DETAILED DESCRIPTION OF THE INVENTION (9-Fluoro-3-methyl-10- (4-methyl-1-piperazinyl) -7-oxo-2,3-dihydro-7H-pyrido [3,2,1, IJ] Esters of (-1,3,4-benzoxadiazine-6-carboxylic acid) and their use as antibacterial agentsThe present invention relates to non-human animals, particularly horses, cows, pigs, sheep, or dogs and cats. A method for treating or preventing bacterial infections in domestic mammals and birds, such as companion animals, poultry including chicks. For the administration of antibacterial agents to livestock and poultry, the oral route, for example, by mixing with animal feed or drinking water, is often convenient. Therefore, it is important that such compounds be palatable to the target animal if therapeutic treatment is to be effective. EP-A-0 259 804 has the structural formula (A), that is, (9-fluoro-3-methyl-10- (4-methyl-1-piperazinyl) -7-oxo-2,3-dihydro-7H-pyrido [ 3,2,1, ij] -1,3,4-benzadiazine-6-carboxylic acid). This compound is stated to have antibacterial activity. A disadvantage of compounds of structural formula (A) is that they are unpalatable and therefore difficult to administer orally to animals. The present invention provides a method for treating or preventing a bacterial infection in a non-human animal, which comprises orally administering a compound of formula (I) in an antibacterial effective amount. (Wherein R 1 is a pharmaceutically acceptable ester group that is hydrolysable in the body.) Suitable ester groups that are pharmaceutically acceptable and hydrolyzed in the body are, for example, readily degradable in the body and Includes those that leave the compound acid or salt thereof. Suitable ester groups of this type include substructures (i), (ii), (iii), (iv). Wherein R a is hydrogen, C 1-6 alkyl, C 3-7 cycloalkyl, methyl or phenyl; R b is C 1-6 alkyl, C 1-6 alkoxy, C 3-7 cyclo Alkoxy, phenyl, benzyl, C 3-7 cycloalkyl, C 1-6 alkyl, C 3-7 cycloalkyl, 1-amino C 1-6 alkyl, or 1- (C 1-6 alkyl) amino C 1-6 Alkyl or R a and R b together form a 1,2-phenylene group, which may be optionally substituted by one or two methoxy; R e is a C 1-6 alkylene R d and R e independently represent C 1-6 alkyl; R f represents C 1-6 alkyl; R g represents a hydrogen atom or optionally halogen, it may indicate a phenyl group substituted by a group of up to 3 selected from C 1-6 alkyl or C 1-6 alkoxy; Q is oxygen or NH Examples of suitable ester groups that are hydrolyzed in the body are, for example, acetoxymethyl, pivaloyloxymethyl, α-acetoxyethyl, α-pivaloyloxyethyl, 1- (cyclohexylcarbonyloxy) prop- Acyloxyalkyl groups such as 1-yl and (1-aminoethyl) carbonyloxymethyl; alkoxycarbonyloxyalkyl groups such as ethoxycarbonyloxymethyl and α-ethoxycarbonyloxyethyl; cycloalkoxy such as cyclohexyloxycarbonyloxymethyl Carbonyloxyalkyl groups; dialkylaminoalkyl such as dimethylaminomethyl, dimethylaminoethyl, diethylaminomethyl or diethylaminoethyl, especially groups such as di-lower alkylaminoalkyl; phthalidyl or dimethoxyphthalidyl Lactone group such as; and a second antibiotic bound ester group, including. Further pharmaceutically compatible esters that are hydrolyzed in the body have the following structural formula: (In the formula, Rh is a hydrogen atom, C 1-6 alkyl or phenyl.) A particularly preferred ester group is pivaloyloxymethyl. A second aspect of the invention is the manufacture of a medicament for the oral administration of a compound of formula (I) for use in the treatment and prevention of bacterial infections in non-human animals. Preferably, the compounds of structural formula (I) are administered in the animal feed or drinking water. Accordingly, a further aspect of the invention provides a feed or drinking water mixed with a compound of formula (I), and a premix comprising a compound of formula (I) together with a veterinarily acceptable carrier. I do. Examples of suitable carriers include a mixture of a binder such as polyvinylpyrrolidone and a filler such as lactose, which can be granulated and mixed with the animal feed or sprinkled. For addition to drinking water, the active form is previously prepared as a concentrate with a liquid carrier such as gluconolactone. The compounds of the present invention may also be in the form of tablets containing excipients such as starch or lactose, capsules or eggs, alone or as a mixture with excipients, as elixirs or suspensions containing flavoring and coloring agents. It may be administered orally. Tablets or capsules for oral administration may be in unit dosage form, for example, with