【発明の詳細な説明】エステルを異性化することによりカルボン酸および/または対応するエステルを 製造する方法
本発明は、nが3より大きいかまたは等しい(n−1)個の炭素原子を含有す
るアルコールおよびギ酸エステルの異性化による、nが3より大きいかまたは等
しいn個の炭素原子を含有するカルボン酸および/または対応するエステルの製
造に関する。
カルボン酸への様々な経路が知られており、そして工業規模にて用いられる。
これらの中に、(n−1)個の炭素原子を含有するアルコールをカルボニル化し
て、該アルコールより1個多い炭素原子を有する酸および/またはエステルを得
る方法がある。これらのカルボニル化反応は、反応体の一つである一酸化炭素の
圧力下、ロジウムおよび/またはイリジウムをベースとした化合物およびヨウ素
含有促進剤を含む均質触媒系の存在下で液相において用いられ得る。
更なる経路は、ロジウムまたはイリジウムをベースとした触媒の存在下での、
(n−1)個の炭素原子を含有するアルコー
ルおよびギ酸エステルの異性化により構成される。
異性化方法の中で、欧州特許EP−A−0,045,637は、イリジウムを
ベースとした触媒の存在下でエステルを用いることを記載する。この方法は、カ
ルボン酸から選択される溶媒、特に、生成された酸を用いて反応が行われること
を特徴とする。更に、該反応は、窒素を含んでなる雰囲気中で行われる。該反応
中一酸化炭素はいかなる特別な利点も追加せず、また異性化反応における或る阻
害の原因とさえなって副反応を助長し得る、ということが分かった。かかる挙動
は、触媒系がロジウムをベースとするときに観察される挙動(この場合、一酸化
炭素の存在は、その金属を均質相に維持することにとって必須である)と完全に
相違する、ということが留意されるべきである。このタイプの方法(その重要性
はここで繰り返さない)は、記載された反応が十分には効率的でないので、工業
にとって真の利益を有さない。その反応速度は、2mol/h.lの生成酸およ
び/またはエステルの程度にすぎない。
先の方法の結果を改善するために、EP−A−0,135,286は、パラ−
トルエンスルホン酸のような強スルホン酸タイプの酸の存在下で異性化反応を行
うことを提案する。この方
法の条件下で、反応は多量の異性化されるべきエステルを用い、しかして該エス
テルは反応溶媒としても用いられる。この改善は反応の活性を改善することに寄
与するけれども、それは補足化合物の使用を必要としてこの方法を単純にしない
不利を有する。
更に、該酸は反応媒質の条件下で分解し得る。
本発明の目的は、この目的を達成するのに補足化合物を必ずしも用いることな
く、上記に記載された二つの異性化変法に関して改善生産性でもって、ギ酸エス
テルを酸および/またはエステルに異性化する方法を提供することである。
これらのおよび他の目的は本発明において満たされ、しかして本発明は、式H
COOR2(III)〔ここで、R2は、nが3より大きいかまたは等しい(n−
1)個の炭素原子を含有する〕を有する少なくとも1種のエステルを、水、溶媒
、並びに少なくとも1種のハロゲン化促進剤および少なくとも1種のイリジウム
をベースとした化合物からなる触媒系の存在下で反応させることにより、式R1
COOH(I)、R1COOR’1(II)〔ここで、基R1およびR’1は、同一
でも異なっていてもよくそしてnが3より大きいかまたは等しい(n−1)個の
炭素原
子を含有する〕をそれぞれ有するカルボン酸および/または対応するエステルを
製造することを提案する。本発明の反応は、0.1×105Paないし25×1
05Paの範囲の一酸化炭素の分圧を維持することにより該反応が行われること
並びに式HCOOR2を有するエステルの量が反応混合物の重量の20%未満に
