Abstract
N-substd. lactams of formula (I) are new (where R1,R2,R3,R4,R5,R6 and R7 are H, 1-4C alkyl or phenyl; R8 singly is H; or R8 and R3 together are -CH2CH2- or -CH2CH2CH2-; n is 3, 4 or 5). A pref. cpd. 2-(2-oxo-1-pyrrolidineacetamido) acetamide was prepd. by reaction of 2-oxo-1-pyrrolidineacetic acid with glycine ethyl ester in CH2Cl2 in presence of NEt3 and treating the N-(2-oxo-1-pyrrolidineacetyl)glycine ethyl ester in CH2Cl2 in presence of NEt3 and treating the N-(2-oxo-1-pyrrolidineacetyl)glycine ethyl ester with NH3 in methanol. Used for treatment of hypoxic aggression and impaired memory in elderly patients. Also used for treatment of cerebro- and cardio-vascular disorders, toxic or posttracimatic coma and lack of concentration. They also inhibit blood platelet aggregation and may be used in treating myocardial infarction and thromboembolic disease. Oral daily dose 20 to 2000 mg.
