)の150mLクロロホルム
溶液に、30mLクロロホルム中の臭素(9.62g)を温度が5℃を超えるこ
とのないように滴下した。添加完了後、混合液を0〜5℃にて4時間撹拌した。
残渣を水で処理した。有機層をMgSO4で乾燥し減圧下で溶媒を除去すると、
ピンクがかった粉末が得られた。それをエーテルで粉砕した後濾過すると、融点
148〜152℃の表題の化合物が得られた。実施例2 3−ブロモ−4−ヒドロキシ−5−メトキシ− α,α−ジメチルベンゼンメタノール
窒素ガスの存在下0〜5℃に保たれた5−ブロモ−4−ヒドロキシ−3−メト
キシアセトフェノン(3.0g)の無水テトラヒドロフラン(60mL)溶液に
、臭化メチルマグネシウム(3Mジエチルエーテル溶液、11.42mL)を温
度が5℃を超えることのないように滴下した。添加完了後、溶液を室温にて2時
間撹拌した。飽和塩化アンモニウムを発泡が止むまで滴下した。混合液を過剰量
の飽和塩化アンモニウムで処理した。有機層をMgSO4で乾燥し減圧下で溶媒
を除去すると粘性オイルとして表題化合物が得られ、これは時間が経過すると固
化して、融点107〜112℃が得られた。
実施例3 3−ブロモ−5−メトキシ−α,α−ジメチル−4−[[6−クロロ− 2−メチル−5−ニトロ−4−ピリミジニル]オキシ]ベンゼンメタノール
3−ブロモ−4−ヒドロキシ−5−メトキシ−α,α−ジメチルベンゼンメタ
ノール(1.16g)を10%NaOH(1.78g)と水5mLに溶解した。
減圧下で溶媒を除去した。その塩をアセトニトリル50mL中に溶解し、すでに
冷却してある4,6−ジクロロ−2−メチル−5−ニトロ−1,3−ピリミジン
(0.92g)の80mLアセトニトリル溶液(0〜5℃)にピペットで滴下し
た。この混合液を0〜5℃にて3時間撹拌した。減圧下で溶媒を除去し、残渣を
塩化メチレンで抽出した。抽出物を合一し、減圧下で蒸発させると表題の化合物
が得られた。実施例4 3−ブロモ−5−メトキシ−α,α−ジメチル−4− [[2−メチル−5−ニトロ−6−[(1−プロピルブチル)アミノ] −4−ピリミジニル]オキシ]ベンゼンメタノール
炭酸カリウム0.50gを添加した3−ブロモ−5−メトキシ−α,α−ジメ
チル−4−[[6−クロロ−2−メチル−5−ニトロ−4−ピリミジニル]オキ
シ]ベンゼンメタノール(1.88g)の無水1,4−ジオキサン(50mL)
溶液に、4−ヘプチルアミン(1.00mL)を添加し、その溶液を室温にて2
時間撹拌した。減圧下で溶媒を除去し、残渣を水に取って塩化メチレンで抽出し
た。抽出物を合一し、MgSO4で乾燥した。この溶媒を減圧下で除去し、酢酸
エチルとヘキサンの1:1の混合物を用いて残渣をシリカゲルのクロマトグラフ
ィーで精製すると、融点138℃の表題の化合物を得た。
融点119℃〜120℃の3−ブロモ−5−メトキシ−α,α−ジメチル−4
−[(6−(1−メトキシプロピル)アミノ−2−メチル−5−ニトロ)−4−
ピリミジニル]−オキシベンゼンメタノールも、同様の方法で調製した。
実施例5 3−ブロモ−5−メトキシ−α,α−ジメチル−4− [[5−アミノ−2−メチル−6−[(1−プロピルブチル)アミノ]− 4−ピリミジニル]オキシ]ベンゼンメタノール
3−ブロモ−5−メトキシ−α,α−ジメチル−4−[[2−メチル−5−ニ
トロ−6−[(1−プロピルブチル)アミノ]−4−ピリミジニル]オキシ]ベ
ンゼンメタノール(0.80g)の50mLエタノール溶液に、白金黒(5%、
0.25g)を添加した。この混合物を圧力41psiで18時間水素化した。
混合物をセライトで濾過した後、減圧下で濾液を除去した。残渣を1Nの
NaOHに溶解し、塩化メチレンで抽出した。合一させた塩化メチレン抽出物を