維持されることを特徴とする。
先行技術において主張されたのとは対照的に、一酸化炭素の存在はイリジウム
の存在下でエステルを異性化するために必須である、ということを我々は確立し
た。
本反応の更なる重要な特徴は、最良の生産性を得るために反応するエステルの
量が多くとも20%でなければならないことである。
かくして、これらの二つの特徴の組合わせは、単に一酸化炭素分圧およびエス
テルの濃度を反応中規定範囲内に維持することにより、公知の異性化方法の生産
性を改善し得る。
式(I)ないし(III)において、基R1、R’1およびR2は同じ意味を有
するが、しかしそれらは同一でも異なっていてもよい。
さて、一層明瞭化のために、反応体の特質が記載される。
本発明の方法において用いられる反応体は式HCOOR2(III)を有する
ギ酸エステルであり、しかしてこの式において基R2は、生成される生成物即ち
カルボン酸および/またはエステルにおいて所望される炭素原子の数nに関連し
てnが3より大きいかまたは等しい(n−1)個の炭素原子を含有する。
基R2は、少なくとも1個の酸素、硫黄、窒素またはハロゲンタイプの原子を
含有する少なくとも1個の官能基を含み得る、ということが留意されるべきであ
る。該官能基の非制限的例は、エーテル、エステル、ニトリルおよび塩化物官能
基である。
一層特に、上記に定められたような少なくとも1個の官能基を随意に含有する
基R2は、2〜20個の炭素原子を含有する脂肪族基、または3〜10個の炭素
原子を含有する環状脂肪族基、または6〜20個の炭素原子を含有するアリール
基、または7〜21個の炭素原子を含有するアリールアルキル基を表す。用語“
脂肪族基”は、線状または分枝状の飽和または不飽和炭化水素基を意味する。
本発明の有利な実施において、上記に定められたような少なくとも1個の官能
基を随意に含有する基R2は、2〜10個の
炭素原子を含有する脂肪族基を表す。
本発明の更なる有利な実施において、上記に定められたような少なくとも1個
の官能基を随意に含有する基R2は、6〜10個の炭素原子を含有するアリール
基を表す。
本発明の新規な実施において、上記に定められたような少なくとも1個の官能
基を随意に含有する基R2は、7〜15個の炭素原子を含有するアリールアルキ
ル基を表す。
反応体として用いられるギ酸エステルは、文献、特に「“アドヴァンスト・オ
ーガニック・ケミストリー(Advanced Organic Chemis
try),第3版”,ジェリー・マーチ著,ウイレー・インターサイエンス出版
,特に第347頁以下」に記載されている慣用の方法を用いて製造され得る。
本発明の方法は、少なくとも1種のハロゲン化促進剤および少なくとも1種の
イリジウムをベースとした化合物からなる触媒系の存在下で行われる。
触媒系の成分の一つを成すハロゲン化促進剤は、好ましくは、ヨウ素含有化合
物から選択される。
ハロゲン化促進剤は、ヨウ素単独の、または水素のような他の元素、C1〜C1 0
アルキル基、C1〜C10アシル基もしくは
C6〜C10アリール基と組み合わせて、あるいはアルカリ金属ヨウ化物、または
遷移金属ヨウ化物もしくはIIIB欄(「サプレメント・アウ・ブレティン・デ
・ラ・ソシエテ・チミック・
France),no.1,1966」において公表されている周期表において
)の金属のような金属のヨウ化物であり得る。
ハロゲン化促進剤は複数種の上記の促進剤の混合物により構成され得る、とい
うことが留意されるべきである。
本発明はまた、適切な前駆体を用いてのハロゲン化促進剤のその場での製造を
包含する。
本発明において用いるのに適当である促進剤の非制限的例は、ヨウ素、ヨウ化
水素酸、メチルヨージド、エチルヨージド、エチル1,1−ジヨージド、アセチ
