Abstract
A process for the rapid detn. of a spermium count of sperm samples and/or the ratio of the living spermia comprises dissolving the fluorescent dyestuff propiduim iodide in a buffer and measuring the intensity (Fo), then adding the sperm sample and, after mesurement of the intensity (F1) and addn. of a cytoplasm membrane-permeability agent, determining the intensity (F2), then adding a buffer and a permeabilising agent to the buffer and measuring the intensity (F3) and subsequently the intensity (F4) of the pure buffer and finally the intensity (F5) of the sperm mixt. added to the buffer. The cell concn. is calculated using the formula (I). (A = the multiplication prod. from the slope of the calibration curve with the predilution ratio) The living cell ratio is calculated using the formula (II). Pref. is saponin, digitonin or nystatin.
