本発明は、プログラムされた細胞死の制御に関する分子生物学の分野である。
背景技術の説明
プログラムされた細胞死
アポトーシスは、プログラムされた細胞死または調節された細胞死とも呼ばれ
、生物体が不要な細胞を排除するプロセスである。このような細胞死は、組織ホ
メオスタシスや加齢と同じく動物発生の正常な様相として起こる(Glucksmann,A
.,Biol.Rev.Cambridge Philos.Soc.26-:59-86(1950);Ellis et al.,Dev.
112:591-603(1991);Vaux et al.,Cell 76:777-779(1994))。プログラムされた
細胞死はまた、細胞数を調節し、形態発生を促進し、有害あるいは異常細胞を除
去し、そして既にこれらの機能を実行している細胞を排除する働きもできる。更
に、プログラムされた細胞死は、低酸素症または虚血などの様々な生理学的スト
レスに応答して起こると考えられている。アポトーシスの形態学的特徴には、原
形質膜の水庖形成、核液および細胞質の凝縮およびヌクレオソーム間の間隔
(inter-nucleosomal intervals)での染色体DNAの劣化がある(Wyllie,A.H.,

アポトーシスは、細胞自殺の内因性機構を介して成され(Wyllie,A.H.,in
Cell Death in Bio1ogy and Pathology,Bowen and Lockshin,eds.,Chapman
and Hall(1981),pp.9-34)、内部または外部シグナルのいずれかの結果により
、細胞がその内部にコードされた自殺プログラムを活性化すると起こる。自殺プ
ログラムは、慎重に調節される遺伝子プログラムの活性化により遂行される
(Wyllie,A.H.,et al.,Int.Rev.Cyt.68:251(1980);Ellis,R.E.,et
al.,Ann.Rev.Cell Bio.7:663(1991))。細胞死はRNAまたはタンパク質合
成の阻害により妨害できることから、多くの場合で遺伝子発現が要求されるよう
である(Coehn et al.,J.Immunol.32:38-42(1984);Stanisic et al.,
Invest.Urol.16:19-22(1978);Martin et al.,J.Cell Bio1.106:829-844
(1988))。プログラムされた細胞死の遺伝子的経路は、最初、線虫C.elegansにお

インターロイキン−1β変換酵素
ced-3遺伝子産物の哺乳動物相同物は、インターロイキン−1β(IL−1β)
の活性化を担うシステインプロテアーゼであるインターロイキン−1β変換酵素
(ICE)である(Thornberry,N.A.,et al.,Nature 356:768(1992);
Yuan,J.,et al.,Cell 75:641(1993);Miura,M.,et al.,Cell 75:653
(1993))。このIce遺伝子は、遺伝子ファミリーの一員である。哺乳動物ICE/C
ed-3ファミリーは、現在、少なくとも6種:ICE、ICH−1/NEDD2、
CPP32/Yama/Apopain、TX/ICEreIII/ICH−2、ICEreIIIIおよ
びMCH2を含む(Yuan et al.,Cell 75:641-652(1993);Wang et al.,Cell

インターロイキン−1β変換酵素(ICE)は、不活性な31KDプロインター
ロイキン−1βをAsp116−Ala117で切断し、カルボキシ末端153アミノ酸ペ
プチドを放出して、成熟17.5kDインターロイキン−1β(IL−1β)を
生成する基質特異的システインプロテアーゼである(Kostura et al.,Proc.
Natl.Acad.Sci.,USA 86:5227-5231(1989);Black et al.,FEBS Lett.
247:386-390(1989);Cerretti et al.,Science 256:97-100(1992);
Thornberry et al.,Nature 356:768-774(1992))。これは、その活性部位システ
イン残基がICE−媒介アポトーシスにとって不可欠であるプロテアーゼファミ

ICEの特異的阻害因子である牛痘ウイルスのcrmA遺伝子産物は、IL−1
βのタンパク質分解活性化を妨害し(Ray et al.,Cell 69:597-604(1992))、宿
主の炎症応答も阻害する(Ray et al.,Cell 69:597-604(1992))。crmA遺伝子の
欠失した牛痘ウイルスは、ヒヨコ胎芽の炎症応答を抑制できず、ウイルス感染細
胞数の低下をもたらし、宿主へ与える損傷も少ない(Palumbo et al.,
Virology 171:262-273(1989))。この観察結果は、炎症性応答を引き起こすIC
Eの重要性を示している。
ICEの過剰発現がアポトーシスを誘導し、細胞死の際に成熟IL−1βが放
出されることも示されている(Miura,M.,et al.,Cell 75:653(1993);Miura,
M.,et al.,Proc.Natl.Acad.Sci.U.S.A.92:8318-8322,(1995))。セルピ
ンファミリーの一員であり、ICEの阻害因子である牛痘ウイルス遺伝子産物C
rmAもまたアポトーシスを妨害する(Miura,M.,et al.,Cell 75:653(1993);

細胞死におけるICEファミリーの重大な役割は、十分に認められているが、
アポトーシスにおける成熟IL−1βの機能は、論争中である。IL−1βは、
幾つかの系ではアポトーシスを誘導し(Onozaki et al.,Immun.135:3962-3968
(1985);Ankarcrona et al.,Exp.Cell Res.213:172-177(1994);Fratelli,
M.,et al.,Blood 85:3532-3637(1995))、その他では妨害する(Be1izario &
Dinarello,Cancer Res.51:2379-2385(1991);Strijbos & Rothwell,J.