ルヨージド、ヨウ化アルミニウム、ヨウ化クロム、ヨウ化リチウムおよびヨウ化
カリウムである。
本発明の特定の実施において、用いられる促進剤は、水素またはC1〜C10ア
ルキル基を含有する。ハロゲン化促進剤は、好ましくは、ヨウ素およびメチルタ
イプの基を含有する。
本発明の方法において用いられる触媒系の第2の成分は、少なくとも1種のイ
リジウムをベースとした化合物により構成される。
第一に、本発明の反応は、一層特には、均質触媒の存在下で行われる。換言す
れば、これは、イリジウムをベースとした化合物が反応混合物に可溶である形態
にあることを意味する。このイリジウムをベースとした化合物の一部が非溶解形
態で存在しても反応を行う際に重大な困難性を呈しない、ということが留意され
るべきである。
本発明の実施条件下で反応媒質に可溶であるかまたは溶解され得るいかなるイ
リジウム化合物も用いられ得る。本発明を行うために特に適している非制限的例
は、イリジウム金属、この金属の単純塩、酸化物または配位錯体である。
イリジウム単純塩の例は、ハロゲン化イリジウムである。一層特に、そのハロ
ゲンは、塩素、臭素またはヨウ素(好ましくは、後者)から選択される。かくし
て、IrI3、IrBr3、IrCl3、IrI3・4H2O、IrI4、IrBr3
・4H2Oのような化合物が、本発明の方法において用いられ得る。
IrO2、Ir203・xH2Oから選択された酸化物もまた、
本発明の方法において用いられ得る。
イリジウムの可溶性配位錯体に関して、最も通常に用いられる化合物は、一酸
化炭素または一酸化炭素/ハロゲンの組合わせから選択された配位子を有するも
のであり、しかして該ハロゲンは塩素、臭素または一層特にヨウ素から選択され
る。しかしながら、配位子が例えば有機リン化または有機窒素化化合物から選択
される可溶性イリジウム錯体の使用は、排除されない。
当業者に知られている配位錯体の非制限的例は、(IrC1COD)2、Ir4
(CO)12、Ir(CO)2I2 -Q+、Ir(CO)2Br2 -Q+、Ir(CO)2
Cl2 -Q+であり、しかしてこれらの式においてQは水素、基NR4、PR4を表
し、基Rは同一でも異なっていてもよくそして水素、炭化水素基、例えばアルキ
ル基(好ましくは、1〜6個の炭素原子を含有する)、シクロアルキル基(好ま
しくは、シクロヘキシル基)、アリール基(好ましくは、フェニル基)およびア
リールアルキル基(好ましくは、ベンジル基)から選択される。
これらの触媒は、当業者に知られているいかなる方法を用いても得られ得る。
かくして、本発明を実施するのに適当なイリジウムをベースとした触媒溶液の製
造のために、EP−A−
0,657,386および国際特許出願WO95/17963が参照され得る。
本発明の反応は、イリジウムに加えて1種またはそれ以上の他のVIII族金
属を含む触媒系でもって行われ得る。一層特に、反応は、ロジウムおよびイリジ
ウムの組合わせでもって、もしくはルテニウムおよびイリジウムの組合わせでも
って、またはこれらの3種の金属をベースとした触媒系でもって行われ得る。
かかる変法が採用される場合、イリジウム対そのほかの関連金属のモル比は、
一層特に、1/10ないし10/1の範囲にある。それは、好ましくは、1/1
を越える。
上記に述べられたように、本発明の異性化反応は、水および溶媒の存在下で行
われる。
一層特に、溶媒は、カルボン酸から選択される。
本発明の第1の実施態様において、カルボン酸は、一層特に、2〜10個の炭
素原子、好ましくは2〜6個の炭素原子を含有する脂肪族カルボン酸から選択さ
れる。
本発明の第2の実施態様において、カルボン酸は、反応中合成されるものに相
当する。
本発明の範囲は、溶媒として2つのタイプの酸の混合物を用いることを包含す