MgSO4で乾燥し減圧下で濾液を除去すると、融点が114〜116℃の表題
の化合物が得られた。
実施例6 3−ブロモ−5−メトキシ−α,α−ジメチル−4−[(5−メチル− 3−(1−プロピルブチル)−3H−1,2,3−トリアゾロ[4,5−d] ピリミジン−7−イル)−オキシ]ベンゼンメタノール
3−ブロモ−5−メトキシ−α,α−ジメチル−4−[[5−アミノ−2−メ
チル−6−[(1−プロピルブチル)アミノ]−4−ピリミジニル]オキシ]ベ
ンゼンメタノール(0.70g)の氷酢酸(35mL)溶液に、0.10g亜硝
酸ナトリウムの1mL水溶液を滴下した。この混合液を室温で20分間撹拌し、
次いで水で希釈し、1NのNaOHで塩基性化した後、酢酸エチルで3回抽出を
行った。合一させた抽出物をMgSO4で乾燥し減圧下で溶媒を取り除くと、粘
性の液体状の表題の化合物が得られた。
実施例7 7−[2−ブロモ−6−メトキシ−4−(1−メチルエテニル)フェノキシ]− 5−メチル−3−(1−プロピルブチル)−3H−1,2,3−トリアゾロ [4,5−d]ピリミジン
3−ブロモ−5−メトキシ−α,α−ジメチル−4−[(5−メチル−3−(1
−プロピルブチル)−3H−1,2,3−トリアゾロ[4,5−d]ピリミジン
−7−イル)−オキシ]ベンゼンメタノール(0.59g)の35mLベンゼン
溶液に、少量のp−トルエンスルホン酸を添加した。この溶液を共沸条件下で
1.5時間還流した。溶液を室温にまで冷却して、ただちに飽和NaHCO3で
洗浄し次いで水で洗浄した。有機相をMgSO4で乾燥し、減圧下で溶媒を除去
した。残渣を酢酸エチルとヘキサン1:1の混合物を用いてシリカゲルのクロマ
トグラフィーにかけると、融点110℃〜115℃の無色固体状の表題の化合物
が得られた。
実施例8 7−[2−ブロモ−6−メトキシ−4−(1−メチルエチル)フェノキシ− 5−メチル−3−(1−プロピルブチル)−3H−1,2,3−トリアゾロ [4,5−d]ピリミジン
5%(0.19g)の白金黒を、7−[2−ブロモ−6−メトキシ−4−(1
−メチルエテニル)フェノキシ]−5−メチル−3−(1−プロピルブチル)−
3H−1,2,3−トリアゾロ[4,5−d]ピリミジン(0.17g)の50
mLエタノール溶液に添加した。混合液を40psiの圧力下で18時間水素化
し、セライトで濾過した。減圧下で濾液から溶媒を除去し、残渣をヘキサンから
再結晶させると、融点が129〜131℃の無色結晶状の表題の化合物が得られ
た。
実施例9 3−(1−エチルプロピル)−5−メチル−7−(2,4,6−トリメチル フェノキシ)−3H−1,2,3−トリアゾロ[4,5−d]ピリミジン
2,4,6,−トリメチルフェノール(0.114)をナトリウムメトキシド
(0.334g)メタノール(10mL)溶液に添加し、その溶液を減圧下で蒸
発させ乾燥させた。このようにして得られた塩を10mLのアセトニトリルに溶
解し、温度が5℃を超えることのないようにしながら、7−クロロ−3−(1−
エチルプロピル)−5−メチル−3H−1,2,3−トリアゾロ[4,5−d]
ピリミジンの35mLのアセトニトリルの冷溶液(0〜5℃)に滴下した。この
混合液を0〜5℃にて3時間撹拌した。次いで、減圧下で混合液から溶媒を除去
し、残渣を水で処理した後塩化メチレンで3回抽出した。合一させた抽出物をM
gSO4で乾燥し、減圧下で溶媒を除去した。この物質を、1%メタノールの塩
化メチレン溶液を用いてシリカゲルで精製すると、融点92℃の無色粉末状の表
題の化合物が得られた。
実施例10 6−(1−メトキシプロピル)アミノ−2−メチル−5−ニトロ− 4−[(2−ブロモ−6−メトキシ−4−(1−メチルエテニル) フェノキシ]ピリミジン