Neurosci.15:3468-3474(1985))ことが示されている。成熟IL−1βは、TN
F−α処理アポトーシス繊維芽細胞の培地にて検出されただけでなく、Shjgella
flexneri感染の後にアポトーシスを受けるマクロファージの培地でも検出され
た(Zychlinsky,A.,et al.,J.Clin.Invest.94:1328(1994))。アポトーシス
中の成熟IL−1β放出の検出は、ICE欠損マウスで示されたようにインビボ
ICEがproIL−1βのプロセシングを担う主たる(そうでなければ唯一の)プ
ロテアーゼであるから、細胞死においてICE自身が活性化されていることの強
力な証拠を提供するものである(Li,P.,et al.,Cell 80:401
(1995);Kuida,K.,et al.,Science 267:2000(1995))。
腫瘍壊死因子
腫瘍壊死因子−α(TNF−α)は、多能性殺腫瘍性サイトカインである
(Tracey,K.J.et al.,Ann.Rev.Cell.Biol.9:317-343(1993))。TNF−
αのすばらしい機能の1つは、形質転換細胞のアポトーシスを誘導することであ
る。非形質転換細胞では、TNF−αは代謝阻害因子の存在下でもアポトーシス
を誘導できる(Tracey,K.J.et al.,Ann.Rev.Cell.Biol.9:317-343
(1993))。TNF−αにより誘導されるアポトーシスは、bc1-2によっても抑制さ
れる。
最も詳細にわたって研究されたTNF−α機能の1つは、広範囲の腫瘍細胞系
に対するインビトロでのその細胞毒性である(Laster,S.M.et al.,J.
Immunol.141:2629-2634(1988))。しかしながら、TNFによって誘導される細
胞死の機構は、あまり知られていない。ヒーラー(HeLa)細胞は、細胞死シグナ
ル形成を担うと考えられているp55TNF受容体を優先的に発現する
(Englemann,H.et al.,J.Biol.Chem.265:14497-14504(1990))。更に、ヒー
ラー細胞は、代謝阻害因子シクロヘキシミド(CHX)の存在下でTNF−αに
より容易に殺される。TNF−α/CHXにより誘導される細胞死は、DNA断
片化および細胞溶解を示し、これは、アポトーシスの典型的な特徴である
(White,E.et al.,Mol.Cell.Biol.12:2570-2580(1992))。アデノウイルス
EIB 19Kタンパク質の発現は、機能的にbcl-2と似ており、ヒーラー細胞
においてTNFにより誘導されるアポトーシスを阻害する(White,E.et al.,Mo
l.Cell.Biol.12:2570-2580(1992))。
発明の概要
この度、IL−1β受容体アンタゴニスト(IL−1Ra)は栄養因子欠乏や
低酸素症により誘導されるアポトーシスを阻害することが分かった。更に、成熟
IL−1β自身は、CrmA感受性遺伝子活性とは無関係の経路により細胞死を誘
導し、アポトーシスにおいてICEおよびICH−1Lと協同する。このように
、本発明は、あらゆるアポトーシス誘導遺伝子の最初の基質としてproIL−1
βを同定するものである。
本発明は、第1に、mIL−β受容体結合をブロックする工程を含む、プログ
ラムされた細胞死の妨害方法に関する。好ましくは、mIL−β受容体結合はI
L−IRAでブロックする。
本発明は、更に、感染細胞におけるアポトーシスを刺激することを含む、発癌
性形質転換の阻害方法に関する。好ましくは、アポトーシスはIL−1βおよび
/またはTNF−αで刺激する。
本発明は、更に、ICE経路およびmIL−1β産生を活性化することを含む
、アポトーシスの調整方法に関する。
本発明は、更に、IL−1がその受容体へ結合する前に細胞をプライミングす
ることを含む、アポトーシスの調整方法に関する。細胞のプライミングには、と
りわけ、栄養因子欠乏、低酸素症、G1/S期停止の使用があり得る。その後、
IL−1β処理する。
本発明は、更に、IL−1受容体ブロッカーを使用する低酸素症誘導細胞死の
阻害方法に関する。好ましくは、IL−1受容体ブロッカーは、IL−1Ra、
抗IL−1ポリクローナル中和抗体および抗IL−1 I型受容体中和モノクロ
ーナル抗体からなる群から選択される。
本発明は、更に、IL−1Raの使用を含む、ICH−1Lから生じる細胞死
の妨害方法に関する。
使用法は提供されている。これらには、とりわけ、ヒトを含む動物の特定の身
体器官の腫瘍を含む様々な病状の処理において細胞死を増大または低下するいず
れかの方法がある。更に、発癌性細胞形質転換を阻害するためや、様々な器官に
おいて低酸素症または虚血に付随するアポトーシス関連の合併症を目的として、
あるいはアポトーシスに影響を及ぼす作用物質をスクリーニングするために本発
明を使用することができる。

詳細な説明
下記の説明では、多くの用語を広義に使用している。明細書のより明瞭かつ矛
盾のない理解を提供するために、下記説明を与える。
定義
Ice、ICEまたはIchなどのイタリック体語は、遺伝子を表すのに対し、“
ICE、 IchまたはICH”は対応する遺伝子によりコードされる遺伝子産物
を表す。
アポトーシスは、生物体が不要な細胞を排除するプロセスを表すと理解すべき
である。このプロセスは、細胞プログラムにより慎重に調節される。アポトーシ