る。
本発明の反応はまた、混合物中に存在するギ酸の存在下で行われる。
無論、本発明の範囲は、反応条件下で不活性である補足溶媒を用いることを包
含する。このタイプの溶媒の例は、エステル(例えば、メチルプロピオネート)
、エーテル(例えば、イソプロピルエーテル、ジフェニルエーテル、テトラヒド
ロフラン、ジオキサン)、ケトン(例えば、メチルイソブチルケトン)、アミド
(特に、ジメチルホルムアミド)、スルホキシド(例えば、ジメチルスルホキシ
ド)、またはハロゲン化されていてもされていなくてもよい脂肪族、環状脂肪族
もしくは芳香族炭化水素、特にペンタン、シクロヘキサン、べンゼン、トルエン
またはジクロロメタンである。
好ましい共溶媒は、本発明の方法により得られるエステルまたは得られる酸に
対応するエステル、即ち式R1COOR’1(II)を有するエステルに相当する
。
共溶媒が用いられる場合、カルボン酸の量は、好ましくは、共溶媒の量より多
い。
本発明の方法を用いて生成されるカルボン酸および/またはエステルは、それ
ぞれ次の式R1COOH(I)、R1COOR'1(II)を有し、しかしてここで
R1およびR’1は、nが3より大きいかまたは等しい(n−1)個の炭素原子を
含有する。
基R2の特質に関しての上記の指摘は、本件において有効なままにある。その
結果、R2について与えられた一般的および特定的定義は、基R1およびR’1に
言及されかつ与えられ得る。
無論、基R1、R’1およびR2は、同一または異なり得る。
本発明の反応が複数個のヒドロキシル基を含有するアルコールまたはエステル
の存在下で行われるとき、形成される生成物は、反応するエステル中にあったヒ
ドロキシル官能基と同じぐらい多いカルボキシル官能基(それらが遊離にあるに
せよまたはエステル化されているにせよ)を含有する。
本発明の方法は、数多くのカルボン酸の製造、一層特に次の酸の製造を可能に
する。即ち、
・プロピオン酸、酪酸、イソ酪酸、吉草酸、イソ吉草酸、ピバル酸、カプロン酸
、カプリン酸、ラウリン酸、ミリスチン酸、パルミチン酸またはステアリン酸の
ような飽和脂肪族モノカルボン酸、
・メチルマロン酸、コハク酸、グルタル酸、アジピン酸、ピメリン酸、スベリン
酸、アゼライン酸またはセバシン酸のような飽和脂肪族ジカルボン酸、
・プロピオル酸、メタクリル酸、クロトン酸、イソクロトン酸、ペンテン酸、セ
ネシオ酸、チグリン酸、オレイン酸、マレイン酸、フマル酸、シトラコン酸また
はメサコン酸のような不飽和脂肪族モノまたはジカルボン酸、
・ショウノウ酸、キク酸またはシクロヘキサンカルボン酸のような飽和または不
飽和炭素環式カルボン酸、
・ニコチン酸、イソニコチン酸またはピコリン酸のような複素環式カルボン酸、
・安息香酸、フタル酸、イソフタル酸もしくはテレフタル酸、ナフタレンカルボ

は4−べンゾイル安息香酸のようなオキソ酸。
一層特に、本発明は、プロピオン酸、酪酸、カプロン酸、カプリン酸、ラウリ
ン酸、フェニル酢酸もしくは吉草酸のような酸またはそれらのエステルの合成の
ために特に適している。
複数個のヒドロキシル基を含有するアルコールおよびギ酸エステルから対応す
る酸またはエステルを製造する場合、本発明の方法は、アジピン酸もしくはその
エステルまたはテレフタル酸もしくはそのエステルが合成されることを可能にす
る。
本発明の方法は、式(IV)
〔ここで、R3は、1個の置換基または複数個の置換基Rを担持し得るフェニル
またはナフチル基を表し、Rは、次の基の1つを表す:
・1〜12個の炭素原子を含有する線状または分枝状アルキルまたはアルケニル
基、好ましくは1〜4個の炭素原子を含有する線状または分枝状アルキル基、
・1〜12個の炭素原子を含有する線状または分枝状アルコキシ基、好ましくは
