La presente invenzione si riferisce a un nuovo metodo per prolungare la vitalità e/o migliorare la funzionalità di spermatozoi in vitro comprendente la preparazione di co-colture di spermatozoi e di cellule del Sertoli e a colture cellulari ottenute secondo il metodo dell’invenzione. STATO DELLA TECNICA ANTERIORE STATE OF THE PRIOR ART
L’infertilità à ̈ un problema che coinvolge un alto numero di coppie, solo negli Stati Uniti stimato oltre i 6 milioni. I problemi d’infertilità per oltre il 30% delle coppie à ̈ causato esclusivamente dall’infertilità maschile dovuta ad una funzionalità ridotta degli spermatozoi come ad esempio una riduzione dei parametri di motilità spermatica (astenozoospermia) o un basso numero di spermatozoi nell’eiaculato (oligozoospermia). Per poter risolvere i problemi d’infertilità sono state sviluppate diverse tecniche di procreazione medicalmente assistita quali ad esempio l’inseminazione intrauterina, la fertilizzazione in vitro, la fecondazione assistita e l’iniezione intracitoplasmatica dello spermatozoo. Tutte queste tecniche prevedono l’uso di spermatozoi in vitro. Infertility is a problem that affects a large number of couples, estimated at over 6 million in the United States alone. È noto che gli spermatozoi appena eiaculati e posti in coltura, seppur incubati in terreni specifici (come ad esempio il terreno BWW), perdono rapidamente e progressivamente le loro caratteristiche funzionali in termini di motilità , vitalità e funzione mitocondriale (Calamela et al., Andrologia 33, 79-86, 2001). Calamera et al. (Andrologia 33, 79-86, 2001), dimostrano, in maniera molto netta, che le colture di spermatozoi umani, come quelle allestite per le metodiche di procreazione medicalmente assistita, oltre le 47 ore subiscono una netta perdita di tutti i parametri funzionali quali motilità , vitalità , frammentazione del DNA e capacità di attuare la reazione acrosomiale influenzando negativamente l’esito della fecondazione dell’ovocita. Gli autori mostrano anche che tale perdita di vitalità e funzionalità non può essere in alcun modo rallentata tramite l’aggiunta in coltura di sistemi di “scavenger†dei radicali liberi d’ossigeno, come la catalasi. Successivamente Chi et al. (Human reproduction 23(5),1023-1028, 2008) sono riusciti a migliorare la “performance†funzionale di spermatozoi umani, tramite l’aggiunta al sistema, oltre alla catalasi, anche dell’EDTA, soltanto però non oltre 24 ore dall’eiaculazione. Allo stato delle attuali conoscenze scientifiche, non esiste alcuna possibilità di prolungare la vitalità di colture di spermatozoi oltre le 48 ore mantenendone inalterate le caratteristiche funzionali. La rapida e progressiva perdita delle caratteristiche di vitalità ed efficienza degli spermatozoi in vitro influenza negativamente l’esito delle tecniche di procreazione medicalmente assistita non consentendo un’accurata selezione degli spermatozoi più vitali o un eventuale pre-trattamento degli spermatozoi con sostanze che ne migliorino i parametri funzionali come la motilità . Era pertanto molto sentito nello stato della tecnica il bisogno di proporre nuovi metodi per prolungare e/o migliorare la vitalità e funzionalità di spermatozoi in vitro. Dopo un certo tempo d’incubazione le cellule del Sertoli iniziano a aderire alle piastre di coltura formando un monostrato cellulare cui sono aggiunti spermatozoi come descritto in dettaglio di seguito. Il tempo d’incubazione prima dell’aggiunta di spermatozoi à ̈ di almeno 72 ore e preferibilmente di 96 ore e comunque non superiore a 144 ore. I tubuli raccolti sono quindi lavati in HBSS e centrifugati a 500 r.p.m. Dopo il lavaggio i tubuli vengono incubati con una soluzione di HBSS contenente tripsina (2 mg/ml) e DNAse I (Sigma). Dopo il completamento della seconda digestione, la soluzione di tripsina à ̈ diluita 1:1 con Hank’s 20% FBS per arrestarne l’attività enzimatica. I tubuli dopo ulteriori lavaggi con HBSS vengono separati dalle cellule peritubulari tramite blanda centrifugazione a 300 rpm. Il “pellet†contenente il tessuto tubulare viene adeguatamente filtrato con un filtro in acciaio inox con apertura di maglia di 500 µm. Infine, per rimuovere le eventuali cellule peritubulari e di Leydig contaminanti il preparato, la sospensione contenente i tubuli viene ulteriormente centrifugata a 800 rpm per 5 min e il pellet risultante viene trattato per 7 min con una soluzione di glicina 1 M ed EDTA 2 mM in HBSS a pH 7.2. A questo punto si eliminare il surnatante e si aggiungono 50-100µl di PBS 1X. A questo punto si striscia il pellet su un vetrino. Dopo averlo lasciato asciugare si fissa il preparato per 60 minuti a temperatura ambiente in camera oscura con Paraformaldeide. A conclusione si lava per 2 minuti con PBS 1X. Successivamente si pongono 100 microlitri di soluzione di permeabilizzazione sui vetrini (Triton, sigma) e si incuba 2 minuti in frigorifero ponendo i vetrini distesi all’interno di una scatola. Si lava 2 volte con PBS 1X contenuto in due giare diverse in modo da eliminare tutta la soluzione di permeabilizzazione. A questo punto ai campioni vengono aggiunti 50 µl di sonda a 37 °C in atmosfera umidificata per 60 minuti e successivamente si lava 2 volte con PBS 1X per 2 minuti ogni lavaggio e con 40 ml di PBS 1X 3 ul DAPI per 40 secondi.