4−トルエンスルホン酸の結晶数個を、3−ブロモ−5−メトキシ−α,α−
ジメチル−4−[[6−(1−メトキシプロピル)アミノ−2−メチル−5−ニ
トロ]−ピリミジニル]]オキシ−ベンゼンメタノール(1.70g)のベンゼ
ン(30mL)溶液に添加し、得られた混合液をディーン−スターク・トラップ
を用いて還流下で一晩加熱した。次いで、それを冷却し、ベンゼン(70mL)
で希釈し、続いて1NのNaOH水溶液および水で洗浄した後、硫酸マグネシウ
ムで乾燥し減圧下で蒸発乾燥させると半固体を生じ、その半固体はエーテルで粉
砕し濾過すると黄色い固体が得られた。エタノールから再結晶化すると、融点1
36〜137℃の淡黄色の結晶性固体状の表題化合物が得られた。
実施例11 2−アミノ−6−(1−メトキシプロピル)アミノ−2−メチル−4− [(2−ブロモ−6−メトキシ−4−(1−メチルエチル)フェノキシ] ピリミジン
白金黒(5%、250mg)を、6−(1−メトキシプロピル)アミノ−2−
メチル−5−ニトロ−4−[(2−ブロモ−6−メトキシ−4−(1−メチルエ
テニル)フェノキシ]ピリミジン(550mg)の酢酸エチル(100mL)溶
液に注意深く添加し、得られた混合液を30p.s.i.下で一晩水素化した。次
いで、混合液をセライトのパッドを通して濾過し、減圧下で濾液を蒸発乾燥させ
ると粘性の強い液状の表題の化合物を得られた。
実施例12 7−[2−ブロモ−6−メトキシ−3−(1−メトキシプロピル)− 5−メチル−4−(1−メチルエチル)フェノキシ]− 3H−1,2,3−トリアゾロ[4,5−d]ピリミジン
亜硝酸ナトリウム(42mg)の水(1mL)溶液を、室温で撹拌しながら、
2−アミノ−6−(1−メトキシプロピル)−アミノ−2−メチル−4−[(2
−ブロモ−6−メトキシ−4−(1−メチルエチル)−フェノキシ]ピリミジン
(270mg)の酢酸(10mL)溶液に滴下した。添加完了後、混合液を室温
で一晩撹拌し、減圧下酢酸のほとんどを除去した。残渣を1NのNaOH水溶液
で処理した後、酢酸エチル(2X)で抽出した。酢酸エチル抽出物を水で洗浄し
た後、硫酸マグネシウムで乾燥し減圧下で蒸発乾燥させると半固体を生じ、この
半固体を、溶離剤として酢酸エチル:ヘキサン1:1を用いてシリカのクロマト
グラフィーにかけると、融点149〜150℃の表題の化合物が得られた。有用性
生理学的活性評価のためのCRF−R1受容体結合アッセイ
以下に、標準的結合アッセイで使用するクローン化したヒトCRF−R1受容
体含有細胞膜の分離について記述し、また、アッセイそれ自体についても記述す
る。
メッセンジャーRNAをヒトの海馬から分離した。オリゴ(dt)12−18
を用いてmRNAを逆転写し、コード領域を開始コドンから終結コドンまでPC
Rによって増幅した。得られたPCR断片をpGEMVのEcoRV部位にクロ
ーン化し、XhoI+XbaIを用いてそこから挿入物を再生して、ベクターp
m3ar(これにはCMVプロモータ、SV40’t’スプライスおよび初期ポ
リAシグナル、エプスタイン・バールウイルス複製起点、ならびにハイグロマイ
シン選択マーカが含まれる)のXhoI+XbaIの中にクローン化した。こう
して得たphchCRFRと呼ばれる発現ベクターを293EBNA細胞にトラ
ンスフェクションし、400μMのハイグロマイシンの存在下で、エピソームを
保持している細胞を選択した。ハイグロマイシンによる4週間の選択に耐えて生
存した細胞をプールし、懸濁液中での生長に適応させた後、後述する結合アッセ
イのための膜生成に使用した。次いで、懸濁細胞約1x108個を含有するそれ