スにより、正常な発生、加齢、組織ホメオスタシスの際に、または低酸素症また
は栄養因子欠乏などの外部ストレスの負荷の後に、細胞を排除できる。
低酸素症は、細胞が利用できる酸素濃度が正常レベルよりも低下する症状を表
すと理解すべきである。最も極度の低酸素症は、殆ど全部の酸素が欠乏する(無
酸素症と呼ぶ)。
ICE経路は、インターロイキン変換酵素がproILβをIL−βに変換し、
最終的にプログラムされた細胞死をもたらす経路を表すと理解すべきである。
IL−1媒介シグナル伝達をブロックするとは、IL−1受容体でIL−1の
作用をブロックするあらゆる化合物または化学を使用することを表すと理解すべ
きである。シグナル伝達は、例えば、抗IL−1ポリクローナル中和抗体または
抗IL−1 1型受容体中和モノクローナル抗体を含む免疫グロブリン(モノク
ローナルまたはポリクローナル抗体またはこのような抗体の活性フラグメントな
ど)によりブロックできる。あるいは、シグナル伝達は、例えば、IL−1受容
体に結合する天然サイトカインであるIL−1Raを含む非免疫グロブリン化合
物(ポリペプチド、有機化合物等など)によりブロックできる。あるいは、シグナ
ル伝達は、IL−1βの競合または非競合阻害因子によりブロックできる。
栄養因子欠乏は、細胞生存に欠かせない因子(例えば、血清またはNGF)の
除去と理解すべきである。このような因子がなければアポトーシス経路は活性化
される。
G1/S期停止は、細胞に起こる事象であり、細胞周期のG1期からS期へ移行
できないようにすると理解すべきである。G1期からS期への移行は、細胞周期
の最も重要な工程と考えられている(Chiarugi et al.,Cell.Mol.Biol.Res.4
0:603-612,1994)。
アポトーシスを調整するとは、正または負方向のいずれかに細胞死のレベルを
変えるあらゆる行為であると理解すべきである。このような変化を測定する方法
は、当業者には容易に知られているが、とりわけ、トリパンブルー排除、クロミ
ウム放出、原形質膜の水庖形成、核液および細胞質の凝縮およびヌクレオソーム
間の間隔での染色体DNAの劣化を含む細胞形態の特定変化を含み得る。別の方
法には、MTT(3−[4,5−D,メチルーチアゾル−イル]−2,5−ジフェニル
テトラゾリウムブロミド;チアゾリルブルー)アッセイなどの代謝アッセイまた
はFACS分析による生存能測定がある。
細胞をプライミングするとは、IL−1βが細胞死プログラムを活性化するの
に必要な栄養因子欠乏、低酸素症またはG1/S期停止などを細胞が受ける事象ま
たは処理であると理解すべきである。インビボでは、これに、細胞を“病気”、
例えば病理学的状態にして、生物体から容易に除去されるようにするプロセスを
含めてもよい。
Ich−1LおよびIceは、細胞死遺伝子であると理解すべきである。Ich−1L
は、図7に示す配列(配列番号1および配列番号2)を有する。Ich−1Lは、Ic
h−1遺伝子のフラグメントである。このIch−1遺伝子は、とりわけ、nedd2
を含むその他の細胞死遺伝子と相同である。Ich−1は細胞死遺伝子のQACR
G配列特性を含有する。ヒトICEの配列は、Thornsberr et al.,Nature 356:
768-774,1992にみることができる。
自然に起こる細胞死は、細胞数を調節し、形態発生を促進し、有害あるいは異
常細胞を除去し、そして既にこれらの機能を実行している細胞を排除する働きを
する。更に、プログラムされた細胞死は、低酸素症または虚血などの生理学的ス
トレスに応答して起こると考えられている。
細胞死が急性的および慢性的に調節されないと、主要な多くのヒト疾患を導く
と考えられている(Barr et al.,Biotech.12:487-493,1995)。これらの疾患に
は、限定ではないが、悪性および前悪性症状、神経学的疾患、心臓病、免疫系疾
患、胃腸疾患、腎臓病および加齢がある。
悪性および前悪性症状には、固腫、B細胞リンパ腫、慢性リンパ球白血病、前
立腺肥大、新生物発生前肝臓病巣および化学療法に対する耐性が含まれ得る。神
経学的疾患には、発作、アルツハイマー病、プリオン関連疾患および毛細管拡張
性運動失調が含まれ得る。心臓病には、虚血性心臓損傷および化学療法誘導心筋
停止が含まれ得る。免疫系疾患には、AIDS、I型糖尿病、紅斑性狼瘉、シェ
ーグレン症候群および糸球体腎炎が含まれ得る。胃腸疾患には、赤痢、炎症性腸
疾患および放射線およびHIV誘導下痢が含まれ得る。腎臓病には、多嚢胞性腎
臓疾患および貧血/赤血球生成が含まれ得る。調節されないアポトーシスを伴う
これらの病理学的症状に関する具体的な文献は、Barr et al.,前掲−表Iにみる
ことができる。
ICE経路に関与する遺伝子や基質を知ることにより、細胞死に介入し、アポ
トーシスを変化させる手段が導かれる。このような知識はまた、アポトーシスプ
ロセスに影響し得る作用物質のアッセイの開発も導くことができる。介入には、
とりわけ、遺伝子産物の活性に影響を与える作用物質(例えば、受容体をブロッ
クする作用物質)、アンチセンスオリゴヌクレオチド法などの遺伝子指向法を用
いる遺伝子産物の調整、転写調節および遺伝子治療があり得る(Karp et al.,
Cancer Res.54:653-665(1994))。従って、アポトーシスは、治療的介入に対し
