Abstract
1411503 Purifying enzymes SNAMPROGETTI SpA 19 June 1973 [7 July 1972] 29131/73 Heading C3H The proportion of an enzyme in a mixture of (a) the enzyme and (b) at least one other compound, viz. protein, nucleo protein or nucleic acid, produced, e.g. by lysing micro-organism cells, is increased by treating a solution of the mixture with a cationic surface active agent to precipitate (at least partially) the other compound(s) and the precipitate is removed from the treated solution. The preferred surface active agents are hexadecyl pyridinium chloride and benzyl-dimethyl-alkyl (C 10 -C 18 ) ammonium chloride, used in concentrations ranging from 0.01 to 0.4 g/dl. Examples 1 to 6 are directed to the treatment of mixtures rich in penicillin amidase, obtained by homogenizing a suspension of Escherichia coli cells and thereafter removing cell fragments by centrifuging, with one or other of the above specified cationic surface active agents at pH's ranging from 4.5 to 7-0. The purified enzyme could be recovered from the clear supernatant product by addition of ammonium sulphate. Examples 7 and 8 relate to the treatment of mixtures rich in #- galactosidase obtained from cells of Escherichia coli and Saccharomyces lactis, respectively.
