Abstract

1415597 Reactions with absorbed enzymes NATIONAL PATENT DEVELOPMENT CORP 1 Feb 1973 [11 Feb 1972] 5197/73 Heading B1X [Also in Divisions C3 and G1] A process for subjecting a substrate e.g. blood or body fluid containing an enzymesusceptible material to the action of an enzyme comprises permitting the enzyme-susceptible material of the substrate to come into contact with the enzyme entrapped or encapsulated within a water-insoluble hydrophilic polymer of a monomer of the group consisting of hydroxy lower alkyl (i.e. C 1 -C 3 ) acrylates, hydroxy lower alkyl methacrylates, hydroxy lower alkoxy lower alkyl acrylates, hydroxy lower alkoxy lower alkyl methacrylates with or without a proportion of vinyl pyrrolidone, acrylamide, methacrylamide, N- lower alkyl acrylamide, N-lower alkyl methacrylamide, N-hydroxy lower alkyl acrylamide, and N-hydroxy lower alkyl methacrylamide. The water insoluble hydrophilic polymer may be in the form of a hollow tube having a relatively thin overall internal semipermeable membrane coating of a polymer as above, but enzyme free, the semi-permeable membrane being permeable to the enzyme-susceptible material but not to the enzyme. In Fig. 3 a tube 18, shown in crosssection, comprises a lumen 20, a urease containing polymer layer 22 containing a reinforcing sheathing of polyethylene terephthalate and, optionally, inner and outer coatings of water insoluble hydroxyethyl methacrylate polymer. When a solution of urea is forced through the lumen of the tube the solution diffuses to the enzyme entrapped tube and ammonia diffuses out of the outer surface of the tube thus enabling the purification of body fluids e.g. blood to eliminate urea. Claims (7)

1. An enzymatically-active, water insoluble, alcohol soluble, hydrophilic product comprised of a polymer and an enzyme permanently entrapped in the cross-linked polymeric matrix thereof, said polymer being a polymer of hydroxy lower alkyl aerylate, hydroxy lower alkyl methacrylate, hydroxy lower alkoxy lower alkyl aer late, and hydroxy lowe alkoxy lower alkyl methacrylate.
2. A product according to claim 1 wherein said polymer is a sparingly cross-linked polymer.
3. A product according : to claim 1 wherein the polymer is a polymer of 2-hydroxyethyl;methacrylate.
4. A product according to claim 3 in the form of hard porous beads.
5. composition comprising the product of claim 1 coated with a relatively thin membrane of a solid, enzyme-free, water- nsoluble, alcohol-soluble hydrophilic polymer of the group consisting of polymers of hydroxy lower alkyl aerylate, hydroxy lower alkyl methacrylate, hydroxy lower alkoxy lower alkyl aerylate, and hydroxy lower alkoxy lower alkyl methacrylate.
6. A composition according to claim 5 wherein said thin membrance is a polymer of 2-hydroxyethyl methacrylate.
7. An electrode having a first coating thereon of an enzymatically-active, solid, water-insoluble, alcohol-soluble, hydrophilic, porous material comprised of a polyme and an enzyme permanently entrapped in the cross-linked polymeric matrix thereof, said polymer being a polymer of hydroxy lower alkyl aerylate, hydroxy lower alkyl methacrylate 41450/2 hydroxy lower alkoxy lower alkyl aerylate, and hydroxy lower alkoxy lower alkyl methacrylate, and having thereover a