1〜4個の炭素原子を含有する線状または分枝状アルコキシ基、
・2〜8個の炭素原子を含有する線状または分枝状アシル基、好ましくはアセチ
ル基またはベンゾイル基、
・2〜8個の炭素原子を含有する線状または分枝状アシルオキシ基、好ましくは
アセトキシ基、
・1〜8個の炭素原子を含有する線状または分枝状アシルアミド基、好ましくは
アセトアミド基。〕
を有するアリールプロピオン酸の製造のために特に適している。
これらの置換基の種類の記載は、例示としてなされておりそしていかなるよう
にも制限的でない。
環上の置換基の数は、当業者により容易に決定され得る。それは、一般に、1
〜3である。
特定の例は、2−(ベンゾイルフェニル)プロピオン酸(ケトプロフェン(K
etoprofene)(登録商標))、2−(4−イソブチルフェニル)プロ
ピオン酸(イブプロフェン(Ibuprofene)(登録商標))および2−
(6−メトキシナフチル)プロピオン酸(ナプロキセン(Naproxene)
(登録商標))である。
式(IV)を有する酸は、次の反応図式:
を用いて製造され得る。
式(D)を有する化合物は、本発明の方法を用いて化合物(C)の異性化によ
り得られる。
式(C)を有する化合物への一つの可能な経路は、ケトン化合物(A)をアル
コール(B)へ還元し、その後ギ酸を用いてエステル化する経路である。
(A)および(B)は、慣用的に、例えば接触水素化により、特に触媒(該触
媒は、支持体、好ましくはカーボンブラック上に随意にデポジットした、周期表
のVIII族の微細貴金属、好ましくはパラジウムである)の存在下で還元され
得る。
エステル化反応に関して、それは、酸触媒、例えば硫酸またはスルホン酸、特
にp−トルエンスルホン酸の存在下で通常のやり方で行われる。
かくして、アリールプロピオン酸は、対応するケトンを還元することによりそ
れ自体得られた対応するアルコールのエステ
ル化により得られた1−アリールエチルホルメートの異性化により得られる。
かくして、本発明の方法により、アリールプロピオン酸が製造される。式CH3
−CHR3−COOR4(こでR4は通常基−CH−(R3)(CH3)を表す)を
有するエステルが付随的に形成し得るということが留意されるべきである。
かくして、本発明の方法は、反応の持続期間じゅう一酸化炭素の特定分圧およ
び反応するエステルの特定濃度を維持することから成る。
かくして、一酸化炭素の分圧は、0.1×105Paないし25×105Paの
範囲に保たれる。
圧力は、パスカル絶対圧にて表されそして熱時に即ち反応温度において測定さ
れる。
本発明の一層特定の実施において、一酸化炭素の分圧は、0.5×105Pa
より高く、好ましくは105Paより高く維持される。
一酸化炭素の分圧は、有利には、15×105Paより小さい。一層特に、そ
れは10×105Paより小さい。
本発明の第2の重要な特徴は、反応するエステルの量が反応
混合物の20重量%未満に維持されることである。
エステルの上記の量は、好ましくは、反応混合物の10重量%を越えない。本
発明の特に有利な具体的態様において、反応するエステルの量は、反応混合物の
5重量%を越えない。
反応が連続的に行われるとき、上記に定められた特徴は、好ましくは、反応の
過程中一定に保たれる。一酸化炭素の分圧は、それが常に上記に定められた範囲
にある限り、反応中変化し得る、ということが留意されるべきである。
反応が回分的に行われるとき、反応するエステルの量は、エステルが反応にお
いて消費されるにつれて経時的に減少するけれども、指摘された値未満に保たれ
る。一酸化炭素の分圧に関して、それは、指摘された範囲の値内にある限り、一
定に保たれても保たれなくてもよい。