て敏感に反応すべきである。これに関し、我々は、プログラムされた細胞死の速
度を増加させたいか、低下させたいかに合わせて、このプロセスを刺激すること
も阻害することもできる。
ICEファミリーによるタンパク質分解性切断は、幾つかの方法でアポトーシ
スを導くこともあり得る。1つの可能性は、多数のタンパク質の切断が細胞機構
を破壊することである。しかしながら、細胞がアポトーシスを受けるとき、殆ど
のタンパク質が無傷のままであるようなので、これは恐らく起こらない
(Lazebnik et al.,Nature 371:346-347(1994))。第2の可能性は、極めて重要
な1つの基質の切断により、細胞死が導かれることである。これもまた、proI
L−1βリボースポリメラーゼ(PARP)、U1−70kDリボ核タンパク質お
よび核ラミンを含む多くのタンパク質がアポトーシス中に切断されるので、起こ
りそうにない(Miura,et al.,Proc.Natl.Acad.Sci.92:8318-8322
(1995);Lazebnik et al.,Nature 371:346-347(1994);Casciola-Rosen et
al.,J.Biol.Chem.269:30757-30760(1994);Lazebnik,Y.A.,et al.,
Proc.Natl.Acad.Sci.92:9042-9046(1995)。これらのタンパク質の切断産物
が、細胞死経路の下流事象を媒介するかどうか、またはこれらが単にアポトーシ
スの最終結果であるのかどうかは、(proIL−1β以外は)明らかでない。第3
の可能性は、ICE経路の活性化であり、そのため、ICEファミリーは、数種
の基質の切断をもたらすこともあり、幾つかは活性化され(細胞死を媒介し)、そ
の他は破壊される(細胞生存に欠かせない)。この経路の活性化は、栄養因子欠乏
、低酸素症、G1/S停止またはTNF−α処理などの事象に起因して起こり得る
。明細書の実施例で得られた結果は、最新の仮説に賛同するものであり、なぜな
らば、このデータは、体内で産生させた成熟IL−1βが細胞死に直接関与する
こと、および、その産物がアポトーシスカスケードの媒介において直接的な役割
を果たす、アポトーシス誘導遺伝子の最初に同定された基質であることを示して
いるからである。しかしながら、この提唱された機構は、いかにしても本発明の
請求の範囲を、このような機構による操作に対して限定するとみなすべきでない
。
更に、シグナル伝達機構の多くは、IL−1βの生物学的作用を媒介する。こ
れらの二次メッセンジャーの幾つかは、アポトーシスに関係しており、ICE活
性化に続き、内因性成熟IL−1β受容体結合後の細胞死を媒介するらしい。そ
のため、受容体結合をブロックすることにより、アポトーシスは調整される。I
L−1βは、EL4胸腺腫細胞におけるセラミド産生を誘導する(Mathias,S.,
et al.,Science 259:519-522(1993))。IL−1βもまた、膵臓R1m5F細胞
において一酸化窒素産生を誘導するその能力に依存するある経路を介してアポト
ーシスを誘導する(Ankarcrona e tal.,Cell Res.213:172-177(1994))。セラミ
ドおよび一酸化窒素はいずれも、アポトーシスの直接的なメディエーターとして
強力な候補である(Ankarcrona et al.,Cell Res.213:172-177(1994);
Haimovitz-Friedman,A.,et al.,J.Exp.Med.180:525-535(1994))。最近の
報告では、PC12細胞のNGF欠乏が、アポトーシスを誘導し、JNKおよび
p38MAPキナーゼの実質的な活性化を導くことが示された(Xia et al.,
Science 270:1326-1331(1995))。IL−1βは、JNK−p38シグナル形成経
路を活性化することが示されており、NGF撤収(withdrawal)により、後にJN
K−p38経路と細胞死を活性化するIL−1βの分泌が誘導できる
(Raingeaud,J.,et al.,J.Biol.Chem.270:7420-7426(1995))。
ここまで、本発明を総括的に説明してきたが、例示目的で与えた、特記しない
限り本発明の限定を意図するものではない、下記実施例を参照すれば、本発明は
より容易に理解されるであろう。
実施例
L929およびヒーラー細胞中の一次脊髄神経節(DRG)ニューロンの栄養
因子欠乏、低酸素症またはTNF−αにより誘導されるアポトーシスにおける分
泌成熟IL−1βの果たす役割を調べた。ICEおよびICH−1Lにより誘導
されるアポトーシスにおけるproIL−1βに対する要件もまた評価した。その
結果は、体内で産生された成熟IL−1βがこれらのアポトーシスモデルにおい
て肝要な役割を果たすことを示しており、ICEがproIL−1βをプロセスす
る主要な(さもなければ唯一の)酵素であるから、これはアポトーシスにおけるI
CEの役割に対する更なる証拠を提供するものである。
実施例1
低酸素症の作用
BCL−2(B細胞リンパ腫−2遺伝子がコードするタンパク質)とp53は、
低酸素症媒介アポトーシスに関係している(Shimizu,S.,et al.,Nature 374:
811-813(1995);Jacobson & Raff,Nature 374:814-816(1995);Graeber,T.
