Abstract

Synthetic vaccine comprises a physiologically acceptable carrier on which is disposed a synthetic peptide residue (I) contg. a sequence of 6 amino acids corresp. to the sequence of such amino acids in a protein antigen or allergen where the greatest local average hydrophilicity is found. This hydrophilicity is determined by assigning relative values to the amino acids as follows: arginine 3.0; aspartic acid 3.0 + or -1; glutamic acid 3.0 + or -1; lysine 3.0; serine 0.3; asparagine 0.2; glutamine 0.2; glycine 0.0; proline-0.5 + or -1; threonine-0.4; alanine -0.5; histidine -0.5; cysteine -1.0; methionine -1.3; valine -1.5; isoleucine -1.8; leucine -1.8; tyrosine -2.3; phenylalanine -2.5; and tryptophan -3.4. Then the repetitive local average of hydrophilicity values at a number of points along the amino acid chain is determined. From the results the points of greatest local average hydrophilicity are determined. The vaccine when free from an amino acid sequence corresp. to the entire protein antigen or allergen evokes a protective immunological response by stimulation of antibody formation against an antigen or allergen when introduced into a host animal. The vaccines are formed without use of natural sera and so are free from contaminating serum proteins, viral residues, lipids, DNA etc.
