Abstract
Procedimento, per la produzione di acilfosfatasi mediante cellule geneticamente modificate, comprendente essenzialmente le seguenti operazioni:- digerire un vettore plasmidico (ad esempio il plasmide batterico pT7.7, la cui mappa genica è mostrata in figura 1) con endonucleasi di restrizione per ottenere un vettore a DNA lineare;- legare il vettore a DNA lineare così ottenuto ad un frammento polinucleotidico che codifica per la sintesi di acilfosfatasi, essendo le estremità di detto frammento compatibili con i siti di restrizione del suddetto vettore a DNA lineare;- clonare ed esprimere il detto frammento nucleotidico, eventualmente come proteina di fusione, in cellule geneticamente come proteina di fusione, in cellule geneticamente modificate in quanto includono detto DNA ricombinante, sotto il controllo di una sequenza promotore inducibile oppure costitutiva, essendo le cellule geneticamente modificate in grado di sintetizzare la acilfosfatasi nello spazio intracellulare ed, eventualmente, di secernerla nello spazio extracellulare oppure periplasmatico;- recuperare la acilfosfatasi dallo spazio intracellulare, dallo spazio extracellulare oppure dallo spazio periplasmatico eventualmente come proteina chimerica; e - eventualmente, scindere la proteina chimerica per ottenere acilfosfatasi tal quale.