conjugates such as syrup, acacia, gelatin, sorbitol, tragacanth, or polyvinylpyrrolidone, or with lactose, sucrose, corn starch, calcium phosphate, sorbitol or glycine. Fillers such as, for example, tableting lubricants such as magnesium stearate, talc, polyethylene glycol or silica, leavening agents such as potato starch, and pharmaceutically acceptable agents such as sodium lauryl sulfate. And a general excipient such as a wetting agent. Tablets may be coated according to conventional methods well known in the pharmaceutical arts. Liquid products for oral use may be in the form of, for example, aqueous or oily suspensions, solutions, emulsions, syrups or elixirs, or they may be provided as a dry product and reconstituted with water or other suitable carrier before use. Is also good. Such liquid products include suspending agents such as sorbitol, methyl cellulose, glucose syrup, gelatin, hydroxyethyl cellulose, carboxymethyl cellulose, aluminum stearate gel or hydrogenated edible fats, for example. An emulsion-forming agent such as lecithin, sorbitan monooleate, or gum arabic; a non-aqueous carrier (including edible oils) such as, for example, almond oil, oily esters (e.g., glycerin), propylene glycol, or ethyl alcohol; It may contain conventional additives such as preservatives such as, for example, methyl or propyl p-hydroxybenzoic acid or sorbic acid, and if necessary, general flavors and coloring agents. The compound of structural formula (I) is typically present in an amount sufficient to provide 0.5 to 500 mg / kg, more specifically 1 to 300 mg / kg, per animal weight. Infections compatible with treatment include infections caused by any of the following microbial groups: Staphylococcus, Streptococcus, Aerococcus, Enterococcus, Micrococcus, Kactobacillus, Bifidobacterium, Clostridium, Eubacterium, Peptococcus, Peptostreptocoterium, Propioniobacter. , Enterobacter, Klebsiella, Proteus, Pseudomonas, Serratia, Salmonella, Shige lla, Vibrio, Aeromonas, Haemophilus, Neisseria, Acinetobacter, Alcaligenes, Bordetella, Bacteroids, Fusobacterium, Mycoplasma, and other microorganisms. Certain compounds of structural formula (I) are novel and form a further aspect of the present invention. One example of such a compound is cyclohexyloxycarbonyloxymethyl 9-fluoro-3-methyl-10- (4-methyl-1-piperazinyl) -7-oxo-2,3-dihydro-7H-pyrido [3,2 , 1-ij] -1,3,4-benzoxadiazine-6-carboxylic acid. Certain compounds of structural formula (I) can be prepared from compounds of structural formula (A) and derivatives thereof by procedures well known in the art. For example, a compound of structural formula (A) or an active derivative thereof can be reacted with a compound of structural formula (v), (vi), (vii), (viii), (ix). Wherein R a to R b are as defined above, and X is a leaving group such as a halogen (preferably chlorine) or a hydroxyl group. Reactions of compounds of structural formula (A) are typical Specifically, the reaction is performed in a non-aqueous solvent such as DMF, DMA, DMSO or acetone in the presence of an acid-binding substance such as an organic or inorganic base. Preferably, an inorganic base such as potassium carbonate or sodium carbonate is used. The temperature is typically on the order of between room temperature and reflux temperature, for example about 50 ° C. Alternatively, a suitable activated derivative of the compound of formula (A) can be used. Such derivatives can be prepared by reacting a compound of formula (A) with dicyclohexylcarbodiimide. The present invention will be described in more detail with reference to the following examples. Example 1 Pivaloyloxymethyl 9-fluoro-3-methyl-10- (4-methyl-1-piperazinyl) -7-oxo-2,3-dihydro-7H-pyrido [3,2,1-ij] -1,3,4 benzoxathiol Jia Jin 6 months Le Bon acid 9-fluoro-3-methyl-4- (4-methyl-1-piperazinyl) -7-oxo-2,3-dihydro -7H- Pyrido [3,2,1-ij] -1,3,4-benzadiazine-6-carboxylic acid (1.0 g, 2.78 mmol), chloromethyl pivalic acid (0.63 g, 4.17 mmol) and potassium carbonate (0.77 g, A solution of 5.56 mmol) in anhydrous DMF (20 ml) was stirred at 45 ° C. under argon gas for 16 hours. The reaction solution was poured into DCM (50 ml), and the organic solution was washed with water (30 ml) and dried over sodium sulfate. Removal of the solvent by vacuum concentration gave a white solid contaminated with DMF. The solid was purified by recrystallization from ethyl acetate to give a white fine crystalline solid of pivaloyloxymethyl ester (0.88 g, 67%); mp: 204-207 ° C, m / z (FAB) value : 477 (M + 1).
