Abstract
To determine the spermine in human serum and other biological liquids a clinical chemistry test has been created, which uses a concentrate enriched in spermine oxidases, partially purified of the other proteins present in the bovine serum, by means of precipitation with ammonium sulphate, and successive passage in ionic exchange resin (Enzyme E). The test permits the activation of the following reactionSperm. ox.Spermine + H2O - Oxidized Spermine + H2OPODH2O2 + phenol + 4-aminoantipyrine - quinone + H2Owith the following enzymatic mixturePeroxidases - 30 μl - (1mg/ml)4-amino antipyrine - 12 mgPhenol - 19.2 mgEnzyme E - 700 μlPhosphate Buffer0.1 pH 7.4 - 8.1 mlThe said enzymatic mixture may also be diluted, using the ratio, 1:5, with phosphate buffer 0.1M pH 7.4, and used according to the following methodology:Enzymatic mixture - 700 μlSample or standard (spermine) - 70 μlIncubation time 20' at 37 degrees CReading at 510nm against white reactives.The concentration of spermine is determined by the following formula:Spermine conc. = Optical density test x standard conc. / Standard optical density