G.,et al.,Nature 379:88-91(1996))。ICEファミリーが低酸素症誘導アポ
トーシスに関与するかどうかを調べるために、CrmAがこのプロセスを阻害でき
るかどうかを試験した。
低酸素症誘導アポトーシスは、下記のようにして研究された。ヒーラーおよび
ヒーラー/CrmA細胞(Miura,M.et al.,Proc.Natl.Acad.Sci.U.S.A.92:
8318-8322,1995)を35mm皿にDMEM/10%FCS中6×104/皿の密度で
接種し、一晩増殖させた。次いで、培地を変え、因子を加えた(IL−1Ra(R&
D,Minneapolis,MN)、IL−1抗体(Calbiochem,San Diego,CA)またはIL
−1 1型受容体抗体(R & D,Minneapolis,MN))。皿をBBL GasPack Plus(B
ecton-Dickenson,USA)付き嫌気性チャンバーに入れ、90分以内に酸素濃度を
100p.p.m.以下に下げた。16時間後、細胞をチャンバーから除き、直ちにト
リプシン処理し、トリパン・ブルー排除により生存能についてスコアした。37
℃で2時間加えたIL−1Raによる125I IL−1βの阻害。培地にBSA(
1mg/ml)を添加後、細胞を4℃で15分間インキュベーションし、次いで、125
IL−1β(100ng/ml)を4℃で1時間加えた。125IL−1β結合の検出用に
、細胞を50mMグリシン−HCl、pH2.6で1分間処理し、γ−カウンティ
ングにより定量した。
低酸素症条件下で16時間培養したヒーラー細胞の生存率は、CrmAを安定に
発現するヒーラー細胞(ヒーラー/CrmA)の生存率69.0%と比べて10.1%
であった(図1a)。従って、ICEファミリーのCrmA阻害可能構成員は、低酸
素症誘導アポトーシスにおいて重要な役割を果たす。体内で産生された成熟IL
−1βが低酸素症誘導細胞死において役割を果たすかどうかを目的として、IL
−1がその受容体に結合するのを妨げる幾つかの方法を採用した。IL−1Ra
(IL−1受容体に結合し、IL−1媒介シグナル伝達をブロックする天然のサ
イトカイン)(Dripps,et al.,J.Biol.Chem.266:10331-10336(1991);
Granowitz,et al.,J.Biol.Chem.266:14147-14150(1991))、抗IL−1ポリクロ
ーナル中和抗体および抗IL−1受容体中和モノクローナル抗体(1型受容体は
IL−1シグナル伝達を媒介する)を使用した。これらの試薬はそれぞれ
低酸素症誘導細胞死を阻害し、このことは、低酸素症がICE様CrmA阻害可能
経路を活性化すること、および体内で産生された成熟IL−1βがIL−1 1
型受容体に結合することにより低酸素症誘導細胞死においてある役割を果たすこ
とを示している(図1a)。IL−1Raが現実に125IL−1β結合をブロック
することも評価および確認された(図1b)(Dripps,e tal.,J.Biol.Chem.
266:10331-10336(1991);Granowitz,et al.,J.Biol.Chem.266:14147−
14150(1991))。
実施例2
脊髄神経節におけるアポトーシスおよび栄養因子欠乏
次に、内因性IL−βのアポトーシスにおける役割を調べた。一次脊髄神経節
(DRG)ニューロンは、NGF撤収の際、培養中アポトーシスを受ける(Davies,
A.M.,Development 100:1019(1987))。栄養因子欠乏によって誘導される鶏のD
RGニューロン死は、CrmAにより阻害され、ICEファミリーの関与が示唆さ
れることは既に示した(Gagliardi,V.et al.,Science 283:826-828
(1993))。体内で産生される成熟IL−1β、培養中ではニューロンによって産
生される(Freidin et al.,Proc.Natl.Acad.Sci.U.S.A.89:10440-10443
(1994))、が栄養因子撤収媒介DRGニューロンアポトーシスにおいて何らかの
役割を果たすかどうかを試験するために、ヒトIL−1受容体アンタゴニスト(
IL−1Ra)を使用した。IL−1RaはI型およびII型IL−1受容体に結
合し、IL−1シグナルをブロックする(Dripps,D.J.,et al.,J.Biol.
Chem.266:10331(1991);Granowitz,E.V.,et al.,J.Biol.Chem.266:
14147(1991))。
ニューロン栄養因子欠乏は、下記のようにアッセイした。出生後1日のマウス
DRGニューロンを単離し、37℃で1時間トリプシンで解離させ、8チャンバ
ーポリリジン/ラミニン(Sigma,St.Louis,MO)コーティングスライドにのせる
。ウェルにおよそ1000ニューロン/ウェル(8ウェル/マウス)で接種する。
ニューロンを、20%FCS(Biowhittaker,Walkesvill,MD)、NGF(200
ng/ml)(Sigma,St.Louis,MO)、BDNF(100ng/ml)(Preprotech,Rocky
Hill,NJ)、グリタミン(2mM)およびペニシリン/ストレプトマイシンを補足し
たHam's栄養素F−12中で培養した。培地は、飽和濃度のマウスNGFモノク
ローナル抗体(100ng/ml)(Boehringer Mannheim,Indianapolis,IN)およびIL
−IRA(特記しない限り100ng/ml)の存在下、栄養因子含有培地(TF(+)=
20%FCSおよびNGF(200ng/ml))か栄養因子欠乏培地(TF(−)=血清
およびNGFなしの培地)のいずれかと毎日取り替えた。培地交換後24および
48時間で、位相差顕微鏡により健常ニューロンをカウントした。
IL−IRa(100ng/ml)は、栄養因子撤収誘導アポトーシスを24および
48時間でそれぞれ69.2%および37.8%まで阻害した(図2)。IL−IR
aによるニューロンアポトーシスの阻害は、用量依存性であった(濃度40ng/ml
、24時間で43.5%)。これらの結果は、体内で産生される成熟IL−1βが
栄養因子撤収後に起こるニューロンアポトーシスにおいて役割を果たすことを示
している。しかしながら、ニューロンが培養中に成熟IL−1βがを産生するこ
とが示されているとしても、この混合細胞集団における成熟IL−1βは非ニュ
ーロン起源のものでなとは言い切れない(Freidin,M.,et al.,Proc.Natl.