記載されたきた二つの特徴は、改善された生産性を有する方法を得るために必
須である。
本発明の異性化方法は、水の存在下で行われる。一層特に、水の量は、反応混
合物の重量として表して0(排除される)〜5%の範囲にある。有利には、該量
は、0(排除される)〜2重量%の範囲にある。
水は本方法において重要な役割を演じる、ということが留意されるべきである
。それは、特に混合物の部分気化帯域(フラッシュ)(下記に述べられる)にお
いて、触媒を溶解状態に保つよう助ける。それはまた、無水条件下で行われる方
法において起こると知られている二次反応を制限し得る。
更に、反応中維持されるハロゲン化促進剤の量は、一層特に、反応混合物の0
.1重量%ないし20重量%の範囲にある。ハロゲン化促進剤の量は、好ましく
は、反応混合物の1重量%ないし15重量%の範囲にある。
上記に指摘された促進剤の量は指摘として与えられている、ということが留意
されるべきである。当業者は、この化合物(触媒の活性および安定性に有益な効
果を有する)の最大効率と該化合物を本方法に再循環するコストに関連した経済
的考慮事項の間の最適妥協を解決するのは自由である。
100%への補足が、反応用の溶媒により構成される。一層特に、溶媒は、脂
肪酸および/または生成された酸、随意に生成されたエステル、およびギ酸から
なり得る。
本発明の特定の変型において、反応媒質中に存在するギ酸の量は、反応混合物
の15重量%未満に保たれる。ギ酸の量は、
好ましくは反応混合物の12重量%未満、一層特に10重量%未満に保たれる。
更に、本発明の有利な実施態様を用いると、反応混合物中に存在する遊離カル
ボン酸の量は、該混合物の25重量%を越え、反応混合物の構成成分の総量が反
応混合物の100重量%となる。一層特に、遊離カルボン酸の量は、反応混合物
の30重量%を越え、好ましくは反応混合物の40重量%を越える。
共溶媒が存在する場合、反応において生成されたエステルまたは反応中に生成
された酸のエステルの重量による量は、好ましくは、生成された酸の量より少な
いかまたは等しい。
モル比HCOOH/R1COOR’1は反応条件下で1以外であり得る、即ち、
この値より大きいかまたは小さくあり得る、ということが留意されるべきである
。反応は、無論、1のモル比でもって行われ得る。
一般に、反応媒質中のイリジウムの総濃度は、0.1〜100mmol/lの
範囲、好ましくは1〜25mmol/lの範囲にある。
本発明の異性化反応は、好ましくは、2000ppm未満のの腐食金属の存在
下で行われる。腐食金属は、特に、鉄、ニッ
ケル、クロムおよびモリブデンである。反応混合物中の腐食金属の量は、選択性
沈殿、液体抽出またはイオン交換樹脂上の通過のような、当業者に知られている
方法を用いて維持される。
反応は、一般に、150℃ないし250℃の範囲の温度にて行われる。一層特
に、反応温度は、175℃ないし210℃の範囲にある。それは、好ましくは、
175℃ないし200℃の範囲にある。
反応が行われる総圧力は、一般に、大気圧より高い。一層特に、それは、10
0×105Paより小さく、好ましくは50×105Paより小さいかまたは等し
い。圧力は、パスカル絶対圧にて表され、そして熱時に、即ち反応温度において
測定される。
反応は、腐食性媒質に抵抗性である装置にて行われる。かくして、ジルコニウ
ムまたはCもしくはBタイプのハステロイ(Hastelloy)(登録商標)
合金が、反応の実施条件について特に適している。
開始中、様々な成分が、反応混合物の適正な均質性を確保するための撹拌手段
を備えている適当な反応器中に導入される。反応器が反応混合物用の機械式撹拌
手段を含む(好ましいの
で)場合、かかる手段なしで操作することは排除されず、何故なら該混合物は該