ぞれのアリコートを遠心分離してペレット状にし、凍結した。
hCRFR1受容体でトランスフェクトされた293EBNA細胞を含有する
前記凍結ペレットを、結合アッセイ用に、氷温の組織緩衝液(50mMのHEP
ES緩衝液、pH7.0、10mMのMgCl2、2mMのEGTA、1μg/
lのアプロチニン、1μg/lのロイペプチンおよび1μg/mlのペプスタチ
ンを含有)10ml中でホモジナイズする。ホモジネートを40,000xgで
12分間遠心分離し、得られたペレットを組織緩衝液10ml中で再度ホモジナ
イズする。さらに40,000xgで12分間遠心分離した後、ペレットをタン
パク質濃度が360μg/mlになるまで再度懸濁し、アッセイに使う。
結合アッセイは96ウエル・プレート内で実施され、各ウエルの容積は300
μlである。各ウエルに、希釈検査薬(薬剤の最終濃度は10-10〜10-5Mの
範囲)50μl、125I−ヒツジ−CRF(125I−o−CRF)(最終濃度15
0pM)100μlおよび前記細胞ホモジネート150μlを添加する。次いで
、プレートを室温で2時間培養し、その後適当な細胞回収器を使用して培養物を
GF/Fフィルタ(予め0.3%ポリエチレンイミンで濡らしておく)で濾過す
る。フィルタを氷温の組織緩衝液で2度ゆすいだ後、各フィルタを取り外し、ガ
ンマ計数器でそれらの放射能を評価する。
検査薬の希釈を変えて、細胞膜への125I−o−CRF結合阻害曲線を、相互
曲線フィットプログラムLIGANDで[P.J.MunsonおよびD.Rodbard,Anal.Bio
chem.107:220(1980年)]分析する。このプログラムによって阻害を表すKi値
が得られ、それらの値は生理活性の評価に使用される。
受容体へのCRFの阻害を表すKi値が約10000nM未満であれば、その
化合物は活性であるとみなされる。CRF−刺激性アデニル酸シクラーゼ活性の阻害
CRF−刺激性アデニル酸シクラーゼ活性の阻害をG.Battagliaらによるシナ
プス(Synapse)1:572(1987年)に記載の通りに実施した。手短に言えば、37
℃で10分間、Tris−HCl(37℃でpH7.4)を100mM、MgC
l2を10mM、EGTAを0.4mM、0.1%BSA、イソブチルメチルキ
サンチン(IBMX)1mM、ホスホクレアチンキナーゼを250単位/ml、
クレアチンリン酸を5mM、グアノシン5’−三リン酸を100mM、oCRF
を100nM、アンタゴニストペプチド(濃度範囲10-9〜10-6m)および元
の湿潤重量で0.8mgの組織(タンパク質約40〜60mg)を含有する緩衝
液200ml中で、アッセイを実施した。反応は、1mMのATP/[32P]A
TP(約2〜4mCi/管)を添加して開始し、50mMのトリス−HCL 1
00ml、45mMのATPおよび2%ドデシル硫酸ナトリウムを添加して終了
した。cAMPの回収率をモニタするために、分離の前に各管に[3H]cAM
P(約40,000dpm)1μlを添加した。[32P]ATPからの[32P]
cAM
Pの分離は、Dowexおよびアルミナコラムで順次溶離して実施した。回収率
は一貫して80%以上であった。
本発明の化合物の一部はこのアッセイで試験し、活性であることが判明した。生体内生物検定
本発明の化合物の生体内活性は、当業界内で利用できかつ容認されるバイオア
ッセイであればいずれを用いても評価できる。これら試験の実例には、聴覚刺激
試験、階段上り試験および慢性投与試験が含まれる。本発明の化合物を試験する
のに有用なこれらおよびその他のモデルについては、C.W.BerridgeとA.J.Dunnの