Acad.Sci.U.S.A.89:10440(1992))。この理由で、幾つかの細胞ライン系を試
験し、IL−1Raが同様の抗アポトーシス特性を持つかどうかを測定した。
実施例3
TNF−αおよびアポトーシス
TNF−αは、CrmA阻害可能経路を経てアポトーシスを誘導する
(Gagliardini,V.,et al.,Science 263:826(1993);Boudreau,N.,et al.,
Science 267:891(1995);Enari,M.,et al.,Nature 375:78(1995);Los,
M.et al.,Nature 375:81(1995);Tewqry,M.,et al.,J.Biol.Chem.
270:3255(1955);Hsu,H.,et al.,Cell 81:495(1995))。更に、成熟IL
−1βは、アポトーシスを受けているTNF−α処理細胞により分泌されること
が示されており、これは、このプロセス中にICEが活性化されることを示唆し
ている(Miura,M.,et al.,Proc.Natl.Acad.Sci.U.S.A.92:8318-8322,
1995)。L929およびヒーラー細胞のTNF−α誘導アポトーシスに役立つ分
泌成熟IL−1βの役割を試験した。
ヒーラー、ヒーラー/CrmAおよびL929細胞を24ウェルプレートに接種(
2×104)し、10%FCSを含むDMEM中で一晩増殖させた。12時間後、
細胞を血清なしのDMEMで3回洗浄し、ヒドロキシ尿素(HU)(2.5m
M)(Sigma,St.Louis,MO)をヒーラーおよびヒーラー/CrmA細胞に加えた
(Meikrantz,W.,et al.,Proc.Natl.Acad.Sci.U.S.A.91:3754(1994))。5
時間後、IL−1Ra(40ng/ml)を適当なウェルに加え、1時間後、TNF−
αまたは成熟IL−1βのいずれかを加えた。24時間後、IL−1Raを適当
なウェルに再度加え、HUの初期添加後60時間でトリパン・ブルー排除により
細胞死を評価した。各条件は、二重に3回実施し、ウェル当り200細胞をカウ
ントした。写真用に細胞を2ウェルスライド上で増殖させ、核形態測定用に細胞
を4%パラホルムアルデヒドに固定し、ヘキスト(Hoechst)染料#33258(10μg/
ml)(Sigma,St.Louis,MO)と共にインキュベーションした。
IL−1Raは、TNF−α誘導死を64.9%まで防御し、これは、成熟I
L−1βの分泌および受容体結合がTNF−α誘導細胞死の肝要な成分であるこ
とを示唆している(図3a)。
更に、ヒドロキシ尿素(HU)処理したG1/S期停止ヒーラー細胞は、TNF−
αにより、プログラムされた細胞死を受けるように誘導される(Meikrantz,W.,e
t al.,Proc.Natl.Acad.Sci.U.S.A.91:3754(1994))。この条件下でIL−
1Raはまた、ヒーラー細胞死を56.0%まで阻害した(図3b)。TNF−α
およびシクロヘキサミドにより死滅するよう誘導されるヒーラー細胞はまた、I
L−1Ra並びに3種の異なる中和IL−1抗体(データ表示せず)により、防御
された。ヒーラー/CrmA細胞は、59.5%までTNF−α誘導アポトーシスか
ら防御され、これは、ICE様活性がこのサイトカインにより媒介される細胞死
シグナル形成経路に関与することを示唆している(図3b)。
実施例4
アポトーシスにおける成熟IL−1β
成熟IL−1β単独は、殆ど健常な増殖細胞(ヒーラーおよびL929を含む)
のアポトーシスを誘導しない。IL−1βがG1/S期停止細胞において細胞死を
誘導するかどうかを試験するために、HU処理ヒーラー細胞をこのサイトカイン
に暴露した。
外来成熟IL−1βで処理したG1/S期停止ヒーラー細胞は、用量依存的(1
00ng/mlで83.7%)に死滅し、これはIL−1Raの添加により阻害された(
図3c)。HU停止成熟IL−1βおよびTNF−α処理細胞は、細胞収縮、核
凝縮および断片化という典型的なアポトーシス変化を受けた(図3d−図3i)。
ヒーラー/CrmA細胞が成熟IL−1βから防御されないことは、それらがTN
F−α殺傷由来であるので、興味深く、これは、成熟IL−1βがICEに対し
て遠位のアポトーシスカスケードを誘導すること、およびHU処理細胞において
このサイトカインがICE依存経路を介して細胞死を引き起こすことを示唆して
いる(図3c)。これは、CrmAは現実にICE様機能をブロックすること、およ
び成熟IL−1βの産生および分泌は、アポトーシスTNF−α/ICEカスケ
ードの下流エフェクターであることを示している。しかしながら、成熟IL−1
β誘導アポトーシス対して過敏にさせる適当な細胞内環境を確立するためには、
ヒーラー細胞を(この場合、HU停止で)プライミングさせる必要がある。HU処
理は、アポトーシスカスケードの一部である細胞内シグナルと似ているようであ
る。
実施例5
proIL−1βプロセシングおよびアポトーシス
次に、proIL−1βプロセシングがICE媒介アポトーシスに必要であるか
どうかを直接調べた。このために、COS細胞を使用した。この細胞は、Iceお
よびIch−1L過剰発現により誘導される細胞死に対して耐性であるので、珍し
い(Wang,L.,et al.,Cell 78:739(1994))。
COS細胞を10%FCSを含むDMEM中、6ウェルプレートに入れた(2
×104)。12時間後、ウェルを血清および抗生物質なしの培地で洗浄し、Ice
−lacZ、Ich−1L−lacZ、βアクチン−lacZ(1μg)またはProIL−1
β(0.5μg)のいずれかで、3時間、リポフェクタミンを用いてトランスフ
ェクションした。Ich−1Lの配列は図7に示しており、ヒトICEおよびpro
IL−1βの配列はそれぞれ、Thornsberry et al.,Nature 356:768-774
(1992)およびJ.Immunol.137:3644-3648,1986にある。次いで、培地を除去し
、10%FCSを含むDMEMを加えた。次いで、適当なウェルにIL−1Ra
(40ng/ml)を加え、1時間後、IL−1β(100ng/ml)を加えた。トランスフ
ェクション後36時間、X−gal反応を実施し、丸い青色(死滅)細胞の割合をス
コアした(Miura,M.,et al.,Cell 75:653(1993))。
