I valori di MFC90 (Minimal Fungicidal Concentration per il 90% delle cellule presenti) mostrano come, nel caso degli olii essenziali di C. aurantium var. amara e della M. didyma, sia necessaria una maggiore concentrazione di olio essenziale per avere effetti citocidi su ceppi benefici e appartenenti al microbiota intestinale (Galactomyces sp. e S. cerevisiae) piuttosto che sui ceppi patogeni che al contrario risultano più sensibili. Non è stato possibile individuare un’analoga concentrazione sub-MIC per l’olio essenziale di C. aurantium var. dolce. Al fine di valutare l'effetto degli olii essenziali su un mix fungino caratterizzato da 1 ceppo di C. albicans e 1 ceppo di S. cerevisiae presenti in co-coltura, in ciascun pozzetto è stata acquisita la DO (Densità Ottica) a 450 nm per identificare la MIC (Minimum Ihnibition Concentration, concentrazione minima di inibizione) e valutata la MFC (Minimum Fungicidal Concentration, concentrazione minima fungicida) con idonea semina del contenuto dei pozzetti degli olii essenziali nr. 4, 6 e 7 di tabella 1 (tabella 4). Contestualmente, per confermare i valori MFC, il contenuto di ciascun pozzetto è stato inizialmente seminato su agar solido e colorato con Alamar blu (figura 1 e figura 2). Per valutare l'effetto immunomodulante degli idrolati (ID) nr 4, 6, 8 e 9 di tabella 1, le PBMC (Periferal Blood Mononuclear Cells) ottenute da due donatori sono state incubate durante la notte in terreno di coltura RPMI (Rosewell Park Memorial Institute) contenente diluizioni scalari degli idrolati (da 1/4 a 1/32). Metodi: sono stati considerati 7 ceppi probiotici: Saccharomyces boulardii CBS5926, Lactobacillus acidophilus LA3SACCO, Lactobacillus acidophilus LA-14, Lactobacillus plantarum ATCC 14917, Lactobacillus rhamnosus, Lactobacillus casei RO215, Lactobacillus helveticus ROO52. L’azione degli olii essenziali di C. aurantium, M. didyma e C. zeylanicum è stata testata sui ceppi probiotici secondo il metodo della micro-brodo diluizione come descritto al punto A, con le seguenti modifiche: per i test è stato usato il terreno liquido BH (Bushnell and Haas) poiché risultato maggiormente adatto alla crescita dei probiotici; i ceppi probiotici sono stati incubati a 37°C per 24 ore in microerofilia. I dati degli esperimenti in vitro nr 1-5 sopra riportati hanno mostrato che Candida spp. e Saccharomyces cerevisiae sono ugualmente modulati da olii essenziali ottenuti da Monarda didyma, Citrus aurantium varietà amara e Cinnamomum zeylanicum. Metodi: dopo idoneo consenso informato, fibroblasti gengivali umani sono stati ottenuti da pazienti sani soggetti a gengivectomia della regione molare. Subito dopo la rimozione i tessuti sono stati messi in soluzione HBSS con penicillina (250 U/mL), streptomicina 0.25 mg/mL) e amfotericina B (0.0025 mg/mL). Lo strato epiteliale è stato staccato meccanicamente e i campioni sub-epiteliali sono stati piastrati in fiasche con DMEM, supplementate con 50% FCS, L-glutamina (2mM), sodio piruvato (1 mM), penicillina (50 UI/mL) e streptomicina (50 μg/mL) a 37°C in un’atmosfera umidificata al 5% in CO2.