、脳研究概説(Brain Research Reviews)15:71(1990年)に略述されている。
化合物は齧歯類動物または小哺乳動物のいずれの種でも試験できよう。本明細
書のアッセイの開示は、本発明の可能性を限定しようとするものではない。
本発明の化合物は、うつ病、情動障害および/または不安に罹患している患者
の、副腎皮質刺激ホルモン放出因子の異常レベルに関連したバランス失調の治療
に有用である。
本発明の化合物は、哺乳動物の身体の有効成分作用部位に有効成分を接触させ
る方法でこれらの異常を治療するために投与することができる。これらの化合物
は、個々の治療薬としての製剤または治療薬の組み合せ製剤とともに使用する方
法に利用できるかぎり、従来のいずれの方法によっても投与することができる。
これらの化合物は単独でも投与できるが、一般には選択した投与経路および標準
的薬学的慣行に基づいて選んだ医薬品担体といっしよに投与される。
投与量は、その使用法ならびに個々の薬剤の薬力学的特性、その投与方式およ
び投与経路、受容者の年齢、体重および健康、症状の性質および程度、併用療法
の種類、治療回数、所望の効果などの既知の要因によって異なる。前記疾患また
は症状の治療に使用する目的で、本発明の化合物を一日当り有効成分0.002
〜200mg/kg体重の用量で経口投与することもできる。所望の薬理学的効
果を得るには、通常、一日に1回〜4回の分割量でまたは持続性製剤で0.01
〜10mg/kg体重が有効である。
投与に好適な製剤(組成物)は、単位当り約1mg〜約100mgの有効成分
を含有する。通常、これらの医薬組成物には、組成物の総重量に対し重量で約0
.5%〜95%の有効成分が存在する。
有効成分は、カプセル剤、錠剤および散剤などの固形製剤で、またはエリキシ
ル剤、シロップ剤および/または懸濁剤などの液剤で経口投与できる。本発明の
化合物は、滅菌液剤で非経口投与することもできる。
有効成分と、これらに限定されるわけではないがラクトース、デンプン、ステ
アリン酸マグネシウム、ステアリン酸またはセルロース誘導体などの好適な担体
とを含むために、ゼラチンカプセルを使用することもできる。圧縮錠剤を作るた
めに同様の希釈剤を使用することもできる。錠剤およびカプセル剤はいずれも持
続性医薬品として製造することができ、一定期間にわたって薬剤を連続的に放出
することが可能となる。圧縮錠剤を糖衣またはフィルムコーティングして、不快
な味を遮断したり、有効成分を空気から保護したり、または胃腸管において錠剤
を選択的に崩壊することが可能となる。
患者が受け入れやすくするために、経口投与用液剤に着色料や着香料を含有さ
せることもできる。
一般に水、薬剤学的に許容可能な油、生理的食塩水、デキストロース(グルコ
ース)水溶液ならびに関連の糖溶液およびプロピレングリコールまたはポリエチ
レングリコールなどのグリコール類は、非経口投与溶液に好適な担体である。非
経口投与溶液は、有効成分の水溶性塩、好適な安定剤、および必要に応じて緩衝
物質を含有していると好ましい。重亜硫酸ナトリウム、亜硫酸ナトリウムまたは
アスコルビン酸などの抗酸化剤は、単独でもまたは組み合せても、好適な安定剤
である。クエン酸およびその塩、ならびにEDTAも使用される。さらに、非経
口投与溶液に、塩化ベンザルコニウム、メチルパラベンまたはプロピルパラベン
、およびクロロブタノールなどの保存剤を含有させることもできる。
好適な医薬品担体については、当分野では標準的な参照文献であるレミントン
医薬品科学、A.Osolに記載がある。
本発明の化合物の投与に有用な医薬品の剤形は、以下のように説明できる。カプセル剤
多数の単位カプセル剤は、標準的なツーピース硬ゼラチンカプセルに各々粉末