Rat−1細胞へのIceまたはIch−1Lのトランスフェクションは、24時間
以内に94.2%および92.1%の細胞死をそれぞれ誘導する(Wang,L.,et
al.,Cell 78:739(1994))。反対に、一過的にIce−lacZ、Ich−1L−l
acZまたはproIL−1β遺伝子を発現するCOS細胞は、それぞれ、9%、2
1%および6.3%死滅した。しかしながら、Ice−lacZおよびproIL−1β
またはIch−1L−lacZおよびproIL−1βを発現するCOS細胞は、それぞ
れ51.0%および57.3%死滅した。更に、Ice−lacZまたはIch−1L−
lacZトランスフェクション細胞を成熟IL−1βまたはTNF−αで処理する
と、効果的に細胞死を誘導した。Ice−lacZトランスフェクション細胞をIL
−1βで処理した結果は、図4に示す。外来成熟IL−1βおよびTNF−αは
、COS細胞におけるアポトーシスを誘導せず、これは、ICEおよびICH−
1Lが細胞死に必要なproIL−1β以外の基質を有すること、およびCOS細胞
では、ICE活性化の後、アポトーシスを誘導するにはIL−1βシグナル伝達
が必要であることを示している。IL−1Raは、細胞外TNF−αまたは成熟
IL−1βの存在下、Ice−lacZおよびproIL−1βまたはIch−1L−lac
ZおよびproIL−1βを発現するCOS細胞の死滅と、Ice−lacZまたはIc
h−1L−lacZの死滅を有意に阻害した。これは、ICEファミリー活性化後の
アポトーシスの誘導における成熟IL−1βの役割を示している。
IceおよびproIL−1βで同時トランスフェクションしたCOS細胞の(抗I
CEおよび抗IL−1抗体での)二重免疫蛍光染色は、ICEおよびproIL−1
βをいずれかのタンパク質単独ではなく両方発現する細胞のみが、アポトーシス
を受けることを示している(図5)。Iceでトランスフェクションした細胞の核は
、対照細胞の核よりも小さいことが一貫して注目された(図5c)。これらの細胞
は、その平坦な形態とプレートへの粘着性が示すとおり(図5d)生存状態であり
、これは、ICEはアポトーシスプロセスを始動するが、細胞死経路を完全に実
行するには更なる因子(即ち、成熟IL−1βまたはTNF−α)を必要とするこ
とを示唆している。
二重免疫蛍光染色法は、下記のとおりであった。COS細胞(1.5×104)を
ポリリジンコーティングした2チャンバースライドに置き、12時間後、上記の
ようにトランスフェクションさせた。細胞を36時間後、4%パラホルムアルデ
ヒド(15分)で固定し、PBS中1%熱不活性化ヤギ血清/2%BSAで(2時間
)ブロックし、ウサギポリクローナルIL−1(1:300)(Calbiochem)および
ハイブリドーマ上清マウスモノクローナルヒトICE抗体でインキュベーション
し、チャンバーをPBSで3回洗浄し、ヤギ抗マウスFITC−標識抗体、ヤギ
抗ウサギRITC標識抗体(1:200)(Cappel)およびヘキスト染料
#33258(10μg/ml)と共に45分間インキュベーションした。細胞をPBSで3
回濯いだ。スライドをaxioplan顕微鏡で調べ、40×対物レンズで写真を撮った
。
実施例6
低酸素症誘導アポトーシスの阻害
外来成熟IL−1βプレインキュベーションは、ICE活性化と成熟IL−1
β受容体結合がアポトーシスにとって重要である系において細胞死を阻害するか
どうかを測定した。
低酸素症は、実施例1に記載のようにして発症させた。細胞を低酸素状態チャ
ンバー(100p.p.m.の酸素濃度に到達するには90分必要である)に入れながら
、IL−1β(100ng/ml)を加えた。IL−1受容体結合アッセイ:ヒーラー
細胞(106)を10cm皿に接種し、一晩増殖させた。次いで、培地を4℃で一時
間、1mg/mlBSAおよび100ng/ml125I IL−1βを入れて交換した。冷
培地で2回洗浄後、細胞を37℃で0、30、60および120分間、新しい温
培地
と共にインキュベーションした。次いで、細胞を上記のようにグリシンで処理し
、放射能をスコアした。
外来IL−1βでプレインキュベーションしたヒーラー細胞は、顕著に低酸素
症誘導細胞死から防御された(生存率10.1%対58.7%)(図6a)。IL−1
βによるアポトーシスの阻害を説明するために、アポトーシス刺激にさらす前に
この系において外来IL−1βと共にインキュベーションするとIL−1受容体
をダウンレギュレーションするかどうかを調べた。実際、受容体結合アッセイは
、外来IL−1βがIL−1受容体を有意にダウンレギュレーションしたことを
示した(図6b)。IL−1受容体のダウンレギュレーションは、アポトーシス誘
導前に加えた場合の外来IL−1βの防御的役割を一部説明している。アポトー
シスに対するIL−1β受容体結合の作用は、ICEが活性である(細胞死を増
強する)かどうか、またはICEが不活性である(IL−1受容体を一部ダウンレ
ギュレーションすることにより胞死を阻害する)かどうかに左右される。
実施例に与えた結果により、proIL−1βは細胞死に直接関与するアポトー
シス誘導遺伝子の第1基質であり、そのプロセシング、分泌および細胞外受容体
結合はICEアポトーシスカスケードにおいて肝要な役割を果たすことが同定さ
れた。IL−1βは、共にアポトーシスに関与することが示されているセラミド
および/または一酸化窒素産生を誘導することにより細胞死を引き起こすと考え
られる(Mathias,S.,et al.,Science 259:519(1993);Haimovitz-Friedman,
et al.,J.Exp.Med.180:525(1994);Ankarcrona,M.,et al.,Exp.Cell
Res.213:172(1994))。これらの結果は、様々なアポトーシス刺激(栄養因子欠乏
、低酸素症およびTNF−α)がICE(またはその他のIL−1β変換酵素)を
活性化すること、および細胞死はICE活性をCrmAでブロックするか、IL−
1β受容体結合をIL−1Raでブロックするかのいずれかにより阻害できるこ
とを再確認するものである。
IL−1Raがアポトーシスを十分に阻害しなかったという事実は、恐らく次
の理由から起こる。ほんの少し(細胞当りおよそ5)のIL−1受容体の占有がI
L−1生物学的応答の完全な活性化に必要である(Dinarello,C.A.,FASEB J.
8:1314(1994))から、競合阻害因子であるIL−1Raは、恐らく、全てのIL
−1βをその受容体から十分にはずすのではなく、細胞の一部を単に保護するだ
けである。あるいは、ICE活性化後、成熟IL−1βは、セラミドおよび/ま
たは一酸化窒素の誘導を介して、細胞死経路を増強し、そしてアポトーシスの遅
延をもたらすこれらのシグナルを排除するという働きをするのかもしれない。更
に、外来成熟IL−1βで処理した殆どの細胞は死滅せず、このことは、更なる
基質のプロセシングを導くICEファミリー活性化が細胞死にとって必要条件で
あることを示唆している。明らかに、成熟IL−1βは、ICEファミリーを活
性化できず、これが、TNF−αとは違う特徴である。しかしながら、細胞を適
切にプライミングする条件下、成熟IL−1β単独は、G1/S期停止ヒーラー/
CrmA細胞において成熟IL−1βにより誘導されるアポトーシスにより示した
ようにICE活性の不在下でさえ、細胞死を誘導する。更に、ICH−1Lは、
成熟IL−1βまたはTNF−αにさらした際、COS細胞において活性化され
るようになると思われる。意外にも、ICH−1Lは、proIL−1βと同時発現
した場合、IL−1Raに感受性の細胞死を誘導し、このことは、ICH−1L
が、それ自身で、またはもう一つのICE様プロテアーゼの両方とも高濃度で存
在する場合は、もう一つのICE様プロテアーゼによるかのいずれかで、proI
L−1βをプロセスすることを示している。
アポトーシスにおけるICEの明確な役割を指摘する上記結果からみて、IC
Eノックアウトマウスが発生的に正常であることは、興味深い(Li,P.,et al.,
Cell 80:401-411(1995);Kuida,K.,et al.,Science 267:2000-2002
(1995))。これまで、このマウスで報告されたアポトーシスに対する唯一の耐性
は、抗Fas媒介胸腺細胞死の場合である(kuida,K.,et al.,Science 267:
2000-2002(1995))。しかしながら、ICE−ced-3ホモロジー数を常に増大して
いる唯一の一員のみをノックアウトすると、顕著なアポトーシス表現型を産生し
ないことは、細胞自殺などの重要かつ最終的なプロセスの過剰さを考慮すれば、
意外なことではない。
IL−1βはまた、インビボでウイルス感染細胞におけるアポトーシスの誘導
に関与し得る。幾つかのウイルスは、IL−1βおよび/またはTNF−α活性
のいずれかのサプレッサーを発現することが同定されている。牛痘CrmA遺伝子
以外の例は、ポックスウイルスによって発現されるTNF−α結合タンパク質で
ある(Smith,C.A.,et al.,Science 248:1019(1990))。ワクシニアおよび牛痘
ウイルスは、分泌されたIL−1β結合タンパク質を発現する(Spriggs,M.
K.,et al.,Cell 71:145(1992);Alcami & Smith,Cell 71:153(1992))。これら
のウイルスタンパク質は、免疫応答を下方調整することが示されており、これを
欠失するとウイルス毒性は低下する。免疫調節作用以外に、これらのモデュレー
ターは、IL−1βおよび/またはTNF−αシグナルを排除して、細胞死前に
ウイルスが複製のために細胞機構を使用できるようにすることにより、感染細胞
におけるアポトーシスを阻害できる。このことはまた、アポトーシスの阻害によ
るウイルス媒介発癌性形質転換にとって起こりうる機構を示唆している。このよ
うな機構を知ることにより、発癌性形質転換細胞を殺す方法を導くことができる
。
本結果の更なる関連性は、高レベルのIL−1βメッセージが大脳虚血のラッ
トモデルで検出されていることである(Lui,T.,et al.,Stroke 24:1746
(1993);Buttini,M.et al.,Molec.Brain.Res.23:126(1994))。別のラット
モデルでは、IL−1Raが虚血後50%まで脳梗塞サイズを小さくすることが
示された(Relton & Rothwell,Brain Res.Bull.29:243(1992))。更に、アルツ
